In this study, we evaluated the efficiency of chemical marking of black rockfish scales by immersion in oxytetracycline hydrochloride (OTC, 500 ppm), alizarin red S (AR, 250 ppm) and calcein (CARL, 250 ppm) diluted rearing water. Immersion treatment of chemicals had no effects on both mortality and growth of black rockfish. Marking sucess was 100% in all treatment durations (24, 48 and 72 hours) with three chemicals and marking quality was higher in 48 and 72 hours than 24 hours treatment. Marking retention rates at 24 weeks after treatment were 100% in OTC and CAL treated group, but marking quality was higher in CAL treated group (brilliant 92%, bright 8% and dim 0%) than in OTC treated group (brilliant 4%, bright 70% and dim 26%). AR treated group had lower marking retention rates and marking quality than OTC and CAL treated group. As a results, immersion treatment with OTC and CAL was effective in marking scales of black rockfish and practical in releasing program and other studies requires same rearing environment.
The residue depletion of ampicillin was investigated in the olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major) after 5 days treatment with medicated feed at a dose of 100 mg/kg bw/day. Fishes were sampled for muscle on 1st, 2nd, 3rd, and 4th day after treatment. Ampicillin concentrations were determined by high performance liquid chromatography after SPE column extraction. The recovery rates of ampicillin in muscle samples ranged 94-98% and 83-88% for the concentration of 0.05 mg/kg and 0.1 mg/kg, respectively. Ampicillin concentrations detected on 1st day after treatment were 0.143, 0.138, and 0.187 mg/kg in the muscle of olive flounder, rockfish, and red sea bream, respectively. After a withdrawal of 3 days, muscle concentrations were 0.016, 0.012, and 0.021 mg/kg in the olive flounder, rockfish, and red sea bream, respectively. Ampicillin was not detectable in muscle samples on 4 days following withdrawal of the medicated feed. From results of the present study, a withdrawal period of ampicillin is proposed on 5 days after 5 days treatment with medicated feed at a dose of 100 mg/kg bw/day to avoid the presence of excessive residues of the edible muscles of olive flounder, rockfish, and red sea bream.
Amoxicillin을 넙치, 조피볼락 그리고 참돔에 각각 400 mg/kg bw/day의 용량으로 7일 동안 사료와 혼합하여 경구 투여한 다음, 휴약기간 동안 근육조직 내 잔류 분포를 조사하였다. 실험어는 해수 중에서 일정한 크기의 케이지에 일반 상업용 사료를 주어 사육하였고, 실험에 사용하기에 앞서 15일 동안 환경에 적응시켰다. 약제 투여 후, 근육시료는 1, 2, 3, 4, 그리고 5일에 각각의 실험어를 대상으로 채취하였다. Amoxicillin의 잔류분석은 형광검출기를 부착한 고속액체크로마토그래피를 이용하여 분석하였다. Amoxicillin의 회수율은, 0.05mg/kg의 농도에서 84.3-101.3%, 0.1mg/kg의 농도에서는 75.0-91.5%를 보였다. 투약 후 1일에는, 참돔의 근육 중 amoxicillin의 잔류농도가 넙치와 조피볼락의 근육 중 잔류농도에 비하여 높았으나 통계적 유의성은 없었으며, 투약 후 4일에는, 모든 근육 시료에서 amoxicillin이 검출되지 않았다. 이상의 결과로부터, amoxicillin의 사료혼합을 통한 경구 투여는 넙치, 조피볼락 그리고 참돔의 근육 중에서 안전휴약기간(7일) 보다도 체래 소실이 빨리 일어나는 것으로 추정되는 바, 안전휴약기간을 준수한다면 amoxicillin의 어류 근육 조직 내 잔류로부터 안전할 것으로 사료된다.
