Effect of Ethanolic Extract of Schizandra chinensis for the Delayed Ripening Kimchi Preparation (오미자(Schizandra chinensis) 추출물이 김치의 과숙억제에 미치는 영향)

  • This study mainly focused on to investigate the effects of Schizandra chinensis on the growth of a bacterium, CS6 which was isolated from kimchi. CS6 was final]y identified to lactobacillus plantarum that caused acidification of kimchi. The ethanolic extract of Schizandra chinensis(EES) inhibited the growth of L. plantarum. Minimum inhibition concentration of crude EES on L. plantarum was 62.5mg/$m\ell$. In broth culture, 5$\mu\textrm{g}$/$m\ell$ of EES completely inhibited the growth of L. plantarum during fermentation. The addition of 0.4% of EES has no apparent effect on quality including the taste and color on kimchi. It was expected that EES-containing kimchi could extend the period of preservation. Analysis of organic acids in water fractions of EES was carried out by HPLC. It is apparent that antimicrobial active fractions contained the highest concentration of succinic acid, a little tartaric acid and malic acid. Among these organic acids, succinic acid showed the strong inhibitory effect against L. plantarum CS6 in vitro. Succinic acid-containing kimchi with a concentration of 0.4 and 0.5% had the inhibitory effect on growth of L. plantarum. Inhibitory effect of EES on amylase, cellulase and pectinase was also tested. In conclusion, the present experiment demonstrated that EES inhibited the growth of L. plantarum, and various enzyme activity. EES-containing kimchi was sustained the hardness, and initial acidity during fermentation. EES was considered as the possible additive of kimchi process and EES added in kimchi increase the quality, and storage period of kimchi.

Changes of Chemical Composition and Microflora in Bottled Vacuum Packed Kimchi during Storage at Different Temperature (진공처리 병포장 김치의 저장조건별 성분과 미생물 변화)

  • Mak-kimchi (shredded kimchi) which was prepared in a commercial factory was packed in bottle (200 g) under vacuum (560 mmHg) or atmosphere, and chemical characteristics and microbiological parameters were monitored during storage at 5, 15 and $25^{\circ}C$, respectively. Optimum ripening time of the kimchi at different temperature were 2 days at $25^{\circ}C$, 5 days at $15^{\circ}C$ and more than 60 days at $5^{\circ}C$. By vacuum treatment pH and acidity changes in kimchi were considerably retarded. The vacuum of each bottle released within 1 or 2 days at 25 or $15^{\circ}C$, respectively but the pack at $5^{\circ}C$ maintained more than 380 mmHg vacuum for 36 days and then the vacuum slowly released. The colour of kimchi (lightness, redness, yellowness) in bottle increased sharply at $25^{\circ}C$ and $15^{\circ}C$ but sustained a stable level with vacuum treatment at $5^{\circ}C$. The range of total viable count of kimchi in bottle was $10^7{\sim}10^{10}/ml$. The number decreased by storage temperature drop to $5^{\circ}C$ and even more vacuum treatment than atmosphere treatment at $5^{\circ}C$. Lactobacillus brevis, L. plantarum, L. acidophilus, Aerococcus viridans and Streptococcus faecium subsp. casseliflavus were identified in bottled kimchi and L. brevis and L. plantarum contributed to the main function during kimchi fermentation. Those main lactic acid bacteria decreased in numbers at $5^{\circ}C$ than 25 or $15^{\circ}C$ and even more declined in case of vacuum treatment.

