Respiratory burst activity of tilapia (Oreochromis mosambicus) phagocytes to Edwardsiella tarda ghosts

  • The respiratory burst activity of tilapia (Oreochromis mosambicus) phagocytes agaisnt Edwardsiella tarda ghosts produced by gene E mediated lysis was investigated. Stimulation with E. tarda ghosts (ETG) showed markedly higher respiratory burst activity than that with formalin killed E. tarda (FKC) in phagocytes. This result may suggest that ETG can induce effective cellular immune responses in fish.

  • While respiratory burst enhances neutrophil glucose utilization, many neutrophil functions are critically influenced by extracellular matrix interaction and phosphoinositide-3-OH kinase (PI3K) signaling. We thus evaluated the role of RGD integrin occupancy and PI3K inhibition on respiratory burst and [18F]FDG uptake of stimulated neutrophils. Human neutrophils were stimulated by 100 ng/mL phorbol-myristate-acetate (PMA), and respiratory burst was measured by cumulative luminescence with lucigenin. [18F]FDG uptake and total hexokinase activity was measured 20 min after PMA stimulation in the presence or absence of soluble RGD peptides (200 g/mL) and/or the PI3K inhibitor wortmannin (200 nM). PMA induced a 71.70.9 fold increase in neutrophil oxygen intermediate generation. [18F]FDG uptake was increased to $194.6{\pm} 3.7%$ and hexokinase activity to $145.0{\pm}2.0%$ of basal levels (both p<0.0005). RGD peptides attenuated respiratory burst activation to $35.6{\pm}0.2%$ (p<0.005), but did not inhibit stimulated [18F]FDG uptake or hexokinase activity. In contrast, without affecting respiratory burst activation, wortmannin inhibited PMA stimulated [18F]FDG uptake to $66.9{\pm}1.6%$ and hexokinase activity to $81.0{\pm}4.2%$ (both P<0.0005), demonstrating its dependence on PI3K activity. Neither RGD nor wortmannin reversed the other's inhibitory effect on stimulated [18F]FDG uptake and hexokinase activity or respiratory burst, which suggests the involvement of distinct signaling pathways. Neutrophil [18F]FDG uptake is enhanced by PMA through a mechanism that requires PI3K activity but is independent of integrin receptor occupancy or respiratory burst activation.

  • 연구배경: 말초혈액내 백혈구의 약 60%를 차지하는 호중구는 세균 및 진균 감염시 일차 방어기전으로 염증부위에 모여 세균 및 진균을 탐식하고 이때 활성화된 호중구는 산화율을 생성하고 이와 함께 탈과립된 효소 등에 의해서 살균작용이 이루어진다. Respiratory burst에 의하여 생성된 산화물은 감염에서의 살균 작용 외에, 염증반응의 중추 역할을 하는 물질로서 조직손상, 약물대사, 노화, 발암과정에 관여하여 각종 질환과의 연관성을 제시하고 있다. 본 연구는 폐렴환자에 있어서 폐렴의 경과 중에 중요한 역할을 하는 호중구로부터 산화물의 생성능력을 보기 위해 Respi ratory burst 활성도를 측정하였다. 방법: 건강인 24명과 폐렴으로 진단된 24명의 환자를 대상으로 혈액내 호중구의 respiratory burst 활성도를 입원 1일, 3일, 5일, 7일, 9일째에 자극하지 않은 상태 및 fMLP(formyl- MethionylLeucyl-Phenylalanine)와 PMA(phorbol myristate acerate)로 자극한 상태에서 DCF-DA를 이용한 유세포분석법으로 측정하였다. 결과: 정상인 호중구는 생리적 자극제인 fMLP에 대해 통계학적으로 의미는 없지만 약간 높은 활성도를 보였고 비생리적 자극제인 PMA에 대해서는 통계학적으로 의미있는 높은 활성도를 나타냈다. 그러나 폐렴환자의 호중구는 PMA에 자극은 되었으나 정상인 호중구보다 활성도가 덜 증가하였다(p<0.01). 호중구의 respiratory burst 활성도를 폐렴과 정상인에서 비교할 때, 자극 전에는 양군간에 차이가 없었고 fMLP로 자극한 경우에도 의미있는 차이는 없었다. 그러나 PMA로 자극한 경우 내원 1일부터 10일까지 폐렴환자는 정상인에 비해 자극에 대한 respiratory burst 활성도의 증가폭이 통계적으로 의미있게 폐렴 초기에 저하되었다가 치료가 진행됨에 따라 정상화되는 양상을 보였다. 결론: 폐렴으로 진단된 환자의 치료과정 중 혈중 호중구로부터의 과산화수소 생성능은 항생제 투여전에는 감소된 상태였으며 이후 치료 개시와 함께 점차 증가하여 9일째에는 정상인 수준으로 회복되었다.

  • The activity of the phagocyte respiratory burst oxidase is regulated by complex and dynamic alterations in protein-protein interactions that result in the rapid assembly of an active multicomponent NADPH oxidase enzyme on the plasma membrane. While the enzymatic activity has been studied for the past 20 years, the past decade has seen remarkable progress in our understanding of the enzyme and its activation at the molecular level. This article describes the current state of knowledge, and proposes a model for the mechanism by which protein-protein interactions regulate enzyme activity in this system.

