The present study was designed to clarify the antidiabetic activity and mechanism of Dioscorea rhizome in diabetic db/db mice. Mice were administered Dioscorea rhizome and rosiglitazone orally for 7 weeks and the effects of these compounds on fasting blood glucose, glucose tolerance and intestinal disaccharidase activity in db/db mice were evaluated. The fasting serum glucose of the D. rhizome treated group was reduced when compared with that of the db/db control group. In addition, the disaccharidase activities in homogenates of the proximal, middle and distal segment of the small intestine were significantly decreased response to D. rhizome treatment, especially in the middle segment. These results suggest that D. rhizome decreases blood glucose via a decrease in the activity of disaccharidase in the mucosa of the middle region of the small intestine in db/db mice.
This experiment was conducted to evaluate the contents of allylisothiocyanate and the change of hardness of the rhizome part of Wasabia japonica Matsum. by months after planting in water cultivation. No difference was found in plant height and leaf length at 10th and 15th months after planting, respectively. The rhizome part of wasabi was cut half and the dry weight was weighed. Dry weight was obtained as 17.3% and 21.6% from fresh lower and upper part of rhizome at 10th month after planting. At 15th month after planting, dry weight was obtained as 21.2% and 22.3% from fresh lower and upper part of rhizome. The contents of allylisothiocyanate were 0.047 mg/g and 0.633 mg/g at 10th and 15th month, respectively.
Wasabis (Wasabia japonica Matsum.) have been used as a spice with its petioles and rhizomes, but the production of its rhizomes in upland cultivation was very difficult for the severe damage of rotting disease during summer season. This experiment was carried out to increase the yield of marketable rhizome in wasabi when cultured in upland. Seedlings were raised in sub-alpine area for 7 and 12 months and then transplanted to experimental area on Sept. 20 and the yield was investigated on May 12 of the next year. The marketable rhizome(above 40g/plant) was produced in seedlings raised for 7 months. Production rate of marketable rhizome was 13.3% and total yield was 72.9 kg/10a. The content of allylisothiocyanate in rhizome of upland wasabi was 0.777 mg/g and its content was equal to wasabi cultured in water condition for 18 months. These results suggested that the upland cultivation of wasabi seedlings raised in sub-alpine area for 7 months was possible to product marketable rhizome.
Process of particle bombardment for efficient transformation of Cymbidium virescence rhizome microcross sections was investigated using Biolistic particle delivery system with pBI121 harboring the ${\beta}-glucuronidase$(GUS) and the neomycin phosphotransferaseII(nptII). The best result was obtained from the combination of $1.11{\;}{\mu}m$ tungsten particles coated with pBl121, $77.33kg/cm^2$ helium pressure, 6.35 mm gap distance, and 7.0 cm target distance. Transient expression of the reporter gene, GUS, bombarded into the rhizome microsections was observed by the histochemical assay. The marker gene, nptII, delivered by bombarding the tungsten particles coated with the plasmid DNA was identified in the transformed rhizome by polymerase chain reaction.
Background: The root and rhizome are historically and officially utilized medicinal parts of Panax notoginseng (PN) (Burk.) F. H. Chen, which in raw and steamed forms are used differently in practice. Methods: To investigate the differences in chemical composition and bioactivities of PN root and rhizome between raw and steamed forms, high-performance liquid chromatography analyses and pharmacologic effects evaluated by tests of anticoagulation, antioxidation, hemostasis, antiinflammation, and hematopoiesis were combined. Results: With the duration of steaming time, the contents of ginsenosides $Rg_1$, Re, $Rb_1$, Rd, and notoginsenoside $R_1$ in PN were decreased, while those of ginsenosides $Rh_1$, $20(S)-Rg_3$, $20(R)-Rg_3$, $Rh_4$, and $Rk_3$ were increased gradually. Raw PN samples steamed for 6 h at $120^{\circ}C$ with stable levels of most constituents were used for the subsequent study of bioeffects. Raw PN showed better hemostasis, anticoagulation, and antiinflammation effects, while steamed PN exhibited stronger antioxidation and hematopoiesis activities. For different parts of PN, contents of saponins in PN rhizome were generally higher than those in the root, which could be related to the stronger bioactivities of rhizome compared with the same form of PN root. Conclusion: This study provides basic information about the chemical and bioactive comparison of PN root and rhizome in both raw and steamed forms, indicating that the change of saponins may have a key role in different properties of raw and steamed PN.
