The experiments were carried out to study the effect of physical forms of hay diet on phosphorus metabolism by offering sheep roughage diets containing a low phosphorus content. The sheep were fed coarse hay, short hay or finely ground pelleted hay. The physical forms of hat diet had little effect on the pathway of phosphorus excretion. In all groups, the most of phosphorus was excreted in feces and urinary phosphorus excretion was negligible. Duodenal fluid flow, rumen fluid outflow and estimated salivary flow appeared to be relatively high in sheep fed the coarse hay diet as compared to those in sheep fed the finely ground diet. The amount of phosphorus flow to the duodenum tended to be high in the coarse diet group as compared to those in other two groups. On the other hand, phosphorus concentrations in the duodenal fluid and the rumen fluid were lower in sheep fed the coarse hay diet than those in sheep fed the ground hay. Net intestinal phosphorus absorption tended to be higher in sheep fed the coarse hay than that in sheep fed the ground hay or pelleted hay. However, a negative phosphorus balance was observed in the coarse hay diet group although a positive phosphorus balance was found in other two groups. It was considered that salivary phosphorus secretion was greater than the amount of salivary phosphorus reabsorbed from the intestine, resulting in the negative of phosphorus balance in sheep fed the coarse hay.
A 98 d feeding trial carried out to study liveweight change and rumen metabolites in heifers weighing initially 275 kg and given either untreated or ammoniated rice straw supplemented with 0, 0.4, 0.8 or 1.2 kg protein meal consisting of cottonseed meal (60). All 32 animals received 0.6 kg rice polishings/hd/d and had continuous access to molasses/urea block-licks containing 15% urea. The effects on growth rates of treatment of the straw with ammonia and of supplementation with bypass protein were additive. The heifers fed ammoniated straw grew 267 g/hd/d (p<0.001) faster and consumed 11% (p<0.05) more straw than the heifers on untreated straw. The mean growth response to bypass protein was 0.37 kg gain/kg protein meal supplied. Supplementation with protein meal tended (p=0.06) to depress intake of straw, but straw intakes of the unsupplemented groups were high. Small changes in the composition of the block-licks that were fed throughout the feeding trial led to changes in block intake and in intake of untreated straw. Increasing quantities of protein meal fed were associated with linear increase in concentrations of ammonia (p<0.05) and in molar percentages of iso-butyrate (p<0.01), iso-valerate (p<0.01) and valerate (p<0.01) in the rumen fluid of the heifers on a basal diet of untreated straw. However, in the rumen fluid of the heifers given ammoniated straw, the levels of these metabolities were not affected by the quantity of protein meal given.
This study was conducted to investigate the combined effects of VFA composition of rumen fluid and heat exposure (30${\pm}$2$^{\circ}C$) on the general clinical view and insulin secretory response to glucose in sheep. The total infusion of nutrients was examined in sheep via the technique of continuous alimentation. Four adult Suffolk sheep fitted with a permanent ruminal cannula and a simple T-shaped duodenal cannula were used. A peristaltic pump was used to infuse the solutions of volatile fatty acid triglycerides (VFA-TG) consisting of 70 triacetin : 20 tripropionin : 10 tributyrin (low propionin division: LP) and 50 triacetin : 40 tripropionin : 10 tributyrin (high propionin division: HP) on the basis of energy and minerals into the rumen, and casein solution into the duodenum. The effects of heat exposure and type of the levels of VFA-TG solutions on the insulin secretory response to glucose in sheep were investigated by using hyperglycemic clamp (HGC) technique. The results obtained are summarized as follows: 1. During the heat exposure (latter half of the infusion period), respiration rate, heart rate and rectal temperature increased (P<0.01, P<0.01, P<0.05), but the levels of VFA-TG solutions (LP and HP division) did not affect the general clinical view except for the heart rate. 2. In the HGC technique, glucose infusion rate (GIR) and mean plasma insulin increments (MPII) tended to be ower in the heat exposure than in the thermoneutral environment, but no significant difference was found among the treatments. GIR and MPII remained unchanged between the levels of VFA-TG solutions. 3. In the HGC technique, ratio of MPII to GIR (MPII/GIR) which represents pancreatic ${\beta}$-cell response to glucose stimulation remained unchanged among the treatments.
