  • 마른 멸치 제조에 있어서 산가용성핵산관련물질의 변화와 또 제조에 따른 변화의 차이를 알기 위하여 생체시료, 자건시료 및 소건시료로 나누어 분석 검토하였다. 1. ATP는 자건 및 소건시료에 있어서는 거의 소실되었고, 어획직후에 채취한 생체시료에 있어서도 대부분 분해되어 함량은 대단히 적었다($1.8{\mu}moles/g$, dry basis). 2. ADP는 ATP와 AMP가 거의 소실된 후에 도 상당량이 장기 잔존하였는데, 이것은 myofbril과 결합한 bound nucleotide인 것으로 추정된다. 3. AMP는 전반적으로 미량 검출되는데 지나지 않았으나 자건시료가 타시료에 비하여 비교적 함량이 높았다. 4. IMP는 자건시료에 있어서 그 축적성이 현저하게 인정되었으나 소건시료에 있어서는 거의 분해되어 자건시료의 1/16의 함량에 지나지 않았다. 5. $H_xR$$H_x$는 소건시료에 있어서의 함량이 월등히 많아 생체시료의 3.5배, 자건시료의 2.7배였고, $H_xR$$H_x$의 함량비에 있어서는 생체시료와 자건시료는 $H_x$ 축적형이였는데 비하여 소건시료는 반대로 $H_xR$ 축적형을 나타내었다. 6. 생체시료를 기준으로 건조후의 nucleotides의 잔존율은 자건시료는 $83\%$이고 그 중의 IMP의 비율이 $73\%$인데 비하여 소건시료는 잔존율이 겨우 $10\%$에 불과하였으며, 이 중의 IMP비율은 $38\%$에 지나지 않았다. 즉, 자건시료에 있어서는 ATP를 제외한 nucleotides, nucleose 및 base가 비교적 고르게 잔존하는 데 비하여 소건 시료에 있어서는 nucleotides는 거의 소실되고 대부분이 nucleoside와 base의 형태로 잔존하였다. 7, 마른 멸치의 flavor quality를 IMP 함량만으로 판단한다면 자건법은 소건법에 비하여 보다 효과적인 제조법이라고 할 수 있다.

효소식품으로서 발아유색미의 마이크로파 진공건조 (Microwave Vacuum Drying of Germinated Colored Rice as an Enzymic Health Food)

  • 품질이 우수하고 경제성이 있는 건강지향성 식품개발 연구의 일환으로 유색미를 발아시킨 후 마이크로파 진공건조를 적용하여 건조하고 그 건조특성과 품질특성을 열풍건조, 진공건조, 또는 동결건조의 경우와 비교해 보았다. 마이크로파 진공건조 1 및 2의 경우 열전달속도가 빨라 건조개시후 5분 이내에 $60^{\circ}C$에 도달한 후 건조시간 내내 $60{\pm}2^{\circ}C$를 유지하였으나 열풍건조의 경우는 건조개시 30분 경과 후, 진공 건조의 경우는 3시간 30분 경과 후에야 $60^{\circ}C$에 도달하였다. 마이크로파 진공건조 1의 경우 건조 3시간 후 수분함량 0.08 kg water/kg solid까지 감소하였으나, 마이크로파 진공건조 2의 경우 총건조시간 2시간만에 수분함량 0.07 kg water/kg solid까지 감소하였다. 열풍건조의 경우 건조시간 4시간만에 수분함량 $0.17{\sim}0.18\;kg\;water/kg\;solid$까지 감소하였으나 최소한 2시간은 더 건조시켜야 할 것으로 예상되었다. 마이크로파 진공건조 1 및 2의 경우는 건조 초기 건조속도는 0.055 kg water/kg solid/min이었으며 처음부터 건조속도가 계속 감소하는 감률건조를 보였다. 열풍건조의 경우는 건조 초기 건조속도가 0.0057 kg water/kg solid/min으로서 마이크로파 진공건조 1 및 2의 경우의 1/10수준이었으며 건조개시 60분까지 항률건조를 보이다가 임계수분함량 0.76 kg water/kg solid 부근부터 감률건조가 시작되었다. 건조시료의 ${\alpha}-amylase$ 함량은 마이크로파 진공건조 2, 마이크로파 진공건조 1, 동결건조, 진공건조 및 열풍건조의 순서를 보여 마이크로파 진공건조의 품질보존효과를 잘 보여 주었다.