Objectives : Kaempferia parviflora Rhizome is black ginger indigenous to Laos and Thailand. It has been used as a folk medicine to improve blood flow and promote vitality and longevity with good health and well being. For these reasons, Kaempferia parviflora Rhizome has been focused on developing it as a food or food supplement. In addition, Kaempferia parviflora Rhizome could be under consideration of new prescription based on its characteristic compounds, polymethoxyflavonoids. However, it needs to be certified as safe before it can be used. Here, a single-oral toxicity test and safety classification was carried out to identity acute information of the toxicity of Kaempferia parviflora Rhizome powder and to make sure of its safety in clinical applications. Methods : Test substance was orally administered to male and female SD-rat at dose levels of 5000 mg/kg to estimate approximate lethal dose(ALD). Based on the acute information of the toxicity, the safety classification was estimated using the HED(human equivalent dose)-based MOS(margin of safety). Results : At 14 days after treatment with test substance. there were no of test substance related with mortalities and clinical signs. In addition, no changes in the body or organ weights and no gross or histopathological findings were observed. Thus, the ALD of Kaempferia parviflora Rhizome powder was considered over 5,000 mg/kg in both female and male mice. Conclusions : Based on the single oral toxicity test using the highest and limit dose, 5,000 mg/kg and the decision guideline for safety classification based on HED-based MOS, it was estimated that Kaempferia parviflora Rhizome powder is classified as "Specified class B" indicating that clinical dose is not limited to patients as safe as food.
The aim of the present study was to evaluate the chemical composition and antioxidative activity of rhizome essential oil of Kaempferia parviflora Wall. ex Baker. The essential oil extracted by hydrodistillation was chemically profiled by GC/MS analysis. The antioxidative activity was determined and evaluated spectroscopically by the 2,2-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays. According to the results, the major essential oil components were camphene (18.03%), β-pinene (14.25%), a-pinene (12.38%), endo-borneol (10.23%), β-copaene (8.38%), and linalool (8.20%). K. parviflora rhizome oil possessed antioxidant potential, exhibiting DPPH and ABTS radical scavenging activities as high as 80.90 and 94.04%, respectively, at a concentration of 10 mg/mL. The corresponding IC50 values were 0.451±0.051 and 0.527±0.022 mg/mL, respectively (IC50 values for ascorbic acid, as the standard, were 0.209±0.016 and 0.245±0.022 mg/mL, respectively). The mycelium of F. oxysporum was distorted and collapsed when treated with 0.5 mg/mL of the EO of K. parviflora rhizome for 3 days treatment, which may provide an important information for exploring the metabolism of the fungicide K. parviflora rhizome and its derived compounds against F. oxysporum. This study provides the chemical properties of the essential oil of K. parviflora rhizome grown in Vietnam and their potential antioxidant and antifungal activities.
