Changes in Nutrient Composition, Antioxidant Properties, and Enzymes Activities of Snake Tomato (Trichosanthes cucumerina) during Ripening

  • Snake tomato (Trichosanthes cucumerina) has been cultivated and used as a replacement for Lycopersicum esculentum in many Asian and African diets. Matured T. cucumerina fruits were harvested at different ripening stages and separated into coats and pulps for analyses to determine their suitability for use in culinary. They were analyzed for the nutritional composition and antioxidant potential using different biochemical assays [1,1-diphenyl-2-picrylhydrazyl, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activities, and ferric reducing antioxidant power] and antioxidative enzymes activities. The nutritional composition revealed that T. cucumerina contains over 80% water and is very rich in fiber, thus it can serve as a good natural laxative. The lycopene and ${\beta}$-carotene contents were especially high in the ripe pulp with values of $21.62{\pm}1.22$ and $3.96{\pm}0.14mg$/100 g, respectively. The ascorbic acid content was highest in the pulp of unripe fruit with a value of $56.58{\pm}1.08mg$/100 g and significantly (P<0.05) decreased as ripening progressed. The antioxidant potential of the fruits for the 3 assays showed that unripe pulp> ripe coat> ripe pulp> unripe coat. There were decreases in the antioxidant enzymes (superoxide dismutase, ascorbate peroxidase, and glutathione reductase) activities, with the exception of catalase, as ripening progressed in the fruits. These decreased activities may lead to the softening of the fruit during ripening. Harnessing the antioxidative potential of T. cucumerina in culinary through consumption of the coats and pulps will alleviate food insecurity and help maintain good health among many dwellers in sub-Saharan Africa and Southeast Asia.

Antioxidant potential of buffalo and cow milk Cheddar cheeses to tackle human colon adenocarcinoma (Caco-2) cells

  • Objective: The aim of present study was to assess the anti-oxidant potential of water-soluble peptides (WSPs) extract derived from buffalo and cow milk Cheddar cheeses at different stages of ripening. Methods: The antioxidant potential of WSPs extract was assessed through 2,2'-azinobis-3-ethylbenzothiazoline-6sulfonic acid (ABTS)-radical scavenging activity. In addition, impact of WSPs extract on cell viability and production of reactive oxygen species (ROS) in human colon adenocarcinoma Caco-2 (tert-butylhydroperoxide-induced) cell lines was also evaluated. Results: The ABTS-radical scavenging activity increased progressively with ripening period and dose-dependently in both cheeses. However, peptide extract from buffalo milk Cheddar cheese demonstrated relatively higher activity due to higher contents of water-soluble nitrogen. Intracellular ROS production in Caco-2 cells decreased significantly (p<0.05) till 150th day of cheese ripening and remained constant thereafter. Additionally, dose-dependent response of WSPs extract on antioxidant activity was noticed in the Caco-2 cell line. Conclusion: On the basis of current in vitro study, the Cheddar cheese WSPs extract can protect intestinal epithelium against oxidative stress due to their antioxidant activity.

고려인삼의 식물부위 및 여러 조건과 관련한 에틸렌가스 방출 (Ethylene Release of Panax ginseng in Relation to Plant Part and Various Conditions)

  • Ethylene was released from leaf and fruit but root of Panax ginseng. Root callus showed higher ethylene release (ER) than fruit ER increased with leaf senesence. Fruit during ripening showed decreasing ER in the order of green stage, early stage of reddening and fully ripened stage. between leaves from the plant with fruits in different stages of ripening showed similar trend of fruit in ER but it was about 10 times higher in leaves than in fruits. Leaves of P. quinquefolius showed about 200 times higher ER than that of P ginseng on 22 July Fruits from the plant treated with ethephon showed higher ER after 109 days. Forty-five day-old seedlings grown with various growth regulators showed a significant decrease of stem length and significant increase of ER only in Uniconazole (0.1 ppm) and H-9 (0.0, 5 ppm) solution.

