The timing for the determination in root formation from nodal and internodal explants of cassava (Manihot esculenta Crantz, cv. MCol 22) was investigated. Nodal explants about 10 mm with an axillary bud formed adventitious roots directly on MS basal medium for 8 days of cultures. But internodal segments without an axillary bud did not develop the adventitious roots on the same medium, and most internodal segments excised from nodal explants after cultures of 5 days on MS basal medium developed adventitious roots. On the other hand, the internodal segments rooted at 90% after cultures on medium with 0.5 mg/L IBA for 5 days, with 1 mg/L IBA for 2.5 days, and with 2 mg/L IBA for 1.5 days respectively. Thus the period of culture on medium with IBA and its IBA concentration affected the rooting rate. Therefore, it is suggested that the determination for root formation occurred before the differentiation of root primordia on medium with IBA, and root inducing factors in medium were absorbed and accumulated during the period of determination for root primordium differentiation in internodal segment of cassava.
Recently, a nursery mat made from rock wool has realized transplanting of the younger seedlings with the ordinary transplanting machines for Chibyo and Chubyo(3 and 4~5 leaved seedling, respectively). The seedlings defined as the 'Nyubyo' or 'Nursling seedlings' became possible to achieve economic profits from the reduction in both working time and costs. It being widely noticed as a strategy to solve the difficulties in current rice cultivation. The nursling seedlings are 1.4 to 2.5 leaves and height at 4.5 to 7cm, grown 4 to 7 days after seeding. They maintain still up to 50 to 80% of their nutrients in the endosperm, and can grow by using only their own nutrients for a certain period of time after transplanting. Nursling seedlings take 2 days in the nursery chamber at 32$^{\circ}C$ after seeding, and 2 days in the greening house at $25^{\circ}C$. This is only 4 days, all together, to make the nursling seedlings of 1.5 leaves which are ready for transplanting. Watering is only needed once at the sowing time. It only takes 1 or 2 waterings even to raise a seedlings for a period of 7 days. The number of nursery boxes can be reduced because it is possible to sow more densely(220 to 240g per box), thus it only needs seedlings of 15 to 16 boxes per 10 a which leads to a reduction in facilities and space needed. Temperature during the nursery period can be artificially adjusted more precisely which may lead to the prevention of temperature stress. The nursling seedlings can root rapid by because the crown roots from the coleoptile node begin to emerge immediately after transplanting. They show strong resistance to low temperature (12$^{\circ}C$) and deep-planting. There is no danger in the rooting of the seedlings even if half of their height is buried into the soil. Moreover, it can root at a rate of up to 65 to 80% even if the full height of the seedlings is buried. They show also strong resistance to submergence (10~15cm). The nursling seedlings tend to grow by producing tillers from lower nodes. It is therefore, necessary to control to keep the proper numbers of tillers per unit area. They have no fear in the delay of heading and their yield components can be so well balanced that the same level of yield was achieved with the nursling seedlings compared to that with Chibyo. It was further suggested that if the surplus tillers can be avoided by such cultivation practices, the number of grain per panicle can be kept greater and higher yield can be realized. Practical experiments with the nursling seedlings conducted in 1989 and 1990 by farmers in various areas showed exciting results. This research is mainly to deal with the effects of submergence treatment on the grain yields of two rice plants, local variety, "Akibare" and Tongil line variety, "Milyang 23". The results obtained are summarized as follows. 1. According to the rice products of each plot the grain yield index was smallest in the plot treated at the early heading stage. The index of the next order became smaller in order of late flowering stage, late reduction division stage, milk ripe stage and dough ripe stage etc. The submerged stage at which the damages were smallest was tillering stage. Under the condition of two thirds or one third submerged depth of plant height, few differences in the grain yield index between Milyang 23 and Akibare was found, but except rooting stage, the damages of milyang 23 by whole submergence during growing period were mostly greater than those of Akibare. Especially the grain yield index of early heading stage at which the damages by whole submergence was most serious showed 45 percentage for one day submergence, 31 percentage for 3 days and 0.7 percentage for 7 days in Akibare plots, and 26.7% percentage for one day submergence, 7.9 percentage for 3 days and none for 7 days in Milyang 23 plots. 2. All the factors such as submerged stage, submerged depth and submerged period in this experimental test were highly recognized significance. The factors of the submerged depth and duration influenced on greater damages than the others. According to the difference in grain yield between plots, the larger the submerged depth and duration were, the larger the significant difference appeared. And between the treated levels at other submerged stages except both early tillering stage and most active tillering stage, the significance in the differences in grain yield was recognized, while only the submergence at early heading stage showed the most serious damages. 3. The decreased rate of grain yield for one day submergence at early heading stage indicated that in case of whole submergence of plant height it was 73 percentage in Milyang 23 plot and 55 percentage in Akibare plot, and in the event of two thirds and one third submergences of it, 20 percentage and 10 percentage in both Mulyang 23 and Akibare plots respectively. Therefore, the current criteria for planning project that restricted allowable submergence duration of more than 30cm submerged depth to 24 hours, should be amended not to exceed the submerged depth of 60cm when the duration of more than allowable submerged depth of 30cm will be limited to 24 hours, or within the limits of 12 hour submerged duration for locai variety and of less than 12 hour duration for Tong-illine variety as long as possible in case that submerged depth will be allowed to more than 60cm depth.ore than 60cm depth.
