The crude extracts from Rhus javanica Linne showed comparatively strong antioxidative activity in test oils. Antioxidative components were isolated and identified by column chromatography, thin layer chromatography, UV, and NMR. These antioxidative components were added to several oils to compare antioxidative activity with several commercial antioxidants, such as BHA, BHT, and tocopherol. After the sixth column chromatography, one fraction (R-18-9-3-2-4-2) was separated from chloroform layer of Rhus javanica Linne. The R-18-9-3-2-4-2 fraction was identified as methyl gallate by $^1H-NMR$ and $^{13}C-NMR$ and confirmed with methyl gallate standard as authentic. The R18-9-3-2-4-2 fraction from chloroform layer of Rhus javanica Linne showed stronger activity than that of the ${\alpha}-,\;{\delta}-tocopherol$, BHT, and BHA at the same concentration.
The free radical scavenging activities and the protective effects of Rhus javanica extracts against oxidative damage induced by reactive oxygen species (ROS) were investigated. n-Hexane, ethyl acetate and water fractions were prepared from a methanol extract. DPPH radical, superoxide anion and hydroxyl radical scavenging activities were estimated. Intracellular ROS formation was quantified using fluorescent probes, 2', 7'-dichlorofluorescin diacetate (DCFH-DA) for hydroxyl radical and dihydroethidium (DHE) for superoxide anion. The oxidative DNA damage was investigated by the comet assay in HepG$_2$ cells exposed either to $H_2O$$_2$ or to menadione. The highest $IC_{50}$/ values for DPPH radical scavenging activity was found in the ethyl acetate fraction with a value of 5.38 $\mu\textrm{g}$/mL. Cells pretreated with $\geq$ 1 $\mu\textrm{g}$/mL of the ethyl acetate extract had significantly increased cell viability compared to control cells, which were not pretreated with the extract. Intracellular ROS formation and DNA damage in HepG$_2$ cells, which were pretreated with the various concentrations of Rhus javanica ethyl acetate extract and then incubated either with $H_2O$$_2$ or with menadione, reduced in a dose-dependent manner. These findings suggest that Rhus javanica might have biologically active components which have strong protective effects against ROS induced oxidative damages to the biomolecules, such as cell membranes and DNA.
This study was carried out to investigate the antioxidative activities of medicinal plants. Through the examination of methanol extracts from 30 species for radical scavenging effects using DPPH method, the extracts from Rhus javanica Linne, Smilax china Linne and Polygonum cuspidatum Siebold et Zucarinii showed strong antioxidative activity. Because of its highest antioxidative activity among 30 medicinal plants, radical scavenging effects of 4 different extract compartments (n- Hexane, EtOAc, BuOH and $H_2O$ extracts) from Galla Rhois MeOH extract of Rhus javanica Linne were examined by DPPH method and antioxidant effects on the 4 different extract compartments were tested by Ferric-Thiocyanate method. Antioxidative activities of n- Hexane, EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioxidants (BHA), suggesting that major fractions for the antioxidative activity of Rhus javanica Linne were the n- Hexane, EtOAc and BuOH extract compartments.
Certain parts of 95 species of edible and medical plants were extracted with water and 75% of ethyl alcohol. After addition of those extracts to palm oil, lard and soybean oil at different level, their antioxidative activities were compared by Rancimat test. Six species among them seemed to have rather strong antioxidative activity and high extracting yields(i.e. Taraxacum platycarpum, Plantago asiatica, Rhus javanica L., Lycopus lucidus, Astragalus membranaceus, Taraxacum platycarpumH). Among them, the Rhus javanica L. ethanol extract retarded greatly the induction period of palm oil and lard. When 600 ppm of Rhus javanica L. extract were added to palm oil and lard, AI(antioxidant index was expressed as induction period of oil containing various plant extracts/induction period of control oil) of each was 1.35 and 3.03 respectively. This result indicated that the Rhus javanica L. extract was more effective on lard than the other oils.
This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.
