The purpose of this study is to estimate the value of the sociocultural multifunctionality of rural areas in Korea. The sociocultural multifunctionality of rural areas consists of emotional comfort, green landscape, cultural heritage, and rural viability values. The value of the sociocultural multifunctionality was assessed by contingent valuation method incoporating preference uncertainty. The log-logit models indicated that households were willing to pay 14,027~26,757 won per month and the model with preference uncertainty gave the highest WTP. WTP was affected by respondent's sex, location, awareness of relation with multifunctionality and others. The total value of sociocultural multifunctionality of rural areas in Korea estimated to 2,691~5,134 billion won per year.

Rural Environmental Planning (REP) is a method used by citizens in small towns and rural areas to plan their own future. REP assumes that the primary social value of rural people is to enhance a community's longterm viability by respecting the carrying capacity of the natural environment. REP derives its strength from the direct participation of those people affected by a plan. They determine its goals, shape its content, and implement its components. And both the rural environmental planner and the citizens involved in planning in their community must have a working knowledge of the legal framework of planning. In the results, REP reduces the cost of planning by drawing public agencies, not-for-profit organizations, volunteers from the private sector, and all vital resources available to rural people when they plan for their future.
Purpose: Conventional methods used to evaluate seeds viability are destructive, time consuming, and require the use of chemicals, which are not feasible to implement to process plant in seed industry. In this study, the effectiveness of Fourier transform near infrared (FT-NIR) spectroscopy to differentiate between viable and nonviable watermelon seeds was investigated. Methods: FT-NIR reflectance spectra of both viable and non-viable (aging) seeds were collected in the range of 4,000 - 10,000 $cm^{-1}$ (1,000 - 2,500 nm). To differentiate between viable and non-viable seeds, a multivariate classification model was developed with partial least square discrimination analysis (PLS-DA). Results: The calibration and validation set derived from the PLS-DA model classified viable and non-viable seeds with 100% accuracy. The beta coefficient of PLS-DA, which represented spectral difference between viable and non-viable seeds, showed that change in the chemical component of the seed membrane (such as lipids and proteins) might be responsible for the germination ability of the seeds. Conclusions: The results demonstrate the possibility of using FT-NIR spectroscopy to separate seeds based on viability, which could be used in the development of an online sorting technique.
The present study was to assess the in vitro viability and sexing rate of bovine embryos. Blastocysts were harvested on day 7~9 day after insemination(in vitro and in vivo), and the sex of the embryos was examined using the LAMP method. Embryo cell biopsy was carried out in a $80{\mu}l$ drop $Ca^{2+}$, $Mg^{2+}$ free D-PBS and, biopsied embryos viability were evaluated after more 12 h culture in IVMD culture medium. The formation of recovered embryo to expanded and hatching stages had ensued in higher of sexed embryo in vivo than in vitro (100% vs. 89%, p<0.05), and in vitro, the rates of degeneration after sexing were significantly (p<0.05) higher in vitro than in vivo(11% vs. 0.0%). The rates of the predicted sex were female 61% vs. 56%, and male 39% vs. 44% in vivo and in vitro, respectively. The rates of survival following different biopsy methods were seen between punching and bisection method in vivo and in vitro (100% vs. 89% and 100% vs, 78% respectively). Biopsy method by punching was significantly (p<0.05) higher than bisection between produced embryos in vivo and in vitro. The present data indicate that with microblade after punching for embryo sexing results in high incidence of survivability on development after embryo biopsy. It is also suggested that LAMP-based embryo sexing suitable for field applications.

