To screen for potential biocontrol agents against postharvest disease of garlics caused by Penicillium hirsutum, a total of 933 isolates (432 fungi and 501 bacteria) were isolated from the rhizoshere or rhizoplane of garlics. Among them, Pantoea agglomerans isolate 59-4 (Pa 59-4) was selected for a potential biocontrol agent by in vivo wounded garlic bulb assay, When the spore suspension (10$\^$5/ spores/$m\ell$) of Penicillium hirsutum was co-inoculated with spore or cell suspension of each fungal or bacterial isolate on wounded garlics, the isolate highly suppressed disease development. Soaking garlic bulbs in the suspension of Pa 59-4 significantly reduced garlic decay from p. hirsutum. However, Pa 59-4 did not inhibit the mycelial growth of P. hirsutum in dual-culture with P. hirsutum on Tryptic soy agar. In order to elucidate mode of action of Pa 59-4 nutrient competition between Pa 59-4 and P. hirsutum was investigated using tissue culture plates with cylinder inserts containing defusing membrane reported by Janisiewicz et al. The results showed that Pa 59-4 effectively suppressed spore germination and mycelial growth of blue mold in the low concentration (0.5%) of garlic juice, but did not suppress those of blue mold in the higher concentration (5%) of garlic juice. This result suggests that the mechanism in biocontrol of garlic blue mold by Pa 59-4 may involve in nutrient competition with P. hirsutum on garlic bulbs.
Number of bacterial isolates were collected from high mountainous areas at various locations in the whole country to select promising biocontrol agents. Most of selected isolates belonged to fluorescent pseudomonads. Population densities of fluorescent pseudomonads were examined by DLF method. Alll selected isolates rapidly multiplied on spermosphere after seed inoculation upto 24, and then the population abruptly declined. When seeds were germinated fully, bacteria moved to newly emerging radicle. The good root colonizing isolates, B16 and V13 proliferated on the growing root and moved down to the root tip and lateral roots. but the poor root colonizing isolates, MC07 and X01 moved much slower. Scanning electron microscopic observations showed that the cells of the good colonizing isolates were arranged linearly on the growing root and parallel to growing root axis and continuously existed on the root tip, Whereas the cells of poorly colonizing isolates were gathered and scattered randomly on the root surface.
Microorganisms that live in the rhizosphere play a pivotal role in the functioning and maintenance of soil ecosystems. The study of rhizospheric cyanobacteria has been hampered by the difficulty to culture and maintain them in the laboratory. The present work investigated the production of the plant hormone indole-3-acetic acid (IAA) and the potential of biofilm formation on the rhizoplane of pea plants by two cyanobacterial strains, isolated from rice rhizosphere. The unicellular cyanobacteria Chroococcidiopsis sp. MMG-5 and Synechocystis sp. MMG-8 that were isolated from a rice rhizosphere, were investigated. Production of IAA by Chroococcidiopsis sp. MMG-5 and Synechocystis sp. MMG-8 was measured under experimental conditions (pH and light). The bioactivity of the cyanobacterial auxin was demonstrated through the alteration of the rooting pattern of Pisum sativum seedlings. The increase in the concentration of L-tryptophan and the time that this amino acid was present in the medium resulted in a significant enhancement of the synthesis of IAA (r > 0.900 at p = 0.01). There was also a significant correlation between the concentration of IAA in the supernatant of the cyanobacteria cultures and the root length and number of the pea seedlings. Observations made by confocal laser scanning microscopy revealed the presence of cyanobacteria on the surface of the roots and also provided evidence for the penetration of the cyanobacteria in the endorhizosphere. We show that the synthesis of IAA by Chroococcidiopsis sp. MMG-5 and Synechocystis sp. MMG-8 occurs under different environmental conditions and that the auxin is important for the development of the seedling roots and for establishing an intimate symbiosis between cyanobacteria and host plants.
