Effects of lactic acid bacteria and molasses on fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro ruminal fermentation of rice straw silage

  • Objective: This study was to evaluate the fermentation dynamics, structural and nonstructural carbohydrate composition and in vitro gas production of rice straw ensiled with lactic acid bacteria and molasses. Methods: Fresh rice straw was ensiled in 1-L laboratory silos with no additive control (C), Lactobacillus plantarum (L), molasses (M) and molasses+Lactobacillus plantarum (ML) for 6, 15, 30, and 60 days. After storage, the silages were subjected to microbial and chemical analyses as well as the further in vitro fermentation trial. Results: All additives increased lactic acid concentration, and reduced pH, dry matter (DM) loss and structural carbohydrate content relative to the control (p<0.05). The highest organic acid and residual sugar contents and lignocellulose reduction were observed in ML silage. L silage had the highest V-score with 88.10 followed by ML silage. L and ML silage improved in vitro DM digestibility as compared with other treatments, while in vitro neutral detergent fibre degradability (IVNDFD) was increased in M and ML silage (p<0.05). M silage significantly (p<0.05) increased propionic acid (PA) content and decreased butyric acid content and acetic acid/PA as well as 72-h cumulative gas production. Conclusion: The application of ML was effective for improving both the fermentation quality and in vitro digestibility of rice straw silage. Inclusion with molasses to rice straw could reduce in vitro ruminal gas production.

METHODS OF UREA INCORPORATION IN STRAW AND THEIR EFFECTS ON PERFORMANCE OF BUFFALO HEIFERS

  • Twelve indigenous buffalo heifers were grouped into 3 and were supplied with following diets-urea-ensiled rice straw + wheat bran + green grass (A); rice straw soaked in urea-water + wheat bran + green grass (B); and untreated straw + wheat bran + green grass (C); so that each group received any one diet. Total dry matter intake was highest in animals on diet C and lowest on B. However, straw DM intake was highest in animals on silage diet and lowest on untreated straw diet. Organic matter and crude fibre digestibility of silage and soaked straw diets were significantly higher (p<0.01) than those of untreated straw diet. Although the values were lower with soaked straw diet than on silage diet and soaked straw diet were significantly (p < 0.01) higher, however, the value for silage diet was not significantly different from that of soaked straw diet.

Development of a new lactic acid bacterial inoculant for fresh rice straw silage

  • Objective: Effects of newly isolated Lactobacillus plantarum on the fermentation and chemical composition of fresh rice straw silage was evaluated in this study. Methods: Lactic acid bacteria (LAB) from good crop silage were screened by growing them in MRS broth and a minimal medium with low carbohydrate content. Selected LAB (LAB 1821) were Gram-positive, rods, catalase negative, and were identified to be Lactobacillus plantarum based on their biochemical characteristics and a 16S rRNA analysis. Fresh rice straw was ensiled with two isolated LAB (1821 and 1841), two commercial inoculants (HM/F and P1132) and no additive as a control. Results: After 2 months of storage at ambient temperature, rice straw silages treated with additives were well-preserved, the pH values and butyric and acetic acid contents were lower, and the lactic acid content and lactic/acetic acid ratio were higher than those in the control (p<0.05). Acidity (pH) was lowest, and lactic acid highest, in 1821-treated silage (p<0.05). The $NH_3-N$ content decreased significantly in inoculant-treated silage (p<0.05) and the $NH_3-N$ content in 1821-treated silage was lowest among the treatments. The dry matter (DM) content of the control silage was lower than that of fresh rice straw (p<0.05), while that of the 1841- and p1174-inoculant-treated silages was significantly higher than that of HM/F-treated silage. Microbial additives did not have any significant (p>0.05) effect on acid detergent fiber or neutral detergent fiber contents. Crude protein (CP) content and in vitro DM digestibility (IVDMD) increased after inoculation of LAB 1821 (p<0.05). Conclusion: LAB 1821 increased the CP, IVDMD, lactic acid content and ratio of lactic acid to acetic acid in rice straw silage and decreased the pH, acetic acid, $NH_3-N$, and butyric acid contents. Therefore, adding LAB 1821 improved the fermentation quality and feed value of rice straw silage.