Domestic and international production of Tricholoma matsutake has decreased owing to matsutake forests being left alone, host plant disease, forest fires, climate change, and so on. In order to identify strains that are suitable for the production of T. matsutake-inoculated seedlings, Pinus densiflora seedlings were inoculated with T. matsutake after in vitro rooting and mycorrhization was examined in the roots of T. matsutake-inoculated seedlings after 6 months. The mycorrhization rate was greater than 80% for 5 strains (NIFoS 421, 434, 1681, 1984, and 2001) out of 19 total strains. Seven strains (NIFoS 434, 441, 561, 562, 1016, 1807, and 1812) showed shoot/root ratios of less than 3.0 and had a seedling shoot biomass of 2.0 to 4.8 times higher than that of the root. Eight strains (NIFoS 441, 561, 562, 1016, 1807, 1812, 1984, and 2001) stimulated increases in shoot volume and three stains (NIFoS 441, 562, and 1812) promoted the growth of root biomass by mycorrhizal formation. In conclusion, 4 strains (NIFoS 434, 561, 1984, and 2001) out of 19 total strains tested showed higher mycorrhization rates and seedling growth than those of the other strains. We expect that the use of these four strains may contribute to T. matsutake-inoculated seedling production.
This study was conducted to establish the in vitro optimal condition for seed germination and organogenesis of wild Angelica gigas. The experiment was evaluated the effects of $GA_3$ for pre-treatment with different periods of time (0h, 24h, 48h, 72h) and followed the treatment of seeds by control, scarification and methanol-heating method. As a result, the highest rates (15%) of seed germination was shown under the treatment without soaking of $GA_3$ and methanol-heating treatment. The seed germination was highly increased 60% under the condition of treatment on ultrasonic waves (frequency 80 KHz) with methanol-heating treatment including 0.1 mg/L $GA_3$. The highest callus induction rate was obtained from in vitro germinated stem, root and hypocotyl on the MS medium with 1.0 mg/L NAA and 0.5 mg/L BA. The highest percentages of shooting (50%) and rooting (85%) induction were observed in hypocotyl and root cultured on PGRs free medium and 0.1 mg/L NAA, respectively. In addition, somatic embryogenesis was observed from stem (1.0 mg/L 2,4-D) and hypocotyl (0.1 mg/L NAA).
The leaf and petiole segments of Farfugium japonica were cultured to investigate the influence of growth regulators on their callus induction and plant regeneration. The callus induction and growth showed a good response both leaf and petiole on MS media supplemented with 1∼2 mg/L 2,4-D and 1∼2 mg/L BA. Callus induction and growth were more effective in petiole segments than leaf one. The highest percentage of plant regeneration was obtained from 60-day-old calli on MS medium supplemented with 1 mg/L NAA and 2 mg/L BA. When subcultured to the same medium for about 60 days, multiple shoots were developed from regenerating callus. The shoots produced roots after transferring to rooting medium containing 0.5 mg/L IAA. The plantlets over 50 mm in height were successfully acclimatized in vermiculite, and the survival rate was over 95%.
The axillary buds of 15-year-old Tilia amurensis were cultured on Saito and Ide (IS), Murashige and Skoog (MS) media and woody plant medium (WPM) to establish an effective micropropagation method. Five levels of 6-benzylaminopurine (BAP) were tested. On IS medium and WPM addition of 1.0/l BAP enhanced shoot development and shoot elongation, whereas addition of 0.5/l BAP was effective on MS medium. A better results were obtained from WPM with 1.0/l BAP and MS with 0.1/l BAP. Developed shoots were subcultured on each basal media but with 0.2/l BAP, Multiple shoots were almost doubled in a month. Root formation could be enhanced at higher concentration of indole-3-butyric acid (IBA). Better rooting rate (83.3%) was achieved on a half-strength MS medium with 3.0 /l IBA. Regenerated plantlets were successfully transferred to soil. To investigate the clonal variation in shoot development and shoot elongation by axillary bud culturing, seven plus tree clones were tested, Clonal variation in tissue culturability among plus trees was recognized by the Duncan's multiple range test at the 5% level. Kang Won No. 12 showed the best response on WPM with 1.0/l BAP.

