Effect of Acaromyces Ingoldii Secondary Metabolites on the Growth of Brown-Rot (Gloeophyllum Trabeum) and White-Rot (Trametes Versicolor) Fungi

  • We investigated the antifungal activities of an endophytic fungus identified as Acaromyces ingoldii, found on a loblolly (Pinus taeda L.) pine bolt in Louisiana during routine laboratory microbial isolations. The specific objectives were to determine the inhibitory properties of A. ingoldii secondary metabolites (crude extract) on the mycelial growth of a brown-rot fungus Gloeophyllum trabeum and a white-rot fungus Trametes versicolor, and to determine the effective concentration of A. ingoldii crude preparation against the two decay fungi in vitro. Results show the crude preparation of A. ingoldii from liquid culture possesses significant mycelial growth inhibitory properties that are concentration dependent against the brownrot and white-rot fungi evaluated. An increase in the concentration of A. ingoldii secondary metabolites significantly decreased the mycelial growth of both wood decay fungi. G. trabeum was more sensitive to the inhibitory effect of the secondary metabolites than T. versicolor. Identification of specific A. ingoldii secondary metabolites, and analysis of their efficacy/specificity warrants further study. Findings from this work may provide the first indication of useful roles for Acaromyces species in a forest environment, and perhaps a future potential in the development of biocontrol-based wood preservation systems.

  • The incitant of soft rot on burley tobacco leaves in the curing vinylhouse was identified as Erwinia carotovra subsp. carotovora on the basis of its physiological characters. The bacterium yew best at $25^{\circ}C$ and $30^{\circ}C$, but no growth was detectable at $40^{\circ}C$ in the nutrient broth for 24 hours period. Burley tobacco leaves inoculated with the bacteria (Ecc) produced typical soft rot lesions when the incubation temperature was 25 to $30^{\circ}C$ and the relative humidity was more than 8075, however, the lesion development was suppressed when the temperature was $40^{\circ}C$ and the relative humidity was below 80%. Significant negative correlation was found between hanging space in the curing vinylhouse and the incidence of soft rot on the tobacco leaves harvested in a rainy day regardless of streptomycin treatment.

소 발굽질병에 대한 水針의 治療效果 (The Therapeutic Effect of Aquapuncture on Bovine Foot Rot)

  • To clarify the therapeutic effect of aquapuncture therapy on bovine foot rot, the experimental cattle were divided into control (conventional treatment), aquapuncture and combined treatment groups. Each group was composed of 6 Holstein cows with bovine foot rot. In the present study, the change in grade of lameness, total leukocyte counts, N/L. ratio, mean recovery rates (days) and histopathological changes of interdigital tissue before and after treatment were evaluated. The grade of lameness was decreased by treatment in each group, with the decreasing in order of combined > aquapuncture > control group. In change of total peripheral blood leukocyte counts, the tendency of decrease was found. Significances were detected on 9th day (P < 0.05) in control, on 6th day (P < 0.01) in aquapuncture group, and on 3rd (P < 0.05) and 6th day (P < 0.05) in combined group, respectively. In addition, significances were detected on 3rd and 6th day in aquapuncture, and on 3rd day in combined group by comparison with that of control, respectively. N/L ratio was decreased by therapy in each group with the decreasing in order of combined > aquapuncture ) control group. Significances were detected on 6th day (P < 0.01) between aquapuncture and combined groups. The mean recovery rates(days) were rapid with the decreasing in order of combined > acuapuncture > control group. In histopathological findings of interdigital tissue, the findings of inflammation and hemorrhage were not observed after treatment in each group. In conclusion, it was thought that aquapuncture and combined therapy was very effective on treatment of bovine foot rot.

Occurrence of Phytophthora Root Rot on Kiwifruit in Korea

  • A severe root rot of kiwifruit caused by a species of Phytophthora occurred in 1-to 5-year-old vines at the south coast region of Korea in 1997. Infected vines exhibited leaf chlorosis, scorch and defoliation, root and stem rot, and eventual death. The disease was relatively severe in poorly drained lowlands, of which 19 out of 23 fields were damaged by the disease. Meanwhile, only one among 58 upland fields was infected by the disease. Incidence of infected vines reached over 80% in heavily damaged fields and a species of Phytophthora was isolated from inner tissues of roots, stems, and rhizosphere soils of the plants. The causal pathogen was identified as P. drechsleri based on its mycological characteristics. Pathogenicity of the fungus was confirmed by artificial inoculation to seedlings of kiwifruit 'Hayward'. The pathogen was re-isolated from the inoculated plants showing symptoms similar to those observed in the fields. Root rot of kiwifruit caused by P. drechsleri has not been reported previously in Korea.

  • In order to establish an appropriate spray schedule to reduce the infection, 11 kinds of chemicals were tested for their properties on the inhibition of spore germination at the surface of apple fruits and the duration of the inhibitory effect after spray of each chemical was examined from late June to early September with basically 10 day intervals. Actual control efficacy of each chemical by the 8 successive spray and the patterns of waterborne spore dispersals during that periods were also examined. Combining those results with the meteorological observation data, actual control efficacy of each chemical in the given periods could be estimated. It was revealed that folpet, Brodeaux mixture, mancozeb, oxine copper and imminoctadine-triacetate could be used at any time during the possible infection periods. Captan and dithianon could also be used except the rainy season due to the short duration of inhibitory efficacy against spore germination under heavy rain. However, the usefulness of propineb, benomyl and chlorothalonil against the apple white rot could not be demonstrated in this experiment. Thiram, even though has not been used for apple white rot, can also be used before or after the rainy season to control not only white rot but also alternaria blotch.