마늘대 사일리지 급여가 한우거세우의 성장 및 도체특성에 미치는 영향 (Effect of Garlic Stalk Silage on Performance and Carcass Characteristics of Hanwoo Steers)

  • 평균체중 120kg 내외의 생후 5개월령 한우 27두를 외과적 수술로 거세를 실시한 후 대조구(볏짚 급여구) 10두와 시험구(마늘대 사일리지 급여구) 17두로 나누어 사양시험을 22개월 동안 실시하였다. 농후사료는 대조구에서 육성기, 비육전기 및 비육후기 동안 각각 체중의 1.34, 1.69 및 1.65%를 급여하였고 시험구에서는 1.47, 1.74 및 1.66%를 급여하였다. 조사료원으로 볏짚을 대조구에서 육성기, 비육전기 및 비육후기 동안 각각 체중의 1.37, 0.65 및 0.43%를 급여하였고 시험구에서는 1.38, 0.63 및 0.43%를 급여하였다. 또한 시험구에서는 마늘대 사일리지를 비육전기 및 비육후기 동안 각각 체중의 0.22 및 0.33%를 급여하였다. 본시험의 결과를 요약하면 다음과 같다. 일당증체량은 전기간 동안 대조구와 시험구간에 각각 0.80 및 0.81kg으로 유의적인 차이가 없었다(P〉0.05). 농후사료 섭취량은 대조구와 시험구 각각 5.02kg 및 5.35kg으로 시험구가 약간 높았지만 유의적인 차이는 없었으며(P〉0.05), 조사료 섭취량은 대조구와 시험구 각각 2.58 및 3.58kg이었고, 사료효율은 대조구와 시험구가 각각 9.50 및 11.05로 차이는 없었다. 육량 등급은 마늘대 급여구가 약간 낮았지만 유의적인 차이는 없었고(P〉0.05), 육질 등급은 유의적인 차이는 없었지만 시험구가 약간 높았다. 쇠고기의 전단력은 마늘대 급여구가 볏짚 급여구보다 낮았다. 도체의 아미노산 조성은 필수 아미노산 및 비필수 아미노산 모두 처리구간에 차이가 나타나지 않았고, 도체의 지방산 조성 중 포화 지방산은 대조구가 약간 높았고, 불포화 지방산은 반대로 처리구가 약간 높았다. $\omega$-6과 $\omega$-3 지방산의 비율은 마늘대 급여구가 볏짚 급여구보다 좁아서 마늘대 급여가 불포화지방산중 oleic acid 함량이 볏짚 급여구에 비하여 더 높아서 근육 중의 지방산의 조성을 좋게 하는 것 같다. 두당 소득은 시험구가 대조구보다 22.54% 증가하였다. 이상의 결과들을 종합하여 볼 때 한우 거세우에 마늘대 사일리지를 비육기간 동안 급여하면 거세우의 증체에는 영향을 주지는 않지만, 한우 사육 농가에는 소득의 증대를 가져오므로 마늘대 사일리지를 조사료 공급원으로 급여하는 것이 바람직하다고 사료된다.

볏짚과 고마리(Persicaria Thunbergii Gross var. Coreana Gross ex Nakai)의 혼합Silage 제조에 관한 연구 (Study on the Silage from Rice Straw and Persicaria Thunbergii)

  • 볏짚의 사료가치(飼料價値)를 증진(增進)하고 아울러 고마리의 사료가치(飼料價値)를 구명(究明)하기 위하여 볏짚과 고마리의 혼합비율(混合比率)을 각각(各各) 10 : 90($T_1$), 20 : 80($T_2$), 30 : 70($T_3$), 및 40 : 60($T_4$)로 처리(處理)하여 혼합(混合)Silage를 제조(製造)한 후(後) 이들 재료(材料)와 Silage의 조성분석(組成分析)하고 Silage의 품질(品質)을 판정(判定)하여 그 결과(結果)를 아래와 같이 요약(要約)하였다. 1. 통일계(統一系)(노풍)볏짚의 조성분중(粗成分中) 조단질(粗蛋質)과 가용무질소물(可溶無窒素物)이 고형물비(固形物比)로 각각(各各) 8.18% 및 44.66%로 일반(一般)볏짚보다 사료가치(飼料價値)가 양호(良好)해 보였고 고마리도 고형물(固形物) 기준(基準)으로 조단백질(粗蛋白質)이 14.47%로 일반적(一般的)으로 화본과목(禾本科牧草)보다 높았고 가용무질소물(可溶無窒素物)은 51.13%로 대략 화본과(禾本科)나 두과목초(豆科牧草)보다 높아 보였다. 2. Silage의 수분함량(水分含量)은 $T_2$가 72.12%, $T_3$가 66.43%로 비교적 수분함량(水分含量)이 적당(適當)했고 모든 처리구(處理區)가 타(他) Silage에 비(比)하여 조단백질량(粗蛋白質量)이 높아 보였다. 3. Silage의 pH는 $T_1$이 pH 3.8로 가장 양호(良好)하였고 $T_4$가 pH 4.5로 가장 높았으며 유기산함량(有機酸含量)은 $T_1$이 가장 적었고 $T_4$가 가장 많았으나 총산(總酸)에 대(對)한 유산비율(乳酸比率)은 $T_1$쪽이 높고 $T_4$가 떨어졌다. 4. 볏짚과 고마리의 혼합(混合)으로 양질(良質)의 Silage를 제조(製造)할 수 있었으며 볏짚과 고마리의 혼합비율(混合比率)은 20 : 80~30 : 70 범위가 적당(適當)했으며, 앞으로 혼합(混合) Silage의 이용가치(利用價値)가 기대(期待)된다.