The objective of this study was to analyze the evaluation of the rural resident's on their community at view of community life satisfaction. The specific purposes of this study were to identify the degrees of community life satisfaction, consciousness of rural community according to the individual variables. The data were gathered from 1,870 rural residents by the structured interview in the 187 communities sampled by multi-staged cluster stratified sampling method in November, 2003. The major findings of this study were as follows; first, the degree of the rural residents'community life satisfaction was generally low, especially the rural residents were dissatisfied with the opportunities of leisure and education environment in their communities, and by half were satisfied with housing environment. Second, as consciousness of rural community, the degree of rural resident's joining at community meeting was high, and openness to the outside residents' was high. third, the most important factor for consciousness of rural community were community organization, good neighbor. As result of this study, the following conclusion and recommend can be drawn. First, the level of rural resident's community life satisfaction is overall low. Especially it is needed policy efforts and priority investment for advancing satisfaction on leisure and education environment in rural community. Second, to foster rural resident's community consciousness, community orgaruzatlon Is first of all supported and activated. The objective of this study was to investigate whether supplementation of pentoxifylline (PTX; phosphodiesterase inhibitor) to thawed boar semen improves the post-thaw motility of sperm and affects the efficiency of artificial insemination (AI) and further development. To determine the concentration of PTX for AI, frozen-thawed semen was incubated with 0, 5, 10, and 20 mM PTX in an extender freezing medium, respectively, after thawing. Kinematic properties of sperm were examined with a computer-assisted semen analysis (CASA) system. In addition, viability and mitochondrial activity were also tested by LIVE/DEAD and a MitoTracker kit. There were no significant differences in the kinetic parameters of thawed sperm between control and treatment groups, but overall assessment parameters such as motility and rapid progressive were higher in the 10 mM PTX group. In the viability and mitochondrial assay, there were no significant differences observed in the PTX treatment, compared to the control. For further analysis, artificial inseminations were performed using frozen semen and 10 mM PTX treated cryopreserved semen, respectively. There were no differences in pregnancy rates and fetus weights among the groups until 30 and 40 days, but litter size was reduced and relatively low-birth weight was observed in the PTX group. In summary, our findings suggest that enhancement of in vitro sperm quality or non-toxicity supplemented by PTX may have detrimental effects on fetus development.
Objective: Dairy cattle nutrient requirement systems acknowledge amino acid (AAs) requirements in aggregate as metabolizable protein (MP) and assume fixed efficiencies of MP used for milk protein. Regulation of mammary protein synthesis may be associated with AA input and milk protein output. The aim of this study was to evaluate the effect of nanoemulsified methionine and cysteine on the in-vitro expression of milk protein (casein) in bovine mammary epithelial cells (MAC-T cells). Methods: Methionine and cysteine were nonionized using Lipoid S 75 by high-speed homogenizer. The nanoemulsified AA particle size and polydispersity index were determined by dynamic light scattering correlation spectroscopy using a high-performance particle sizer instrument. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was performed to determine the cytotoxicity effect of AAs with and without nanoionization at various concentrations (100 to $500{\mu}g/mL$) in mammary epithelial cells. MAC-T cells were subjected to 100% of free AA and nanoemulsified AA concentration in Dulbecco's modified Eagle medium/nutrient mixture F-12 (DMEM/F12) for the analysis of milk protein (casein) expression by the quantitative reverse transcription polymerase chain reaction method. Results: The AA-treated cells showed that cell viability tended to decrease (80%) in proportion to the concentration before nanogenesis, but cell viability increased as much as 90% after nanogenesis. The analysis of the expression of genetic markers related to milk protein indicated that; ${\alpha}_{s2}$-casein increased 2-fold, ${\kappa}$-casein increased 5-fold, and the amount of unchanged ${\beta}$-casein expression was nearly doubled in the nanoemulsified methionine-treated group when compared with the free-nanoemulsified methionine-supplemented group. On the contrary, the non-emulsified cysteine-administered group showed higher expression of genetic markers related to milk protein ${\alpha}_{s2}$-casein, ${\kappa}$-casein, and ${\beta}$-casein, but all the genetic markers related to milk protein decreased significantly after nanoemulsification. Conclusion: Detailed knowledge of factors, such nanogenesis of methionine, associated with increasing cysteine and decreasing production of genetic markers related to milk protein (casein) will help guide future recommendations to producers for maximizing milk yield with a high level of milk protein casein.

