An 8-week feeding trial was conducted to investigate the effects of dietary taurine supplementation on the growth performance of juvenile rock bream, Oplegnathus fasciatus. Triplicate groups of 25 fish averaging $2.72{\pm}0.04$ g ($mean{\pm}SD$) were fed one of six experimental diets prepared by adding taurine at 0%, 0.25%, 0.5%, 1.0%, 1.5% and 3.0% (Control, $Tau_{0.25}$, $Tau_{0.5}$, $Tau_{1.0}$, $Tau_{1.5}$ and $Tau_{3.0}$, respectively). At the end of the feeding trial, the weight gain, specific growth rate and protein efficiency ratio of fish fed the $Tau_{0.5}$, $Tau_{1.0}$ and $Tau_{1.5}$ diets were significantly higher (P < 0.05) than those of fish fed the Control and $Tau_{0.25}$ diets. The feed efficiency of fish fed the $Tau_{0.5}$ diet was significantly higher than that of fish fed the Control, $Tau_{0.25}$ and $Tau_{3.0}$ diets. Fish fed the $Tau_{1.0}$ diet had higher whole-body crude protein content than fish fed the Control diet, while the crude lipid content of fish fed the $Tau_{1.5}$ and $Tau_{3.0}$ diets was significantly lower than that of fish fed the Control and $Tau_{0.25}$ diets. An ANOVA suggested that the optimum level of dietary taurine supplementation to improve growth and reduce the body lipid contents of juvenile rock bream, O. fasciatus, was 0.5%, while a broken line analysis of weight gain indicated a level of 0.62%.
Oxybenzone (Benzophenone-3; BP-3) is a compound used in sunscreens and enters the oceans due to improper disposal. It is known to disrupt the endocrine signaling in marine organisms, leading to immune and reproductive abnormalities in corals, shellfish and fishes. In this study, we investigated the effects of different concentrations of BP-3 (0, 10, 100 and 1,000 ㎍/L) on the fertilized eggs and hatched larvae of Oplegnathus fasciatus. The morphological changes and hatching and survival rates during embryo development were assessed. In addition, the levels of triiodothyronine (T3) were also measured. The hatched larvae exposed to BP-3 at concentrations of 10 and 100 ㎍/L for 24 h displayed malformation of the tail. After 36 h of exposure to BP-3, spinal deformity was observed at all tested concentrations. The hatching rate was significantly low when exposed to 100 ㎍/L of BP-3. A high levels of T3 was observed when the larvae were exposed to BP-3 at a concentration of 1,000 ㎍/L for 96 h (the end of the experiment). This may be related to increased size of larvae at 1,000 ㎍/L BP-3. In conclusion, our results suggested that BP-3 may interfere with embryo development, resulting in a reduction in hatching rate and malformation of larvae.

In a 12-week experiment, the rock bream, Oplegnathus fasciatus, and olive flounder, Paralichthys olivaceus, were investigated to determine the effects of starvation on their physiological parameters. The protein and DNA contents of the starved fish were significantly higher than the initial values and those of the fed fish. The RNA contents and RNA/DNA ratios of the fed fish were significantly higher than those of the other groups (P<0.05). The hematocrit, hemoglobin, red blood cells (RBC), and mean corpuscular volume (MCV) of the fed rock bream were significantly higher than at baseline (P<0.05), whereas the mean corpuscular hemoglobin concentration (MCHC) of the fed fish was lower than at baseline (P<0.05). The hematocrit, hemoglobin, RBC, and MCHC of the starved group were significantly lower than the baseline values, whereas the MCV of the starved group was significantly higher than the baseline value (P<0.05). No significant difference in alanine aminotransferase was observed between the fed fish and baseline, whereas the starved fish value was significantly higher than the baseline value (P>0.05). There were no significant differences in cortisol levels. However, the glucose level in the fed group was significantly higher than the baseline level and that in the starved group was significantly lower than the baseline level (P<0.05).