  • The changes in pectinesterase (PE) and polygalacturonase (PG) activities were investigated during maturation and ripening of tomato fruits. PG was isolated from ripe tomatoes and purified by using DEAE-sephadex A-50 column chromatography. Its property was examined by general methods. PE activity was increased during maturation and then peaked at the on set of color change. PG activity was not detectable in mature green tomato fruits but increased during softening. Slab electrophoresis of the crude enzyme isolated from ripe tomatoes shoed 6 bands. Two polygalacturonase (PGI, PGII) were separated from crude enzyme of ripe tomatoes by chromatography on DEAE-sephadex A-50. PGI and PGII were purified by 61 fold and 98 fold by the present procedure, respectively. The Rm values of partially purified PGI and PGII were estimated to be 0.25 and 0.31, respectively. The optimum temp, and pH of PGI activity were $37^{\circ}C$ and pH 4.5, while those of PGII activity were $40^{\circ}C$ and pH 4.7, respectively. Isozyme PGI activity was the most stable at pH 4.3 and retained 50% of its activity when exposed at $78^{\circ}C$ for 5 min.

Effect of the Gibberellin Treatment on Enlargement and Mature Promotion in 'Niitaka' Pear (Pyrus pyryfolia L.) (지베렐린이 '신고'배의 비대와 성숙촉진에 미치는 영향)

  • The object of this study was to evaluate the effect of the gibberellin treatment on fruit enlargement and ripening promotion in 'Niitaka' pear(Pyms pynfolia). Fruit weight was similar between $GA_{4+7}$ treatments and the gibberellin-paste control, but fairly increased of fruit weight compared to non-treatment Most effective time for $GA_{4+7}$ treatment to increase diameter and length was on 35 and 40 days after full bloom, respectively. Shape index was similar in all treatments. Fruit enlargement at the period of early growth, $GA_{4+7}$ 2.4% treatment was remarkably effective than $GA_{3}+GA_{4+7}$ 2.7% treatment or non-treatment However at the period of maturity, $GA_{4+7}$ 2.4% treatment and $GA_{3}+GA_{4+7}$ 2.7% treatment showed little differences in fruit enlargement and coloring. In maturing promotion effect, young fruit treated with $GA_{3}+GA_{4+7}$ showed similar fruit coloring to ethephon treatment on 35 days after full bloom, and both of those treatments promoted fruit coloring than non- treatment about 6 days. $GA_{3}+GA_{4+7}$ treatments resulted higher solid content and decreased acidity than non-treatment However, there was no differences in fruit hardness comparing to non-treatment As a result, gibberellin was most effective in fruit enlargement, so as all $GA_{3}+GA_{4+7}$ treatments were more effective on fruit enlargement than ethephon treatment or non-treatment Specifically, when $GA_{3}+GA_{4+7}$ was treated 35days after full bloom of flower, the diameter, the length and the shape of fruit index were best, and fruit coloring was good as well.

A Medium-Maturing and Good Quality Japonica Rice Variety, "Cheongan" (벼 중생 고품질 신품종 "청안")

  • "Cheongan" is a new japonica rice variety developed from a cross between SR15225-B-22-1-2-1 and Iksan431 in summer season, 1997 by National Institute of Crop Science, RDA. The line SR15225-B-22-1-2-1 has good canopy architecture and multi-disease and insect resistance, and Iksan431 has translucent milled rice and good eating-quality. Heading date of Cheongan is August 13 in central lowland and mid-mountainous areas. "Cheongan" having culm length of 84 cm shows relatively semi-erect pubescent leaf blade and rigid culm, tolerance to lodging with and good canopy architecture. This variety has 14 tillers per hill and 126 spikelets per panicle. It shows tolerance to heading delay and spikelet sterility comparable to Hwaseongbyeo when exposed to cold stress. Leaf senescence of Cheongan progresses slowly during the ripening stage and the viviparous germination ratio was 59 %, similar to that of Hwaseongbyo. "Cheongan" shows moderately resistance to blast disease, but susceptible to stripe virus and brown planthopper. The milled rice of "Cheongan" exhibits translucent, clear non-glutinous endosperm and medium short grain. It shows similar amylose content of 18.7%, gelatinization temperature, and similar palatability of cooked rice compared to Hwaseongbyeo. The milled rice yield of this cultivar is about 5.54 MT/ha at ordinary season culture in local adaptability test for three years. Especially, "Cheongan" has better milling properties of higher 98.4% and 73.9% in the percentage of head rice in milled rice and milling recovery of head rice, respectively, than those of Hwaseongbyeo. "Cheongan" could be adaptable to the central and mid-southern plain area, and mid-western coastal area of Korea.