Occurrence of Sclerotinia Rot in Cruciferous Crops Caused by Sclerotinia spp.

  • Cruciferous crops grown in greenhouses and fields in Korea were surveyed from 1995 to 2000. Sclerotinia rot most severely occurred up to 30% in cabbage. Incidence of the disease was as high as 20% at its maximum in Chinese cabbage and rape and 10% in radish, but as low as less than 1 or 2% in broccoli and kale. Symptoms of Sclerotinia rot commonly developed on loaves and stems of the crucifers, but rarely on rachises of broccoli. A total of 112 isolates of sclerotinia species was obtained from the diseased crucifers. Out of the isolates, 103 isolates were identified as S. sclerotiorum, and the rest as 5. minor based on their morphological and cultural characteristics. S. sclerotiorum was isolated from all the crucifers, while S. minor was isolated from Chinese cabbage, broccoli, and kale. Six isolates of S. sclerotiorum and three isolates of S. minor were tested for their pathogenicity to the crucifers by artificial inoculation. All the isolates of the two Sclerotinia spp. induced rot symptoms on the plants of the crucifers tested, which were similar to those observed in the fields. The pathogenicity tests revealed that there was no significant difference in the susceptibility of the crucifers to the isolates of S. sclerotiorum. However, in case of S. minor, radish was relatively less susceptible to the pathogen.

  • 인삼 근부병의 생물학적 방제를 위해 인삼 재배토양으로부터 분리된 길항미생물 Bacillu subtilis B-4228을 사용하여 방제효과를 검정하였다. 공시 균주 B-4228과 C. destructans의 대치배양결과, C destructans의 균사가 비정상적인 형태로 팽창하였으며 강한 생장억제력을 보였다. C. destructans에 의해 오염된 이병토를 이용한 pot시험결과, 건전근율이 무처리구에서 20%였으나 공시 균주 B-4228의 처리구는 82%로 높게 나타났고, 이병율(개체 인삼에 대한 뿌리썩음 이병정도의 총합비율)은 무처리의 경우 50.4%였으나 공시 균주 B-4228의 처리구는 6.0%로 낮았다.

Stem Rot of Tatarian Aster(Aster tataricus) Caused by Sclerotium rolfsii in Korea

  • In July 2001, a destructive stem rot of tatarian aster(Aster taturicus) was occurred sporadically in exhibition farm of Gyeongsangnam-do Agricultural Research and Extension Services, in Hamyang, Korea. The fungus also caused collar and crown rot and systemic wilt or blight of whole plant. White mycelium spread over stems and petioles of infected plants and sclerotia formed on the old lesions and near the soil surface. The fungus showed maximum mycelial growth was obtained around $30^{\circ}C$ but did not grow below $5^{\circ}C$ or above $45^{\circ}C$. The mycelial width ranges $4.2{\sim}10.4{\mu}m$. and the color is white, usually many narrow mycelial stand grow in the aerial mycelium and formed clamp connection. Numerous sclerotia were formed in artificial media like PDA at $30^{\circ}C$. The shape of sclerotia were sphere and $1.0{\sim}3.2{\mu}m$ in diameter. The fungus was isolated repeatedly from the infected, tissues and confirmed its pathogenecity to aster and identified as Sclerotium rolfsii. This is the first report that Sclerotium rolfsii causes stem rot of tatarian aster in Korea.

Control of Root Rot and Wilt Diseases of Roselle under Field Conditions

  • Roselle (Hibiscus sabdariffa L.) is one of the most important medicinal crops in many parts of the world. In this study, the effects of microelements, antioxidants, and bioagents on Fusarium oxysporum, F. solani, and Macrophomina phaseolina, the causal pathogens of root rot and wilt diseases in roselle, were examined under field conditions. Preliminary studies were carried out in vitro in order to select the most effective members to be used in field control trials. Our results showed that microelements (copper and manganese), antioxidants (salicylic acid, ascorbic acid, and EDTA), a fungicide (Dithane M45) and biological control agents (Trichoderma harzianum and Bacillus subtilis) were significantly reduced the linear growth of the causal pathogens. Additionally, application of the previous microelements, antioxidants, a fungicide and biological control agents significantly reduced disease incidence of root rot and wilt diseases under field conditions. Copper, salicylic acid, and T. harzianum showed the best results in this respect. In conclusion, microelements, antioxidants, and biocontrol agents could be used as alternative strategies to fungicides for controlling root rot and wilt diseases in roselle.

Effect of Brown-rotted Wood on Mechanical Properties and Ultrasonic Velocity

  • Artificial brown-rot decay was induced to two wood species, Pinus densiflora and Pinus radiata. A modified direct inoculation method was used and the decay indicators of mass loss and two compressive mechanical properties, maximum compressive strength (MCS) and compressive stiffness, were estimated over the period of 8 weeks of fungal exposure. Measurable mass loss occurred 2 weeks after the fungal attack, with 15% to 22% of the loss occurring 8 weeks after fungal exposure with Fornitopsis palustris and Gloeophyllurn trabeurn. Mechanical properties proved to be far more sensitive than mass loss detection: approximately five to six times by quantity. Of the two mechanical properties, MCS was more sensitive to and consistent with progressive brown-rot decay. An ultrasonic test was performed to determine the feasibility and accuracy of this method for nondestructive detection of brown-rot decay. The ultrasonic test is highly sensitive at qualitative detection of the early stages of brown-rot decay.