Water temperature is a key environmental factor controlling the epizootics of viral diseases in fish. High water temperature is associated with the rapid spread of rock bream iridovirus (RBIV) disease and with high mortality of RBIV infected fish. Although protection of fish against iridoviral disease by active immunization has been reported, little information is available concerning whether fish survived from an epizootic of iridoviral disease can naturally acquire resistance against the viral disease. In the present study, we have demonstrated that juvenile rock bream, which survived from a natural epizootic of RBIV, acquired resistance against recurrence or reinfection of RBIV, and this resistance was established during the subsequent low water temperature period. Furthermore, the possible involvement of the adaptive humoral immune response in the resistance of the juvenile rock bream was suggested by in vivo neutralization experiment.

Monitoring fish movement is important to understand how physiology adapts to environmental change. To explore the applicability of a video tracking system for determining if chemical cues attract or repel aquatic animals, the movement patterns of rock bream, Oplegnathus fasciatus, were analyzed upon exposure to various materials, including extracts of sea urchin, rock worm, bait worm, krill, barley kernel, and commercial fish feed. Pellets were prepared by mixing freeze-dried tissues with a cellulose and corn flour mixture. Behavioral analysis was carried out with five fish that had been acclimated in the adaptation zone of a Y-shaped tank. Preference toward chemical cues was quantified by assessing the frequency rock bream were observed in a discrete zone around the test material located at the end of each arm and the duration each fish stayed in each zone. The analysis of fish movement upon exposure to commercial feed and barley kernel at each end, respectively, indicated a clear preference toward the feed relative to the barley kernel. Movement responses were further tested with pellets containing extracts of sea urchin, one of the species collected on a large scale, and other materials including krill and worms. A stronger preference toward sea urchin (100%) was observed based on the duration of stay in the test zone, compared to krill (90.1 ± 44.2%), bait worm (81.1 ± 39.1%), rock worm (73.7 ± 28.9%), and barley (63.9 ± 25.9%), under the conditions tested. A detailed comparison of rock bream movements toward each material revealed significant differences in frequency and duration, respectively, between pairs of test materials including krill (74 ± 29.8 and 375.6 ± 118.9) vs. rock worm (41.5 ± 18.7 and 160.2 ± 42.6), krill (86.3 ± 22.9 and 477.1 ± 84) vs. bait worm (36.2 ± 5.5 and 166.1 ± 50.7), and rock worm (45.9 ± 26.2 and 213.7 ± 100.1) vs. bait worm (34.6 ± 21.7 and 159.5 ± 98.5). Rock bream exhibited preference for the test materials in the following order: commercial fish feed > sea urchin > krill > rock worm > bait worm > barley. The results suggest a higher potency of sea urchin extract as a rock bream fishing bait compared to the other materials that are used as commercial bait.

EST analysis was performed to identify stress-responsive and immune-related genes from rock bream (Oplegnathus fasciatus). cDNA libraries were constructed with liver and randomly chosen 624 clones were subjected to automated sequence analysis. Of 624 clones sequenced in total, approximately 15% of ESTs was novel sequences (no match to GenBank) or sequences with high homology to hypothetical/unknown genes. The bioinforamtic sequence analysis including functional clustering, homology grouping, contig assembly with electronic northern and organism matches were carried out. Several potential stress-responsive biomarker and/or immune-related genes were identified in all the tissues examined. It included lectins, ferritins, CP450, proteinase, proteinase inhibitors, anti-oxidant enzymes, various heat-shock proteins, warm temperature acclimation protein, complements, methyltransferase, zinc finger proteins, lysozymes, macrophage maturation associated protein, and others. This information will offer new possibilities as fundamental baseline data for understanding and addressing their molecular mechanism involved in host defense and immune systems of this species.
Crytocaryon irritans infection of rock bream was investigated in embankment fish farm from July to December 2009. Prevalence variation was 20.0~88.0% that was high in september and was low in July. While no mortality was found in July and August, it occurred after mid september. Environmental conditions during the survey period were water temperature $19.3{\sim}24.3^{\circ}C$, dissolved oxygen $5.0{\sim}7.1\;mg\;L^{-1}$ and salinity 31.9~33.7 psu, and the water temperature during mortality season was $24.0^{\circ}C$. External symptom of the rock bream was secretion mucus but swimming showed fine in the early infected period. While, the symptoms in the severe outbreak season were excess of mucus, falling off tail and congestion, bleeding spot the surface and weakened swimming. We observed free living and parasitic stages in the gills and body surface of rock bream. Hepato somatic index (HSI) was $1.9{\pm}1.1{\sim}3.5{\pm}1.7%$, which was low in the September being mortality season but was high in November. The percentages of hematocrit infected with Crytocaryon irritans were 37.3% and 41.0% in July and August, respectively while they gradually decreased to 32.1% and 24.2% in september and October, respectively. Total cholesterol and Triglyceride values rapidly decreased by October. After mortality, AST and ALT were 7 and 5 folds higher compared to non-mortality season.