Estimation of Paddy Rice Growth Increment by Using Spectral Reflectance Signature (분광반사특성을 이용한 벼의 생장량 추정)

  • To have a basic idea on the spectral reflectance signature in paddy rice canopy, we measured spectral reflectance from paddy rice canopy(Ilpumbyeo) using spectroradiometer (GER Inc. SFOV : 0.35~2.50 ${\mu}{\textrm}{m}$) in situ weekly or biweekly from transplanting to ripening stage. Spectral reflectance of the visible range (0.4~0.7 ${\mu}{\textrm}{m}$) was decreased to below 5% and then slightly increased again after heading stage in rice canopy. Meanwhile spectral reflectance of the near-infrared range (0.7~1.1 ${\mu}{\textrm}{m}$) was increased to 40~50% and then decreased a great deal after panicle initiation stage in rice canopy. Landsat TM equivalent band set ($\bar{p}$$_{TMi}$) was created by averaging spectral reflectance values to the real TM bands. Correlation analysis between the rice crop variables (LAI, total dry matter) and TM equivalent band set ($\bar{p}$$_{TMi}$) showed that LAI and total dry matter of rice were highly correlated with visible bands such as $\bar{p}$$_{TM1}$, $\bar{p}$$_{TM2}$, and $\bar{p}$$_{TM3}$. Ratio values ($\bar{p}$$_{TMi}$/$\bar{p}$$_{TMi}$) such as $\bar{p}$$_{TM4}$/$\bar{p}$$_{TM2}$ were also highly correlated with rice crop variables such as LAI and total dry matter.

  • The results of the investigation on the effects of plant nutrio-physiological status and reemergence of sudden wilting in rice on the occurrence of sudden wilting were summarized as follows: 1. The occurrence of sudden wilting, degree of damage and occurring time in the investigated variety showed to concur with the results of the previous experiment. 2. Nitrogen and Potassium contents in leaves related an occurrence of sudden wilting at heading stage have a very high negative co-relation (the value is -0.9224 at Yushin and -0.8243 at Milyang 23). 3. Regarding the relation between occurrence of sudden wilting and root activity capacity at the late growth stage, the root activity capacity at productive growth stage showed higher than at reproductive growth stage, which is the same in the two varieties but Milyang 23 is higher than Yushin. It is confirmed that root activity capacity in the two varieties has become very low at reproductive growth stage. 4. The toot activity capacity in the field with potassium showed a small occurrence of sudden wilting and the root activity capacity in the field with no potassium showed a very high co-relation value (the value is 0.8947 at Yushin and 0.7301 at Milyang 23). 5. Regarding the relation between the occurrence of sudden wilting and water evaporation quantity, water evaporation quantity after the heading date has a more direct relation to sudden wilting than that which occurrs prior to the heading date. Water evaporation quantity is higher in the field with no potassium, low leaves and Yushin, than in the field with potassium high leaves and Milyang 23. 6. The starch content in culm of Yushin and Milyang 23 at ripening stage showed high value in proportion to potassium quantity, and the starch content is lower as internode is lower. However, in fourth internode, the starch content is greatly increased in the field with potassium contrasted to the field with no potassium.

