The experiment was carried out in a 10 $\ell$ vessel in order to evaluate the growth and nutritional quality of rotifer, Brachienus rotundiformis fed by different diets (Freshwater Chlorella, Marine Chlorella and $\omega$-yeast) for the high density cultivation. The maximum densities for the rotifer fed on the marine Chlorella, freshwater Chlorella and $\omega$-yeast were $10,900\~12,400,\;9,190\~10,600$ and 2,390$\~$2,750 inds./ml, respectively. Therefore, the maximum densities for the rotifer fed on the marine Chlorella and freshwater Chlorella were higher than that for rotifer fed on the $\omega$-yeast The essential n-3 highly unsaturated fatty acid in rotifer fed on the marine Chlorella was $8.71\%$ which was slightly lower than that in rotifer fed on the $\omega$-yeast, $9.14\%$, while it was higher than that in the rotifer fed on freshwater Chlorella, $4.45\%$. This result indicated that marine Chlorella could be appropriate diet for the high density cultivation of rotifer.
The growth of rotifer, Brachionus rotundiformis was evaluated at different culture conditions. Rotifer was fed on condensed freshwater Chlorella. The productivity of rotifer in the high density culture system was compared to that of rotifer in the batch culture system, in which rotifer was fed on baker's yeast. The growth rate of rotifer increased as temperature increased in the culture system supplied with air or oxygen gas. The maximum density of rotifer in the culture systems supplied with air was in range of 16,300$\~$17,000 ind./ml at $24^{\circ}C$. In the culture systems supplied with oxygen gas, it ranged 26,300$\~$30,500 ind/ml at $28^{\circ}C$. When the concentration of dissolved oxygen in the culture system supplied with air reached to below 1 ppm or when the concentration of undissolved ammonia in the culture system supplied with oxygen gas reached 16.6$\~$22.6 ppm, the growth of rotifer decreased. When oxygen gas was supplied and pH was adjusted to 7, the maximum density of rotifer reached to 43,000 ind/ml at $32^{\circ}C$. The production costs for 10 billion rotifer in the high density culture and batch culture were 693,000 and 961,000 won, respectively. Therefore, it was concluded that the productivity of rotifer in the high density culture was higher than that in a batch culture.

This study was carried out to compare the growth and fatty acids composition of the rotifer (Brachionus rotundiformis) cultured in high density by the various enrichments and culture methods. The rotifer fed on condensed freshwater Chlorella was enriched with $\omega$-yeast, Algamac, Super Selco and marine Chlorella. In another culture method, the rotifer was cultured with enrichment supplements for 6 hours after feeding with condensed freshwater Chlorella supplement for 18 hour. The rotifer fed with condensed marine Chlorella for 24 hours without fieshwater Chlorella was used as a control group. Culture tanks (5 $\ell$ working volume) was immersed in a water bath ($28^{\circ}C$). The density of rotifer and dissolved oxygen level in water was stable in control group of rotifer cultured with condensed marine Chlorella for 24 hours and the n-3 HUFA content of rotifer was the highest among the rotifer culture methods. However, the density of rotifer and dissolved oxygen level in the groups of rotifers enriched with $\omega$-yeast, Algamac and Super Selco by methods were drastically decreased, The n-3 HUFA contents of rotifers enriched by Super Selco were higher than those of rotifer enriched by either $\omega$-yeast or Algamac in both methods. The results from this experiment indicated that supplementation of condensed marine Chlorella for 24 hour by the semi-continuous culture was effective for the improvement of the nutritional value of rotifer and it could provide the stable growth condition for rotifer culture in high density.
Production of resting egg from the Korean rotifer, Brachiunus plicatilis (L and S-type) was investigated at different temperatures (L-type : 20, 24, $28^{\circ}C$, S-type : 28 32, $36^{\circ}C$) and salinities (10, 20,30 ppt). The rotifer was cultured in 25 ml test tube and fed on Nannochloris oculata. With regard to mixis rate, L-type rotifer showed higher rate at lower temperature, and the highest rate was observed at 20 ppt of salinity at each temperature of the experiment. However, for S-type rotifer, the optimum temperature and salinity were $28\~32^{\circ}C$ and 20 ppt, respectively. The highest number of resting egg was 173 eggs/ml in 16 days at $24^{\circ}C$, 10 ppt for L-type rotifer and 410 eggs/ml in 14 days at $28^{\circ}C$, 10 ppt for S-type rotifer. The maximum number of resting egg produced per 10,000 rotifers was 8,122 eggs at $20^{\circ}C$, 20 ppt for L-type rotifer and 8,700 eggs at $28^{\circ}C$, 20 ppt for S-type rotifer. The maximum number of resting egg produced $10^8$ cells of N. oculata was 50.7 eggs for L-type rotifer ($24^{\circ}C$, 20 ppt) and 79.6 eggs for S-type rotifer ($32^{\circ}C$, 10 ppt). The number of resting egg produced per day was $1\~11$ eggs/ml for L-type rotifers and $21\~35$ eggs/ml for S-type rotifer in 9 combination experiments. In this study, S-type rotifer is better than L-type rotifer in resting egg production, and the optimum temperature and salinity for resting egg production were $20^{\circ}C$, 20 ppt for L-type rotifer and $28^{\circ}C$, 20 ppt for S-type rotifer. This result shows the difference of Korean rotifer in the optimum condition for resting egg production from other rotifers reported earlier.
