A series of experiments was planned for practical application of rhizobia in grass lands in Korea. This is the report for the studies mainly on the isolation and characterization of alfalfa nodule bacteria, and on the assessment of their nodulation abilities and nitrogen fixation capacities. 1. Total number of 47 strains was isolated from nodules which were taken from alfalfa grown in Daekwanyong, Cheju and other places. 2. Morphological and cultural characteristics of the strains were studied, and attempts. were also made to investigate their antigenic properties and to demonstrate lysogenic strains. The results were; i) the isolates varied in their cultural characteristics on yeast mannitol broth and agar, and in degree of congo red absorption; ii) similarities in their antigenic prorerties were found between the strains: SU 47/M-11, M-13/M-15, and M-3/M-5; iii) no lysogeny was found in the strains. 3. Plant infection test by test tube method in light room were carried out to elucidate the ability of the strains to nodulate Luna alfalfa and of the capacity of such nodules to fix atmospheric nitrogen. The isolates were grouped info non-invasive, ineffective, or effective to the legumes. Those strains which produced effective nodules, supporting similar/higher level of growth as nitrate control were: M-8, 9, 14, 20, 21, 25 and 34.
BACKGROUND: For sustainable agriculture, there are various agricultural practices including low input. Over the last few decades high input of chemical fertilizer and compounds results in environmental pollution and deterioration of soil fertility. Soybean (Glycine max L.) is well known eco-friendly crop due to their symbionts. Soybean has a relationship with nitrogen fixation bacteria called rhizobia. In this research work, we investigated effects of soybean cultivation on soil microorganism activities. METHODS AND RESULTS: Experiments were conducted in pots and potato cultivation was used as reference. Soil chemical properties were analyzed considering soil nutrient over cropping period. For the soil microbial community analysis, dehydrogenase activity analysis (DHA) analyzed along with denaturing gradient gel electrophoresis. The results showed that higher soil organic matter in the soybean cultivation soil than in the potato cultivation soil. Available $P_2O_5$ concentration increased gradually in both pots but showed higher value in the potato cultivation soil. DHA value implying microbial activities showed higher value in the soybean cultivation soil over all cropping period. CONCLUSION: The cause of high microbial activity in the soybean cultivation soil was considered to the effects of some specific microorganisms related to soybean cultivation. Therefore, the availability of soybean cultivation for sustainable agriculture should be encouraged in terms of microorganism community activity in soil.
Symbiotic effectiveness and significance of Bradyrhizobium japonicum strains in five serogroups which were consisted of three corresponding serotype strains, respectively, indigenous to Korean soils were evaluated in terms of utility of strain serogroup for symbiotic improvement on soybean plant. The nodulation by rhizobia of each serogroup on four soybean cultivars(Danweonkong, Kwangkyo, Pangsakong, and Eunhakong) was good in order of USDA 123 > YCK 150 > YCK 117 > YCK 141 > USDA 110 serogroup members. Shoot dry weight of soybean was relatively high with USDA 110 serogroup members as well as with YCK 141 serogroup members, whereas the effectiveness of USDA 123 serogroup members was the lowest among the serogroups examined. In particular, Pangsakong among soybeans inoculated with five-serogroup members was positively outstanding on nodulation and shoot dry weight of the plant. Overall, symbiotic parameters of serogroup members associated with soybean plant such as nodule number, nodule mass, $N_2$ase activity, and shoot dry weight showed significantly different responses at level of 1% probability among both rhizobial serogroups and soybean cultivars, respectively. The rate in symbiotic similarity of the members of each serogroup from F-test ($$P{\leq_-}0.05$$) was 100% for nodule No., 90% for $N_2$ase activity. and 80% for soybean shoot dry weight. Taken together, the results indicated that the serological grouping of B. japonicum could be strongly useful for improving the symbiotic effectiveness hetween soybean and Rhizobium.
Effects of paddy-upland rotation system on nutrients distribution in soil profile and nutrient balance were studied in paddy fields from 1989 to 1993. The obtained results are summarized as follows. 1. $NO_3{^-}-N$, Av.$P_2O_5$, Ex.-K, and EC were high by small extent in surface layer of 0~20cm soil depth without the sign of salt movement to deeper layers. On the contrary Ex.-ca, Ex.-Mg, and pH became high with increase of soil depths. 2. $NO_3{^-}-N$, Av.$P_2O_5$, Ex.-K, and EC in surface soil were high in the order of Converted, Paddy-Upland Rotation, Potato-Chinese Cabbage>2 Year, Rotation, Potato-Chinese Cabbage>Converted, Paddy-Upland Rotation, Soybean>Continous Paddy, which responded well to fertilizer application rates. On the other hand Ex.-Ca, Ex.-Mg, and pH in whole layers were high in the order of Converted, Paddy-Upland Rotation, Soybean>Converted, Paddy-Upland Rptation, Potato-Chinese Cabbage>2 Year, Rotation, Potato-Chinese Cabbage>Continuous Paddy, which largely depended on plant absorption. 3. Nutrient balance in upland cropping system cultivating potato and Chinese cabbage showed that the input of chemical fertilizer of nitrogen and potassium was less than the plant uptake, while it was reverse for phosphorus with much gap between fertilizer input and plant uptake. Therefore, phosporous was expected to be accumulated by 27kg/10a every year. 4. Nutrient balance in upland cropping system cultivating soybean showed that nitrogen was not deficient to soybean crops even the chemical fertilizer input was less than plant uptake because of nitrogen fixation by rhizobia. However, there was about 1kg/10a deficit of potassium, which resulted dificiency symptom in the middle stage of soybean growth. For phosphorous there was excess of 4kg/10a, which was expected to be short for maintaining phosphorous fertility of upland soils.