다. 순으로 나타났으며 $FB_2$의 경우도 비슷한 경향을 나타내었다.EX> 수준이었으며, 전발효 종료시 알코올 생성은 모두 14% (v/v) 정도로 비슷하였다. 포도주의 숙성기간 중 잡균 오염 방지를 위해 potassium metabisulfite를 40 ppm 이상 첨가하였을 때에는 포도주의 색이 탈색되었다.Gram (-)균으로는 Aeromonas hydrophila, 효모로는 Candida pelliculosa가 각각 분리 동정되었다. 배추 물김치의 경우 Lactobacillus plantarum, Leuconostoc citrum, Leu. mes.ssp.mesenteroides/dextranicum, Streptococcus facium이 동정되었으며, Pediococcus는 역시 미확인되었다. Gram (-)균은 Pseudomonas fluorescens, Pseu. aureofaciens가 각각 분리 동정되어 주재료간 차이를 보여주었다.되고 있는 국제환경 속에서 소비자들에게 방사선조사 식품에 대한 올바른 정보를 효과적으로 제공함으로써 그들의 식품구입 의사결정에 도움을 주는 것이 매우 필요하다. 더욱이 본 연구결과 우리 나라 소비자들은 방사선조사 식품에 대한 낮은 인지도를 가지고 있어 소비자로서의 알권리를 제대로 누리지 못하고 있는 것으로 나타나 앞으로 국내에서 방사선조사 식품이 상업적으로 널리 보급되기에 앞서 방사선조사 식품에 대한 소비자교육이 활발하게 이루어질 필요가 있다고 본다., 년령군별 발생양상은 외국의 그것과 마찬가지이다. 따라서 무과립구증 발생양상의 기준상황 자료(baseline data)로써 유용하리라고 판단 된다. 한편 본 조사연구에서 적용한 방법론은 앞으로 특정 질병, 특히 희귀한 중증질환의 전국적인 발생율을 타당하고 신뢰성있게 추정할 수 있는 적절한 방법의 하나라고 평가된다. 그리하여 본 연구에서 적용하였던 방법을 앞으로 우리

절단 대파의 품질특성에 미치는 세척 및 포장재의 효과 (Quality Attributes of Fresh-Cut Green Onion as Affected by Rinsing and Packaging)

  • 국내 생산과 소비가 많은 대표적 조미 채소 가운데 하나인 대파를 원료로 절단 가공 후 세척 및 포장재 적용에 따른 저온저장 중 품질 특성의 변화를 살펴보았다. 전처리 방법으로서 무수세, 냉수세, 염소수 처리 등을 적용한 후 $63\;{\mu}m$ 두께의 폴리에틸렌 필름에 포장하여 $10^{\circ}C$에 저장하면서 생균수 및 관능적 특성을 측정한 결과, 무처리 대조구와 비교했을 때 냉수세나 염소수 처리는 절단 대파의 미생물학적 품질에는 그다지 큰 영향을 미치지 않았으나 외관의 관능평가에 있어서는 냉수세나 염소수 처리구가 상대적으로 우수하게 나타났다. 또한 절단가공 및 냉수세 처리후 각기 두께가 다른 폴리에틸렌 필름(22, 36, $63\;{\mu}m$)에 밀봉 포장하여 $10^{\circ}C$에 저장한 결과, 대조구인 통기성 천공 필름 포장구에 비해 밀봉 포장구가 품질유지에 유리 하였으며 필름의 기체투과도가 낮은 두꺼운 필름일수록 저장 중 이화학적, 미생물학적, 관능적 측면에서 시료의 품질변화가 적게 나타났다. 저장 13일째 중온 호기성 세균수는 대조구가 $3.07{\times}10^6}$ CFU/g인데 반해 밀봉 포장구는 필름 두께별로 $2.02{\times}10^5,\;1.75{\times}10^5,\;1.74{\times}10^5\;CFU/g$을 나타내었고, 관능적 평가에서도 두께 $63\;{\mu}m$의 polyethylene 필름(600 $O_2\;mL/day{\cdot}m^2{\cdot}atm;\;2,500\;CO_2\;mL/day{\cdot}m^2{\cdot}atm$)으로 밀봉한 포장구가 가장 우수하여 저장 18일까지 초기 품질을 거의 그대로 유지할 수 있었다.