This study investigated the effects of acarbose addition on changes in ruminal fermentation characteristics and the composition of the ruminal bacterial community in vitro using batch cultures. Rumen fluid was collected from the rumens of three cannulated Holstein cattle fed forage ad libitum that was supplemented with 6 kg of concentrate. The batch cultures consisted of 8 mL of strained rumen fluid in 40 mL of an anaerobic buffer containing 0.49 g of corn grain, 0.21 g of soybean meal, 0.15 g of alfalfa and 0.15g of Leymus chinensis. Acarbose was added to incubation bottles to achieve final concentrations of 0.1, 0.2, and 0.4 mg/mL. After incubation for 24 h, the addition of acarbose linearly decreased (p<0.05) the total gas production and the concentrations of acetate, propionate, butyrate, total volatile fatty acids, lactate and lipopolysaccharide (LPS). It also linearly increased (p<0.05) the ratio of acetate to propionate, the concentrations of isovalerate, valerate and ammonia-nitrogen and the pH value compared with the control. Pyrosequencing of the 16S rRNA gene showed that the addition of acarbose decreased (p<0.05) the proportion of Firmicutes and Proteobacteria and increased (p<0.05) the percentage of Bacteroidetes, Fibrobacteres, and Synergistetes compared with the control. A principal coordinates analysis plot based on unweighted UniFrac values and molecular variance analysis revealed that the structure of the ruminal bacterial communities in the control was different to that of the ruminal microbiota in the acarbose group. In conclusion, acarbose addition can affect the composition of the ruminal microbial community and may be potentially useful for preventing the occurrence of ruminal acidosis and the accumulation of LPS in the rumen.

This study evaluated the in vitro effect of medicinal plant extracts on ruminal methanogenesis, four different groups of methanogens and ruminal fermentation characteristics. A fistulated Holstein cow was used as a donor of rumen fluid. Licorice and mugwort extracts (Glycyrrhiza uralensis and Artemisia capillaris, 0.5% and 1% of total substrate DM, respectively), previously used as folk remedies, were added to an in vitro fermentation incubated with buffered-rumen fluid. Total gas production in Glycyrrhiza uralensis extract treatment was not significantly different between treatments (p<0.05) while total gas production in the Artemisia capillaris extract treatment was lower than that of the control. Artemisia capillaris extract and Glycyrrhiza uralensis extract reduced $CH_4$ emission by 14% (p<0.05) and 8% (p<0.05), respectively. Ciliate-associated methanogens population decreased by 18% in the medicinal plant extracts treatments. Medicinal plant extracts also affected the order Methanobacteriales community. Methanobacteriales diversity decreased by 35% in the Glycyrrhiza uralensis extract treatment and 30% in the Artemisia capillaris extract treatment. The order Methanomicrobiales population decreased by 50% in the 0.5% of Glycyrrhiza uralensis extract treatment. These findings demonstrate that medicinal plant extracts have the potential to inhibit in vitro ruminal methanogenesis.
Napier grass (Pennisetum purpureum Schum.) harvested at an early growth stage was ensiled with 4% of molasses in a polyethylene bag silo which contained 15 kg of chopped forage each. Dry matter (DM) content of the silage was so low as 14.75%, although chemical quality of the silage was very high. Ratio of ammonia nitrogen to total nitrogen was 6.59%, and the pH value of the silage was 3.79. Nutritive value of the silage was estimated using goats and compared to that of a timothy hay as a reference ration. Feeding level of each rations was adjusted to a level of nitrogen (N) recommendation. DM and N digestibilities of the silage were 65.0 and 54.5%, respectively, but those of the timothy hay were 37.6 and 37.2%. Feeding of the napier grass silage maintained body weight and kept positive N retention. Ammonia N concentration in the rumen fluid in goats fed the napier grass silage increased after feeding, but blood urea concentration was constant. Feeding of the timothy hay did not increase ammonia N concentration in the rumen fluid, but increased blood urea concentration. These facts indicated that the napier grass silage had enough digestible DM and N for maintenance ration to goats.
Performance and production of twenty lactating Holstein cows fed diets containing either soybean meal or fish meal as the primary protein source were compared in a continuous or split feeding scheme. At 1 wk prepartum four groups of five animals were placed on each experimental diet. Animals assigned to the continuous feeding scheme were continued on these diets for 10-wk postpartum. At 4 wk postpartum, the diets for the groups assigned to switching protein sources were changed. These treatments were continued for another 6 wk. Milk production and dietary intakes were recorded daily. Milk constituents were measured every 2 d. Cows weights, rumen fluid samples and jugular blood samples were collected weekly. Data showed no effect of early lactation diet on cow performance or milk characteristics. Overall, compared to the soybean meal diet, the fish meal diet lowered the milk fat percent and increased production of milk per unit of dry matter ingested. No differences were observed for volatile fatty acid content of rumen fluid, blood mineral content, milk protein, somatic cell count, 4%-fat corrected milk, dry matter intake, or body weight.

