Effect of environmental conditions on the stock structure and abundance of the Pacific saury, Cololabis saira in the Tsushima Warm Current region (대마난류계 꽁치의 자원구조와 풍도에 미치는 해양환경의 영향)

  • Interannual and decadal scale changes in body size of Pacific saury, catch and catch per unit effort were examined to investigate the environmental effects on the stock structure and abundance in the Tsushima Warm Current region. Interannual changes in thermal conditions are responsible for the different occurrence (catch) rates of sized group of the fish. Changes in body size due to environmental variables lead the stock to be homogeneous during the period of high abundance, while one of the reminder cohorts supports the stock during the period of low level of abundance. Migration circuits of two cohorts of saury stock are hypothesized on the basis of short life span and spatio-temporal changes of the stock structure in normal environmental conditions. Changes in upper ocean structure and production cycles by the decadal scale climate changes lead changes in stock structure and recruitment, resulting in the fluctuation of saury abundance. Hypothesized mechanism of the effects of climate changes on stock structure and abundance is illustrated on the basis of changes in thermal regime and production cycle.

Paralytic Shellfish Toxins in the Mussel, Mytilus edulis, Caused the Shellfish Poisoning Accident at Geoje, Korea, in 1996 (96년 거제에서 패류 중독 사고를 유발한 진주담치의 마비성 독소)

  • Food poisoning accident occurred on May, 1996 at Oepo, Geoje County, Kyeongnam Province, Korea, and two persons were died within a few hours after ingestion of the soup prepared with wild mussel, Mytilus edulis, harvested on the sea rock. Paralytic shellfish poisons (PSP) were elucidated as the responsible toxins for the food poisons accident because the wild mussels caught after three days at the near place from the accident contained high toxicity of PSP ranged $650\~1000MU/g$ of edible meat by mouse bioassay. Gonyautoxin-1+4 $(42.7\%)$ and C1+C2 $(40.0\%)$ were detected as the major toxins in the mussels by fluorometric HPLC method. Although, the poison extracted out with drip during freezing and thawing, and the toxicity gradually decreased by boiling for 20 minutes, over 30 MU/g of toxins remained in the soup and meat, which indicated that they could be able to make food poisoning.

Muscle Intoxication of 'Gukmeri-bok' Puffer (Takifugu vermicularis radiatus) by Freezing and Thawing (냉동과 해동에 따른 국매리복 근육의 독화)

  • We here report that the muscle of 'gukmeri-bok' puffer fish (Takifugu vermicularis radiatus) is intoxicated after freezing and thawing processes even though it is not toxic when it is fresh. This study was carried out to investigate the effect of different freezing and thawing conditions as well as the effect of the presence of skin or viscera on the intoxication of muscle of 'gukmeri-bok' puffer during freezing and thawing process. As a result of this investigation, thawing condition, but not freezing conditions or periods played an important role in muscle intoxication. Tetrodotoxin secretory grands exist in the skin of toxic puffer fish, the skin was more responsible for the muscles intoxication than the viscera during the freezing-thawing process. In other words, no toxicity was measured in the muscle of skinned-frozen specimens even when thawed. According to this result, it is recommended that the skin and viscera must be removed before being frozen for edible purpose. Otherwise, when a whole fish should be frozen, alternative half-thawing and removal of skin from frozen specimens is recommended.

  • The dinoflagellate Alexandrium tamiyavanichii Balech, a producer of toxins causing paralytic shellfish poisoning (PSP), has recently been considered as one of main organisms responsible for toxication of shellfish in Japan. In this study, A. tamiyavanichii was subjected to a molecular phylogenetic analysis inferred from 28S rDNA D1-D2 sequences and a species-specific LSU rRNA-targeted oligonucleotide DNA probe was designed to identify A. tamiyavanichii using the whole cell-FISH (fluorescence in situ hybridization). The sequences of the 28S rDNA D1-D2 region of A. tamiyavanichii showed no difference from A. cohorticular AF1746l4 (present name A. tamiyavanichii) and formed a distinct clade from the 'tamarensis species complex'. The probe, TAMID2, reacted specifically with A. tamiyavanichii cultured cells, without any cross-reaction with other species belonging to the same genus, including A. tamarense, A. catenella, A. affine, A. fraterculus, A. insuetum and A. pseudogonyaulax. In a test of cross-reactivity with a field sample, TAMID2 reacted consistently with only A. tamiyavanichii, indicating that the present protocol involving the TAMID2 probe might be useful for detecting toxic A. tamiyavanichii in a simple and rapid manner.