총정맥영양법의 간담도 합병증에 대한 Ursodeoxycholic Acid 조기투여의 이중맹검 위약대조군 연구 (A Randomized, Double-Blind, Placebo-Controlled Trial of Early Ursodeoxycholic Acid Administration for Prevention of Total Parenteral Nutrition-Induced Hepatobiliary Complications)

  • 목 적: 총정맥영양법의 합병증인 간담도계 장애시 ursodeoxycholic acid (UDCA) 투여는 독성이 있는 내인성 담즙의 분비를 촉진하여 간손상을 감소시키는 역할을 하는 것으로 알려져 있다. 그러나 대부분의 연구는 합병증 발생 후 치료에 관한 것이어서 본 연구자들은 UDCA를 총정맥영양법의 시작과 동시에 조기 투여할 경우 그 예방효과를 이중맹검 위약대조군 연구로써 알아보고자 하였다. 방 법: 2000년 5월부터 2002년 5월까지 10일 이상의 총정맥영양을 받은 13명의 환아를 대상으로 하였다. 총정맥영양 시작과 동시에 UDCA를 투여받는 시험군 7명, 위약을 투여받는 대조군 6명을 이중맹검법으로 구분하였다. 연령은 생후 1일부터 13세까지이고 환아들의 진단은 경관영양이 불가능한 미숙아와 뇌성마비아, 만성설사, 거식증, 췌장염, 주기성 구토증 등이었다. 총정맥영양의 기간은 10일에서 70일까지였다. 주기적으로 간기능을 비롯한 검사항목등을 측정하였으며 총정맥영양의 기간, 조성, 투여속도, 열량 등이 조사 기록되었고 total bilirubin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase에 대하여 군간 비교하였다. 결 과: Mixed procedure model을 이용한 분석에서 시험군을 reference로 하였을 때 대조군의 autoregressive coefficient 값은 total bilirubin의 경우 0.4419 (p=0.0651), AST는 -0.0431 (p=0.7923), ALT는 0.2398 (p=0.2416), 그리고 alkaline phosphatase는 0.2459 (p=0.1922)였다. 결 론: 총정맥영양과 UDCA를 초기부터 동시 투여하였을 때 total bilirubin은 대조군에 비하여 상승하지 않는 것으로 나타났으나 통계적으로 유의하지는 않았다.

