Effects of Sambutang and Consitituent Herbs on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (삼부탕(蔘附湯) 및 구성약물(構成藥物)이 백서(白鼠)의 국소뇌혈류량(局所腦血流量)과 혈압(血壓)에 미치는 영향(影響))

  • This experimental studies were done to investigate the effects of Sambutang(SBT) and consitituent herbs on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(BP) in rats. The results are as follows: SBT and consitituent herbs(SBT omitting Zingiberis Rhizoma Recens, Zingiberis Rhizoma Recens. Ginseng Radix and Aconiti iateralis preparata Radix) was increased rCBF significantly in a dose-dependent manner. SBT omitting Zingiberis Rhizoma Recens and Ginseng Radix were decreased significantly BP in rats. But, SBT, Aconiti iateralis preparata Radix and Zingiberis Rhizoma Recens were not affected BP in rats. According to the above findings. it is suggested that SBT causes a diverse response of rCBF, and specially the rCBF by increasing SBT is mediated action of Zingiberis Rhizoma Recens.

A Review on Two Fructus Jujubae & Three Rhizoma Zingiberis Rencns in Prescription of "Dongeuibogam" ("동의보감(東醫寶鑑)"처방(處方) 중(中) 강삼조이(薑三棗二)에 관한 고찰(考察))

  • Two Fructus Jujubae & three Rhizoma Zingiberis Rencns are usually used in parts of prescriptions in oriental medicine. In "Dongeuibogam", those are prescribed as drugs. However, those are not exactly same with original books, which "Dongeuibogam" had quoted. In original books, those are sometimes mentioned just as 'Fructus Jujubae & Rhizoma Zingiberis' or different amount of them. Therefore, authors try to compare with "Dongeuibogam" and its original books to find out the reason of discordance. As a result, we could conclude that Two jujubes & three pieces of ginger are more commonly prescribed than original books and are fixed to use more conveniently and to give standard for doctor.

  • On the quality control of commercial Zingiberis Rhizoma and its processed product. quantitative determination of 6-gingerol using HPLC method has been conducted. Quantitative analysis of 6-gingerol in Zingiberis Rhizoma showed average 0.359% in 14 samples collected throughout the regions of Korea. The contents of 6-gingerol in Zingiberis Rhizoma were decreased during the processing procedure(0.306%). (omitted)

  • Zingiberis Rhizoma and Dioscoreae Rhizoma were processed according to chinese pharmacopoeia and literatures. The content of 6-gingerol in processed Zingiberis Rhizoma was significantly increased than that of unprocessed one. The content of allantoin in unprocessed Dioscoreae rhizoma through HPLC was $0.32{\pm}0.06%$ and after processing was not significantly decreased to $0.23{\pm}0.12%$.

Angiogenic inhibitory effect of Zingiberis Rhizoma (건강(乾薑)의 혈관신생(血管新生) 억제효과(抑制效果)에 관한 연구(硏究))

  • This study was conducted to investigate angiogenic inhibitory effect of Zingiberis Rhizoma methanol extract using ECV-304 cells and HT1080 fibrosarcoma cells. The viability of ECV-304 was 30% at 50${\mu}g/m{\ell}$ of Zingiberis extract and that of HT1080 was 30% at 100${\mu}g/m{\ell}$. Using the BrdU incorporation assay, Zingiberis inhibited the DNA synthesis of ECV-304 and HT1080 by 70% and 50 % at 200${\mu}g/m{\ell}$. In tube formation assay, at 10${\mu}g/m{\ell}$ of Zingiberis, tube network began to degrade and at higher doses, it was completely destroyed. Zymography demonstrated that Zingiberis extract decreased MMP-9 at 10${\mu}g/m{\ell}$ and higher doses remarkably inhibited the expression of MMP-9. These data indicate that Zingiberis Rhizoma has angiogenic inhibitory effects and shows the possibility of future anti-metastatic drug.

Studies on the Germanium Components of Zingiberis Rhizoma (생강중 게르마늄 성분에 관한 연구)

  • Germanium (Ge) content in Zingiberis Rhizoma was determined by flameless atomic absorption spectrometry with a graphite tube atomizer. The Ge content of the cortical layer was 65.mu.g/g, the central cylinder, $98\mu$g/g and the total, $87\mu$g/g. In the effect of the extraction of Ge by various solvents, polar solvent was more effective than the nonpolar one, Especially, water was the most effective solvent. The water extract of Zingiberis Rhizoma, separated by sephadex G-25 gel filtration, was fractionated into 2 peaks at 254nm. Both the peaks showed atomic absorptions of Ge. It may be concluded that the Ge components of Zingiberis Rbizoma exist as the form of an aqueous organic compounds or associated forms with proteins.

