경북 울릉도와 경남 욕지도 야생화들로부터 효모의 분리 및 종 분포 특성 (Isolation and Diversity of Yeasts from Wild Flowers in Ulleungdo and Yokjido, Korea)

  • 경상북도 울릉도와 경상남도 욕지도의 야생화들에 분포하고 있는 효모 종 다양특성을 알아보고자 이들로부터 효모를 분리하고 분자생물학적인 26S rDNA의 D1/D2 영역 염기서열을 확인, 비교하여 동정하였다. 울릉도 야생화들에서는 22종 48균주, 욕지도에서 야생화들로부터는 25종 60균주 효모들을 분리, 동정하였다. 두 섬에서 분리한 효모들 중 Cryptococcus albidus, Cryptococcus laurentii, Metschnikowia reukafii, Pichia scolyti, Rhodotorula glutinis, Rhodotorula graminis and Rhodotorula mucilaginosa 등 7종이 공통으로 분리되었고 나머지 33종이 두 섬에서 특이적으로 분리되었다.

  • Large-scale resolution of epoxides by the yeast Rhodotorula glutinis was demonstrated in an aqueous/organic two-phase cascade membrane bioreactor. Due to the chemical instability and low solubility of epoxides in aqueous phases, an organic solvent was introduced into the reaction mixture in order to enhance resolution of epoxide. A cascade hollow-fiber membrane bioreactor was used (i) to minimize the toxicity of organic solvents towards the epoxide hydrolase of Rhodotorula glutinis, and (ii) to remove inhibitory amounts of formed diol from the yeast cell containing aqueous phase. Dodecane was selected as a suitable solvent and 1,2-epoxyhexane as a model substrate. By use of this membrane bioreactor, highly concentrated (0.9 M in dodecane) enantiopure (>98% ee) (S)-1,2-epoxyhexane (6.5 g, 30% yield) was obtained from its racemic mixture.

  • The development of hollow fiber reactor system for the production of chiral 1,2-epoxy-7-octence by epoxide hydrolase for Rhodotorula glutinis was investigated. Dodecane with high solubility of the racemic substrate passed through the lumen side of the hollow fiber reactor and cell suspension was recirculated through the shell side. The 2nd hollow fiber reactor was coupled to the production reactor to extract the diol byproduct which was the inhibitor of epoxide hydrolase. Optically pure (S)-1,2-epoxy-7-octene (0.6 M in dodecane) could be obtained using hollow-fiber reactor system.

한국 목포 연해안해수내의 효모 분포 및 집단밀도와 해조류상의 효모상 (The Distribution and Population Densities of Yeasts and their Epiflora on Seaweeds in Inshore Waters of Mok-po, Korea)

  • The distribution and population densities of yeasts including their epiflora on seaweeds were estimated over two months in inshore waters of Mok-po, Korea. Nine species of 48 isolates were obtained from this area. Rhodotorula, Torulopsis and Debaryomyces that are widespread in estuaries were of common occurrence in this water body. The highest counts belonged to Torulopsis candida which was predominated in temperate estuarine zone. The distribution of sea weeds seems to be correlated with the population densities of yeasts. Two species of marine algae harbored yeasts during May and July, 1975. Among seaweeds isolates, the highest numbers werre of the species Rhodotorula glutinis var. glutinis. A dominant colonization of this strain on Chlorophyceae and Phaeophyta and a role of water temperature for the growth of yeasts are discussed.

인삼과 당귀 재배 토양으로부터 야생효모들의 분리 및 미백성 Tyrosinase 저해활성 (Isolation and Tyrosinase Inhibitory Activity of Wild Yeasts Obtained from Soil in the Fields of Medicinal Plants, Ginseng and Korean Angelica)

  • 약용식물 재배지 토양의 야생효모 분포특성을 조사하고 이들 야생효모들의 산업적 응용성을 위한 자료를 얻고자 먼저 약용식물 집단 재배지인 충남 금산의 인삼과 당귀 재배지의 토양들을 2018년 3월에 50점을 채취하여 야생효모들을 분리, 동정하였다. 이들 시료들로부터 21종 43균주의 야생효모들이 분리되었고 인삼재배 논 토양에서는 Rhodotorula glutinis 균주가 6주, 당귀 재배 밭 토양에서는 Cyberlindnera saturnus 균주가 역시 6균주로 가장 많이 분리되었다. 이들 야생효모들을 이용하여 미백성 기능성 화장품 소재를 개발하고자 이들의 무세포추출물을 제조하여 tyrosinase 저해활성을 측정한 결과 저온성 야생효모인 Naganishia globosa G1-7가 28%의 가장 높은 저해활성을 보여 우수효모로 선발하였다.

발효차로부터 효모의 분리 및 동정 (Isolation and Identification of Yeast Strain from Fermented Tea)

  • 발효차로부터 분리 동정한 JY-1 효모는 18S rDNA 유전자 분석에서는 Rhodosporidium azoricum과의 유의성 99%, 생화학적 분석 VITEK system에서는 Rhodotorula glutinis/Rhodotorula mucilaginosa와 93%의 유의성을 나타내였다. 배양조건에서 최적의 온도는 $25-30^{\circ}C$, pH는 5.0이었으며 염의 농도는 2%, 최적의 에탄올 농도는 4%이었다. 일반적인 실험결과에서도 Rhodosporidium 속의 특이성과 비교하여 보았을 때 형태적, 생리적, 생화학적 유사성을 나타내었다. 이상의 결과에서 앞으로의 연구과제는 많이 음용되고 있는 발효차와 관련한 또 다른 미생물군을 분리 동정하여 우수한 균주를 확보하는데 있으며 이를 이용한 발효차 음료산업 개발에 목적을 두고 있다.

