자궁경부암세포에서 방사선조사시 차등 발현되는 유전자 동정 (Identification of Differentially Expressed Radiation-induced Genes in Cervix Carcinoma Cells Using Suppression Subtractive Hybridization)

  • 목적 : 자경경부암세포에서 polymeric chain reaction (PCR)원리를 이용한 suppression subtractive hybridization (SSH) 방법으로 방사선조사 시 차등 발현되는 유전자를 동정하고자 하였다. 대상 및 방법 : 자궁경부암세포주인 HeLa세포주에 방사선조사 전과 후 총 RNA와 poly $(A)^+$ mRNA를 분리였다. SSH방법으로 forward 및 reverse-subtracted cDNA libraries를 만들었다. 차등 발현된 유전자를 screening하기 위해 reverse Northern blotting (dot blot analysis)을 이용하여 각각의 library에서 88개의 클론을 선택하였고 Nothern blotting으로 확인 후 sequencing하였다. 결과 : screening상 176개 클론 중 forward-subtracted library에서 10개의 유전자가 reverse-subtracted library에서 9개의 유전자가 동정되었다. forward-subtracted library로부터 3개의 유전자가 Northern blotting에 의하여 확인되었고 이중 telomerase catalytic subunit and sodium channel-like protein 유전자와 1개의 ESTs (expressed sequence tags) 유전자가 방사선선량에 따라 증가하쳐다. 결론 : 본 연구를 통해 자궁경부암세포주에서 방사선에 의해 유도되는 유전자를 SSH 방법을 통해 동정할 수 있었다. 그러나 이러한 유전자가 어떤 생물학적인 기능을 갖고 있는지에 대한 계속적인 연구가 필요하다

Identification of Proapoptopic, Anti-Inflammatory, Anti-Proliferative, Anti-Invasive and Anti-Angiogenic Targets of Essential Oils in Cardamom by Dual Reverse Virtual Screening and Binding Pose Analysis

  • Background: Cardamom (Elettaria cardamom), also known as "Queen of Spices", has been traditionally used as a culinary ingredient due to its pleasant aroma and taste. In addition to this role, studies on cardamom have demonstrated cancer chemopreventive potential in in vitro and in vivo systems. Nevertheless, the precise poly-pharmacological nature of naturally occurring chemo-preventive compounds in cardamom has still not been fully demystified. Methods:In this study, an effort has been made to identify the proapoptopic, anti-inflammatory, anti-proliferative, anti-invasive and anti-angiogenic targets of Cardamom's bioactive principles (eucalyptol, alpha-pinene, beta-pinene, d-limonene and geraniol) by employing a dual reverse virtual screening protocol. Experimentally proven target information of the bioactive principles was annotated from bioassay databases and compared with the virtually screened set of targets to evaluate the reliability of the computational identification. To study the molecular interaction pattern of the anti-tumor action, molecular docking simulation was performed with Auto Dock Pyrx. Interaction studies of binding pose of eucalyptol with Caspase 3 were conducted to obtain an insight into the interacting amino acids and their inter-molecular bondings. Results:A prioritized list of target proteins associated with multiple forms of cancer and ranked by their Fit Score (Pharm Mapper) and descending 3D score (Reverse Screen 3D) were obtained from the two independent inverse screening platforms. Molecular docking studies exploring the bioactive principle targeted action revealed that H- bonds and electrostatic interactions forms the chief contributing factor in inter-molecular interactions associated with anti-tumor activity. Eucalyptol binds to the Caspase 3 with a specific framework that is well-suited for nucleophilic attacks by polar residues inside the Caspase 3 catalytic site. Conclusion:This study revealed vital information about the poly-pharmacological anti-tumor mode-of-action of essential oils in cardamom. In addition, a probabilistic set of anti-tumor targets for cardamom was generated, which can be further confirmed by in vivo and in vitro experiments.

Lactococcus sp. HY449가 생산한 Bacterisocin의 정제 (Purification and Partial Amino Acid Sequence of a Bacteriocin Produced by Lactococcus, sp. HY449)

  • Lactrococcus sp. HY449균줄르 M17-glucose broth에 배양하여 배양 상등액으로부터 propanol-actone 침전 ion-exchange chromatography gel-filtration chromatography 및 reverse-phase chroamtography 등을 통하여 비활성 25,600,000 BU/mg 인 순수한 bacteriocin 을 정제하였다. 정제 과정 주에서 ion-exchange chromatography 단계에 서는 35.3%의회수율이 7.3%로 감소하였다. Reverse-Phase chromatography에선 3.3%의 회수율을 보였고 활성도는 413.5배로 증가하였다. Tricine-SDS 전기영도 결과 bacteriocin 은 단일 밴드로 나타났으며, N-말단 아미노산 서열 분석을 수행한 결과 $NH_2$-IIe-Leu-Pro-GIn로 확인되었다. 아미노산조정 분석결과를 바탕으로 분자량을 예측한 결과 본 bacteriocin은 32개의 아미노산으로 이루어져 있으며 분자량은 3.6kDa인 것으로 추정되었다.