The root-mat formation of infant rice seedlings (8- to 10-day-old) is sometime not enough for machine transplanting because of the too-short nursery period. This experiment was conducted to elucidate the effect of metalaxyl seed-soaking treatment in the mixed solution of metalaxyl, seed disinfectant and insectcides on the root-mat formation of infant rice seedling in machine transplanting. The rice seeds of Odaebyeo were soaked in the mixed solution of metalaxyl, prochloraz and insectcides. with the recommended concentrations for 24 hours at room temperature. Seeding rate was 220g per seed tray (30 x 60 x 3cm). Metalaxyl (25% wettable powder), a fungicide. was used in 1. 000 times dilution as a promoting substance on the root-mat formation. Generally, the metalaxyl-treated seeds markedly increased the root number and length, and rooting activity of infant rice seedlings as compared with the control, thus the root-mat formation was excellent. When the rice seeds were sterilized by the mixed solution of prochloraz and insectcides, metalaxyl could be used together for increasing root-mat rormation of infant rice seedlIngs due to no interaction among agro-chemicals used. Seedling damping-off and physiological seedling rot were also controled in the seedlings of metalaxy-treated seeds. The root-mat of metalxyl-treated seeds had higher hardness and tension than control in terms of physical properties.
The duration of raising seedling of infant rice seedling(IRS) in machine transplanting is 8 to 10 days. This experiment was conducted to investigate the minimum duration of IRS's raising seedling and its method by the combination of several treatments such as plant growth regulator, root-break-seat and temperature. The rice seeds of Hwaseongbyeo was soaked in the solution of metalaxyl for 24 hours at room temperature. Metalaxyl (25% wettable powder), a fungicide, was used in 1, 000 times dilution as a promting substance on the root-mat formation of IRS. The application of fungicide in the nursery soil for the controlling of damping-off and physiologyical seedling rot was omitted due to the metalaxyl seed-soaking treatment. Seeding rate was 220g per seed tray (30$\times$60$\times$3cm). To promote the root-mat formation of IRS, the sheets of polyethylene vinyl and absorptive paper were placed bottom the seed tray. The root-mat formation of IRS was promoted at higher temperature and longer duration of raising seedling. The metalaxyl-treated seeds markedly increased the root length and rooting activity of IRS as compared with the control, thus the root-mat formation was excellent. The absorptive paper with polyethylene vinyl as root-break-seat showed a better root-mat formation compared with control, polyethylene vinyl, alone. The minimum duration of raising seedling of IRS was 5 days after sowing based on the root-mat formation and seedling height under the condition of metalaxyl seed treatment, absorptive paper with polyethylene vinyl as a root-break-seat and the raising seedling temperature 30/2$0^{\circ}C$ (day /night).

Calystegia soldanella L. is a perennial herbaceous halophyte belonging to the convolvulaceae family, which mainly grows in coastal sand dunes in Korea. Shoots and rhizomes are edible, and roots called 'Hyoseon Chogeun' are known to have medicinal effects such as antipyretic, sterilization, and diuretic. In addition, physiological activities of antioxidant, anti-inflammatory, antiviral, antifungal and PTP-1B (protein tyrosine phosphate-1B) inhibition have been reported. In this study, in vitro induction cell lines of C. soldanella L. collected from the coastal sand dunes in Jeju island was redifferentiated into adventitious roots that can be used as medicinal resources. Also the biomass of mass-proliferated adventitious roots using a bioreactor were evaluated. Plants of C. soldanella L. were collected from the crevice of the seashore in the coastal area of Taeheung 2-ri, Namwon-eup, Seogwipo-si. Then, it was separated into leaves, stems, rhizomes, and roots, and surface sterilized with 70% ethyl alcohol and 2% NaOCl (sodium hypochlorite). After washing with sterilized water, each organ section was cultured in Hormone-free MS medium (Murashige & Skoog Medium). As a result, the induction response rates were evaluated at 85% and 55%, respectively, in terms of callus formation and shoot generation in the rhizome segment. In the case of the adventitious roots morphological characteristics induced by single-use treatment of auxin-based plant growth regulators IBA and NAA from redifferentiated shoots were compared. Most efficient adventitious root culture method as a rooting rate, number, length, and biomass proliferation in the bioreactor system was confirmed when treated by culturing in MS salts, Sucrose 30 g·L-1, and IBA 1mg·L-1 for 4 weeks. In this study, the medium composition and culture period were confirmed using a bioreactor system to mass-proliferate adventitious roots derived from C. soldanella L. in Jeju island. Also this adventitious root line developed a new medicinal material could increase value of the bio-industry ingredient through quantitative and qualitative screening of phyto-bioactive compounds.