Alarm substance Detection and Fright Reaction in Giant Danio (Danio malabaricus)

  • A series of experiments were conducted to examine on what stimulus giant danio (Danio malabaricus) were mainly responsible for detecting alarm substance. When 0.15 ml alarm substance (10 ppm) was introduced into the tank, fish showed no notable change in swimming pattern. However, the introduction of 1.5 ml alarm substance (100 ppm) could induce fright reaction about in 6 minutes. Further, when 15 ml alarm substance (1,000 ppm) was introduced into the tank, fish showed the following fright reaction in a few seconds; suppressing to feed diet, no swimming, strong chasing, and visual alertness. In detecting alarm substance, fish were mainly depending on the chemical stimulus (nares) rather than the vision stimulus (eyes) because fish detected alarm substance by the chemical cue showed the significantly stronger fright reaction than by the visual cue. The time for fish to show the initial fright reaction after detecting alarm substance by the chemical cue was shorter than by the visual cue. Also after alarm substance was introduced into the tank, olfaction-deprived fish showed significantly weaker fright reaction and less frequency of chasing than the normal fish which detected alarm substance by both olfaction and gustation stimulus. These results indicated that chemical stimulus, especially olfaction might be the primary sensory modality used in the detection of the alarm substance for giant danio.

The Typology of Childhood Obesity of Fathers with Preschool Children (학령전기 아동의 비만에 대한 아버지의 인식 유형)

  • Traditionally, fathers have been a bread earner and responsible for external matters as the head of household. However, the interest of today's fathers in their children has changed from that of the traditional fathers due to the increased employment of female, decreased birth rate and the trend of shared role in parenting. The purpose of this study was to identify the perception type of fathers of preschool children regarding obesity and the characteristics of the type by applying Q-methodology, and to create the basic data for the development of education programs for fathers or policy-making to prevent child obesity. Data were collected by applying 38 Q statements to 24 fathers of preschool children. Collected data were analyzed with the PC QUANL program. Results indicate that fathers' perception of obesity was divided into three types: 'management of physical activity and diet,' 'generous acknowledgment of reality' and 'compromised emphasis on homemade meal.' The 'compromised emphasis on homemade meal' type is those who consider homemade food important, although they sometimes suggest conditions to make their children cooperative for food. It was found that the subject of the study uniformly recognized that processed foods, fast foods or chocolates are harmful to health. The type of fathers' perception of obesity in preschool children was discussed, focusing on the perception type identified in this study.

In situ Particle Size and Volume Concentration of Suspended Sediment in Seomjin River Estuary, Determined by an Optical Instrument,'LISST-100' (현장입도분석기를 이용한 섬진강하구 부유퇴적물의 특성 연구)

  • In situ particle size and volume concentration of suspended sediment was measured at the mouth of Seomjin River Estuary In February 2001, using an optical instrument, 'LISST-100'. Time variation of in situ particle size and concentration shows: (1) during ebb tide, Seomjin River supplies relatively fine-grained particles with less-fluctuated, compared to during flood tide, and well-behaved concentrations following the tidal cycle; and (2) during flood tide, relatively coarse-grained particles with highly variable in size distribution and concentration flow upstream from Kwangyang Bay. This explains a poor correlation $(r^{2}=0.10)$ between sediment concentration and beam attenuation coefficient during flood and a high degree of correlation $(r^{2}=0.80)$ during ebb tide. Relatively fine grained and well defined, monotonous size distribution may promote the correlation between concentration and beam attenuation coefficient due to optical homogeneity of particles during ebb tide. Abundance of large aggregates with time-varying size and shape distributions may be mainly responsible for variations in optical properties of the sediment during flood tide, and thus may confound the relationship between the two variables. The difference in particle sizes and shapes between flood and ebb tides can also be observed on SEM images.