한국의 지역개발과 댐건설 (Regional Development And Dam Construction in Korea)

  • Because of differences in thoughts and ideology, our country, Korea has been deprived of national unity for some thirty years of time and tide. To achieve peaceful unification, the cultivation of national strength is of paramount importance. This national strength is also essential if Korea is to take rightful place in the international societies and to have the confidence of these societies. However, national strength can never be achieved in a short time. The fundamental elements in economic development that are directly conducive to the cultivation of national strength can be said to lie in -a stable political system, -exertion of powerful leadership, -cultivation of a spirit of diligence, self-help and cooperation, -modernization of human brain power, and -establishment of a scientific and well planned economic policy and strong enforcement of this policy. Our country, Korea, has attained brilliant economic development in the past 15 years under the strong leadership of president Park Chung Hee. However, there are still many problems to be solved. A few of them are: -housing and home problems, -increasing demand for employment, -increasing demand for staple food and -the need to improve international balance of payment. Solution of the above mentioned problems requires step by step scientific development of each sector and region of our contry. As a spearhead project in regional development, the Saemaul Campaign or new village movement can be cited. The campaign is now spreading throughout the country like a grass fire. However, such campaigns need considerable encouragement and support and the means for the desired development must be provided if the regional and sectoral development program is to sucdceed. The construction of large multipurpose dams in major river basin plays significant role in all aspects of national, regional and sectoral development. It ensures that the water resource, for which there is no substitute, is retained and utilized for irrigation of agricultural areas, production of power for industry, provision of water for domestic and industrial uses and control of river water. Water is the very essence of life and we must conserve and utilize what we have for the betterment of our peoples and their heir. The regional and social impact of construction of a large dam is enormous. It is intended to, and does, dras tically improve the "without-project" socio-economic conditions. A good example of this is the Soyanggang multipurpose dam. This project will significantly contribute to our national strength by utilizing the stored water for the benefit of human life and relief of flood and drought damages. Annual average precipitation in Korea is 1160mm, a comparatively abundant amount. The catchment areas of the Han River, Keum River, and Youngsan River are $62,755\textrm{km}^2$, accounting for 64% of the national total. Approximately 62% of the national population inhabits in this area, and 67% of the national gross product comes from the area. The annual population growth rate of the country is currently estimated at 1.7%, and every year the population growth in urban area increases at a rising rate. The population of Seoul, Pusan, and Taegu, the three major cities in Korea, is equal to one third of our national total. According to the census conducted on October 1, 1975, the population in the urban areas has increased by 384,000, whereas that in rural areas has decreased by 59,000,000 in the past five years. The composition of population between urban and rural areas varied from 41%~59% in 1959 to 48%~52% in 1975. To mitigate this treand towards concentration of population in urban areas, employment opportunities must be provided in regional and rural areas. However, heavy and chemical industries, which mitigate production and employment problems at the same time, must have abundant water and energy. Also increase in staple food production cannot be attained without water. At this point in time, when water demand is rapidly growing, it is essential for the country to provide as much a reservoir capacity as possible to capture the monsoon rainfall, which concentarated in the rainy seaon from June to Septesmber, and conserve the water for year round use. The floods, which at one time we called "the devil" have now become a source of immense benefit to Korea. Let me explain the topographic condition in Korea. In northern and eastern areas we have high mountains and rugged country. Our rivers originate in these mountains and flow in a general southerly or westerly direction throught ancient plains. These plains were formed by progressive deposition of sediments from the mountains and provide our country with large areas of fertile land, emminently suited to settlement and irrigated agricultural development. It is, therefore, quite natural that these areas should become the polar point for our regional development program. Hower, we are fortunate in that we have an additional area or areas, which can be used for agricultural production and settlement of our peoples, particularly those peoples who may be displaced by the formation of our reservoirs. I am speaking of the tidelands along the western and southern coasts. The other day the Ministry of Agriculture and Fishery informed the public of a tideland reclamation of which 400,000 hectares will be used for growing rice as part of our national food self-sufficiency programme. Now, again, we arrive at the need for water, as without it we cannot realize this ambitious programme. And again we need those dams to provide it. As I mentioned before, dams not only provide us with essential water for agriculture, domestic and industrial use, but provide us with electrical energy, as it is generally extremely economical to use the water being release for the former purposes to drive turbines and generators. At the present time we have 13 hydro-electric power plants with an installed capacity of 711,000 kilowatts equal to 16% of our national total. There are about 110 potential dams ites in the country, which could yield about 2,300,000 kilowatts of hydro-electric power. There are about 54 sites suitable for pumped storage which could produce a further 38,600,000 kilowatts of power. All available if we carefully develop our water resources. To summarize, water resource development is essential to the regional development program and the welfare of our people, it must proceed hand-in-hand with other aspects of regional development such as land impovement, high way extension, development of our forests, erosion control, and develop ment of heavy and chemical industries. Through the successful implementation of such an integrated regional development program, we can look forward to a period of national strength, and due recognition of our country by the worlds societies.