Studies on Adaptability by Rice Heading Ecology Type in the Central Northern Mid-Mountainous Cultivation Zone of Chungbuk Region (충북지역 중북부 중산간지 벼 출수생태형별 적응성 검토)

  • In recent years, air temperature has been increasing rapidly compared to the 1980s because of global warming. This increase in temperature reduces the yield and quality of rice; therefore, measures are needed to prevent such effects and ensure food security. The early maturing type (EMT) of rice is mainly cultivated in the central northern mid-mountainous area (CNMA). This study was conducted to shift the transplanting date of EMT and to examine the adaptability of the mid-maturing type (MMT) or mid-late maturing type (MLMT) in the Jecheon region of the CNMA to address global warming. The air temperature increased by 0.7-0.9℃ in the 2010s, compared to that in the 1980s, and was similar to other decades during the ripening period. Over the past 35 years, considering rice quality, the heading date of the Odae variety has arrived sooner by approximately 10 days, the ripened grain ratio has increased by more than 10%, and the thousand grain weight; however, the mean temperature at 40 days after heading has increased by more than 2℃. The late marginal heading date in the Jecheon region was determined as August 11 based on the accumulated temperature of 880℃ and August 15 based on 840℃ for 40 days after heading. According to different transplanting dates, milled rice yield per 10 a was the highest at 567 kg with June 10 in EMT, 595 kg with June 10 in MMT, and 572 kg with May 30 in MLMT. Considering the late marginal heading date, rice yield, and quality, the optimum transplanting date was June 15 in EMT, June 5 in MMT, and May 30 in MLMT in the Jecheon region of CNMA. Owing to global warming, MMT and MLMT are expected to be reliably cultivated in the CNMA.

A New Rice Cultivar 'Manjong', Suitable to the Rice-Cash Crop Double Cropping System (중생 소득작물 후작지 적응 벼 품종 '만종')

  • 'Manjong' is a new japonica rice cultivar developed in 2009 derived from a cross between 'Yeongdeog34' and 'Nampyeongbyeo' at the Department of Functional Crop, NICS, RDA. This cultivar is suitable for the double cropping system after the cash crop cultivation. Heading date of 'Manjong' is later than 'Keumobyeo' under the late transplanting cultivation on July 10. It has high grain fertility under cold conditions and low premature heading. One of the distinguishing characteristics of this variety is its resistance to major diseases like leaf blast and rice stripe virus disease. However, it shows susceptibility to major insect pests and bacterial blight disease. Milled rice kernels are translucent with non-glutinous endosperm and have 7.2% protein and 18.7% amylose contents. Ripening ratio and head rice ratio is very higher than those of 'Keumobyeo'. The palatability of cooked rice is also better than 'Keumobyeo'. The milled rice yield of 'Manjong' in local adaptability tests after harvest of the cash crop is $4.81MT\;ha^{-1}$. This cultivar is suitable for planting in the plain paddy fields of Honam and Yeonnam regions in Korea.

A Very Early-Maturing, Cold Tolerant and High Quality japonica Rice Variety 'Hanseol' (극조숙 고품질 내냉성 벼 신품종 '한설')

  • 'Hanseol' is a new very early-maturing, cold tolerant and high quality japonica rice variety developed from a cross of 'Jinbu24' and 'Jinbu25' by the rice breeding team of National Institute of Crop Science (NICS), Rural Development Administration (RDA) in 2009. The heading date of this variety is July 25, which is four days earlier than check variety, 'Jinbubyeo'. 'Hanseol' has 65 cm of culm length, 99 spikelets per panicle, 82.9% of ripened grain rate, and 21.5 g of 1,000 grain-weight of brown rice. This variety shows susceptibility to bacterial leaf blight and virus diseases, and insect pests. It is tolerant to cold stress in terms of less heading delay and high fertility in cold water irrigated cultivation. This variety shows delayed leaf senescence and considerable tolerance to viviparous germination at ripening stage. The milled rice of this variety exhibits translucent, clear non-glutinous endosperm and medium-short grain. 'Hanseol' showed low gelatinization temperature and 6.1% protein content, 19.1% amylose content and good palatability of cooked rice. The milled rice yield of this variety is about 5.43 MT/ha at ordinary culture in local adaptability test for three years. 'Hanseol' would be highly adaptable to mid-north and mid-mountainous areas, and mid-northern alpine area in Korea.