Cyperus rotundus L. is a perennial herb that was found to be dominating an area in northeast Brazil previously contaminated with petroleum. In order to increase our knowledge of microorganism-plant interactions in phytoremediation, the bacterial community present in the rhizosphere and roots of C. rotundus was evaluated by culture-dependent and molecular approaches. PCR-DGGE analysis based on the 16S rRNA gene showed that the bacterial community in bulk soil, rhizosphere, and root samples had a high degree of similarity. A complex population of alkane-utilizing bacteria and a variable nitrogen-fixing population were observed via PCR-DGGE analysis of alkB and nifH genes, respectively. In addition, two clone libraries were generated from alkB fragments obtained by PCR of bulk and rhizosphere soil DNA samples. Statistical analyses of these libraries showed that the compositions of their respective populations were different in terms of alkB gene sequences. Using culturedependent techniques, 209 bacterial strains were isolated from the rhizosphere and rhizoplane/roots of C. rotundus. Dot-blot analysis showed that 17 strains contained both alkB and nifH gene sequences. Partial 16S rRNA gene sequencing revealed that these strains are affiliated with the genera Bosea, Cupriavidus, Enterobacter, Gordonia, Mycoplana, Pandoraea, Pseudomonas, Rhizobium, and Rhodococcus. These isolates can be considered to have great potential for the phytoremediation of soil with C. rotundus in this tropical soil area.
This experiment was carried out to investigate the distribution of associative nitrogen fixers, Azospirillum spp. and their nitrogenase activities measured by ARA in the rhizosphers of rice, soybean, and weed grown in the rice paddy field at ear formation stage of rice. Nitrogenase activities produced by Azospirillum spp. enriched from histophere ranged from 16 to 53 n mole/tube/hr.. High nitrogen fixing activities more than 30 n mole/tube/hr. were observed in the histophere of the Echinochloa crus-galli L., Finbristylis miliacea L., and Monochoria vaginalis var.. Nitrogen fixing activities of Azospirillum spp. obtained from single colonies which originated from the rhizoplane of rice (pot-kwang var.), Finbris tylis miliacea L., Monochoria vaginaliz var., Glycine max L. were higher over 100 n mole/tube/hr. than those histophere. Genus of Azopsirillum isoltated from roots of the Graminease (Oriza sativa L., $C_3$-plant, Echinochloa crus=galli L., $C_4$-plant, Cyperus difforuis L.. $C_4$-plant), and Aeschynomene indica L. (Leguminosae, $C_3$-plant) was identified as A. brasilense. However, both strains, A. lipoferum and A. brasilense ($nir^-$ or $nir^+$ strain) were isolated from other plant roots, Both $nir^-$ and $nir^+$ strains of A. brasilense were associated with the same host plant.
A series of field and laboratory experimentes were conducted to find out the effects of kinds of crops residues and the different amount of fertilizer to the microbial interaction, chemical properties, plant growth and their interaction under continous cultivation of hot pepper, soybean, sesame and upland rice. The results obtained were summerized follows : 1. Total number of bacteria and actinomycetes were enhanced by cultivation of upland rice and soybean while no appreciable effects were obtained by the cultivation of hot pepper and sesame. 2. The number of bacteria, actinomycetes and fungi were increased by return of crop residues when the cultivation of hot pepper, soybean, sesame and upland rice. Specially, actinomycetes was remarkably increased by upland rice cultivation. 3. Increased amount of fertilizer were remarkably affected to decrease of number of soil microorganisms. Specially, actinomycetes succession was appearently affects while plant growing time. 4. The number of identified soil bacterial species were obtained high in order of Bacillus sp.>Rhizobium sp.>Agrobacterium sp.>Pseudomonas sp. The number of Gram positive bacteria were superior than that of Gram negative bateria. 5. Interaction between microbial succession and crops cultivation, the number of Bacillus sp. was increased in hot pepper, Rhizobium sp. was in soybean, and sesame, and Agrobacterium sp. were increased in soybean, respectively. 6. Survival and habitate of soil microorganisms were differ with kinds of crop, application of residue and fertilizers. Most high number of Bacillus sp. Rhizobium sp. and Pseudomonas sp. were obtained on the rhizoplane and rhizosphere while Agrobacterium sp. and Fusarium sp. were high in rhizosphere. 7. Factors in relation to change of soil microbial population was obtained high in order of climates>crops>organic>matter>fertilizer.
Lesion enlargement of ginger rhizome rot was most rapid at 35~40 C, but delayed greatly as temperature decreased. Time needed for a killing a ginger plant, 22~25 cm long, was about 5 days at 35~40 C, but was 15 days at 15 C in a growth chamber test. Higher RH above 90%, higher soil moisture level above 80% of maximum soil moisture capacity, and deeper planting below 4cm enhanced the lesion development on ginger stems and rhizomes. Pythium myriotylum existed in field soil as forms of hyphal portion, hyphal swelling body, or oospore- or zoospore-like bodies, and served as the origin of its colonization. Inocula of P. myriotylum was randomly distributed in soil surface around ginger plants, but its density was decreased as increasing soil depth with the highest density at 0~10 cm soil depth. Population density of P. myriotylum did not vary significantly between the rhizoplane and the rhizosphere soil of a ginger plant, but differed greatly between the disessed and healthy plants with several to several hundreds times higher population in the diseased plants. A positive curvilinear relationship was found between P. myriotylum density and ginger rhizome rot severity.

