Studies on the Selection of Microorganism for Food Wastes and Optimization of Fermentation Process (음식물찌꺼기 소멸효율 재고를 위한 발효균 및 발효 공정 최적화 연구)

  • For the effective disposal of organic food wastes, we seleted 4 strains of microorganism from 186 microbial candidate via enzyme activity test, salt tolerance, food decomposition rate, stability and safety of strains. The identity of these 4 strains are as follows : Fungi is Rhizopus sp., yeasts are Galactomyces sp., Pichia sp. and Hyphopichia sp., In the 50L fermenter scale, we tested various fermenting factor for the optimization of conditions of food waste decomposition using 4 selected strains. The optimum fomenting conditions were as follows : BIO-CHIP Volume 25-30 L, BIO CHIP size 2.0-6.0mm, air flow 200-280L/min, mixing intensity 2-4rpm, temperature $30-45^{\circ}C$. In these fermenting conditions, the efficiency of decomposition(rate of weight loss of food wastes) were 93%. Also the quality of fermenting output were assayed at the basis of fertilizer, and the results were as good as general compost.

The Quantitative Change of Chitin as a Criterion to Indicate Fungal Invasion to Rice Storage (미곡저장중 Fungus 침해 판정에 관한 연구)

  • In order to find out a reliable chemical indicator which can be used to tell whether rice, during storage, is invaded by fungi or not, 90 percent milled rice (Tongil) was stored at 26$^{\circ}C$ for 4 weeks and weekly analyzed for changes in several chemical components. The results were as follows : 1) Of several genera of fungi (Aspergillus sp., Penicillium sp., and Rhizopus sp.) observed in stored rice after surface disinfection, Aspergillus sp. were the most frequent fungi. 2) Chitin content in the rice was 25 ug/g at the beginning of the experiment and increased proportionly with moisture content of the storedrice to become 1980 ug/g at 26.9% moisture after 4 weeks. 3) Non-reducing sugartended to decrease with increasing moisture content and or prolonged period. 4) Fat acidity showed a tendency of increase with rise in moisture content of the stared rice. Consequently, changes in chitin content can be mployed as a reliable measure to evaluate the quality of stored rice in connection with fungal invasion.

Taxonomy and Identification of Fungi Isolated from Round Bale Silage (원형 곤포사일리지에 발생한 곰팡이의 분류 동정)

  • To identification of fungi that occurs round bale silages, 253 fungal contaminated samples were collected from 2009 to 2011. Total 253 silage samples from Italian ryegrass, sudan grass, rye, corn, barley and oat were analysed. Total 270 strains were purely isolated from contaminated round bale silages. The fungi were identified with morphological characteristics and rDNA sequence analysis. Nineteen species of fungi(Rhizopus sp., Fusarium spp., Coprinus sp., Blastomyces sp., Aureobasidium sp., Polypaecilum sp., Botryoderma sp., Mucor sp., Scytalidium sp., Sphaeropsis sp., Aspergillus spp., Trichocladium sp., Humicola sp., Staphylotrichum sp., Periconia sp., Verticillium sp., Diplococcium sp., Penicillium spp. and Trichoderma spp.) were identified by morphological characteristics. On the other hand, fungi isolated from silage were identified to Acremonium strictum, Aspergillus tubingensis, Bionectria ochroleuca, Dipodascaceae sp., Fusarium proliferatum, Fusarium oxysporum, Fusrium solani, Gelasinospora reticulata, Gibberella moniliformis, Gibberella zeae, Nectria mauritiicola, Penicillium paneum, Pseudallecheria boydii, Schizophyllum commune, Scopulariopsis brevicaulis and Simplicillium lamellicola by rDNA sequence analysis. Penicillium sp. and Trichoderma sp., were isolated 74 and 64 strains, respectively. Humicola sp., Aspergillus sp., Coprinus sp., and Fusarium spp. were identified 10 to 30 strains. Most fungi were isolated together with more than one species in a sample looked like one species with the naked eyes.

Screening of Functional Rhizopus stolonifer for Alcohol Fermentation and Production of High Quality Korean Traditional Rice Wine

  • Different strains of mold were screened for the production of high quality Korean traditional rice wine with anti-hypertension and good acceptability. We isolated 867 nuruk mold strains and selected 24 for further study based on measurement of amylase activity. Among them, mold No. 17 showed high ethanol production upon fermentation with Saccharomyces cerevisiae as well as anti-hypertensive properties. The No. 17 strain was therefore selected as the functional mold and later identified as Rhizopus stolonifer based on molecular biological characteristics. Optimal fermentation conditions for the brewing of anti-hypertensive traditional rice wine comprised the addition of R. stolonifer No. 17 koji at a concentration of 35 sp/g and a fermentation period of 10 days at $25^{\circ}C$ using S. cerevisiae.