The clarify the ecological properties, and to predict change of understory vegetation of mt. Nam Park, population dynamics and interspecific competition of D. smilacinum and D. viridescens, which grow in understory of deciduous broad-leaved forest and pseudo-annuals, were studied from May 20 to May 30 1998. The depth of litter layer, soil moisture content, soil organic matter and soil texture were surveyed in 18 populations (15 D. smilacinum populations and 3 D. viridescens populations). Mean litter layer of d. smilacimum population was thinner than that of D. viridescens populations). Mean litter layer of D. smilacnum population was thinner than that of D. viridescens population. The contents of soil moisture and organic matter of D. smilacinum population were lower than that of D. viridescens population. The D. smilacinum growed in broad range of soil texture but D. viridescens in loamy soil. Because D. smilacinum could tolerate more broad range of soil moisture and soil texture than D. viridescens, the former covered the herb layer in earlier stage and the latter introduced in later stage when rhizome could grow easily. The numbers of individual in two marginal parts were smaller than that in center in same D. smilacinum patch. And the total numbers of individuals grown in (10 ${\times}$ 10)cm were from 0 to 12. The rhizome (subterranean runner) weight, rhizome length, root weight, shoot weight, lea weight and leaf number per subquadrat (cell) increased along the number of individual, that is, increased from marginal part to center. But rhizome weight and rhizome length per individual were vice versa. Therefore, the individuals in marginal part reproduced longer and stronger asexual propagules than that in center. The distribution pattern of D. smilacinum was contageous and that of D. viridescens was random or regular. Therefore, population growth of former was independent on density and that of latter was dependent on density. The distributions of size-class showed normal curves in two population, but the curves based on data of total dry weight showed positive skewness and those of leaf number showed negative skewness The correlation coefficient (CC) values between the properties of each organ were high in two population and significant at 0.1% level. The CC values of D. viridescens were higher of the two. Therefore, the former allocated the energy to each organ stable. The rhizome depth of d. viridescens was 2 times deeper than that of D. smilacinum. And rhizome length and weight of D. viridescens were longer (2 times) or heavier (4 times) than those of D. smilacinum. The patch size of D. viridescens increased 60 cm per year and that of D. smilacinum 30 cm. On this results, the intrinsic increase velocity of d. viridescens patch was 2 times faster than that of d. smilacinum, therefore, on the competition, the former had an advantage over D. smilacinum. The reason why d. viridescens defeated D. smilacinum resulted from that the leaf area of former was 4 times broader than that of latter. in Mt. Nam Park, it was thought that two disporum Population would change with the 3 thpes of environmental change as followings. First, no human impact and increase of soil moisture content resulted in increase of D. viridescens population. Second, mild human impact and similar condition of soil moisture content resulted in slow increase or no changes of D. smilacinum and d. viridescens population. Third, severe human impact and dry condition resulted in decrease or vanishment of two disporum populations.

Studies were carried to evaluated the influence of storage method by temperatures and fillers on yield and quality of seed rhizome in turmeric. Seed rhizome was stored at styrofoam box filled with rice hull and sand (3:1) or vermiculite for 30, 60 and 90 days at different temperatures (5, 10 and $15^{\circ}C$. compared to traditional method (rhizome only). Parameters were obtained for weight loss, cold injury, percentage of decayed in stored rhizome during storage periods. Also, the germination, growth pattern and yield from stored rhizome has been investigated. It was confirmed that storage of turmeric in stored with filled with vermiculite helps in prevention of rhizomes from microbial and fungal attack. The storage of rhizomes in styrofoam box without any filler at low temperature below $10^{\circ}C$. is not advocated due to heavy losses weight and decayed in management of postharvest for turmeric rhizome. Germination percentage, growth pattern and yield was maximum for rhizomes stored at styrofoam box filled with vermiculite for 90 days at $15^{\circ}C$. The paper outlines a brief attempt to assess the efficacy of non-chemical methods including optimal storage method (temperature and filler) of control of decay and moisture losses during storage of turmeric.

Conventionally, the seeds of Rehmannia glutinosa (Gaertn.) Libosch. ex Steud. have been directly sown at the field without using the plug seedling method. Plug seedlings have the advantage of promoting germination and convenient transplanting. However, there is little information about propagation of R. glutinosa using the plug seedling method. This study was conducted to investigate the optimal seed rhizome length, diameter, and sowing direction of R. glutinosa for establishing the plug seedling method. Seed rhizome length and diameter were separated by 1, 2, 3 cm and 0.3-0.5, 0.6-1.0, 1.1-1.5 cm, respectively. And seed rhizomes were sown in vertical and horizontal directions. The survival rate in 1 cm length of seed rhizomes was lower than in other treatments. The leaf length, leaf width, number of leaves, SPAD, leaf area, and fresh and dry weights of shoot and roots were the greatest in 3 cm length of seed rhizomes. As the seed rhizome diameter decreased, the growth characteristics of R. glutinosa tended to increase. When R. glutinosa was sown horizontal direction, the leaf length, leaf width, number of leaves, and leaf area were significantly higher than in the vertical direction. In conclusion, when sowing seed rhizome in a plug tray, using a length of 3 cm, a diameter of less than 1 cm, and sowing in a horizontal direction is considered an appropriate sowing method for R. glutinosa.