EFFECT OF ZINGIBERIS RHIZOMA AND ASIASARI RADIX EXTRACTS ON THE JAW OPENING REFLEX PROVOKED BY NOXIOUS STIMULI AND THE PERIPHERAL NERVE CONDUCTION (유해자극에 의한 개구반사와 말초신경 흥분전도에 미치는 건간과 세신 추출물의 영향)

  • The aim of this study was to investigate the effect of herbal organic extracts on the pain response provoked by noxious stimuli on dental nerve and the peripheral nerve conduction. Cats (2-2.5Kg regardless of sex) that were chosen as experimental animals were classified into control group, Asiasari radix application group and Zingiberis rhizoma application group. They were anesthetized with ${\alpha}$-chloralose, then anterior belly of digastric muscle of both sides were exposed and wire electrodes were inserted for recording of Electromyogram (EMG). Cavities were prepared on canines until pulp of the teeth were exposed. And after the drugs solubilized for 2% and 4% concentration (W/V) in vehicle were applied, their effects were compared through the recording of EMG immediately after drug application, 30 minutes, 60 minutes, 120 minutes and 5 days after, respectively. And after both inferior alveolar nerves were exposed, 4% organic extracts of Zingiberis rhizoma and Asiasari radix were applied for 30 minutes then the change of jaw opening reflex provoked by noxious stimuli on pulpal nerves were observed immediately after washing out, at 30, 60 and 90 minutes after drug had been washed out. After saphenous nerve of both sides were exposed, one side of nerve was used for vehicle application and the other side was used for drug application for 30 minutes. Then conduction of action potential of A-${\delta}$ and C-fiders of saphenous nerves, which have changed with time, was recorded. With analysis of these records, the following results were obtained: 1. Organic extract of Zingiberis rhizoma (2% or 4% concentration) greatly suppressed EMG of digastric muscle provoked by noxious stimuli on pulpal nerve at five days after application, the suppressive: effect was greater than that of organic extract of Asiasari radix. 2. Organic extract of Asiasari radix (2% or 4% concentration) suppressed jaw opening reflex provoked by noxious stimuli on pulpal nerve, at 5 days after drug application. 3. Organic extract of Zingiberis rhizoma and Asiasari radix (immediately after 30 minutes application) suppressed neural conduction of A-${\delta}$ and C-fibers, the suppressive effect was greater on A-${\delta}$ fibers than on C-fibers. 4. Jaw opening reflex provoked by noxious stimuli on pulpal nerve in inferior alveolar nerve was greatly suppressed 30 minutes after drug application, this effect was greater by Zingiberis rhizoma than by Asiasari radix.

  • This study was designed to investigate the effects of Sagunja-tang Extract & Zingiberis rhizoma recens Juice (SEZJ) and Zingiberis rhizoma recens Juice (ZRJ) on the changes in regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats. The results were as follows ; SEZJ and ZRJ significantly increased rCBF in a dose-dependent manner, while it did not change MABP. This results suggest that SEZJ and ZRJ significantly increased rCBF by dilating pial arterial diameter. Increase of SEZJ-induced rCBF was significantly inhibited by pretreatment with methylene blue (10 ${\mu}g/kg$, i.p.), an inhibitor of guanylate cyclase, and indomethacin (1 mg/kg, i.p.), an inhibitor of cyclooxygenase. SEZJ-induced MABP was significantly increased by pretreatment with indomethacin but was not changed by methylene blue. These results suggested that the action of SEZJ was mediated by cyclic 3',5'-guanosine monophosphate.

Metabolism of Glycyrrhizin in Polyprescriptions Containing Glycyrrhizae Radix by Human Intestinal Bacteria and Their Inhibitory Effects on Some Enzymes (감초 함유 처방의 글리치리진 대사와 몇가지 효소저해효과)

  • To analyze scientifically the prescription principle of polyprescriptions (Gamchotang, Daewhanggamchotang, Jakyakgamchotang, Gamchogungangtang and Gilkyungtang) containing Glycyrrhizae Radix, the transforming rate of glycyrrhizin in these polyprescriptions to 18 ${\beta}-glycyrrhetinic$ acid and their inhibitory effect on ${\beta}-glucuronidase$, hyaluronidase, phosphodiesterase and trypsin were investigated. When Glycyrrhizae Radix containing polyprescriptions were extracted with water, the contents of glycyrrhizin in water extract of Glycyrrhizae Radix with Rhei Rhizoma or with Zingiberis Rhizoma were higher than that of Glycyrrhizae Radix only, but that in water extract of Glycyrrhizae Radix with Platicodi Radix was lower than that of Glycyrrhizae Radix only. By human intestinal bacteria, glycyrrhizin was metabolized to 18 ${\beta}-glycyrrhetinic$ acid. These metabolism of glycyrrhizin in polyprescriptions containing Glycyrrhizae Radix was inhibited by Rhei Rhizoma, Paeoniae Radix and Platicodi Radix, but was not affected by Zingiberis Rhizoma. The inhibitory activity of Glycyrrhizae Radix on hyaluronidase and ${\beta}-glucuronidase$, was synergistic with Rhei Rhizoma or Zingiberis Rhizoma, but was antagonistic by Platicodi Radix.

Antigastritic and Antiulcerative Action of the Extract of Zingiberis Rhizoma (생강추출물의 항위염 . 항궤양 작용)

  • The rhizoma of Zingiber officinale has been used as antiemetic, expectorants, stomachache relieving drugs and digestive accelerators. From the observation of antigastritic action of the methanol extract of the rhizoma, it was fractionated with hexane, chloroform, ethyl acetate and butanol, followed by bioassay on antigastritic and antiulcerative activity. The hexane and the chloroform fraction reduced significantly HCl ethanol induced gastric lesion at the dose of 370 and 210 mg/kg, p.o., respectively. On the gastric ulceration and gastric secretion in pylorus-ligated rats, the hexane fraction decreased the volume of gastric secretion and acid output, and also increased pH at the dose of 370 mg/kg, i.d.. It showed considerable curative ratio of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000 mg/kg, p.o. in mice. In conclusion, Zingiberis rhizoma exhibited antigastric and antiulcerative activity which might be attributable to inhibition of gastric secretion. It is revealed that the active component may be present in the hexane fraction.