본 연구는 감성돔(Acanthopagrus schlegeli)에 기생한 Amyloodinium sp.의 성상을 알기 위하여 이 기생충의 형태 계측학적인 특징을 조사하는 한편, 지속적인 감염을 유도하여 충체의 발달과 번식 특성 및 병리 조직상을 확인하고자 하였다. 이 기생충의 형태는 크게 기생기(parasitic phase)와 비기생기(nonparasitic phase)로 구분할 수 있었고, 영양형(trophont), 분열형(tomont), 자충(dinospore)의 3단계의 발달과정을 거치는 것을 알 수 있었다. 방추형 또는 난형인 기생 충체는 색깔이 없었으며, 직경 30-$80{\mu}m$ 크기였다. 성숙한 충체는 크기가 약 $80{\mu}m$ 내외로 체내에 $15{\mu}m$의 둥근 핵과 다수의 식포 및 전분성 과립을 함유하고 있으며, 가근상 돌기가 있는 부착기로 아가미 기저막에 부착해 있었다. 비기생기인 분열형은 80-$90{\mu}m$의 크기로 어체로부터 이탈된 직후 구형의 Cyst wall을 가지고 있던 것이 분열과 함께 Cyst wall의 모양이 사각형으로 변하였으며, 지속적인 2분열로서 감염형을 생성하였다. 8회의 분열로서 생성된 감염형은 $10-15{\times}10-14{\mu}m$의 크기로 전체 형태가 둥글고 양끝이 편평하였으며 체내에 다수의 과립을 가지고 있었다. 그리고 자충은 2개의 편모(충체 중앙부에 허리띠 모양의 가로 편모와 뒤로 길게 늘어뜨린 세로 편모)를 가지고 있는 것을 볼 수 있었다. 또한 안점은 기생 초기의 충체와 자충에서 관찰되었다. 조피볼락(Sebastes schlegeli)에 인위 감염을 유도하여 발달과 번식 특성을 확인한 결과, 이 충은 수온 24-$26^{\circ}C$의 어체에서 3-5(평균, 3.75)일만에 성숙하며, 이탈하여 수중에서 3-4(평균, 3.3)일만에 분열하여 256개의 자충을 생성하는 것으로 조사되었다. 한편, 이들 감염어의 아가미 조직은 감염 초기에 뚜렷한 병변을 나타내지 않지만, 감염이 진행되면서 지속적인, 재감염으로 인하여 아가미 상피세포의 과잉 형성을 일으켜 새판이 비후되어 곤봉화되고 결국 감염어는 폐사되는 것으로 판단되었다.
Two feeding experiments were conducted to investigate the effects of dietary genetically modified (GM) soya and com on growth performance, feed utilization and body composition of juvenile olive flounder, Paralichthys olivaceus and rockfish, Sebastes schlegelii. For each fish species, four isonitrogenous (50% crude protein) and isocaloric (4.1 kcal/g) diets (designated as nGM soya, GM soya, nGM com and GM com) were formulated to contain 20% non-GM (nGM) and GM soya and com. Thirty olive flounder (initial body weight, 15.4${\pm}$0.4 g) and fifty rockfish (initial body weight, 3.1${\pm}$0.02 g) were distributed in each 400 L tank (200 L water) in a flow through system. Each experimental diet was fed to triplicate groups of fish to visual satiation, twice a day (9:00 hand 17:00 h) for 6 weeks. Growth performance was measured every three weeks. No effects of GM feedstuffs on survival were observed. Dietary inclusion of GM feedstuffs did not affect growth performance and feed utilization of fishes, except for rockfish fed GM com. Rockfish fed the GM com diet showed higher weight gain, daily feed intake and daily protein intake than did fish fed the nGM com diet, but no significant differences were observed in final body weight between the dietary treatments. Condition factor, hepatosomatic index, visceral somatic index and body composition were not altered by the inclusion of GM feedstuffs. These results indicate that dietary inclusion of GM soya and com could have no effects on growth performance and feed utilization of juvenile flounder and rockfish. Lower weight gain and feed intake in flounder and rockfish fed the diets containing 20% soya were likely due to anti-nutritional factors, rather than transgenic factors in the feedstuffs. Dietary inclusion of GM soya and com at the level tested did not alter the body composition of fishes. Further studies to investigate the effects of GM feedstuffs on health conditions and the development of fishes, as well as those of residue of transgenic fragments in ambient environments and in animals are necessary for safe use of the ingredients in aquaculture.
이 실험에서는 조피볼락을 대상으로 중성 formalin과 formalin으로 약욕하여 잔존량과 잔존기간 및 병리 조직학적 변화를 관찰하고 온도가 미치는 영향에 대해서 알아보았다. formalin 과 중성 formalin의 잔존량은 $15^{\circ}C$일 때 약욕 직후 약간의 차이를 제외하고는 별다른 차이를 보이지 않았다. 잔존기간은 수온 $15^{\circ}C$와 $25^{\circ}C$은 조건에서 각각 72 hr과 24 hr으로 온도의 영향을 많이 받는 것으로 나타났다. 어체에 미치는 독성은 중성 formalin보다 formalin이 강하게 나타나고 수온과 농도가 증가 할수록 조직에 미치는 영향이 심해졌다. formalin 약욕으로 인하여 아가미 조직에서는 부종과 상피세포의 박리, 뒤틀림, 괴사증상이 그리고 간에서는 부종과 핵농축, 공포화 현상이 나타나고 신장에서는 수증성 퇴행적 병변과 세뇨관 상피세포의 과립화 중상, 괴사가 일어나고 피부에서는 점액 세포의 증가와 표피의 괴사 및 피하조직과 진피층의 균열이 나타났다.
