Lipid Lowering Effect of Anthocyanin-Pigmented Rice Bran in Streptozotocin-Induced Diabetic Male Rats

  • Oryza sativa cv. Heugjinjubyeo, an anthocyanin-pigmented rice variety, is well known to contain high levels of bioactive phytochemicals, anthocyanin, quinolone alkaloids and phenolic acids. Here, we studied the inhibitory effect of Oryza sativa cv. Heugjinjubyeo bran on the absorption of dietary fat in streptozotocin-induced diabetic Sprague-Dawley male rats. For these experiments, experimental animals were divided into four groups: normal, diabetic-control and two experimental groups that were fed 1.0 g or 2.0 g/kg body weight/day of Oryza sativa cv. Heugjinjubyeo bran supplement for 14 days. As a result, liver glycogen levels increased significantly by 65% and 32% in groups receiving 1.0 g and 2.0 g/kg body weight/day, respectively, compared to diabetic-control. Liver cholesterol levels were significantly lower by 8.3% and 14.5% in the groups fed 1.0 g and 2.0 g of anthocyanin- pigmented rice extracts, respectively.

A Study on the Source of Reductants for Nitrate Reduction in Rice (Oryza sativa cv. Tongil) Roots (벼(Oryza sativa cv. Tongil) 뿌리에 있어서 Nitrate 환원에 필요한 환원력의 공급원에 관한 연구)

  • There was a decrease in nitrate reductase activity (NRA) measured in vivo in rice roots (Oryza sativa cv. Tongil) grown in anaerobic culture solution. But it was reversed by addition of malonate to the in vivo nitrate reduction assay medium. Malonate increased the in vivo NRA during 2-5 hours incubation and decreased it in longer incubation hours. In vivo NRA was stimulated by addition of NaHCO3 to the assay medium, but not by Na2CO3. The stimulation of NRA by NaHCO3 was not observed in shoot removed rice roots. It is suggested that CO2 from NaHCO3 is carboxylated by phosphoenol pyruvate carboxylase, results in increasing the malate contents in the roots, and stimulates the in vivo NRA. NADH needed in nitrate reduction is supported by malate oxidation. In rice roots, it seems probable that malate oxidation in the mitochondria is more important to nitrate reduction than malae oxidation in cytoplasm.

Genetic Variation of Rice Populations Estimated Using nrDNA ITS Region Sequence

  • The rice belonging to Oryza sativa is not only has significant economic importance, for it is the major source of nutrition for about 3 billion all around the world. But also plays a vital role as a model organism, because it has a number of advantages to be a model plant, such as efficient transformation system and small genome size. Many methods and techniques have been conducted to attempt to distinguish different Oryza sativa species, such as amplified fragment length polymorphism (AFLP), random amplified polymorphic DNA (RAPD), simple sequence repeat (SSR) and so on. However, studies using sequence analysis of internal transcribed spacer (ITS), a region of ribosomal RNA has not been reported until now. This study was undertaken with an aim to understand the phylogenetic relationships among sixteen isolates of Oryza sativa collected from abroad and fifteen isolates collected from Korea, using ribosomal RNA (rRNA) internal transcribed spacer (ITS) sequences to compare the phylogeny relationships among different Oryza sativa species. The size variation obtained among sequenced nuclear ribosomal DNA (nrDNA) ITS region ranged from 515bp to 1000bp. The highest interspecific genetic distance (GD) was found between Sfejare 45 (FR12) and Anapuruna (FR15). Taebong isolate showed the least dissimilarity of the ITS region sequence with other thirty isolates. This consequence will help us further understanding molecular diversification in intra-species population and their phylogenetic analysis.