Ethrel의 농도, 처리시기 및 질소농도별 시비의 잎담배 성숙에 미치는 영향 (Effects of Ethrel on Tobacco-Leaf Maturity -Influences by Different Levels of the Chemical, Soil Nitrogen and Time of the Chemical Application-)

  • The most commonly grown economical and flue-cured tobacco cultivar Yellow Special A was used in pot-culture tests in order to study Ethrel (2-chloroethyl phosponic acid) effects on accelerating maturity of tobacco leaves in relation to the most adequate level of the chemical useful for field growing, nitrogen level in soil for the most pronounced response, and the most suitable spray period during the growth stages of pre-, post- and topping periods. The following conclusions, thus, were obtained from the present studies; 1. 500ppm Ethrel spray was reconfirmed to be adequate in the practical applications, although the extent of yellow-ripening of tobacco leaves was increased as the Ethrel level increased. The highest leevel treated resulted in causing chemically damaged lesions on leaves and early defoliation. 2. Ethrel-treated leaves showed deeper yellowish tinge to them than those without treatment, while different levels of the chemcial had less influence on the tinge. 3. An adequate level of nitrogen supply to plants favored the Ethrel response, whereas either very low or high level of nitrogen in the soil lowered the chemical effect on accelerating the yellow-ripening. When carbohydrates versus total nitrogen ratio became relatively high, the condition brought out some outstanding Ethrel effects. 4. Chlorophyll level of leaves increased as soil applications of nitrogen level increased, and that also increased carotenoid level of the tobacco leaves. Ethrel-treated leaves showed deeper orange tinge than those without treatment, while the highest level of nitrogen application showed the deepest orange tinge to tobacco leaves. 5. Pre-topping treatment (12 days before topping and flowering) resulted in almost no Ethrel response, and that treatment right on the day of topping, showed response of yellow-leaf ripening at nearly bottom-half leaves of a tobacco plant. The post-topping treatment (12 days after topping) made plants showing full response of Ethrel from bottom to the top leaves of tobacco plant in accelerating the leaf maturity. 6. The extent of Ethrel responses on accelerating yellow-ripening of tobacco leaves was discussed for the modifying influences brought about by certain environmental factors. Discussions were also made about the possible practical applications (particularly for pre-rice planting) and quality difference that may be caused by such growth environments.

  • Changes in the phosphorylation of Chlorella cells during the life cycle the aulotrophic and micotrophic synchronous culture were followed under the light and dark. 1. In the autotrophic culture of Chlorella the amounts of esterified phosphate compounds of the algal cell under the light increased during the growing period and decreased strikingly in the ripening period showing a peak at the $L_1$ i/-cell stage. 2. TRhe amount of total esterified phosphate compounds of the cell under the dark, however, decreased during the growing period and then kept fairly constnat during the ripening nad division periods showing the greates activity of the oxidative phosphorylation in the early growing stage. 3. It is presumed that the energy requirement of the dividing algal cell in the autotrophic culture is fulfilled prior to the nuclear division mostly by the photosynthetic phosphorylation. 4. In the mixotrophic culture, the amount of esterified phosphate compounds of the algal cells under the light increased during the growing period and decreased during the late ripening and early division periods showing a peak in the $L_2$-cell stage as in the case of the phosphorylation under the dark. 5. The phosphorylation of the fell grown in the glucose medium is more active under the dark than under the light in the stages of the growing and early ripening periods. 6. It is considered that the excess glucose in the algal cell not only promotes the oxidative phosphorylation but also inhibits the photosynthetic phosphorylation of the cell. 7. It is presumed that the energy requirement of the dividing algal cell in the glucose medium is fulfilled prior to the nuclear division by the combined action of oxidative and photosynthetic phosphorylation, mostly by the oxidative phosphorylation.

온주밀감 '하례조생'과 '부지화' 과실의 착색 단계별 고온에 의한 성숙 관련 유전자의 발현 변화 (Gene Expression as Related to Ripening in High Temperature during Different Coloration Stages of 'Haryejosaeng' and 'Shiranuhi' Mandarin Fruits)