Survival rate of larval red spotted grouper, Epinephelus akaara is very low, because mouth size of the larvae is too small to eat normal sizes of rotifers. In order to enhance larval survival, optimal culture conditions for larvae using different live foods and water temperatures were studied. Bigger live food produced larger size of rotifer, when the rotifer was cultured with three different live foods such as Tetraselmis tetrathele, marine Chlorella and Nannochloris oculata. Also, the size of rotifer produced was temperature-dependent showing that higher water temperature induced smaller rotifer. Survial rate of the larvae increased with the increase of water temperature up to $29{\circ}C$. Also, higher larval survials were found at the food density of 15 to 20 individuals per milliliter. The larvae fed L-type or small individuals of L-type rotifer died all within a week, although the larvae fed S-type or small individuals of S-type one survived successfully during this period. The size of small individuals of S-type rotifer collected through fine net became larger in a few hours within larval culture tank. Survival rate of grouper larvae with initial stocking density of 40 thousands in 6-ton concrete tank was 6.3% in 10 days and 0.2% in 53 days after hatching, respectively. Total length of the larvae reached around 29.5 mm in 53days after hatching.
To reduce the production cost of the rotifer, different combinations of Chlorella and Baker's yeast were used to mass culture of Brachionus calyciflorus. Rotifer density tended to decrease abruptly at 100% Chlorella-feeding, when unionized ammonia level ranged from 14.1 to 29.6 ppm and DO level from 0.2 to 0.3 ppm. Hence DO level > 0.3 ppm and unionized ammonia level < 14 ppm should be maintained. Production cost of the rotifer was the highest, when a combination of 100 % Chlorella was chosen as food source, but it was about 1.8 times less, when a combination of 70 % Chlorella and 30% yeast was used as food source. However, the highest rotifer density remained almost at the same level, when either of the combination was chosen as food source. A combination of 70 % Chlorella and 30 % yeast is recommended for maximum production of the rotifer at the lowest feed cost.

현재, 해수어와 해산 갑각류 종묘생산에 있어 해수 자치어와 유생의 초기먹이로 rotifer, Brachionus plicatilis와 B. rotundiformis를 사용한다. 또한 담수산 rotifer, calyciflorus는 담수산 자치어의 먹이로 이용이 가능하며 특히, 입이 작은 열대 관상어의 초기 먹이로 이용되고 있다. 그러나 rotifer 생산에 있어 일반적인 batch 배양 방법은 많은 공간을 차지하면서 rotifer 생산 기간 중 갑자기 전량 폐사하는 등 배양이 불안정하고 생산성을 고려할 때 양적확보를 위한 생산단가가 비교적 높다. Sixteen strains of marine rotifer, Braohionus plicatilis were isolated from salt pond, estuary and lagoon. Among 16 strains, 2 strains were large (L)-type and the others were small (S) or ultra small (US)-type. Four strains of fresh water rotifer, B. calyciflorus were isolated from commercial fish ponds. The size of lorica and resting egg were measured. In B. plicatilis, the range of lorica length from S-type and S-type were $244.3{\~}255.3\;{\mu}m$ and $131.0{\~}165.8\;{\mu}m$, respectively. The major axis of resting egg in the marine rotifer were $93.7\~116.4\;{\mu}m$ for S-type and $142.4{\~}145.5\;{mu}m$ for L-type, respectively. In freshwater rotifer, B. calyciflorus, the size range of lorica and major axis of resting egg were $211.8\~229.9\;{\mu}m$ and $126.8\~140.2\;{mu}m$, respectively. The size of freshwater rotifer was larger than that of S-type marine rotifer, but smaller than that of L-type one. Growth and formation of resting egg of B. plicatilis were different among the strains. The maximum density of S-type and L-type rotifer was 753.3 inds./ml for H-S strain and 220 inds./ml for O-L strain, respectively. The largest production of resting egg of S-type and L-type rotifer were 86.7 inds./ml for YY-S strain and 45.8 inds./ml for O-L strain, respectively.
Mass production of resting egg of the rotifer, Brachionus plicatilis was carried out in 1 $m^3$ tank for L-type rotifer and in 1 $m^3$ and 4 $m^3$ tank for S-type rotifer. L-type rotifer was fed on concentrated Chlorella + baker's yeast and only baker's yeast for 15 days. S-type rotifer was fed on concentrated Chlorella in 1 $m^3$ tank for 7 days and frozen Chlorella + baker's yeast in 4 $m^3$ for $6\~8$ days. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for L-type rotifer fed on Chlorella + baker's yeast were $54.5{\times}10^6$ eggs, $30.5{\times}10^6$ eggs and $100{\times}10^3$ eggs, respectively. These were higher than those of rotifer fed on baker's yeast only. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for S-type rotifer fed on concentrated Chlorella in 1 $m^3$ tank were $50\~104{\times}10^6$ eggs, $47\~82{\times}10^6$ eggs and $136\~260{\times}10^3$ eggs, respectively. In 4 $m^3$ tank with frozen Chlorella + baker's yeast, these were $149\~567{\times}10^6$ eggs, $36\~123{\times}10^6$ eggs and $131\~338{\times}10^3$ eggs, respectively. This result suggests that S-type rotifer are better than L-type rotifer for the mass production of resting egg of the Korean rotifer.
We studied new observation method about take process of rotifer by marine fish larvae. Till now, we can not accurate observation and count of first rotifer feeding day and/or feeding numbers of rotifer by marine fish larvae. Because take rotifer is ingested and disappeared in the digestive system of fish larvae. However we suggest possible observation method for these problems. The trophi is mastication organ of rotifer, and has only one in each rotifer individual. The trophi is left in the mastication organ because sole indigestible organ of rotifer. Therefore we can accurate observation and count of first rotifer feeding day and/or feeding rotifer numbers of marine fish larvae by trophi observation method (RTCM; Rotifer Trophi Counting Method).