  • This study was performed to elucidate the mechanisms involved in efflux process of phosphate from intact and excised roots of Oryza sativa (Tongil). The rapid loss of phosphate during the first 40-minutes of treatment is mainly from the cytoplasmic fraction, and the gradual loss taking place afterwards is possibly reflecting the losses at the tonoplast. During the first 60 minutes, loss of phosphate from the excised roots treated with KCN was more rapid than the control but slower thereafter. The effect of $CaCl_2$ and KCl appeared to decrease the rate of phosphate efflux in excised roots of rice. The mechanism of phosphate efflux by rice roots seems to be passive and oscillatory in the short period. Key words: Oryza sativa(Tongil), Phosphate efflux.

  • Three phytosterols of rare occurrence, schleicheol 2 (1), $7{\beta}$-hydroxysitosterol (2), and $7{\alpha}$-hydroxysitosterol (3), were isolated from the n-hexane fraction of rice (Oryza sativa) bran, for the first time. Some nuclear magnetic resonance (NMR) assignments in the literatures are inaccurate. This study employed two-dimensional NMR experiments to identify exact peak assignments.

  • Telomeres are specialized nucleoprotein complexes that function to protect eukaryotic chromosomes from recombination and erosion. Several telomere binding proteins (TBPs) have been characterized in higher plants, but their detailed in vivo functions at the plant level are largely unknown. In this study, we identified and characterized OsTRFL1 (Oryza sativa Telomere Repeat-binding Factor Like 1) in rice, a monocot model crop. Although OsTRFL1 did not directly bind to telomere repeats $(TTTAGGG){_4}$ in vitro, it was associated with telomeric sequences in planta. OsTRFL1 interacted with rice TBPs, such as OsTRBF1 and RTBP1, in yeast and plant cells as well as in vitro. Thus, it seems likely that the association of OsTRFL1 with other TBPs enables OsTRFL1 to bind to telomeres indirectly. T-DNA inserted OsTRFL1 knock-out mutant rice plants displayed significantly longer telomeres (6-25 kb) than those (5-12 kb) in wild-type plants, indicating that OsTRFL1 is a negative factor for telomere lengthening. The reduced levels of OsTRFL1 caused serious developmental defects in both vegetative and reproductive organs of rice plants. These results suggest that OsTRFL1 is an essential factor for the proper maintenance of telomeres and normal development of rice.

Brassinosteroid substances in immature oryza sativa seeds (벼종자의 brassinosteroid 활성물질)

  • To investigate the presence of the brassinosteroid substances in immature Oryza sativa L. cv Tongjinbyeo seeds, the methanol extract was purified by the sequential use of solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 chromatography and charcoal adsorption chromatography. The activity of brassinosteroid was monitored by the rice inclination test and its presence could be confirmed in each purification step. The purified active components were separated by silica gel adsorption chromatography and Sephadex LH-20 chromatography. Brassinosteroid substances in separated active fractions were identified as castasterone, teasterone and 6-deoxocastasterone by HPLC. Our work is probably the first report of endogenous brassinosteroids in Oryza sativa seeds. The content of brassinosteroid in Oryza sativa seeds as converted into brassinolide was $0.5{\sim}1.5\;ng/g$ fresh weight.

Phytolith analysis and rice cultivation possibility from Myosa-ri Archeological Site, Haman-gun, Gyeongnam Province (경남 함안 묘사리 식물규소체 분석과 농경가능성)

  • Phytolith analysis of sediments at the river mouth of valley bottom plains in Myosa-ri, Haman is performed. By the phytolith results, Phragmites and Oryza sativa are dominated in the lower horizon. An abundance of Paniceae and Andropogoneae indicates the active agricultural activities under wet conditions. Moreover, the results of archeological studies suggest the paddy fields in the Three States Age. The phytolith assemblages are dominated in the middle horizon and they gradually increase in the upper horizon. The rice cultivation layer is not proportional to total phytolith yield due to the dominance of Oryza sativa in the lower horizon. However, because the Oryza sativa are continuously observed over the entire sediments, it is suggested that there had been the agricultural activities for the entire periods.