  • 본 연구에서는 기후변화로 인한 온도상승에 따른 과실 착색 불량 등의 문제를 해결하는데 필요한 기초자료를 제공하고자 고온에 의해 과피에서 발현되는 유전자들의 발현 양상 특성을 분석하였다. '하례조생'과 '부지화' 감귤 과실을 숙기 별로 수확하여 온도 조건(25, 30, $35^{\circ}C$)을 처리하고 당대사, 과피 착색, 세포벽 연화에 관련된 유전자들의 발현을 확인하였다. 유전자들은 '하례조생'과 '부지화'에서 각각 다른 양상으로 발현하였는데, beta-amylase(BMY), phenylalanine ammonia-lyase(PAL), chalcone synthase(CHS), flavanone 3-hydroxylase(F3H) 등의 유전자 발현은 대체적으로 유도되었고, polygalacturonase(PG) 유전자는 발현이 감소되는 경향이었다. '하례조생'은 과피 착색과 관련된 유전자인 CHS와 F3H는 성숙이 진행된 2-3단계에서 $25^{\circ}C$에 비해 고온에서 유전자 발현이 감소하였으며, PAL과 stilbene synthase(STS) 유전자는 $25^{\circ}C$에 비해 $30-35^{\circ}C$ 처리구에서 유전자 발현이 증가하였다. 2-3단계의 '부지화'에서는 BMY 유전자가 $25^{\circ}C$에 비해 $30-35^{\circ}C$에서 유전자 발현이 증가하였으며, F3H와 STS 유전자의 발현은 과실 성숙단계에서 모두 감소하는 경향이었고 온도의 영향은 크지 않았다. 성숙 1, 2단계에서 유전자의 발현 양상은 두 품종 모두에서 대체로 비슷한 경향이었는데, 3단계에서 '부지화' 과실의 유전자 발현은 '하례조생'과는 다르게 감소하는 경향이었다. 성숙이 진행되는 감귤류인 '하례조생'에서 '부지화'에 비해 7종류의 유전자의 발현이 많았으며, 고온에 따른 반응의 차이도 크게 나타났다. 본 연구에서 도출한 결과를 바탕으로 과실의 전사체를 분석함으로써 고온에 의한 감귤 과실의 성숙불량 문제를 이해하는 주요한 정보를 획득할 수 있을 것이다

최근의 기상환경 변화에 따른 가을보리의 수량구성요소 및 생육단계 변화 (Impact of Recent Weather Variation on Yield Components and Growth Stages of Winter Barley in Korea)

  • 근래 지구 온난화 현상과 더불어 우리나라에 '춥지 않은 겨울' 날씨가 지속되어 가을보리를 비롯한 겨울(월동)작물의 생육과 수량에 큰 변동이 있을 것으로 예상되는 바, 최근의 기상환경 변화에 따른 가을보리의 생육시기, 생육기간, 수량, 수량구성요소의 변동양상을 분석하였으며, 각 생육단계별 기상요소가 보리의 수량과 관련되는 재배적 형질에 미치는 영향을 분석하였다. 그 결과는 다음과 같다. 1. 수원에서 14년(1987∼2000년)동안 올보리 품종의 평균수량은 지난 21년(1974∼2000년) 평균보다 헥타르(ha)당 0.42톤이 증수되었다. 증수의 요인은 단위면적당 수수 확보이고, 1수입수의 증가도 다소 기여하였으나, 천립중은 평균보다 오히려 0.6그램(g)정도 가벼워졌다. 2. 풍년에는 흉년보다 단위면적당 수수는 많았고, 1수입수는 비슷하였으나, 천립중은 가벼웠다. 그리고, 월동기간은 짧았고, 유묘기간과 분얼기간이 길어졌다. 출현기간에 기온이 높아 발아율이 높았으며, 단위면적당 수수가 증가되었다. 반면에 흉년에는 월동기간의 저온과 분얼 및 등숙기간의 많은 강수량으로 단위면적당 수수가 감소하였다. 3. 올보리 품종에서 수량은 단위면적당 수수와는 밀접한 정의 상관이었지만, 1수입수 및 천립중 하고는 유의성이 보이지 않았다. 반면에 단위면적당 수수는 천립중 및 수장과 부의 상관을 나타냈다. 4. '춥지 않은 겨울' 날씨로 지속된 14년(1987∼2000년) 동안에 생육재생기의 출현초일은 평균(1974∼2000년) 출현초일 보다 5일 일찍 나타났다. 또한 생육재생기는 출수기 및 성숙기의 출현초일과 높은 정의상관을 보였다. 그리고, 이 기간의 등숙기간은 평균 등숙기간과 비슷하였으나 월동기간은 평균보다 9일 짧아졌다. 반면에, 유묘기간과 분얼기간은 평균보다 3일 길어졌다. 5. 올보리 품종의 수량에 영향을 주었던 생육단계별 기상요소는 월동기간의 기온(+상관)과 일조시간(-상관), 분얼기간의 강수량(-상관), 등숙기간의 증발산위(+상관) 였다. 천립중에 유의적 영향을 주는 생육단계별 기상요소는 발견되지 않았으며, 분얼기간의 고온과 건조한 대기는 줄기의 신장에 불리하였지만, 이삭의 신장에는 유리하였다. 즉, 올보리 품종의 수량에는 월동기간과 그 이후 생육단계에서 기상요소의 영향이 크게 작용하였다.