An 8 week feeding trial was conducted to evaluate whether methionine and cysteine would effectively spare taurine supplementation on growth performance in juvenile rock bream Oplegnathus fasciatus. Triplicate groups of 25 fish averaging $2.74{\pm}0.04g$ ($mean{\pm}SD$) were fed one of the experimental diets. Five experimental diets including a fish meal based control diet were prepared by adding sulfur amino acid at fixed level of 1.0% and taurine at graded levels of 0%, 0.25%, 0.5% and 1.0% ($S+Tau_0$, $S+Tau_{0.25}$, $S+Tau_{0.5}$ and $S+Tau_{1.0}$, respectively). After the feeding trial, growth performance of fish fed the $S+Tau_{0.25}$, $S+Tau_{0.5}$ and $S+Tau_{1.0}$ diets were significantly higher (P < 0.05) than those of fish fed the Control diet. ANOVA test suggested that when sulfur amino acid were supplemented to the diets, the optimum taurine supplementation level could be 0.25% in the diet, and broken line analysis of weight gain indicated a level of 0.33%, for positive effects on growth and feed utilization. Fish whole-body protein content and taurine concentration steadily increased with the increase of dietary level in the presence of sulfur amino acid in the diets. On the other hand, whole-body lipid content significantly decreased with the incremental levels of dietary taurine. In conclusion, the results of the present study clearly indicated that dietary supplementation of methionine and cysteine at a level of 1% could spare 0.25 to 0.33% of taurine in juvenile O. fasciatus diets.
The susceptibility of five different marine fish to iridovirus IVS-1 infection was analyzed and found a higher the cumulative mortality in the order of rock bream (Oplegnathus fasciatus), red sea bream (Pagrus major), sea perch (Lateolabrax sp.), rockfish (Sebastes schlegeli) and black porgy (Acanthopagrus schlegeli). However, the concentrations of virus in the infected spleens of these species did not differ significantly. To determine the release of iridovirus from infected fish into culturing seawater, rock bream were challenged with iridovirus IVS-1 and the concentration of virus in the water was analyzed using PCR. Over the 10 days of the analysis, the linear relationship between the number of dead fish and viral DNA concentration found in culturing seawater should be considered direct evidence of horizontal iridovirus transmission.
C-type lectins are crucial for pathogen recognition, innate immunity, and cell-cell interactions. In this study, a C-type lectin gene was cloned from the rock bream. The full-length RbCTL cDNA was 729 bp with a 429 bp ORF encoding a 164-residue protein. The deduced amino acid sequence of RbCTL had all of the conserved features crucial for its fundamental structure, including the four cysteine residues involved in sulfide bridge formation and potential $Ca^2+$/carbohydrate-binding sites. RbCTL contains a signal peptide one single carbohydrate recognition domain. It showed 29.4% similarity to the C-type lectin of rainbow trout. RbCTL mRNA was predominately expressed in gill and head-kidney tissue and expressed less in peripheral blood leukocytes, trunk-kidney, spleen, liver, intestine and muscle. Expression of RbCTL was differentially upregulated in rock bream stimulated with LPS, Con A/PMA and poly I:C.
