Effect of Carbon Source on the Hydrolytic Ability of the Enzyme from Fomitopsis pinicola for Lignocellulosic Biomass

  • In this study, effect of carbon source on the hydrolytic ability of the enzyme from Fomitopsis pinicola, a brown rot fungi, for lignocellulosic biomass were examined on two lignocellulosic biomasses (rice straw and wood) without any pretreatment. Cellulase activities of crude enzyme from F. pinicola, which was cultured on softwood mixture as a carbon source, were 19.10 U/$m{\ell}$ for endo-${\beta}$-1,4-gulcanase (EG), 36.1 U/$m{\ell}$ for ${\beta}$-glucosidase (BGL), 7.27 U/$m{\ell}$ for cellobiohydrolase (CBH), and 7.12 U/$m{\ell}$ for ${\beta}$-1,4 xylosidase (BXL). Softwood mixture as a carbon source in F. pinicola comparatively enhanced cellulase activities than rice straw. The optimal pH and temperature of the cellulase was identified to pH 5 and $50^{\circ}C$for the hydrolysis of rice straw. Under these condition rice straw was hydrolyzed to glucose by the cellulase up to $32.0{\pm}3.1%$ based on the glucan amount of the rice straw for 72 h, while the hydrolytic capability of commercial enzyme (Celluclast 1.5${\ell}$) from rice straw to glucose was estimated to $53.7{\pm}4.7%$ at the same experimental condition. In case of addition of Tween 20 (0.1% w/w, substrate) to the cellulase the hydrolysis of rice straw to glucose was enhanced to $38.1{\pm}2.0%$.

Effects of Base Papers Containing Nonwood Pulp on the Properties and Printabilities of Coated Papers(II) -Rice Straw Bleached Kraft Pulp- (비목재펄프를 이용한 도공원지의 특성이 도공지의 물성 및 인쇄적성에 미치는 영향(II) -볏짚 표백 크라프트펄프-)

  • This study was carried out to investigate the effect of the type of base papers containing rice straw-BKP on the properties and printabilities of coated papers. Also, it was intended to evaluate the effect of coated paper prepared with anionic and amphoteric latex based coating color. The results obtained from this study were as follows. The fiber length of rice straw-BKP was observed much shorter than those of the wood pulps(Sw-BKP, Hw-BKP). This has effect on physical properties. Therefore, the results of measured physical properties were much lower than those of wood pulps. Also, physical properties tended to increase slightly with the increase of the mixing ratio of Sw-BKP. Opacity of the base paper containing rice straw-BKP, was higher than that of the wood pulps. As the mixing ratio of Sw-BKP increases the opacity tended to decrease. On the other hand, smoothness and roughness to rice straw-BKP were similar with Sw-BKP and its air permeability was lower than those of wood pulps. The optical properties of coated papers containing rice straw-BKP tended to be similar to those of other base papers. The ink receptivity of coated papers containg rice straw-BKP was higher than those of wood pulps and printing gloss was similar to wood pulps. The mixing ratio of Sw-BKP, the ink receptivity and printing gloss of rice straw-BKP were showed no difference irrespective the mixing ratio of Sw-BKP. Meannwhile, amphoteric latex improved the optical properties and printability of coated papers.

The Effect of Addition of Grape Pomace on Chemical Composition and Quality of Silage (포도박 첨가가 사일리지의 화학적 성분과 발효품질에 미치는 영향)

  • The qualities of the rice straw, black locust and orchardgrass silage added with grape pomace were investigated in this study and the amount of grape pomace added in different treatments were 0, 20, 40 and 50%, respectively. Crude protein contents of rice straw(6.5-10.6%) and orchardgrass silage(13.2-14.8%) added with grape pomace were significantly higher(p<0.05) than that of 100% rice straw(5.3%) and orchard grass silage(12.8%), however the crude protein contents(14.9-15.6%) of black locust silage in supplementation were lower than that of 100% black locust silage(16.3%). With higher amount of addition of grape pomade to rice straw, black locust and orchardgrass silages, ADF, NDF and crude ash contents decreased significantly(p<0.75). Moisture contents decreased according to the higher proportion of grape pomace in the rice straw silages, but increased according to the higher proportion of grape pomace in the black locust and orchardgrass silage. Values of pH in 20∼60% addition of grape pomace in rice straw(4.0∼4.2), black locust(4.1∼4.3) and orchardgrass silages(4.2∼4.4) were lower than that of 100% in rice straw(4.6), black locust(5.4) and orchardgrass silages(4.7). The contents of acetic acid and total volatile fatty acid significantly increased according to higher levels of addition of grape pomace compared to the respective values of silage. However the contents of lactic acid in supplementation of grape pomace were lower than that of 100% black locust silage.