  • The primary focus of this study was on the issues associated with family and economic decision-making, in particular for the elderlyhood which might have to (or could be) share the family resources. This study regarded willingness to consider applying for Reverse Mortgage as an economic decision-making. The data was collected by interviewing 320 persons (over age 55) living in metropolitan areas in 2008. We selected 227 cases to find the influence of family, having both spouse and adult-children. We have used a theory of Planned Behavior by Fishbein to analysis three research questions. The results are as follows : first, the results partially explain willingness to consider applying for reverse mortgage using Fishbein's theory. Second, several results indicated that economic decision-makings within the family are influenced by several factors relating to other family member's view. They means that some of Subjective Norm variables, of Attitude Behavior ones, and of Perceived Behavior Control ones. Third, the result shows men and women respond differently to consider to applying for that policy. Men are conscious of the other people, i.e., friends, relatives except close family members, and, on the other hand, women are mindful of close family members, i.e., adult children. Forth, the group who have lower income showed higher intentions to consider that policy, when they have adult-children. Fifth, according to age group, pre-elderly group are influenced by consent of adult-children, a level of education, and on the other hand, elderly group are influenced by the other people, i.e., friends, relatives, a level of education, and so on. These results meaned that these distinct characteristics should be considered to establish Income Security policy for the pre-elderly and the elderly.

배전계통에서 변압기 결선에 의한 역 조류현상에 관한 연구 (A Study on the Reverse-Power-Flow Phenomenon due to Transformer Wiring Types in Distribution System)

  • 본 논문에서는 최근 분산전원 확대보급에 따라 계량오차 발생, 변전소 CB 또는 Recloser, 수전설비 VCB의 OCGR 오동작, 배전자동화 시스템 거짓FI 정보발생 등 한전 배전계통에서 발생되는 오동작 현상에 대해서 원인분석을 수행하였고 그 원인은 변압기 결선에 의한 역 조류현상임을 규명하였다. 이러한 역 조류현상의 영향으로 배전계통의 3상 기본파 전류가 N상에서 동상이 되어 3배의 전류로 합산되었으며, 이 현상을 근거로 새로운 계통해석 방법을 제시하였다. 새로운 계통해석 방법을 적용하여 구간별 고장전류를 해석하였고, 역 조류현상 규명을 위해 변압기 결선형태 별로 역 조류 발생여부를 실험하였다. 또한 PSCAD/EMTDC로 실제 계통을 모의함으로써 역 조류에 의한 오동작 현상을 줄이기 위한 방법으로 중성선의 영상임피던스를 조절하는 방법을 제시하였다.

  • In the construction industry, there has been a surge in the implementation of high-tech equipment in recent years. Various technologies are being considered as potential solutions for future construction projects. Building information modeling (BIM), which utilizes advanced equipment, is a promising solution among these technologies. The need for safety inspection has also increased with the aging structures. Nevertheless, traditional safety inspection technology falls short of meeting this demand as it heavily relies on the subjective opinions of workers. This inadequacy highlights the need for advancements in existing maintenance technology. Research on building safety inspection using 3D laser scanners has notably increased. Laser scanners that use light detection and ranging (LiDAR) can quickly and accurately acquire producing information, which can be realized through reverse engineering by modeling point cloud data. This study introduces an innovative evaluation system for building safety using a 3D laser scanner. The system was used to assess the safety of an existing three-story building by implementing a reverse engineering technique. The 3D digital data are obtained from the scanner to detect defects and deflections in and outside the building and to create an as-built BIM. Subsequently, the as-built structural model of the building was generated using the reverse engineering approach and used for structural analysis. The acquired information, including deformations and dimensions, is compared with the expected values to evaluate the effectiveness of the proposed technique.

역분석공학기법을 이용한 불연속면 분석 프로그램 개발 (Discontinuity Analysis Method using Reverse Engineering)

  • 3차원 레이저 스캐너를 이용하여 사물의 형상을 측정하고, 이로부터 얻어진 데이터를 이용해서 측정된 사물의 형상을 재현하는 기술을 역분석공학이라고 한다. 최근 암반공학분야에서도 사람 손이 닿지 않는 암반사면이나 넓은 지역에 대한 신속한 불연속면 조사를 위해 이러한 역분석공학 기술을 응용하고자 하는 연구가 활발히 진행되고 있다. 이러한 기술을 이용하여 불연속면을 조사하고 분석하기 위해서는 레이저 스캐너로부터 얻어진 3차원 점군데이터를 처리하고 이로부터 불연속면을 추출하고 분석하는 프로그램이 필수적이다. 그러나 기존 프로그램들은 불연속면 조사에 필요한 기능을 충분히 갖추었다고 보기 어려운 실정이다. 따라서 이 연구에서는 3차원 레이저 스캐너를 이용하여 암반면을 측정한 점군데이터로부터 불연속면을 추출하고 통계분석하는 일련의 과정을 자동화된 하나의 프로세스로 처리하는 프로그램을 개발하였으며, 개발된 프로그램을 암반 사면과 터널 벽면의 불연속면 조사에 적용하여 조사기법과 프로그램의 적용성을 검증하였다. 3차원 레이저를 이용한 불연속면 조사와 개발된 프로그램을 이용함으로써 부지 조사시 접근이 곤란한 지역의 조사 용이성이 증대될 수 있을 것으로 기대된다. 또한 관련 기술의 국제적인 수준이 초보 단계인 것을 감안하면 앞으로 해외시장 선점 가능성도 클 것으로 기대된다.