볏짚의 화학적 처리와 사일리지 제조가 화학성분 변화 및 한우 반추위 분해율에 미치는 영향 (Effects of Chemical Treatments and Ensiling on the Chemical Composition and Degradation Rate in the Rumen)

  • This study was conducted to examine of rice straw after chemical treatments and ensiling on its feeding value, in situ studies using a rumen fistulated Korean cow and nylon bag technique. NaOH treatment greatly improved the degradation the Dry matter and Neutral detergent fiber degradation in the rumen but the intake was not affected. Ammonia treatment did not improve the degradation rate of rice straw in the rumen, but remarkably increased the rice straw digestibility and intake by sheep. Making silage of rice straw did not affect its rumen degradation rate, but the digestibility and its take by sheep were greatly improved especially when a little molasses together with Lactobacillus were supplemented. Degradation rate of rice straw in the rumen measured by nylon bag technique was influenced by various treatments but did not appear to coincide with digestibility by sheep. This would be due to the fact that feed intake affect digestibility as well as the degradation in rumen. Therefore, it can be said that making silage with some molasses and Lactobacillus is one of the easest way of using rice straw for animal feed. (Key words : NaOH, Digestibility, Silage, Molasses )

볏짚, 클로버 및 고구마덩굴 Silage 조제(調製)를 위한 생고구마 첨가(添加)의 효과(效果) (Effects of Adding Crushed Fresh Sweet Potatoes on the Quality of Rice Straw, Clover and Sweet Potato Stalk Silages)

  • 볏짚, 클로버 및 고구마 덩굴 silage의 조제(調製) 시(時)에 고구마의 첨가(添加)가 이들 silage의 품질(品質)에 미치는 영향(影響)을 구명(究明)하기 위하여 silage 조제시(調製時)에 파쇄(破碎)한 생 고구마를 클로버 및 고구마 덩굴 silage에는 각각(各各) 5~15%, 볏짚 silage에는 10~30%로 첨가(添加)하여 조제(調製)한 후(後)에 완성(完成)된 silage의 일반조성분(一般組成分), 유기산(有機酸) 및 pH를 측정(測定)하여 품질(品質)을 평가(評價)한 결과(結果)는 다음과 같다. 1. Silage의 일반(一般) 조성분(組成分) 중(中)에서 수분(水分) 함량(含量)은 고구마덩굴 silage에서, 조단백질(粗蛋白質) 함량(含量)은 클로버 silage에서 그리고 가용무질소물(可溶無窒素物)과 조직유(粗織維)의 함량(含量)은 볏짚 silage에서 가장 높았다. 모든 silage에서 고구마의 첨가수준(添加水準)이 높아 질수록 가용무질소물(可溶無窒素物)은 증가(增加)되었고 조직유(粗織維)는 감소(減少)되었다. 2. Silage의 총산함량(總酸含量)은 클로버 silage에서 1.96~2.75%로 가장 높았고, 총산(總酸)에 대(對)한 유산(乳酸)의 비율(比率)은 고구마덩굴 silage에서 81~85%로 가장 높았다. 모든 silage에서 고구마의 첨가(添加)수준(水準)이 높을 수록 총산함량(總酸含量)에 대(對)한 유산함량(乳酸含量)의 비율(比率)이 증가(增加)되었다. 3. Silage의 pH는 고구마덩굴 silage에서 4.10~4.20으로 가장 낮았고 볏짚 silage에서 4.75~5.22로 가장 높았다. 모든 silage에서 고구마의 첨가(添加) 수준(水準)이 높을 수록 pH가 낮아졌다. 4. 고구마 첨가시(添加時)에 고구마의 시중가격(市中價格)을 고려(考慮)한다면 고구마덩굴 silage에서는 첨가효과(添加效果)가 크게 기대(期待)되지 않으나 클로버 silage에서는 5%, 볏짚 silage에서는 10% 정도(程度)가 적당(適當)하였다.

