Synthesis and Characterization of Novel Fullerene($C_{60}$) Derivative with Photoresponsive Azobenzene Group (광감응형 아조벤젠기를 갖는 신규 플러렌 유도체의 합성과 물성분석)

  • A novel fullerene derivative with photoresponsive azobenzene group was designed and synthesized, and its photoresponsive properties were reported. Starting from 4-nitrophenol, compound 1, which is containing fullerene moiety connected to azobenzene group through covalent linkage was synthesized by 5 steps. All the intermediates and the final compound were characterized by $^1H$, $^{13}C$-NMR, FAB-Mass or elemental analysis. Compound 1 exhibited the expected photoresponsive behavior. Chloroform solution($10^{-5}M$) of it served to maximize the absorption at 351 nm corresponding to the trans-azobenzene chromophore. Irradiation of this solution with 365 nm light resulted in photoisomerization to cis-azobenzene, as evidenced by decrease in the absorbance at 351 nm and an increase in absorbance at 450nm. A photostationary state was reached within about 150 s. Thermal reversion to the original spectrum was observed over the course of about 6 h at room temperature in the dark. However, exposure to bright sun light for about 5 s also effect almost complete reversion to the trans-isomer. This indicates that there is no strong steric influence on the trans-cis reversible isomerization of compound 1.

Induction and Chatacterization of pKM101 Mutants in Salmonella typhimurium (Salmonella typhimurium내로의 pKM101 돌연변이체의 유도와 그 특성에 관한 연구)

  • Mutants of plasmid pKM101 modified to enhance mutagenesis were induced and characterized in Salmonella typhimurium. The pKM101 mutant plasmid were transferred normally and stably maintained in cells. They had modified in their ability (i) to enhance the reversion of both point and frameshift mutations, (ii) to protect the cell against UV-irradiation and chemical mutagen treatment, (iii) of ampicillin resistance. A similar modification in enhancement of reversion was also observed in a $uvrB^-$ strains. These results indicated that mutator effect of pKM101 was coded by one plasmid gene.

  • Cure characteristics of carbon black-filled rubber compounds with different rubber composition were studied using a rheometer. The carbon black-filled rubber compounds with single, binary, and ternary rubber compositions of natural rubber (NR), styrene-butadiene rubber (SBR), and butadiene rubber (BR) were used. Delta-torques of the NR/BR- and SBR/BR-based compounds with a high BR content were higher than those of the single rubber-based compounds. For ternary rubber-based compounds, the delta-torques of the compounds were lower when the difference in the rubber content ratios was small than when it was big. Scorch and optimum cure times of the rubber compounds became shorter by increasing the content of NR in the compounds while those became longer by increasing the SBR content. Cure rates of the rubber compounds increased with a decrease of the SBR content in the rubber compounds. Reversion ratios decreased with an increase of the SBR content in the rubber compounds.

Genotoxicity Evaluation Using Reversion Mutation Test of SU-Eohyeol Pharmacopuncture (SU어혈약침의 복귀돌연변이시험을 이용한 유전독성평가)

  • SU-Eohyeol pharmacopuncture(SUEP) was developed by adding Cervi Pantotrichum Cornu to Jungsongouhyul pharmacopuncture. This genotoxicity evaluation was performed to evaluate the mutagenic potential of the test substance SUEP agent using histidine, which requires strains of Salmonella typhimurium (TA98, TA100, TA1535, TA1537), and tryptophan, which requires Escherichia coli (WP2uvrA) strain in the presence and absence of metabolic activation. According to the results of the dose range finding study conducted prior to the main study, the dose levels of the test substance in the main study were determined as 100, 50, 25, 12.5, 6.25%, and positive and negative controls were established. As a result of the main study, the mean number of revertant colonies compared to negative controls was less than 2-fold at all dose levels of SUEP in all strains with and without metabolic activation. In the positive control group, the mean number of revertant colonies for each strain was markedly increased by more than two times compared to the negative control group. Based on the result of this study, the test substance, SUEP did not show any indication of mutagenic potential under the conditions of this study.

Inhibitory Effect of Rice Extract on the Chemically Induced Mutagenesis (쌀 추출물의 돌연변이 억제효과)

  • The inhibitory effects of rice extract on mutagenicity induced by 3-amino-1,4-dimethyl-5H-pyrido [4,3-b]indole(Trp-P-1), 3-amino-1-methyl-5H-pyrido [4,3-b]indole(Trp-P-2), sodium azide(SA), 2-nitrofluorene(2NF), mitomycin C(MMC), aflatoxin $B_1(AFB_1)$ and 4-nitroquinoline oxide(4-NQO) were investigated using Salmonella typhimurium reversion assay, SOS chromotest and spore rec-assay. In Salmonella typhimurium reversion assay, methanol extract from brown rice (Illpumbyeo, Japonica variety) showed the highest inhibitory effect among other extracting solvent including hexane, chloroform and water. Methanol extract showed stronger inhibitory effect, above 85%, on indirect-acting mutagens(Trp-P-1, Trp-P-2 and $AFB_1$) than those on direct-acting mutagens(4-NQO, 2NF). In SOS chromotest, methanol extracts showed $77.6{\sim}88.9%$ effects on SOS function induced by Trp-P-1, Trp-P-2, $AFB_1$ and 4-NQO. In spore rec-assay, methanol extracts inhibited the mutagenicity induced by $AFB_1$ and MMC. As the concentration of methanol extract increased, inhibitory effect on mutagenicity increased but reached at steady state as inhibition rate of 90% when the concentration was above 5 mg/plate. In inhibitory effects of methanol extracts by various rice varieties, all of 11 varieties turned out to have inhibitory effect on mutagenicity. There was no significant difference (p>0.05) in inhibitory effect of methanol extracts between brown and white rice against Trp-P-1, but showed difference (p<0.05) against 4-NQO.