Development of fragility curves for RC bridges subjected to reverse and strike-slip seismic sources

  • This paper presents a probabilistic fragility analysis for two groups of bridges: simply supported and integral bridges. Comparisons are based on the seismic fragility of the bridges subjected to accelerograms of two seismic sources. Three-dimensional finite-element models of the bridges were created for each set of bridge samples, considering the nonlinear behaviour of critical bridge components. When the seismic hazard in the site is controlled by a few seismic sources, it is important to quantify separately the contribution of each fault to the structure vulnerability. In this study, seismic records come from earthquakes that originated in strike-slip and reverse faulting mechanisms. The influence of the earthquake mechanism on the seismic vulnerability of the bridges was analysed by considering the displacement ductility of the piers. An in-depth parametric study was conducted to evaluate the sensitivity of the bridges' seismic responses to variations of structural parameters. The analysis showed that uncertainties related to the presence of lap splices in columns and superstructure type in terms of integral or simply supported spans should be considered in the fragility analysis of the bridge system. Finally, the fragility curves determine the conditional probabilities that a specific structural demand will reach or exceed the structural capacity by considering peak ground acceleration (PGA) and acceleration spectrum intensity (ASI). The results also show that the simply supported bridges perform consistently better from a seismic perspective than integral bridges and focal mechanism of the earthquakes plays an important role in the seismic fragility analysis of highway bridges.

Prognostic Relevance of Human Telomerase Reverse Transcriptase (hTERT) Expression in Patients with Gall Bladder Disease and Carcinoma

  • Background: Gallbladder carcinoma (GBC) has been stated as an Indian disease, with the highest number of cases being reported from certain districts of northeast India, which has an ethnically distinct population. Unfortunately there are no scientific reports on the underlying molecular mechanisms associated with the pathogenesis of the disease from this region. Aim: The present study evaluated the role of differential expression of human telomerase reverse transcriptase (hTERT) in the development of gall bladder anomalies. Materials and Methods: Blood and tissue samples were collected from patients undergoing routine surgical resection for clinically proven cases of gallbladder disease {cholelithiasis (CL, n=50), cholecystitis (CS, n=40) and GBC (n=30) along with adjacent histopathologically proved non-neoplastic controls (n=15)} with informed consent. Whole blood was also collected from age and sex matched healthy controls (n=25) for comparative analysis. Differential hTERT mRNA expression was evaluated by semi-quantitative rt-PCR and real-time PCR based analysis using ${\beta}$-actin as an internal control. Evaluation of differential hTERT protein expression was studied by Western blot analysis and immunoflourescence. Statistical analysis for differential expression and co-relation was performed by SPSSv13.0 software. Results: Gallbladder anomalies were mostly prevalent in females. 대형 캐비테이션터널에서 펌프젯 추진기 단독성능 시험 및 해석 기법 연구 (Study of the Open-Water Test and Analysis for a Pumpjet Propulsor in LCT)

  • In order to study the open-water test and analysis techniques for pumpjet propulsors in the Large Cavitation Tunnel (LCT), at the Korea Research Institute of Ships and Ocean Engineering, a set of test equipment was designed and manufactured. The pumpjet propulsor is composed of rotor, stator and duct resulting in the strong interaction between the components. A ring-shaped sensor was developed to measure the thrust and torque for duct and stator. The test equipment including the pumpjet is installed on an existing POW dynamometer in the reverse direction. The results from the reverse POW test setup were validated against those from the conventional POW test setup in the Towing Tank (TT) as well as in the LCT. The pumpjet open-water test was conducted at the Reynolds number of around 1.0×106, at which the obtained experimental data became stable in the Reynolds number effect test. The open-water test for the rotor (rotor-only) was conducted to study whether the duct and stator should be considered as a part of the hull or the propulsor. On the basis of the test results, it was shown that the duct and stator could be included in the propulsor. The total thrust, combined thrust of rotor, duct, and stator was used for the pumpjet open-water test analysis. As the whole pumpjet is defined as a propulsor, it is thought that the self-propulsion test and analysis could be conducted in the same way as that of the conventional propeller.