The quality and yield of crops produced using field cultivation are expected to decrease due to the recent global climate change caused by extreme weather. The plant reproductive stage associated with crop yields is a highly vulnerable period to global warming caused by high temperatures. This study analyzed the adzuki bean's yield properties, antioxidant contents, and pollen viability of adzuki bean (Vigna angularis L.) under high-temperature stress and growth periods in a temperature gradient greenhouse that forms 0 to 4℃ above the outside temperatures. As the main variety of red beans cultivated in Korean farms, the "Arari" red bean was grown in the rain shield greenhouse and the temperature gradient greenhouse from 2021 to 2022 in Milyang, Korea. Compared to 2022, it showed a 0 - 1.0℃ lower temperature during the whole growth period in 2021. However, its average temperatures were 0 - 3.7℃ higher in the vegetative stage and 0.4 - 2.4℃ higher in the anthesis stage in 2021. The lowest yield (6.8 ± 0.7 g) was at the highest temperature (T4: low, 23.6℃; average, 28.5℃; high, 35.8℃) during the anthesis stage in 2021. The temperatures of the mature stage were 1.7 - 3.9℃, which were higher in 2022 than in 2021, although the low temperatures of 2022 were lower than in 2021. The yields of the mature stage in 2022 increased more than in 2021 because of the high temperature of the mature stage. The growth and yield were good at 40.5℃ in the vegetative stage. However, growth was poor when the average temperature was 27.0℃ or higher, and yields decreased during the flowering period. Total polyphenol and flavonoid contents were increased, and the pollen viability was 40.75% in the whole growth period at high temperature (T4: low, 22.9℃; average, 28.8℃; high, 36.9℃). These results showed that the antioxidant levels increased when the antioxidant component was affected at higher temperatures than at normal. In contrast, the pollen viability-related yield decreased as the temperature increased. Our results are the basic data for field growers and the breeding of thermos-tolerance in adzuki beans to prepare for the changeable future climate.

This research is related to the evaluation of rural amenities. Since the UR negotiations, the wave of market liberalization has brought many difficulties to the rural areas. This paper serves to identify a number of important elements such as socialization, cultural and ecological resources and investigate the viability of rural areas. The purpose of this study is to evaluate rural amenities by evaluation indices and establish planning direction of amenities for rural areas. The data have been collected by methods of a field survey and a questionnaire survey. The evaluation indices have been defined as socialization, safety, amenity and convenience by several researches. Villages have been divided into two groups. Seven(Shimchon, Ibaek, Muan, Dokpyong-li, Imchin-li Hyangyang-li, Wonwol-li) have been newly constructed, six(baeksa, Mokmyon, yongho-li, shindyae-li, hwangdun·songgae) have been redeveloped. There are considerable differences in the degree of satisfaction between he two groups. The results are as follows: 1) socialization is very necessary for amenities in rural villages. Especially the values of the resident's interactions and management of the village are most important in the preparation of an amenity plan in rural areas. So the plans and establishment of public facilities are requisite for the residents community. 2) The convenience of rural public facility plans is necessary for the improvement of the farmers'living conditions. For planning the rural villages, special regards are paid to characteristics of the village, such as the former place of residence and occupations. 3) Newly constructed village should improve their socialization and the redeveloped types should try to get a better life for amenity and safety, The residents of the redeveloped types show relatively high degree of satisfaction with indices of resident community, living convenience facilities, and management of village. On the contrary, amenity and safety are good in newly constructed types. This study has taken into consideration the characteristics of rural villages. It makes a contribution to the redevelopment of rural villages and improves amenities in rural villages. It is recommended that more studies classify the resources of rural areas and measure amenities which are significant to city dwellers in the future.
Seed viability testing provides valuable information for assessing seed lot germinability. However, most testing methods require destruction of seed prior to test. Because the dissected seeds for viability test cannot be used further evaluation, the nondestructively X-ray photography technique that can be applied for the evaluation of seed quality has been developed. In order to know the validity and accuracy of X-ray photography technique in seed evaluation test that conducted to remove the abnormal seed from a seed lot, we have compared the results from tetrazolium viability test, germination test and X-ray contrast method in cotton. Metallic salts treatment increased the efficiency of X-ray photographic method by enhancing the penetration of X-ray in abnormal or damaged seeds rather than normal seeds that have strong and well-organized tissues in seed. Cotton seeds presoaked for 16 hr in distilled water followed by soaking into metallic salt solution (5% NaI in water) for 60 min were easily classified seeds into dead seed and viable seed based on the radiography images obtained by X-ray radiation. We concluded that soft X-ray photography was reliable to find out the various defective characters due to heat and mechanical damage of seeds.