Cefoperazone(T-1551)의 약리학적 연구 (Pharmacological Studies of Cefoperazone(T-1551))

  • The pharmacological and microbiological studies of Cefoperazone (T-1551, Toyama Chemical Co., Japan) were conducted in vitro and in vivo. The studies included stability and physicochemical characteristics, antimicrobial activity, animal and human pharmacokinetics, animal pharmacodynamics and safety evaluation of Cefoperazone sodium for injection. 1) Stability and physicochemical characteristics. Sodium salt of cefoperazone for injection had a general appearance of white crystalline powder which contained 0.5% water, and of which melting point was $187.2^{\circ}C$. The pH's of 10% and 25% aqueous solutions were 5.03 ana 5.16 at $25^{\circ}C$. The preparations of cefoperazone did not contain any pyrogenic substances and did not liberate histamine in cats. The drug was highly compatible with common infusion solutions including 5% Dextrose solution and no significant potency decrease was observed in 5 hours after mixing. Powdered cefoperazone sodium contained in hermetically sealed and ligt-shielded container was highly stable at $4^circ}C{\sim}37^{\circ}C$ for 12 weeks. When stored at $4^{\circ}C$ the potency was retained almost completely for up to one year. 2) Antimicrobial activity against clinical isolates. Among the 230 clinical isolates included, Salmonella typhi was the most susceptible to cefoperazone, with 100% inhibition at MIC of ${\leq}0.5{\mu}g/ml$. Cefoperazone was also highly active against Streptococcus pyogenes(group A), Kletsiella pneumoniae, Staphylococcus aureus and Shigella flexneri, with 100% inhibition at $16{\mu}g/ml$ or less. More than 80% of Escherichia coli, Enterobacter aerogenes and Salmonella paratyphi was inhibited at ${\leq}16{\mu}/ml$, while Enterobacter cloaceae, Serratia marcescens and Pseudomonas aerogenosa were somewhat less sensitive to cefoperagone, with inhibitions of 60%, 55% and 35% respectively at the same MIC. 3) Animal pharmacokinetics Serum concentration, organ distritution and excretion of cefoperazone in rats were observed after single intramuscular injections at doses of 20 mg/kg and 50 mg/kg. The extent of protein binding to human plasma protein was also measured in vitro br equilibrium dialysis method. The mean Peak serum concentrations of $7.4{\mu}g/ml$ and $16.4{\mu}/ml$ were obtained at 30 min. after administration of cefoperazone at doses of 20 mg/kg and 50 mg/kg respectively. The tissue concentrations of cefoperazone measured at 30 and 60 min. were highest in kidney. And the concentrations of the drug in kidney, liver and small intestine were much higher than in blood. Urinary and fecal excretion over 24 hours after injetcion ranged form 12.5% to 15.0% in urine and from 19.6% to 25.0% in feces, indicating that the gastrointestinal system is more important than renal system for the excretion of cefoperazone. The extent of binding to human plasma protein measured by equilibrium dialysis was $76.3%{\sim}76.9%$, which was somewhat lower than the others utilizing centrifugal ultrafiltration method. 4) Animal pharmacodynamics Central nervous system : Effects of cefoperazone on the spontaneous movement and general behavioral patterns of rats, the pentobarbital sleeping time in mice and the body temperature in rabbits were observed. Single intraperitoneal injections at doses of $500{\sim}2,000mg/kg$ in rats did not affect the spontaneous movement ana the general behavioral patterns of the animal. Doses of $125{\sim}500mg/kg$ of cefoperazone injected intraperitonealy in mice neither increased nor decreased the pentobarbital-induced sleeping time. In rabbits the normal body temperature was maintained following the single intravenous injections of $125{\sim}2,000mg/kg$ dose. Respiratory and circulatory system: Respiration rate, blood pressure, heart rate and ECG of anesthetized rabbits were monitored for 3 hours following single intravenous injections of cefoperazone at doses of $125{\sim}2,000mg/kg$. The respiration