Gene Expression Profiles of Rainbow Trout Oncorhynchus mykiss after Salinity Challenge (염분 변화에 따른 무지개송어(Oncorhynchus mykiss)의 삼투조절 유전자 발현변화)

  • Euryhaline teleost have extraordinary ability to deal with a wide range of salinity changes. To study the seawater adaptability of rainbow trout Oncorhynchus mykiss (body weight 638±54 g, length 38.6±2 cm) to salinity increase fish were transferred from freshwater to 7, 14, 21, 28 and 32 psu and checked for mortality over 5 days. No mortality was observed in 0-32 psu. In fish transferred to 0-32 psu, blood osmolality was maintained within physiological range. The changes of serum enzyme activities (aspartate transaminase, AST and alanine transaminase, ALT) showed no significant level during experimental period. To explore the underlying molecular physiology of gill and kidney responsible for body fluid regulation, we measured mRNA expression of five genes, Na+/K+/2Cl- cotransporter1 (NKCC1), aquaporin3 (AQP3), cystic fibrosis transmembrane conductance regulator (CFTR), glucocorticoid receptor (GR) and growth hormone receptor (GHR) in response to salt stress. Based on our result, rainbow trout could tolerate gradual transfer up to 32 psu for 5 days without mortality under physiological stress. This study suggests to alleviate osmotic stress to fish, a gradually acclimation to increasing salinity is recommended.

Effects of different heating conditions on protein composition in each muscle type of yellowtail (Seriola quinqueradiata)

  • To clarify the factors influencing the physical properties of fish after heat treatments, we investigated changes in the properties of proteins in the dorsal ordinary and dark muscle of yellowtail (Seriola quinqueradiata) heated under different conditions commonly used for the purposes of food hygiene. High-temperature/short-time heating (85℃ for 90 s and 75℃ for 60 s) affected the protein solubility more than low-temperature/long-time heating (63℃ for 30 min). Sodium dodecyl sulphate-polyacrylamide gel electrophoresis and differential scanning calorimetry showed that low-temperature/long-time heating reduced the degree of actin denaturation in fish compared with that by other heating conditions. In addition, collagen solubility was enhanced with low-temperature/long-time heating. Therefore, these results suggest that differences in the degree of actin and collagen denaturation are responsible for the enhanced meat tenderness and diminished meat shrinkage, resulting from low-temperature/long-time heating.

A Study on the Meteorological Disaster of Fisheries and Ocean Institution in Jeju Island (제주도 수산해양시설의 기상재해에 관한 연구)

  • The typhoon, heavy rain, blizzard, storm and heavy snowfall had the main caused of natural disasters occurred in Korea from 1993 to 2002. Among them, typhoon has responsible for biggest disaster, recording about 47.4% of total economic damage. Typhoons concentrated mostly in the months from June to October. The average occurrence number in those months ranged from 3.9 to 5.5 based on 30 years of record(1971-2000). However, the numbers increased from 4.0 to 6.2 during the most recent 10 years(1991-2000). Jeju province, among others in Korea, was most frequently affected by typhoon which occurred 23 times during the period of 1991-2000. Typhoons which occurred from July to early August have passed mostly through the west of Jeju, whereas, those of late August to September have passed through the center and eastern sea area of Jeju. The typhoons 'Ramasun' and 'Rusa' caused severe damage in Jeju area in September 2003 and the surge heights were +39cm and +77cm, respectively. The main cause of the damage was surge height which was highly associated with the tidal phase at the time of typhoon passage. The damage caused by typhoon on the aquaculture, fishing boat and harbor cosatline wall around Jeju Island which was amounted to be 417 billion won(\) during the recent 3 years(2002-2004)