EFFECT OF ADDITION OF INTACT OR ALKALIZED LUCERNE JUICE AT ENSILING ON THE NUTRITIVE VALUE OF RICE STRAW SILAGE

  • This experiment was conducted to study the effect of addition of Lucerne juice (LJ) obtained by mechanical extraction of freshly harvested crop on the nutritive value of rice straw silage. Rice straw (RS) was ensiled with intact, NaOH or $NH_3$ treated LJ at 3:7 ratio on fresh weight basis (LJ RS, LJ NaOH RS and LJ $NH_3$ RS, respectively). Each alkali was mixed with fresh juice at a level of 4% of rice straw dry matter just before ensiling. Rice straw ensiled with water was prepared as the control (W RS). In the digestion trial, goats were allocated in a $4{\times}4$ Latin-square design and fed the diet containing three parts of RS silage and one part of wheat bran (DM basis). For the goats receiving the control silage, urea was supplemented at feeding time so as to adjust the nitrogen intake except for goats on LJ $NH_3$ RS silage. Crude protein content of RS silage was increased from 5.2 to 9.1% (DM basis) by the addition of intact LJ and to about 24% by $NH_3$ treated LJ. The control W RS silage contained only trace amount of lactic acid and was dominated by acetic and butyric acid. The addition of intact LJ reduced butyric acid content and $NH_3-N/TN$ of the silage whereas the addition of alkalized LJ increased those values and shifted to a butyrate type fermentation. Nutrient digestibilities and nitrogen balance of goats were almost the same when they were fed W RS and LJ RS silage indicating the addition of intact LJ did not improve the nutritive value. The addition of alkalized LJ significantly increased the fiber digestibilities of RS silage and $NH_3$ treatment was more effective than NaOH treatment. Postprandial ruminal $NH_3-N$ and blood urea nitrogen (BUN) concentrations were decreased by feeding LJ NaOH RS silage suggesting ruminal protein synthesis was enhanced along with the increase of energy supply for supply for rumen microbes by the alkali treatment. The advantageous fiber digestibilities of LJ $NH_3$ RS silage compared with those of LJ NaOH RS silage might be attributable to a sufficient nitrogen supply for microbial fiber digestion in the rumen.

  • The objective of this study was to determine the effect of various roughage sources on nutrient digestibility and enteric methane ($CH_4$), and carbon dioxide ($CO_2$) production in goats. Four castrated black goats ($48.5{\pm}0.6kg$) were individually housed in environmentally controlled respiration-metabolism chambers. The experiment design was a $4{\times}4$ balanced Latin square design with 4 roughage types and 4 periods. Alfalfa, tall fescue, rice straw, and corn silage was used as representative of legume, grass, straw, and silage, respectively. Dry matter digestibility was higher (p < 0.001) in corn silage than in alfalfa hay. Dry matter digestibility of alfalfa hay was higher than those of tall fescue or rice straw (p < 0.001). Neutral detergent fiber digestibility of tall fescue was lower (p < 0.001) than those of alfalfa, rice straw, or corn silage. Daily enteric $CH_4$ production and the daily enteric $CH_4$ production per kilogram of $BW^{0.75}$, dry matter intake (DMI), organic matter intake (OMI), digested DMI, and digested OMI of rice straw did not differ from those of tall fescue but were higher (p < 0.001) than those of alfalfa or corn silage. Roughage type had no effect on enteric $CO_2$ emission in goats. Straw appeared to generate more enteric $CH_4$ production than legume or silage, but similar to grass.

Effects of Lactic Acid Bacteria Inoculants on Fermentation of Low Moisture Fresh Rice Straw Silage at Different Storage Periods

  • The purpose of this study was to analyze the effectiveness of different storage periods of lactic acid bacteria (LAB)-fermented low moisture fresh rice straw silage. The low moisture fresh rice straw sample was inculcated with LAB and stored for different storage periods such as 45, 90, and 365 days, respectively. The low moisture fresh rice straw (LMFRS) silage inoculated with LAB exhibited reduction in pH throughout the fermentation as compared with the control (P<0.05). The lactic acid content was increased at the late fermentation period (90 and 365 days, respectively) in LAB inoculated LMFRS silage as compared with the control (P<0.05). In contrast, the acetic acid and butyric acid concentrations were slightly reduced in the LAB inoculated LMFRS silage sample at 90 and 365 days fermentation, respectively. Meanwhile, the non-inoculated LMFRS silage showed higher amounts of acetic acid and butyric acid at an extended fermentation with low bacterial population as compared with the LAB inoculated LMFRS silage. However, lactic acid concentration was slightly high in the non-inoculated LMFRS silage at early 45 days fermentation. Additionally, the nutrient profile such as crude protein (CP), acid detergent fibre (ADF), neutral detergent fibre (NDF), and total digestibility nutrients (TDN) were not significantly different in control and LAB inculcated samples during all fermentation. Though, the microbial population was greater in the LAB inoculated LMFRS silage as compared with the control. However, the massive population was noted in the LAB inoculated LMFRS silage during all fermentation. It indicates that the inoculated LAB is the main reason for increasing fermentation quality in the sample through pH reduction by organic acids production. Overall results suggest that the LAB inoculums are the effective strain that could be a suitable for LMFRS silage fermentation at prolonged days.
