Evaluation of Disease Resistance of Rice Cultivar Developed in North Korea (북한에서 육성된 벼 품종의 병 저항성 검정)

  • Almost 30% of arable lands of North Korea are covered with paddy rice. In rice cultivation of North Korea, rice blast disease is the most important fungal disease and bacterial leaf blight is the most important bacterial disease. Seven North Korean rice cultivars had been tested for the disease resistance against rice blast pathogen, Magnaporthe oryzae and bacterial leaf blight pathogen, Xanthomonas oryzae pv. oryzae. The responses of seven cultivars against 17 different M. oryzae races from South Korea had been quite different. Among seven cultivars, Giljoo1ho was very resistant to all 18 different M. oryzae isolates from South Korea, nevertheless KI or KJ. Pyungdo5ho was very susceptible, it showed susceptible responses to 8 out of 10 KI races and 7 out of 8 KJ races of M. oryzae isolated in South Korea. However, the response to bacterial leaf blight was different from the response to rice blast pathogen. Gijoo1ho, Wonsan69ho, Onpo1ho, and Pyungdo15ho were susceptible to KXO42 (K1) and KXO90 (K2), respectively. Pyungdo5ho was resistant to KXO85 (K1) and KXO19 (K3), and Pyungyang21ho was resistant to K1 races. Based on these results, Giljoo1ho can be a good resource for the breeding of resistant rice cultivar against M. oryzae isolates from South Korea.

Screening of Gamma Radiation-Induced Pathogen Resistance Rice Lines against Xanthomonas oryzae pv. oryzae (방사선을 이용한 벼 흰잎마름병 저항성 돌연변이 벼 계통의 선발)

  • Bacterial blight is one of the most serious diseases of rice (Oryza sativa L.), and it has been known that Xanthomonas oryzae pv. oryzae (Xoo) causes this disease symptom. To develop resistance rice cultivars against Xoo, 3,000 lines of $M_3$ mutants, which were irradiated with gamma ray, were tested by 'scissor-dip method' primarily, and 191 putative resistant lines were selected. In $M_4$ generation, these lines were screened again with various ways such as measuring of symptom of bacterial blight in leaf, number of tiller, fresh weight, and phenotypic segregation ratio in next generation. Finally, six resistance lines were selected. RT-PCR analysis revealed that these lines displayed high level of R-genes such as Xa21, Pi36, and Pi-ta. These results indicate that mutations by gamma ray cause disruptions of regulatory signal transduction systems of these R-genes. Furthermore, these selected mutants could be useful for the development of rice cultivar resistant to Xoo.

  • The experiments were conducted to examine the formation of chemical resistance of Xanthomonas oryze, the causal organism of rice bacterial leaf blight, to Agrepto in the succeeding geerations. The measurements of the bacterial growth were determined with the optical density as indirect method, and the results obtained were summarized as follows. 1. The Agrepto was effective in hindering the bacterial growth in the treatment of first generation, which was added in the broth even as low concentration as 30ppm. 2. when the successive four generations were treated with same concentrations, the bacterial growth increased conspicuously with succeeding generations and this phenomenon showed remarkable in the plots treated with 30ppm than in that of 10ppm. 3. When selections were repeated at treatment of 20 ppm, increasing the concentrations of the treatment in successive generations, the resistance increased with succeeding generations. 4. There was a tendency to acquire the higher rate of resistance in the progenies which selected from the treated plot of higher concentrations in previous generation than that of low concentrations.

Simultaneous Detection of Three Bacterial Seed-Borne Diseases in Rice Using Multiplex Polymerase Chain Reaction

  • Burkholderia glumae (bacterial grain rot), Xanthomonas oryzae pv. oryzae (bacterial leaf blight), and Acidovorax avenae subsp. avenae (bacterial brown stripe) are major seedborne pathogens of rice. Based on the 16S and 23S rDNA sequences for A. avenae subsp. avenae and B. glumae, and transposase A gene sequence for X. oryzae pv. oryzae, three sets of primers had been designed to produce 402 bp for B. glumae, 490 bp for X. oryzae, and 290 bp for A. avenae subsp. avenae with the $63^{\circ}C$ as an optimum annealing temperature. Samples collected from naturally infected fields were detected with two bacteria, B. glumae and A. avenae subsp. avenae but X. oryzae pv. oryzae was not detected. This assay can be used to identify pathogens directly from infected seeds, and will be an effective tool for the identification of the three pathogens in rice plants.