Genotoxicological Safety of Hot Water Extracts of the γ-Irradiated Astragali Radix, Atractylodes Rhizoma, and Cimicifugae Rhizoma in Vitro (감마선 조사 황기, 백출 및 승마 열수 추출물의 in vitro 유전독성학적 안전성 평가)

  • As the utilization of medicinal herbs in food and bio-industry increases, safe hygienic technologies for them are demanded. To consider the possibility of application of radiation technology for this purpose, the genotoxi-cological safety of three r -irradiated medicinal herbs were studied. Astragali Radix, Atractylodes Rhizoma and Cimicifugae Rhizoma were irradiated at 10 kGy, and then were extracted with hot water. The genotoxicity of the extracts was examined in two short-term in vitro tests: (1) Salmonella reversion assay (Ames test) in strains of TA98 and TA100; (2) Micronucleus test in cultured Chinese hamster ovary (CHO) cells. The extract was treated at maximum doses of 5 mg/plate in Salmonella reversion assay, and 1 mg/mL in micronucleus test where growth of CHO cells was inhibited by 50%. In Salmonella reversion assay with or without metabolic activation, both ex-tracts of irradiated and non-irradiated herbs showed no significant differences in formation of revertant colonies compared with the negative control. And also in micronucleus test, the incidences of micronucleus in CHO cells cultured with extracts of irradiated herbs were almost same as negative control in less than 3%. These results of two in vitro tests suggest that ${\gamma}$-irradiated herbs do not show mutagenicity and cytogenetic toxicity. Further tests of in vivo genotoxicity and chronic toxicity are needed to ascertain the safety of ${\gamma}$-irradiated herbs.

The Protoplast Fusion of Aspergillus niger (Aspergillus niger의 원형질체 융합에 관한 연구)

  • Conditions for production, fusion and reversion of protoplasts of Aspergillus niger were investigated, and an attempt was made to enhance fusion frequency. Auxotrophic mutants and morphological mutants were induced by U.V. irradiation $(9.9\;erg/mm^2,\;13min)$ on Aspergillus niger. Maximum yield of protoplasts was obtained from 21 hr cultured mycelia by using 1% driselase in 0.6 M KCl or 0.6 M $NH_4Cl$ as osmotic stabilizer. The optimal temperature for mycelium digestion was $30^{\circ}C$, and the optimal pH was 6.0. Protoplasts produced at different digestion period showed heterogeneity in size and vacuole content. Maximal frequency of protoplasts reversion was obtained on 0.6 M KCl stabilized agar medium at pH 5.0. Reversion frequencies of protoplasts produced for 3 hr and 1 hr mycelial digestion were 8.0% and 15.3%, respectively. The optimal concentration of PEG(m.w. 6000) for protoplast fusion was 30%, and that of $CaCl_2$ was $1{\sim}50\;mM$. The optimal pH and period for the reaction of PEG solution were 8.0 and 10 minutes, respectively. Fusion frequencies between auxotrophic protoplasts produced for 3 hr-mycelial digestion were $0.06{\sim}0.42%$, and those for 1 hr-mycelial digestion were $0.09{\sim}0.54%$.

Effect of Virus-free Plant and Subsoiling Reversion Soil for Reduction of Injury by Continuous Cropping of Sweet Potato (고구마 연작장해 경감을 위한 바이러스 무병묘 재배와 심토반전 효과)

  • To reduce the injury by continuous cropping of sweet potato (Ipomoea batatas (L.) Lam.), the farmer's plant and virus-free plant were cultivated with the density of $70{\times}25cm$ (June 10, 2011) in continuous cropping soil (CCS) and subsoiling reversion soil (SRS). Fertilizer was applied at the rates of 55-63-156 $kg\;ha^{-1}$ ($N-P_2O_5-K_2O$) and 10 $ton\;ha^{-1}$ of cattle manure in CCS, and it was applied the 50% increased cattle manure compost and nitrogen in DRS. Symptoms of viral infection were revealed in the farmer's plant at 30 days after planting, but there were no symptoms in virus-free plant. The yield of virus-free plant was more increased 15% and 10.5% than that of farmer's plant in DRS and CCS, respectively. The yield of sweetpotato in SRS was more increased 8.8% and 3.2% in farmer's plant and virus-free plant compared to CCS, respectively. In DRS, the rate of marketable tuber of virus-free plant was increased by 80% compared to the farmer's plant (60.1%). The virus-free plant was produced the tuber with more brilliant peel color and well-formed shape compared to the farmer's plant. The increased yield of virus-free plant and in SRS soil condition showed a positive relationship (p=0.05) with the number of leaf per plant at 30 days and the number of branch per plant at 120 days after planting. The results showed that the early growth after planting was very important for the development of storage root. Therefore, the deep-subsoil reversion and cultivation of virus-free plant could be reduced the injury by continuous cropping of sweet potato, and increased farm income.

  • As preliminaries for the study of Plasmid pKM101 functions nad their interaction with the host DNA repair genes, seven mutants of pKM101, visualized on tetrazolium-galactose plates, deficient in their abitity to enhance mutagenesis were isolated and paritally characterizee. They all have altered functions not only for mutagenesis against MMS and 4-NQO but for the spontaneous reversion of host cell.

Are Korean Industry-Sorted Portfolios Mean Reverting?

  • This paper tests the weak-form efficient market hypothesis for Korean industry-sorted portfolios. Based on a panel variance ratio approach, we find significant mean reversion of stock returns over long horizons in the pre Asian currency crisis period but little evidence in the post-crisis period. Our empirical findings are consistent with the fact that Korea accelerated its integration with international financial market by implementing extensive capital liberalization since the crisis.
