Shell waste from the butter clam Saxidomus purpuratus and littleneck clam Ruditapes philippinarum is a large by-product of shellfish aquaculture, and it is desirable to convert it into value-added products for industrial applications. In this study, calcium carbonate (CaC) polymorphs from butter clam (BCSP) and littleneck clam (LCSP) shell powders and commercial CaC were characterized using Fourier transmission infrared spectroscopy (FT-IR), X-ray diffraction (XRD), and scanning electron microscopy (SEM). The results revealed that the optimal calcination conditions to eliminate organic substances and improve solubility for both BCSP and LCSP were $800^{\circ}C$ for 8 h in an electrical furnace. Calcination improved the white index of the butter clam (BCCP) and littleneck clam (LCCP) calcined powder compared with shell powders. The calcium content in BCCP (51.1%) was higher than that of LCCP (44.9%) or commercial calcium oxide (CaO, 44.7%). The XRD patterns of BCCP and LCCP were similar to that of CaO. Cubic-like crystals of CaC and irregular crystals of BCCP and LCCP were observed by SEM. The FT-IR and XRD analyses revealed the presence of calcite and aragonite in the BCSP and aragonite in the LCSP, whereas the CaC contained calcite. These results indicate that butter and littleneck clam shells are potential biomass resources for calcium carbonate and calcium oxide.
This study aimed to evaluate the effects of shaking stress in the hemolymph of the Manila clam Ruditapes philippinarum by quantification of nitric oxide (NO) levels. The clams were divided into 3 groups as follows: clams placed in a plain container (control), clams injected with nitro-L-arginine methyl ester (NAME, an NO inhibitor), and clams in a container filled with nylon fiber at a density of $1kg/m^3$. Subsequently, each group was placed in sea water and shaken at 100 rpm for 6 h. The concentration of NO was quantified by using DAF assay and Griess assay. Both the assays showed that while shaking significantly increased the NO concentration, the NO inhibitor reduced the NO concentration in the hemolymph of the clams tested. In addition, the nylon fiber, which was used as a filler, effectively prevented the increase in NO concentration. This result suggests that measurement of NO concentration is a useful tool for evaluation of physiological stress in marine bivalves. In addition, it should be considered that a filler is necessary when dredge fishing or the suspended clam culture method is developed.
Gonadal developmint, age and growth of Ruditapes philippinarum were investigated using samples from the intertidal zone of Simpo on the coast of Kimje, Korea, which were collected onthly for one year from Februaty 1993 to January 1994.Ruditapes philippinarum is diecious in sex. The gonads are located between the subregion of the midintestinal glands and reticular connective tissue of the foot. The ovary is composed of a number of ovarian sacs, and the testis is composed of numerous seminiferous tubules. The clam spawns once a year from early June to darly October, and the main spawning occurred between July and August when the water temperature went above 23$^{\circ}C$. Ripe oocytes are about 65-70${\mu}{\textrm}{m}$ in diameter. Gonadal phases of this species can be divided into five successive stages; multiplicative(February to March), growing (April to May), mature(Aprilto Septimber), spent(June to October), and degenerative and resting(july to March). Spawning is closely related to the sea water temperature. Based on the monthly variations of marginal index (MI')of the shell, it was suggested that the annual ring mark formation occurred in March once a year and took approximately 8 months (0.67 year) for first ring to be formed on the shell. The relationship between the shell length(SL) and the total weight (TW) was represented by nonlinear equation; TW=1.208 x 10/ sup -4/ S $L^{3.158}$, and also in the relationship be-tweenthe shell length (SL) and the shell height(SH), the shell length and the shell width (SW) were represented by the linear equations; SH=0.726 SL-0.483, SW=0.542 SL-0.803.Growth curves for shell length and total weight fitted to von Bertalanffs equation were expressed as: S $L_{t}$ =68.34(1- $e^{0.221}$(t+0.418)) T $W_{t}$ =75.16(1- $e^{0.221}$(t=0.418))$^{3.158/3.158}$
Prevalence of a protozoan parasite Perkinsus olseni in Manila clam Ruditapes philippinarum was surveyed from July to December 2009 on the west and south coast of Korea. P. olseni infection was diagnosed using two primer sets, P.olseni NTS Forward/P.olseni NTS Reverse set and PolsITS-140F/PolsITS-600R set in polymerase chain reaction(PCR). The results using PolsITS-140F and PolsITS-600R primer set was retained up to 60% at all stations from July to December, except for Padori. Especially, Goheung showed 100% prevalence from October to December. The results about comparison of the 4 station's DNA sequences which were analyzed from PCR products(457bp) using PolsITS-140F and PolsITS-600R primer set, there were only 2base differences at Sunjedo.
Ultrastructural characteristics of the germ cells and accessory cells in testis during spermatogenesis and taxonomic values of mature sperm morphology of Ruditapes philippinarum were investigated by the transmission electron microscope and scanning electron microscope observations. The testis is the diffuse organ that consists of branching acini containing developing germ cells and accessory cells associated with spermatogenesis. The morphology of the spermatozoon is of the primitive type and is somewhat different to those of other bivalves. The morphologies of the sperm nucleus type and the acrosome shape of this species have a cylinderical type and a modified cone shape, respectively. As some ultrastructural characteristics of the acrosomal vesicle, the peripheral parts of two basal rings show electron opaque part, while the apex part of the acrosome shows electron lucent part. These characteristics of sperm belong to the family Veneridae in the subclass Heterodonta, unlike a characteristic of the subclass Pteriomorphia showing all part of the acrosome being composed of electron opaque part. In particular, a cylinder-like nucleus of the sperm is curved. The spermatozoon is approximately $48-51{\mu}m$ in length, including a long acrosome (about $2.4{\mu}m$ in length), a curved sperm nucleus (about $3.40{\mu}m$ in length), and a tail flagellum. The axoneme of the sperm tail shows a 9+2 structure.