Effects of Dietary Cadmium on the Respiratory Burst of Phagocytes and the Antioxidant Defense in Cultured Red Seabream (Pagrus major)

  • To examine effects of cadmium on the respiratory burst of kidney phagocytes and antioxidant defense in liver, juvenile red seabream Pagrus major were fed a cadmium-incorporated diet $(1g\;CdC1_2/kg\;diet)$. The respiratory burst activity measured by chemiluminescence (CL) was significantly reduced by oral intake of cadmium. Lipid peroxidation in liver expressed as thiobarbituric acid reactive substances (TBARS) was significantly higher in the fish fed a cadmium-incorporated diet than that of the fish fed a control diet both on Day 3 and Day 9. Liver Glutathione S-transferase (GST) activitiy was significantly increased both on Day 3 and Day 9 by feeding a cadmium-incorporated diet, when compared with the controls. From the present results, it can be concluded that oral intake of cadmium in red seabream is associated with marked reduction of respiratory burst capacity of kidney phagocytes which can elevate susceptibility of fish against infecting pathogens. Cadmium administration also elicits significant increment of lipid peroxidation in liver, and fish try to detoxify cadmium by increasing GST activity.

알로에가 in vitro에서 넙치 백혈구의 호흡폭발에 미치는 영향 (In vitro Effect of Aloe on the Respiratory Burst Activity of Olive Flounder (Paralichthys olivaceus) Leucocytes)

  • 알로에가 in vitro에서 넙치의 비특이적 면역반응에 미치는 영향을 조사하기 위하여 신장으로부터 분리된 백혈구의 호흡폭발을 chemiluminescent(CL) 반응을 이용하여 측정하였다. 알로에 분말을 0.001, 0.01, 0.1, 0.5, 1.0, $10.0{\mu}g/ml$의 농도로 MEM에 희석하여 넙치의 식세포와 함께 배양한 후 3, 6, 12, 24, 48시간째에 CL 반응을 이용하여 호흡폭발능을 측정한 결과, 알로에 $0.5{\mu}g/ml$$1.0{\mu}g/ml$의 농도에서 24시간 동안 배양한 실험구만이 알로에를 첨가하지 않은 대조구에 비해서 유의성 있게 호흡폭발능이 증가하였다. 이러한 결과는 알로에가 양식 넙치의 비특이적 면역능을 증강시킬 수 있음을 시사하며, 앞으로 in vivo 실험을 통한 검증이 필요할 것으로 사료된다.

The Effects of Acute Osmotic Stress on Innate Immunity of Nile Tilapia (Oreochromis niloticus)

  • The effects of osmotic stress on the non-specific immune response of Nile tilapia, Oreochromis niloticus, were investigated. Osmoregulatory mechanism of tilapia has been studied, but less information is available about innate immune response of O. niloticus faced with hyperosmolality. Acute osmotic stress was elicited by transferring tilapia from freshwater (FW) to 24 psu seawater (SW). Non-specific immune parameters including lysozyme activities of plasma and head kidney (HK), alternative complement pathway (ACP) activity in plasma, phagocytic capacities of spleen and HK immune cells, and respiratory burst activity of immune cells in both HK and spleen were analyzed. Lysozyme activities were increased at 1 h and 30 h after transfer to SW, but decreased at 10 h after SW transfer. Conversely, ACP activity increased 10 h after SW transfer. Phagocytic capacity increased slightly at 1 h and 5 h after SW transfer, and respiratory burst activity showed an increase in superoxide release at 10 h after SW transfer. Taken together, these results indicate that the exposure of tilapia to hyperosmotic conditions has immunostimulatory effects on cellular and humoral immune reactions.

  • The information obtained from the analysis of respiratory muscle elecromyographic (EMG) activities provides a mean for studying muscular activity in relation to the ventilatory process. Thus, in order to comprehend the airflow pattern and its brain control, signal processing is required to characterize respiratory muscle activity. This paper presents a computerized method for the analysis of the electrical activity of the respiratory muscles of premature lambs, and focuses upon the automatic determination of respiratory timing points such as onset and cessation points of the burst activity. Based on experimental results, reliable timing points can be obtained using the proposed methodology.

Non-specific Immune Response of Rainbow Trout (Oncorhynchus Mykiss) by Dietary Heat-inactivated Potential Probiotics

  • Background: The effects of the dietary administration of two heat-inactivated whole bacteria from the Vibrionaceae family, singly or combined, on innate immune response of the rainbow trout were studied. The two bacteria (Pdp11 and 51M6), which were obtained from the skin of rainbow trout, showed in vitro characteristics that suggested they could be considered as potential fish probiotics. Methods: The fish were fed four different diets: control (non-supplemented), or diets supplemented with heat-inactivated bacteria at $10^8$ cfu/g Pdp11, $10^8$ cfu/g 51M6 or with $0.5{\times}10^8$ cfu/g Pdp11 plus $0.5{\times}10^8$ cfu/g 51M6 for 4 weeks. Six fish were sampled at weeks 1, 2, 3 and 4, and then the main humoral (natural haemolytic complement activity and serum peroxidase content) and cellular innate immune responses (leucocyte peroxidase content, phagocytosis, respiratory burst and cytotoxicity) were evaluated. Results: The serum peroxidase content and the natural haemolytic complement activity increased with time, reaching the highest values in the third and fourth weeks of feeding, respectively. The phagocytic ability of specimens fed the mixture of the two inactivated bacteria was significantly higher than in the controls after 2 and 3 weeks of treatment. The same activity increased significantly in rainbow trout fed the Pdp11 diet for 2 weeks or the 51M6 diet for 3 weeks. Respiratory burst activity was unaffected by all the experimental diets at all times assayed. Cytotoxic activity had significantly increased after 3 weeks in fish fed the 51M6 diet. Conclusion: Our results demonstrated the usefulness of incorporating inactivated probiotic bacteria into fish diets.