본 연구에서는 우리나라 연안의 대표적인 저서단각류인 Monocorophium acherusicum과 대표적인 어류인 조피볼락 Sebastes schlegeli에 대한 TBT와 여러 대체 방오도료(antifouling agent)의 급성독성영향을 평가하였다. 과거에는 방오도료로서 tributyltin(TBT)이 광범위하게 이용되어 왔으나, TBT는 매우 미량의 농도에서도 다양한 생물학적 저해영향을 일으키는 것이 알려져 이를 대체하기 위한 여러 화학물질들이 개발되어 왔다. 이중에서 Sea-Nine 211, Cu-pyrithione, 그리고 Zn-pyrithione 등은 최근에도 널리 이용되는 방오도료이다. 하지만 아직까지 이들 물질에 의해 국내 연안의 생물과 생태계가 어떤 저해 영향을 받을 수 있는지에 대한 충분한 연구는 부족하다. 실험결과 개별 화학물질의 독성영향은 농도와 노출시 간에 비례하여 증가하였다. 물질별로 비교해보면, Cu-pyrithione는 조피볼락에 대한 96시간 반수치사농도(LC50)가 $56{\mu}g{\cdot}1^{-1}$으로 독성영향이 가장 큰 것으로 나타났고, $TBT(73{\mu}g{\cdot}1^{-1};96-h\;LC50),\;Sea-Nine(184{\mu}g{\cdot}1^{-1}),\;Zn-pyrithione(l707{\mu}g{\cdot}1^{-1})$의 순으로 독성영향이 감소하였고, 단각류에 대해서는 $TBT(26{\mu}g{\cdot}1^{-1}$가 가장 독성영향이 크고, 이어 Sea-Nine($49{\mu}g{\cdot}1^{-1}$), Cu-pyrithione($119{\mu}g{\cdot}1^{-1}$), Zn-pyrithione($334{\mu}g{\cdot}1^{-1}$) 의 순으로 영향이 감소하였다. 이와 같은 물질별, 생물별 급성독성영향 에 대한 평가결과는 향후 이들 물질의 생태계위해성을 평가하기 위한 기초자료로서 활용되어질 수 있으나, 향후 보다 다양한 생물에 대해 급만성 독성영향을 평가하는 연구가 필요하다.
1998년 3월부터 2000 4월까지 한국 남해안과 서해안에서 채집된 해산어 5종을 대상으로 어류의 위 및 장내에 기생하는 흡충류 종들을 형태학적 특징에 의해 분류 기재하였다. 본 실험을 위해서 사용된 어종은 조피볼락(Sebastes schlegeli), 쥐노래미(Hexagrammos otakii), 문절망둑(Acanthogobius flavimannus), 도다리(Pleuronichthys cornutus), 수조기(Nibea albiflora)의 5 어종으로, 그 중 4 어종은 금강하구입구 근처 고군산 군도에서 채집하였고, 쥐노래미는 고군산 군도와 완도에서 채집하였다. 흡충류 4종이 해산어류에서 검출되었다: 1)고군산 군도에서 채집된 조피볼락, 도다리, 쥐노래미의 위와 장에서 Opecoelus lobatus Ozaki, 1925가 검출되었고, 2)고군산 군도에서 채집된 문절망둑의 장내에서 Coitocoecum glandulosum Yamaguti, 1934 이 검출되었고, 3)고군산 군도에서 채집된 수조기의 장 내에서 Phyllodistomun sp. Braun, 1899 가 발견되었으며, 4)완도에서 어획된 쥐노래미의 위와 장에서 Pharyngora sp. Lebour, 1908 가 검출되었다. 해산 어류에서 검출된 4 종의 흡충류 중 3종(Opecoelus lobatus Ozaki, 1925, Phyllodistomun sp., Pharyngora sp.)은 국내에서 처음으로 발견된 종들이였다.
색광에 대한 조피볼낙 Sebastes schlegeli의 행동을 조사하기 위하여, 2가지씩의 서로 다른 색광을 조합하여 수조의 양단에서 동시에 광자극을 가했을 때의 반응을 주간과 야간으로 구분 조사한 결과는 다음과 같다. 1. 수조내에서의 조피볼낙의 구간별 분포곡선은 대체로 L 자형과 U 자형으로 나타났다. 2. 양색광원쪽에서의 평균분포차는 $13.50\%(0.70{\sim}31.65\%)$였고, 주간($17.03\%$) 보다 야간($9.98\%$)에 적었다. 3. 조피볼낙이 잘 모이는 색광은, 주간에는 청색, 백색, 황색, 적색의 순이며 야간에는 황색, 청색,n 백색, 적색의 순으로 나타났다. 4. 조명시간의 경과에 따른 집어율의 변화는 심했으며 계속 불안정하게 변동했다. 5. 두가지 색광에 대한 집어율의 차는 비교적 뚜렷하였는데 야간보다 주간에 더 뚜렷하였다.