  • Rice is an important food crop for most of the population in the world and it is largely cultivated in Pakistan. It not only fulfills food demand in the country but also contributes to the wealth of Pakistan. But its production can be affected by climate change. The irregularities in the climate can cause several diseases such as brown spots, bacterial blight, tungro and leaf blasts, etc. Detection of these diseases is necessary for suitable treatment. These diseases can be effectively detected using deep learning such as Convolution Neural networks. Due to the small dataset, transfer learning models such as vgg16 model can effectively detect the diseases. In this paper, vgg16, inception and xception models are used. Vgg16, inception and xception models have achieved 99.22%, 88.48% and 93.92% validation accuracies when the epoch value is set to 10. Evaluation of models has also been done using accuracy, recall, precision, and confusion matrix.

  • 1. Kum Nam Poong which is highly susceptible to bacterial leaf blight was used as the host plant throughout this experiment. Xanthomonas oryzae 6526 was inoculated on the top of upper leaves by single needle inoculation method. After 14 days, the enlarged spots were examined in the experimental pots. Each of 3 levels of nitrogen, phosphate and potassium pots were arranged at random with three replications. 2. The amounts of nitrogen fertilizer applied and the lesion development of bacterial leaf blight were positively correlated regardless of application of phosphate and potassium fertilizers. 3. The effect of phosphate fertilizer on the lesion development was not significantly different from standard level. The lesion development was stimulated when the amount of phosphate fertilizer was increased as twice as standard level. 4. The inhibitory effect of potassium fertilizer on leaf blight was maximum by applying standard level. The stimulative effect of potassium fertilizer on the lesion development, however, was noticed. when potassium fertilizer applied was increased as twice as the standard level. 5. The heading date and spike number of rice plant were significantly correlated with the lesion development, and such phenomena were depended on the amount of nitrogen fertilizer applied regardless of the other fertilizers applied in this experiment.

The transition of noteworthy rice diseases and their control in Korea

  • In Korea the most important plant industry is the rice-growing but she has not yet attained self-sufficiency with regard to rice which is the basic food of Korean. In the past, various measures of increasing rice production have been tried with some success but the increasing rate of did not show the sufficient result. Judging from experiences, disease control may be the most important factor which has influence on increasing of rice production in Korea. During the past 60 years three periods divided by the transition of rice diseases were noted, and especially the recent transition attracted much attention . That is, sheath blight and stripe which were minor diseases in the past have promptly spread all over the country and caused heavy losses to rice plant, and also local outbreak of bacterial leaf blight and dwarf were noticed. Various environmental factors are related to such transition of rice disease. Above all, cultivated varieties susceptible to these diseases, increase of application of nitrogenous fertilizer, earlier transplanting and density of transplanting are considered to be lucre closely related to the development of these diseases and successive development of blast. On the other hand. application of forage fungicides such as organic mercury compounds specific to blast contributed to reduce prevalent area if blast in spite of wide spread of susceptible varieties and increase of application of nitrogenous fertilizer. These facts were confirmed by Korean investigators At the present which various diseases to control coexist in the same region, no one variety resistant to all of these diseases is provided and cultural practices for the control of these diseases often do not produce sufficient yield of rice. it is emphasized that a role of pesticides is very great.

  • Rice bacterial leaf blight (BLB) by Xanthomonas oryzae pv. oryzae (Xoo) is considered to be one of the major rice diseases steadily occurring around the rice-producing countries. In this study, we developed a recombinase polymerase amplification (RPA) assay for the rapid, convenient and specific diagnosis of Xoo by targeting Xoo-specific transposase A gene. As the target gene can be amplified in 10 min without DNA extraction process and special equipment for temperature control, RPA for BLB can be useful and practical component for on-site diagnosis.