Yeast Diversity of Wild Flowers in Islands, Jejudo, Ulleungdo and Yokjido of Korea

  • We isolated various yeasts from wild flowers in main islands, Jejudo, Ulleungdo and Yokjido of Korea and their yeasts were identified by comparison of their PCR-amplified D1/D2 regions of 26S rDNA using the BLAST database. Thirty two yeast strains of fourteen species were isolated from wild flowers of Jejudo. Forty eight yeast strains of twenty two species were isolated and identified from wild flowers of Ulleungdo, Korea. Sixty yeast strains belonged to twenty five species were isolated identified from wild flowers of Yokjido in Tongyeong, Korea. Only Metschnikowia reukaufii was overlapped from the three different islands areas. Two species overlapped from Jejudo and Ulleungdo: Pichia guilliermondii, Metschnikowia reukaufii. Seven species were overlapped from Ulleungdo and Yokjido: Cryptococcus albidus, Cryptococcus laurentii, Metschnikowia reukafii, Pichia scolyti, Rhodotorula glutinis, Rhodotorula graminis and Rhodotorula mucilaginosa. Four species were overlapped from Jejudo and Yokjido: Candida sp. Cryptococcus aureus, Metschnikowia reukafii and Pseudozyma sp.

미생물에 의한 Glutathione 생산에 관한 연구 (제 1보) 생산균주의 선정 및 배양 (Studies on the production of Glutathione by Microorganism)

  • 효모의 균체생산에 관한 연구중 glutathione 함량이 다량 축적되는 효모를 검색한 결과, Rhodotorula glutinis 균주가 높은 균체량과 g1utathione 을 함육하였다. 이 균주에 대한 배양시간에 따른 glutathione의 함량증가를 검토한 결과 pH 6.0, 3$0^{\circ}C$에서 왕복진탕배양으로 48시간 배양하였을 때 가장 좋은 결과를 보였다. Glutathione 함량 증가를 보기 위해 기본배지에 아미노산을 첨가하여 glutathione 함량을 측정한 결과, 명아미노산의 최적농도는 0.7%, 최적 시기는 배양 후 36시간째 였다. Glutathiohe의 구성아미노산인 glutamic acid, cysteine, glycine을 조합하여 첨가할 때 219$\mu\textrm{g}$/ml으로 무첨가 배지의 73$\mu\textrm{g}$/ml에 비해 약 3배의 증가를 보였다.

Property Characterization and Lipid - Compositional Analysis of Lipid Granules Isolated from an Oleaginous Yeast Rhodotorula glutinis

  • Preparative isolation of lipid granules from Fhodotorula glutinis, which has been studied for long time to produce edible lipids, was carried out by flotation method in Ficoll-Linear density gradient. When the isolated lipid granules were suspended in a series of solutions containing varying concentration of osmotic stabilizer (sorbitoal and mannitol) ranging from 0.8M to 0M, the lipid granules appeared to be disrupted at a concentration between 0.8M and 0.7, and again at a concentration below 0.1M, suggesting that lipid granules have a membraneous structure and that at least two types of lipid granules are present. Compositional analysis of lipids from lipid granules revealed that lipids are composed mainly of neutral lipids (87.8% of total lipids), predominantly as triacylglycerols (71.89%). Marked differences were observed inphospholipids between lipids of lipid granules and those of whole cells . The major components of phospholipids in lipid granules and inwhole cells are phosphatidylcholine(38.6%) and phosphatidylserine(42.8%), respectively. In addition, significant differences were also observed in the fatty acid composition of phospholipids. As phospholipids are important structural components of membranes, these differences lead to the suggesting that the membrane of lipid granules may be distinct functionally and structurally from other membranes of yeast cells. The major fatty acid components of neutral lipidss of whole cells and lipid granules are palmitic , oleic and linoleic acid. However , degreeof fatty acid unsaturation of neutal lipids of lipid granules was much lower than that of neutral lipids of whole cells.

Phylogenetic Analysis of Genus Sporobolomyces Based on Partial Sequences of 26S rDNA

  • The sequences of the D1/D2 region of 26S rDNA from seven Sporobolomyces species, Bensingtonia subrosea, and Rhodosporicium toruloides were determined and compared with those from representatives of the genera Leucosporidium, Rhodosporidium, Rhodotorula, and Sporidiobolus. The five species of Sporobolomyces analyzed were distantly related to a monophyletic clade consisting of species of Sporidiobolaceae and Sporobolomycetaceae. Sporobolomyces falcatus was found to be closely related to Tremella exigua. The members of Sporidiobolaceae and Sporobolomycetaceae were divided into four groups. Group 1 was composed of Leucosporidium scottii and two Rhodotorula species, and group 2 contained three Rhodotorula species. Group 3 was designeate as the Sporobolomyces/Sporidiobolus core group, as it contained Sporidiobolus johnsonii, the type species of Sporidiobolus and the teleomorphic state of Sporobolomyces salmonicolor (the type species of Sporobolomyces). Group 4, named the Rhodotorula/Rhodosporidium core group, included Rhodosporidium toruloides and Rhodotorula glutinis, the type species of the genera Fhodosporidium and Rhodotorula, respectively. The four groups were differentiated on the basis of their physiological characteristics including the assimilation of D-glucosamine, glucuronate, 2-keto-gluconate, L-arabinitol, raffinose, methyl-$\alpha$-glucoside, and satrch. The taxonomy of the genera Leucosporidium, Rhodosporidium, Rhodotorula, Sporidiobolus, and Sporobolomyces will require a major revision when more data becomes available.