In this study, the influence of water temperature and salinity on sand ejection of Manila clams, Ruditapes philippinarum, was investigated. The result showed that, under different water temperature, the highest quantity of sand ejection was at $20^{\circ}C$, the value of which was 0.091 g, while the smallest one was only 0.058 g at 0$^{\circ}C$. The highest releasing rate, 95.79%, could be seen in $20^{\circ}C$ group, while $0^{\circ}C$ group was the lowest one, 85.29%. Under different salinity, 30 psu group had the highest value, 0.057 g, and 0 psu (freshwater) group was the lowest one, only 0.026 g. At the same time, the highest releasing rate appeared at 25 psu, which was 90%. 0 psu group was the lowest one in releasing rate, 60.47%. According to these results, we recommend that the clams should be placed into the clear seawater with about 25 psu of salinity at near $20^{\circ}C$ to let the clams perform the self-depuration. Better quality of clams containing less impurity in the aquaculture and commerce could be obtained from this pre-treatment.
In order to develop the sustainable ecological management, short-necked clams, Ruditapes philippinarum, were collected from Yoengi coast in Tongyoeng, Korea. The growth of the clam was estimated as: $L_f=68.08{\cdot}(1-e^{-0.145(t+0.324)})$ from ring radius composition of shell. Instantaneous co-efficiency of total mortality and natural mortality were calculated as: 0.991/year and 0.494/year, respectively. The age of the clams from the first capture was estimated to be 3.28. The total biomass was estimated to be 212 MT in the fisheries area (6.4 ha). Applied by these parameters, the annual recruit biomass and the current yield per recruit were calculated to be 649.5 individual/$m^2$ and $0.7\;g/m^2$, respectively. The current fishing intensity was much lower for maximum sustainable yield and acceptable biological catch. Although higher yield per recruit could be achieved by increasing fishing intensity, it is favorable to retain the current fishing intensity because of the unique fishing attitude on Yeongi coast in Tongyoeng, Korea.
In order to understand the effect of introduction of oyster rack culture on natural short-necked clam, Ruditapes philippinarum, stock, we investigated the status of natural clam stock prior to introduction of oyster rack culture at Yeongheung Coast of Wando Island. The growth equation of the clam was estimated as: $L_t=61.46{\cdot}(1-e^{-0.172(t+0.155)})$ from ring radius composition of shell. Instantaneous coefficient of total mortality and natural mortality were calculated as: 2.4087/year and 0.478/year, respectively. The age at first capture was estimated to be 2.55 year. The total biomass was estimated to 3.23 ton in the bed (0.8 ha). Applied by these parameters, the annual recruit biomass and the current yield per recruit (Y/R) was corresponded to $114.7individuals/m^2$ and $92.0g/m^2$, respectively. Maximum sustainable yield (MSY) was estimated 0.77 ton/year which was close to annual catches, 0.74 ton/year, in the area.
In the present study, we investigated temporal variation in the reproductive effort and biochemical contents in adult Manila clam Ruditapes philippinarum at Shi-Heung-Ri beach on the east coast of Jeju Island. Gonad-somatic index (GSI), a ratio of the egg mass to the total biomass determined using ELISA increased dramatically from late May to June (7.1% to 19.5%). In mid-July, GSI dropped to 15.1%, indicating spawning on a small scale during this period. GSI also declined dramatically from late August (15.5%) to September (4.3%), suggesting that massive spawning occurred during this period. A positive correlation was observed between clam size and potential fecundity, while potential fecundity ranged between 1.19-8.40 million eggs/clam. Total proteins and lipids in the tissue increased from late spring to mid-summer, coinciding with an increase in GSI. Protein, lipid and carbohydrate contents in the tissue were lowest in January, indicating that clams were suffering from poor nutritional circumstances, which may be associated with poor food supply from the environment. Monthly increase and/or decrease in the body weight were closely related to gonad maturation and subsequent spawning, which was also linked to temporal changes in the contents of protein, lipid and carbohydrates in the tissue.
A comparative analysis of size and age structures of coastal subfossil shell assemblages of the shortnecked clam Ruditapes philippinarum from open and protected bays of Cheju Island (Korea) was carried out. On the whole, taking into account the damage of small fragile shells, size and age structures of the shell assemblages corresponded to the classical curve of bivalve population distribution when its mortality diminishes with age increase up to a certain threshold. It was found that shell samples from open bays of the western, southern and eastern coasts included shells of smaller and younger individuals (L ${\leq}$ 40 mm, ${\leq}$ 4 years) than samples from the eastern protected bay (L ${\leq}$ 54.5 mm, ${\leq}$ 6 years). Evidently, strong wave activity was the reason for a short life-span of the clams from the open areas. Growth was investigated retrospectively by annual growth rings on the shells. Growth rates of the clams from the various coasts of Cheju Island differed. However, growth rates of the clams from different biotopes at the same (eastern) side of the Island were similar. Shell height/length and width/length ratios statistically significantly increased with the clam age increase. Most likely, the reason for such shell shape alteration is that more conglobated individuals more survive being more energy-optimal than oblong specimens.