Effects of Rice Straw Amendment and Nitrogen Fertilization on Rice Growth and Soil Properties in Reclaimed Tidal Paddy Field

  • Farmers with forage barley (Hordeum vulgare L.)-rice (Oryza sativa L.) cropping system at reclaimed tidal lands burn crop residues to facilitate seedbed preparation or remove them for feed stock. This study was conducted to investigate the effect of rice straw amendment and N fertilization on soil properties and N uptake of rice under forage barely-rice cropping system at reclaimed tidal paddy field. Rice straw was applied at the rates of 0, 2.5 and $5.0ton\;ha^{-1}$ and N was fertilized at 0, 100, 200 and $400kg\;ha^{-1}$. Although there was no significant difference in the growth and yield of rice, fresh and dry weight of forage barely increased with increasing the amount of rice straw. The amount of N uptake of rice at harvesting stage was $65.8-69.2kg\;ha^{-1}$ by the amount of rice straw amendment, but there were no significant differences among rice straw amendment levels. After harvesting the rice, the soil salinity decreased with rice straw amendment compared to the control. After forage barely and rice cultivation, soil organic matter contents increased to $2.6-2.8g\;kg^{-1}$ and $3.2-3.5g\;kg^{-1}$, respectively. The amount of N uptake of rice at harvesting stage increased up to $82kg\;ha^{-1}$ in $400kg\;ha^{-1}$ N applied plots which were $37.8kg\;ha^{-1}$ higher than the control. Nitrogen fertilization decreased N recovery efficiency. The highest yield of rice was observed at $244kg\;ha^{-1}$ N fertilization level, but the optimum N level was estimated at $168kg\;ha^{-1}$ in order to keep the protein content of rice under 6.5%. Further researches on N uptake and application of organic matter according to soil salinity will be necessary to increase N use efficiency at reclaimed tidal paddy field.

EFFECT OF ALKALINE HYDROGEN PEROXIDE TREATMENT OF RICE STRAW ON IN SACCO RUMINAL DIGESTIBILITY

  • The objective of this experiment was to compare the effect of pH-regulated alkaline hydrogen peroxide (AHP) treatment of rice straw with those of sodium hydroxide (NaOH) and anhydrous ammonia ($NH_3$) treatments on in sacco digestivility. Three non-lactating ruminally cannulated Holstein cows were fed a diet containing 90% forage and 10% concentrate on a dry matter (DM) basis. The AHP treatment significantly (p<0.05) reduced acid detergent lignin content of the straw, resulting in significant (p<0.05) increase of neutral detergent fiber (NDF), acid detergent fiber (ADF) and cellulose concentrations. Disappearance rates of DM and NDF of the straw significantly (p<0.05) increased at the incubation time of 24 h. On the other hand, those of ADF and cellulose were significantly (p<0.05) higher at the incubation time of 12 h than those of the others. The effective degradability of DM(EDDM), NDF(EDNDF), ADF(EDADF) and cellulose (EDCE) were determined using in sacco nylon bag technique on the basis of 0.05/h solid outflow rate. The greater differences (p<0.05) of EDDM, EDNDF, EDADF and EDCE were found between AHP treated straw and the others. In general, AHP treatment of the straw recorded higher digestion coefficients than untreated straw as well as NaOH and $NH_3$ treated straws. The results of this study demonstrate that AHP treatment can be used as a effective method for improving the nutritive value of rice straw for ruminants.

  • This experiment was on purpose to estimate freezing and thawing resistance concering with developing rice straw ash concrete which were mixed rice straw ash to cement as ratio of cement weight. Freezing and thawing test was done by Method A of KS F 2456. It could estimate change of original mass, pulse velocity and dynamic modulus of elasticity during test. Test results showed that 5% filled rice straw ash concrete had the highest durability factor(DF) as 86 and from 5% to 7.5% filled rice straw ash concrete showed higher DF than normal cement concrete.

  • In this Experiment three wether male Matou goats (♂), all fitted with permanent rumen fistulae, were used to study the rumen degradabilities (incubation time 48h) of dry matter (DM), neutral detergent fibre (NDF), acid detergent fibre (ADF), acid detergent ligin (ADL), cellulose (CEL) and hemicellulose (HC) and their contents of wheat and rice straws were determined for the different morphological fractions and segments. The results showed that the variation of different fractions for wheat and rice straw is the true resources of their nutritive value variation and the cell wall contents of wheat and rice straw are also obviously different. The rumen degradabilities of different straw fibre are significantly different (p<0.01), mean while the effect of straw variety is also significant (p<0.05). The aim was to set up a foundation for studying the rumen degradation of the roughage resources.

Acetone-Butanol Fermentation of Rice Straw by Simultaneous Saccharification and Fermentation (동시당화 발효법에 의한 볏짚의 Acetone-Butanol 발효)

  • Rice straw was used in the acetone-butanol fermentation by semultaneous saccharification and fermentation (SSF) using Clostridium acetobutylicum and cellulolytic enzyme. Over 230 mM of solvent was produced from alkali treated rice straw of from ball-milled microcrystalline cellulose whilst only acidic fermentation products were formed from ball-milled rice straw. From the results it is concluded that rice straw used in the study contained an inhibitor for the solventogenesis by the organism which is insoluble in water and some organic solvent and destroyed by alkaline treatment.

  • This paper surveys the effect of rice straw ash solution to the physical properties of Sappan Wood dyeing fabrics. In the quantitative analysis of rice straw ash solution, the quantities of absorbed ingredients in fabrics were increased by bath pull treatment but the amount of absorption(K/S value) was increased by bath pH4.5 treatment. This is related to the metal ion. Among the metal ion, effect of Fe iou and Al ion were related. In case added extracted dye solution to mordants, the color dye solution became dark and increased reddish. The changes of mechanical properties of fabrics tensile resilience, bending rigidity(B), compressional resilience(RC) were increased. Generally mechanical properties were increased by rice straw ash solution treatment, specially bath pH9 treatment. Rice straw ash solution treatment of dyeing fabrics made the improvement in tensile strength and elongation and in the amount of absorption, dye ability, color fastness, mechanical properties, tensile strength, elongation.