rate decreased by $3{\sim}l7%$ at all the doses of cefoperazone administered. Blood pressure did not show any changes but slight decrease from 130/113 to 125/107 by the highest dose(2,000 mg/kg) injected in this experiment. The dosages of 1,000 and 2,000 mg/kg seemed to slightly decrease the heart rate, but it was not significantly different from the normal control. All the doses of cefoperazone injected were not associated with any abnormal changes in ECG findings throughout the monitering period. Autonomic nervous system and smooth muscle: Effects of cefoperazone on the automatic movement of rabbit isolated small intestine, large intestine, stomach and uterus were observed in vitro. The autonomic movement and tonus of intestinal smooth muscle increased at dose of $40{\mu}g/ml$ in small intestine and at 0.4 mg/ml in large intestine. However, in stomach and uterine smooth muscle the autonomic movement was slightly increased by the much higher doses of 5-10 mg/ml. Blood: In vitro osmotic fragility of rabbit RBC suspension was not affected by cefoperazone of $1{\sim}10mg/ml$. Doses of 7.5 and 10 mg/ml were associated with 11.8% and 15.3% prolongation of whole blood coagulation time. Liver and kidney function: When measured at 3 hours after single intravenous injections of cefoperaonze in rabbits, the values of serum GOT, GPT, Bilirubin, TTT, BUN and creatine were not significantly different from the normal control. 5) Safety evaluation Acute toxicity: The acute toxicity of cefoperazone was studied following intraperitoneal and intravenous injections to mice(A strain, 4 week old) and rats(Sprague-Dawler, 6 week old). The LD_(50)'s of intraperitonealy injected cefoperazone were 9.7g/kg in male mice, 9.6g/kg in female mice and over 15g/kg in both male and female rats. And when administered intravenously in rats, LD_(50)'s were 5.1g/kg in male and 5.0g/kg in female. Administrations of the high doses of the drug were associated with slight inhibition of spontaneous movement and convulsion. Atdominal transudate and intestinal hyperemia were observed in animals administered intraperitonealy. In rats receiving high doses of the drug intravenously rhinorrhea and pulmonary congestion and edema were also observed. Renal proximal tubular epithelial degeneration was found in animals dosing in high concentrations of cefoperazone. Subacute toxicity: Rats(Sprague-Dawley, 6 week old) dosing 0.5, 1.0 and 2.0 g/kg/day of cefoperazone intraperitonealy were observed for one month and sacrificed at 24 hours after the last dose. In animals with a high dose, slight inhibition of spontaneous movement was observed during the experimental period. Soft stool or diarrhea appeared at first or second week of the administration in rats receiving 2.0g/kg. Daily food consumption and weekly weight gain were similar to control during the administration. Urinalysis, blood chemistry and hematology after one month administration were not different from control either. Cecal enlargement, which is an expected effect of broad spectrum antibiotic altering the normal intestinal microbial flora, was observed. Intestinal or peritoneal congestion and peritonitis were found. These findings seemed to be attributed to the local irritation following prolonged intraperitoneal injections of hypertonic and acidic cefoperazone solution. Among the histopathologic findings renal proximal tubular epithelial degeneration was characteristic in rats receiving 1 and 2g/kg/day, which were 10 and 20 times higher than the maximal clinical dose (100 mg/kg) of the drug. 6) Human pharmacokinetics Serum concentrations and urinary excretion were determined following a single intravenous injection of 1g cefoperazone in eight healthy, male volunteers. Mean serum concentrations of 89.3, 61.3, 26.6, 12.3, 2.3, and $1.8{\mu}g/ml$ occured at 1,2,4,6,8 and 12 hours after injection respectively, and the biological half-life was 108 minutes. Urinary excretion over 24 hours after injection was up to 43.5% of administered dose.