In this study, the comparative effects of transgenic corn (Mon 810 and Event 176) and isogenic corn (DK729) were investigated for their influence on in vitro rumen fermentation. This study consisted of three treatments with 0.25 g rice straw, 0.25 g of corn (Mon810/Event176/DK 729) mixed with 30 ml rumen fluid-basal medium in a serum bottle. They were prepared in oxygen free conditions and incubated at $39^{\circ}C$ in a shaking incubator. The influence of transgenic corn on the number of bacterial population, F. succinogenes (cellulolytic) and S. bovis (amylolytic), was quantified using RT-PCR. Fermentative parameters were measured at 0, 2, 4, 8, 12 and 24 h and substrate digestibility was measured at 12 and 24 h. No significant differences were observed in digestibility of dry matter, NDF, ADF at 12 and 24 h for both transgenic and isogenic form of corns (p>0.05) as well as in fermentative parameters. Fluid pH remained unaffected by hybrid trait and decreased with VFA accumulation as incubation time progressed. No influence of corn trait itself was seen on concentration of total VFA, acetic, propionic, butyric and valeric acids. There were no significant differences (p<0.05) in total gas production, composition of gas (methane and hydrogen) at all times of sampling, as well as in NH3-N production. Bacterial quantification using RT-PCR showed that the population number was not affected by transgenic corn. From this study it is concluded that transgenic corn (Mon810 and Event 176) had no adverse effects on rumen fermentation and digestibility compared to isogenic corn. However, regular monitoring of these transgenic feeds is needed by present day researchers to enable consumers with the option to select their preferred food source for animal or human consumption.
Dietary fatty acid including mainly palmitic acid and stearic acid was fed to fattening cattle and its effect on body weight gain, plasma lipid contents and rumen liquid fermentation in vitro was examined. In expt. 1, the effect of dietary fatty acid on body weight gain and plasma lipid concentrations was examined. In the control diet group, cattle were fed 1 kg/day of rice straw and concentrate which satisfied the requirement. In the fatty acid group, cattle were given 250 g/d of fatty acid with the same diet of the control diet group. In the excess concentrate group, cattle were given the same diet of the control diet group plus 735 g/d of concentrate corresponding to the same TDN of 250 g/d of fatty acid. Diets were given for 7 days. Body weight gain of cattle given dietary fatty acid was significantly greater than that of cattle fed only rice straw and concentrate. When dietary fatty acid was added to cattle feed, plasma NEFA and HDL-cholesterol concentrations increased. In expt. 2, the influence of dietary fatty acid on gas production and VFA profile in the rumen liquid was investigated in vitro. In the control group, 10 mg of rice straw and 90 mg of concentrate were incubated in the rumen fluid. In the excess concentrate group, 10 mg of rice straw and 97.5 mg of concentrate were incubated. In the fatty acid group, 10 mg of rice straw, 90 mg of concentrate and 2.5 mg of fatty acids were incubated. The rumen liquid mixed with feed materials was incubated for 24 h and the cumulative gas volume was measured. The VFA profile was also measured. Cumulative gas volume in the rumen liquid with fatty acid was equal to the control. Excess concentrate increased cumulative gas volume compared to the fatty acid group. There was no significant difference in total VFA concentration between experimental diet groups. It is suggested that dietary fatty acid has the potency to improve growth performance in fattening cattle without failure in rumen fermentation.
The study was conducted to investigate the effect of soybean meal (SBM) supplementation on the voluntary intake of rice straw and ruminal fermentation characteristics. Balance trials were conducted with three Hapanese Corriedale wethers fed a rice straw alone (control), rice straw supplemented with 75 and 150 g of SBM/day in a $3{\times}3$ latin square design. Voluntary intake of rice straw in sheep fed both levels of SBM supplemented diets was significantly higher (p<0.05) than that in sheep fed control diet. Crude protein digestibility was significantly increased (p<0.05), but organic matter, crude fibre, neutral detergent fibre and acid detergent fibre digestibilities were not affected by SBM supplementation. Nitrogen balance was positive in sheep on both levels of SBM supplemented diets, but negative in animals on the control diet. Rumen ammonia and blood urea-nitrogen concentrations increased (p<0.05) as increasing level of SBM. Total volatile fatty acids, acetate, propionate, butyrate and valerate concentrations in rumen fluid were also significantly increased (p<0.01), but ruminal pH was decreased (p<0.05) by SBM supplementation.
