Production of Salidroside in Rhodiloa sachalinensis A. Bor Callus by the Elicitation and Precursor (참돌꽃 (Rhodiola sachalinensis A. Bor) 캘러스에서 elicitor와 전구체에 의한 Salidroside 생산성의 변화)

  • The effect of elicitor and precursor on salidroside production from Rhodiola sachalinensis A.Bor callus cultures was investigated. Callus cultures were treated with yeast extract, soft-ferrite ceramics powder, methyl jasmonate, ascorbic acid, jasmonic acid and $CuCl_2$/$CdCl_2$ as an elicitor. When callus cultures were treated with $0.2g/\ell$ of yeast extract, salidroside production from callus treated with yeast extract is 3.45 times higher than that of the controlled group. Among of them, callus cultures treated with yeast extract produced the highest salidroside. Callus cultures were treated with L-phenylalanine and L-tyrosine as a precursor for 4 days. The result of salidroside content analysis showed that all feeding of precursors not affected salidroside production from callus cultures. In case of L-tyrosine fed into callus cultures, both callus growth and salidroside production decreased at all concentrations.

  • The acetone extract of the roots of Rhodiola sachalinensis has furnished six phenolic compounds which exhibited significant scavenging effects against DPPH free radical. The structures of these compounds were identified and determined as gallic acid (1), (-)-epigallocatechin 3-O-gallate (2), kaempferol (3), kaempferol 7-O-$\alpha$ -L-rham nopyranoside (4), herbacetin 7-O-$\alpha$ -L-rhamnopyranoside, (5) and rhodiolinin (6) by physico-chemical and spectral evidences.

Effect of Macro-nutrients for the Salidroside Production from Callus Cultures of Rhodiola sachalinesis A. Bor (참돌꽃의 캘러스로부터 salidroside 생산에 미치는 배지성분의 영향)

  • Rhodiola sachalinensis A. Bor is a Chinese herb containing the natural compound salidroside, which has been known to possess medical properties such as enhancing bodies' ability to survive in adverse environments and extending human life. To improve a metabolite production from Rhodiola sachalinensis callus cultures, the concentrations of macro nutrients were investigated. To investigate the salidroside production in Rhodiola sachalinensis callus cultures, we analyzed salidroside content in each callus which was cultured in 30 $m{\ell}$ of $2\;{\times}\;B_5$ liquid medium for 4 weeks. The optimal concentrations of macro nutrients for salidroside production ($67.96\;{\pm}\;3.41\;mg/{\ell}$) were found to be 99 mM $KNO_3$, 1 mM $(NH_4)_2SO_4$, 2 mM $NaH_2PO_4{\cdot}2H_2O$, 2 mM $CaCl_2{\cdot}2H_2O$ and 1 mM $MgSO_4{\cdot}7H_2O$. From these results, we determined the more upgraded culture condition for industrial production of salidroside.

  • To enhance salidroside productivity from a cell suspension cultures of Rhodiola sachalinensis, various combinations of auxin (NAA) and cytokinins (BA, Kinetin) concentration and addition of $GA_3$, TDZ, zeatin, spermine and spermidine at a hormone combination to be established were examined. NAA/BA combination is superior to NAA/kinetin combination in biomass and salidroside content. Maximum salidroside production ($64.6{\pm}8.9\;mg/L$ medium) was obtained from $2B_5$ medium with 1 mg/L NAA and 5 mg/L BA. The adding of $GA_3$ ($0.01\;mg/L{\sim}5\;mg/L$) was beneficial for salidroside accumulation and the highest productivity of salidroside, $90.3{\pm}8.34\;mg/L$, was obtained from $2B_5$ medium supplemented with 1 mg/L NAA, 5 mg/L BA and 0.1 mg/L $GA_3$. On TDZ, zeatin, spermine and spermidine, expectant results were not obtained except a little affirmative effect of spermidine ($69{\pm}2.88$ at 1 mg/L).

  • Callus of Rhodiola sachalinensis A. Bor were induced from leaf explant on l/2MS solid supplemented with combination of auxin (2.4-D, NAA: 0.1∼2mg/L) and cytokinin (BA: 0.1∼0.2mg/L). The effects of various medium, culture conditions and phytohormones on the growth of callus were investigated. MS, WPM, B$_{5}$ medium and diluted or concentrated media (1/2X, 2X, 3X) were used to investigate the growth of callus on each media. Among these, the highest growth was observed when cultured in in 2B$_{5}$ medium containing 0.5mg/L NAA and 1mg/L BA, 3% sucrose and 49.6 mM KNO$_3$ as nitrogen source, and 2.16mM NaH$_2$PO$_4$ as phoshate source at $25^{\circ}C$ in the dark. The calli cultured with 5% sucrose produced high salidroside content (0.41% on the basis of dry wt) than normal root (0.17%).

Efficient Plant Regeneration from Shoot Tip and Young Leaf in Rhodiola sachalinensis A. Bor.

  • The shoot tip and young leaf of Rhodiola sachalinensis were cultured to invest the plant growth regulator condition for callus induction, shoot and root regeneration. When the shoot tip was sterilized in 2.0% of NaOCl for 20min., the contamination rate was the lowest. And the survival rate of the culture material was good in carbenicillin 500mg/L treatment group. Callus was obtained from shoot tip and young leaf segments. NAA 0.1-1.0mg/L and 2,4-D 0.1-0.5mg/L alone treatment were shown to have a good response on callus induction from shoot tip culture. In the case of young leaf culture, NAA and 2,4-D 0.1-0.5mg/L alone treatment were good in callus induction. In culturing shoot tip NAA 0.5mg/L and BA 0.5mg/L, NAA1.0mg/L and BA 0.lmg/L combination treatment was good in shoot regeneration. The regenerated shoots were rooted on MS medium supplemented with NAA and BA combination treatment. Especially, NAA 1.0mg/L and BA 0.1mg/L combination treatment was effective for root regeneration.

  • This study was performed to compare effect of immune activities of Rhodiola sachalinensis by various extraction process with different temperature and extraction solvents. Experiments were performed for investigate the immune activities on human B and T cell growth and secretion of their cytokines. Also, antibodies in serum were investigated in female ICR mouse by feeding the extracts of R. sachalinensis at doses of 40, 120 and 360 mg/kg orally for 15 days. The immune cell growth and secretion of cytokines (IL-6, TNF-$\alpha$) on human B and T cells were increased by adding R. sachalinensis extracts compare to the control. Also, total serum IgG levels increased by feeding R. sachalinensis extracts. It can be conclude that optimum condition for efficient extraction of R. sachalinensis as functional material is slovent extraction process using water with ultrasonification at below $100^{\circ}C$ than typical process.