Mycelial Production and Amylase Activity of Fungi for Brewing in Different Submerged Culture Conditions (액체배양에서의 양조용 곰팡이의 균체 생산 및 전분분해효소 활성)

  • In this study, twelve strains of brewing fungi were individually cultivated on wheat extract broth (WEB), potato dextrose broth (PDB) and malt extract broth (MEB) in order to determine the microorganism with good culture characteristics as well as with amylolytic activity. The strain cultured in PDB exhibited mycelia production from 12.6 g/L (Rhizopus oryzae KACC 45714) to a maximum of 48.0 g/L (Aspergillus oryzae KACC 46959), which was 2.3~9.2 times lower than that of the strain cultured in WEB and 1.7~14.6 times lower than that of the strain cultured in MEB. Accorfing the results, We found that the commercial strains of A. oryzae Suwon, CF1001 and CF1003 had a higher dry cell mass than the wild-type strains KACC 46421, 46423, 46424 and 46959. For Rhizopus sp., the acidity levels in WEB, PDB and MEB were 0.12~0.47%, 0.22~1.0% and 0.16~0.68% (equivalent lactic acid concentration) respectively. For A. oryzae, the acidity levels were 0.06~0.11%, 0.03~0.04% and 0.06~0.08% (equivalent lactic acid concentration), respectively. Amylase enzyme from Rhizopus delemar KACC 46419 exhibited an enzyme activity of 0.013 U and 0.019 U in WEB and MEB cultures, respectively. The enzyme activity of the amylase enzyme from A. oryzae was 0.019~0.037, 0.017~0.033 and 0.028~0.046 U in WEB, PDB and MEB cultures, respectively.

Manufacture of Koji Using fungi Isolation from Nuruk and Identification of Koji Molds (전통누룩 진균류를 이용한 입국의 제조 및 입국곰팡이의 동정)

  • Various koji were prepared by fungi isolated from traditional nuruk and their quality characteristics were investigated. Acidity and saccharification power of their koji were ranged in 5.0~6.8 and 128sp~241sp. Nine fungi which were showed good quality and sensory evaluation were identified by analysis of their nucleotide sequences with PCR-amplified 18S rDNA internal transcribed spacer-1(ITS-1) and ITS-4 genes. Among them, six strains were identified as Aspergillus oryzae and the other strains were identified as Mycocladus corymbiferus, Rhizo-pus oryzae, Lichtheimia corymbifera.

Takju Brewing of Uncooked Rice Starch Using Rhizopus Koji (Rhizopus Koji를 이용한 무증자 쌀탁주 양조)

  • The Takju brewing of raw rice starch was carried out by the simultaneous saccharification- fermentation using Rhizopus sp. koji and yeast, and compared with the Takju mash brewed by the conventional method. Rhitopus koji was prepared with uncooked rice for Takju brewing without cooking of rice starch. Alcohol concentration of Takju mash brewed with uncooked rice was slightly higher of 1.8% than that with cooked one. Amino acid contents was almost double and fuse1 oil contents was lower in uncooked brewing. The Takju mash prepared after fermentation without cooking of rice had a characteristic odor of raw material and a good quality of taste.

  • Rhizopus oryzae CJ-2114 was selected for its strong polygalacturonase activity among various strains of mold found in soil. It was found that the production of polygalacturonase reached to maximum when the wheat bran medium containing 1% albumin, 1% sorbitol and 0.2% (NH$_4$)$_2$C$_2$O$_4$was cultured for 96 hrs at 3$0^{\circ}C$. Polygalacturonase was purified 11.13 fold from Rhizopus oryzae CJ-2114. The purification procedures include ammonium sulfate treatment, gel filtration on Sephadex G-75, G-150 and DEAE-cellulose ion exchange chromatography. Yield of the enzyme purification was 40.3% .Purified enzyme was confirmed as a single band by the polyacrylamide gel electrophoresis. When the purified enzyme was applied to SDS-poly-acrylamide gel electrophoresis, the molecular weight was estimated to be 47,000. The amino acid composition indicated relatively high contents of glutamic acid and glyrine.

  • Among 12 kinds of ginsenosides in ginseng saponin, ginsenoside-Rb$_1$was contained the most abundantly. But ginsenoside-Rd which is similar to ginsenoside-Rb$_1$in structure, was known to be superior to ginsenoside-Rb$_1$pharmaceutically. In order to convert ginsenoside-Rb$_1$into ginsenoside-Rd by microbial enzyme treatment, a Rhizopus sp. was selected among various strais of molds found in rotten ginseng roots. Enzyme was prepared from the extract of wheat bran koji culture by ammonium sulfate precipitation (1.0 sat'd) and succeeding ammonium sulfate fractionation method (0.6-0.9 sat'd). For the purpose of use as substrate, saponins were purified by the several purification steps from alcohol extract of red ginseng roots. We obtained the total saponin which was composed of 36.5% of ginsenoside Rb$_1$, 12.2% of ginsenoside-Rd and other ginsenosides. For increase of ginsenoside-Rb$_1$ component ratio, we also obtained further purified ginsenoside-Rb group saponin containing 54.5% of ginsenoside-Rb$_1$, 1.1% of ginsenoside- Rd and other ginsenosides from purified the total saponin. In the enzymatic reaction system including the total saponin or the ginsenoside-Rb group saponin, we confirmed the specific conversion of ginsenoside-Rb$_1$to ginsenoside-Rd proportionally and no change of any other ginsenoside patterns by thin layer chromatography and high performance liquid chromatography.