In order to obtain promising rice growth-promoting microbial strains that can be used as substitutes for chemical fertilizers, 172 bacterial strains were isolated from rice roots grown in Korean and Russian soils. Out of them, the strains KR076, KR083, KR181 and RRj228 showed plant growth-promoting activities on maize seedlings. Bacillus megaterium KR076 and Bacillus sp. KR083 showed both nitrogen-fixing and plant growth-promoting activities, while Rhizobium sp. KR181 and Pseudomonas sp. RRj228 appeared to support only plant growth-promotion, but not $N_2$ fixation. Especially, RRj228 showed high growth promoting activity at low concentrations. Inoculation studies with KR083 and RRj228 revealed a high affinity to the Japonica rice variety such as Junambyeo than the Korean Tongil type variety such as Arumbyeo. Both KR083 and RRj228 strains showed rhizoplane and/or endophytic colonization in Japonica and Tongil types rice when soaked with the bacterial suspension of $1.1{\times}10^5cfu\;ml^{-1}$ for six and twelve hours. However, the total bacterial cell numbers were higher in the roots of Japonica variety than in the Tongil type. In inoculation trials with Daesanbyeo rice variety, the seedlings inoculated with KR181 and RRj228 at the rate of $2.0{\times}10^6cfu\;ml^{-1}$ showed yield increment of 35% and 33% (p < 0.01), respectively, so that they contributed to the replacement of chemical fertilizer at half doses of N, $P_2O_5$, and $K_2O$ in pots. In Junambyeo rice seedlings, the strain RRj228, when inoculated with a cell suspension of $1.8{\times}10^6cfu\;ml^{-1}$, promoted 3.4% higher yield at 70% dose than at a full dose level of N $110kg\;ha^{-1}$ in field. These results suggest that the rhizobacteria KR181 and RRj228 are prospective strains for enhancing rice performance.
The study was performed to investigate the property of rhizosphere microorganisms, and community structure during GMO, and Non-GMO rice cultivation. In the dilution plate technique, there were no significant differences in microbial populations of rhizosplane with genetically modified, and non-genetically modified rice cultivation, and rhizosphere were also the same results. Dominant bacterial genera were Afipia 12.5%, Spingomonas 10.0%, Ramlibacter 10.0%, Mycobacterium 7.5%, and Tetrasphaera 7.5% in rhizosphere soil of genetically modified rice plant, while Afipia 7.3%, Spingomonas 12.2%, Ramlibacter 7.3%, Mycobacterium 17.1%, Tetrasphaera 14.6% in non-genetically modified cultivated at Suwon test fields in 2006. Majorgenera isolated from root surface cultivated in Yesan fields were Arthrobacter 12.7% in rhizoplane of genetically modified plant, and Burkholderia 22.2% of non-genetically modified plant in 2007, Paucimonas 26.6% of genetically modified plant, Chryseobacterium 15.4% of non-genetically modified plant in 2008. Also the microbial communities in rhizosphere soils of genetically modified, and non-genetically modified plants were characterized using phospholipid fatty acid, and denaturing gradient gel electrophoresis. The phospholipid fatty acid profiles of soils in this condition showed different pattern, but did not show significant differences between soils cultivated with genetically or non-genetically modified rice plants.
Among the fluorescent pseudomonad isolates from soil- root system of red pepper in Chinju, Kyunsangnam-Do, the phylogenetic analysis for 35 isolates were conducted. The partial 16S ribosomal DNA sequences were used as taxonomic key for phylogenetic analyses, and these sequences were enabled to identification of the fluorescent pseudomonad isolates on the species level. The 17 isolates among them were classified into Pseudomonas putida group, and consisted of the strains isolated mainly from soil. This group were subdivided into 4 subgroups (I, II, III, and IV). The subgroup I and IV were unique ones which were relatively remotely related with subgroup II and III including the type strain of P. putida. The 15 isolates among 35 isolates were grouped along with the type strain of Pseudomonas fluorescens, and 3 isolate were characterized as intermediates of P. fluorescens and Pseudomonas chlororaphis. Most of strain isolateds from the rhizosphere soil and rhizoplane of red pepper were identified as P. fluorescens and closely related with each other. In this study, root of red pepper was supposed to be colonized by a specific strain or strains of P. fluorescens.