Rhizobium meliloti populated in five Korean pasture soils were characterized by symbiotic effectiveness and intrinsic antibiotic resistance using whole-soil inoculum and 11 antibiotics, respectively. Most probable number (MPN) of naturalized rhizobia counted with alfalfa Vernal[Medicago sativa (L.)] as a host ranged $1.7{\times}10^2\;cells/g$. soil(Chunghyo, Kyeongiu)-$1.0{\times}10^5\;cells/g$. soil(Gampo, Kyeongiu) and ended to be positively associated with soil pH. On the whole, the effectiveness of population as compared to TAL mix inoculum (TAL 380+TAL 1372+TAL 1373) was very low. Nevertheless, there were two highly effective strains, YCK 539 and YCK 542, which were not inferior to TAL 1372, from Ogpo, Dalseong among the total of 30 of 6 isolates per each soil. As long as mean $N_2$ fixing ability of each soil isolate, the isolates from Hyeongog, Kyeonju were outstanding and the rest were in order of Ogpo, Dalseong>Chunghyo, Kyeongju>Hwaweon, Dalseong>Gampo, Kyeongiu. Isolates as a whole were resistant to erythromycin(67崙), nalidixic acid(77%), and streptomycin sulfate(8051), which had the concentration of $100\;{\mu}g/ml$, $160\;{\mu}g/ml$, and $10\;{\mu}g/ml$, respectively and divided into 14 patterns of resistance. Association between resistances in each soil was not clear. And there was no relationship of resistance pattern to effectiveness. The best effective strain YCKa 542 exclusively fell into No. X pattern having resistance to erythromycin, nalidixic acid, and neomycin sulfate.
This experiment was conducted to isolate the mutants from S118 and to investigate the physiological characteristics of R. japonicum mutants. The results obtained were as follows; Based on nodulation and acetylene reduction, nodulation of rhizobia was divided into 4 groups, i.e. slow-nodulation, earlier-nodulation, infrequent-nodulation and non-nodulation. At 5% significant level, the growth of inoculated plant with SM255 was bad, but that of HP277 was good. Root-hairs curling was induced by strains S118 and HP277 on soybean, but not by strain SM255. S118 and SM255 were found to be slow-gorwers and produced alkali, whereas strain HP277 was fast-grower and produced acid in YEM broth. In litmus milk reaction, all strains indicated alkaline reaction, and serume-zone was induced weakly by HP277. All of the strains tested in this experiment utilized sucrose. HP277 and LP268 utilized xylose, whereas S118 and SM255 did not. SM255 showed bad growth in nitrogen carriers however utilization of $Ca(NO_3)_2{\cdot}4H_2O$ by HP277 was possible at 25mM and 10mM level. To compare with S118, the protein band of SM255's cell protein electrophoresis was not developed at 0.62 Rm position.

Morphological and cultural characteristics of the strains were studied, and attempts were also made to investigate their antigenic properties and to demonstrate lysogenic strains in these groups. The results were : i) the isolates varied in cultural characteristics on yeast mannitol broth and agar, and in degree of congo reel absorption ; ii) similarities in their antigenic properties were found between/among the strains: G-3/G-9/D216, G-20/G-52 in soybean group; iii) no lysogeny was found in the strains of these groups. 3. Plant infection tests by test tube and bottle method in light room were carried out to elucidate the ability of the strains to nodulate specific legumes and of the capacity of such nodules to fix atmospheric nitrogen. The isolates were grouped into non- invasive, ineffective, or effective to the legumes. Those strains which produced effective nodules, supporting similar level of growth as nitrate control, were: P-3, 4 and 8 in pea and vetch group; G-23, 27, and, D-216 in soybean group.
This experiment was conducted to find out the effects of fertilizer-N and co-inoculation of the Rhizobium japonicum and the Azospirillum lipoferum on nodulation, $N_2$-fixation, and growth of soybean under in situ conditions. The results obtained were summarized as follows: 1. The yield of soybean dry matter was significantly greater in the R. japonicum alone, and the mixed inoculation of R. japonicum and A. lipoferum than those of un-inoculation. But inoculum applied by different strains did not significant effect on plant growth. The effects of nitrogen applied on soybean dry matter were higher in the ammonium sulfate than potassium nitrate, and decreased with increasing rates of two forms of nitrogen applied regardless of nitrogen source. 2. Acetylene redution activity was more increased in a single inoculation of R. japonicum than those of the mixed inoculation of the R. japonicum and the A. lipoferum, in cases of Danyeup cultivar, regardless of the form of combined nitrogen used. 3. Nodule mass and total nitrogenase activity per plant showed the positively significant effect in the interrelationship between dry matter of soybean and some factors related to nitrogen fixation efficiency. 4. The highest symbiotic effect in Danyeup cultivar was obtained when a single R. japonicum 84 Dy-1 strain was inoculated and fertilized with 18 mM potassium nitrate.