Antioxidative effects of crude ethanol extract of the Rhus javanica L. and its fractionates with various synergists on the oxidation of palm oil, lard and soybean oil were compared with induction period(IP) using a Rancimat test. Addition of 200 ppm of the crude extract with phosphoric acid to palm oil extended IP 2.89 times as much as that of control and ethyl acetate fractionate extended IP 4.18 times at the same condition. To lard, 600 ppm of chloroform fraction with 200 ppm of ${\delta}-Tocopherol$ extended IP 13.42 times as much as that of control. 200 ppm of each fraction with various synergists were added to palm oil and lard, and oxidative stability of the oils were monitored by measuring POV, AV, and TBA value. The POV of palm oil containing 200 ppm of ethyl acetate and chloroform fraction with 200 ppm of phosphoric acid after 27 days storage at $60^{\circ}C$ were 8.9 meq/kg and 9.4 meq/kg respectively while the POV of control was 98.3 meq/kg at the same condition. AV and TBA value were also lower than that of control. The POV value of lard containing same amount of ethyl acetate and chloroform fraction with ${\delta}-Tocopherol$ after 12 day storage at $60^{\circ}C$ were 20.0meq/kg and 10.7 meq/kg respectively while the POV of control was 161.1 meq/kg at the same condition. AV and TBA value were also lower than that of control.
This reserch was carried out to investigate the nitrite scavenging abilities (NSA) of 7 kinds of tea extracts. Nitrites are used as additives of cured meat preperation and exist in plants, foods, and medicines, so we intake it very often easily. Nitrites can act with amines to produce nitrosamine which is known to be a carcinogen. It is known that the pH, concentration of amines, and amounts of nitrite are three important conditions of nitrosamine occurence. 7 kinds of tea used in this experiment were Persimmon tree (Diospyroo kaki Thiunb, Per.), Mulberry tree (Morus alba Linne, Mul.), Rubber tree (eucommia ulmoi-des Oliver, Rub.), Solomon's-seal (Bolygonatu Morr, Som.), Chicory (Cichorrium intybus L, Chi.), Sumach (Rhus javanica L., Sum.), Docwood (Cornus officinale Sieb, Doc.) and they were extracted with methanol (MeOH), ethyl ether (EtEt), ethyl acetate (EtAc), and also the extract existed in the aqueous layer II (Aq L. II) was used.
The major components were isolated from the n-hexane, EtOAc and BuOH extract of Galla Rhois(Rhus Javanica Linne). Their structures were characterized as syringic acid, gallic acid methylester, protocatechuic acid, gallic acid and 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$. This study was carried out to investigate the biological activities of isolated compounds. Five compounds were tested for hepatoprotective effects on CCl4-induced cytotoxicity in primary cultured rat hepatocytes and antioxidative effect on Ferric-Thiocyanate method and TBA method. As a result, isolated five compounds showed stronger antioxidative activity than tocopherol, and the antioxidative activity of gallic acid methylester, protocatechuic acid and syringic acid were similar to that of BHA on Ferric-Thiocyanate method. Specially 1, 2, 3, 4, $6-penta-O-galloyl-{\beta}-D-glucose$ showed stronger effect of lipid-peroxidation inhibition than BHA. Gallic acid appeared stronger inhibitory effect of malondialdehyde on TBA method. Hepatoprotective effect of 1, 2, 3, 4, $6-penta-O-galloyl -{\beta}-D-glucose$ was similar or even higher than that of glycyrrhizin on primary cultured rat hepatocyte cytotoxicity.
Hexane(Hx), ethyl acetate(EtoAc) methanol(MeOH) and 99% ethanol(EtOH) extract of Rhus javanicus Lines with synergists e.g. ascorbic acid(AA), citric acid(CA) and ${\delta}-tocopherol$(TO) were tested their antioxidative effect on palm oil and lard by Rancimat. The methanol showed the highest extraction yield as 14.53%(w/w). When each 600 ppm of Hx, EtOAc, MeOH and EtOH extract with 200 ppm of AA was added to palm oil, the antioxidative index(AI: induction time of oil containing of each extract/induction time of test oil) were 1.83, 2.25, 2.81 and 2.85 respectively which were higher than other treatments and 600 ppm of each extract with 200 ppm of TO to lard, the AI were 3.64, 7.83, 7.34 and 9.30 respectively. Each solvent fractionate of EtOH and EtOAc extracts resulted no higher antioxidative effect than crude whole extract. Palm oil and lard containing 600 ppm of methanol extract were very stable comparing with the control by POV and TBA at oven test($60^{\circ}C$).
