Molecular Cloning and Characterization of Chitosanase Gene from Bacillus amyloliquefaciene MJ-1 (Bacillus amyloliquefaciens MJ-1 유래의 chitosanase 유전자의 클로닝 및 특성)

  • In order to develop chitosanase for the production of chitosan oligosaccharides, a chitosanase-producing bacterium was isolated from the traditional fermented soybean, Meju, and identified as Bacillus amyloliquefaciene MJ-1. The cloned chitosanase gene, 825 bp in size, encoded a single peptide of 274 amino acids with a estimated molecular mass of 30.9 kDa. The deduced amino acid sequence showed significant homology with microbial chitosanases. The recombinant chitosanase was expressed in Escherichia coli upon induction with isopropyl-D-thiogalactopyranoside, and purified using $Ni^{2+}-NTA$ agarose column chromatography. The maximal activity of the recombinant chitosanase is at pH 5.0 and $60^{\circ}C$. The recombinant chitosanase is stable between pH 5.0 and pH 7.0 at $37^{\circ}C$ for 30 min, and more than 75% of the activity still remain at $80^{\circ}C$ for 30 min incubation.

  • The iSTMS is a web-based integrated information system supporting Pre-Service and In-Service Inspection(PSI/ISI) processes for the nuclear power plants of KHNP(Korea Hydro & Nuclear Power Co. Ltd.). The system provides a full spectrum coverage of the inspection processes from the planning stage to the final report of examination in accordance with applicable codes, standards, and regulatory requirements. The major functions of the system includes the inspection planning, examination, reporting, project control and status reporting, resource management as well as objects search and navigation. The system also provides two dimensional or three dimensional visualization interface to identify the location and geometry of components and weld areas subject to examination in collaboration with database applications. The iSIMS is implemented with commercial software packages such as database management system, 2-D and 3-D visualization tool, etc., which provide open, updated and verified foundations. This paper describes the key functions and the technologies for the implementation of the iSIMS.

The Study on the Characteristics of Technology Innovation Activities of High Growth Firms (고성장기업의 기술혁신활동 특성에 대한 연구)

  • This paper explores the characteristics of innovation activities in high growth firms that contribute to national and regional economic growth and job creation. The analysis is based on the 2016 KIS data to analyze the difference in innovation activities between high growth firms and general firms. The main results are as follows: First, high growth firms have a higher proportion of R&D personnel than general firms. Second, high growth firms are actively introducing product innovation, process innovation, and organizational innovation as compared to general firms. In the innovation activities related to product innovation and process innovation, there is no statistically significant difference between high growth companies and general companies except for external R&D. Third, High growth firms are more likely to cooperate with other technology partners than general firms. But, there is no statistically significant difference between high growth firms and general firms in the external knowledge search and the diversity of cooperating partners. Fourth, in terms of protecting innovation, high growth firms are more likely to use all kinds of innovation protection method, such as 'utilizing intellectual property rights', 'maintaining confidentiality', 'adopting complex design methods', 'market preemption ahead of competitors', and the most important means is the intellectual property rights. Fifth, government innovation policies that high growth firms chose as important are 'innovation subsidies and loans', 'acquirement, utilization and protection of intellectual property rights' and 'human resource support'.

A Study on the Effectiveness of Rehabilitation by Virtual Reality Program: Systematic Review (가상현실 프로그램을 사용한 재활치료의 효과성 연구: 체계적 고찰)

  • The purpose of this study is to provide a basic data about the virtual reality rehabilitation therapy to the occupational therapist in the domestic clinical field by systematically analyzing the types and effects of the rehabilitation therapy using the virtual reality program. Research articles were retrieved from January 2007 to December 2017 using the Research Information Sharing Service (RISS), Google Scholar, and Pubmed database. The main search terms were 'Virtual Reality AND Rehabilitation' and 'Virtual reality AND Effect'. Finally, 10 foreign studies were selected as the subjects of this study. Selected studies were organized using PICO (Patient, Intervention, Comparison, Outcome) format. The results were as follows: The subjects were divided into 4 types, stroke, brain tumor, Parkinson 's disease, and kidney disease. The tools used for arbitration were three IREX (30 %), two X-box Kinect (20 %), two Nintendo Wii (20 %), one [Existing tool + VR] (10 %), one [Mobile application + VR] (10 %). As a result of the intervention, improvement of function was observed in all 10 studies from overseas, and physical function was 24 times (66 %), mental function 6 times (17 %), cognitive function 5 times (14 %), Activity of daily living 1 time (10 %). this study could be used as a basic resource to enhance the professionalism and quality of rehabilitation services and expand the scope by organizing virtual reality-based rehabilitation and its effects.

Effects of an Anti-cancer Drug, Tubastatin A, on the Growth and Development of Immature Oocytes in Mice (항암제 tubastatin A에 의한 생쥐 미성숙 난모세포의 성장과 발달에 미치는 효과)

  • In recent years, progress has been made in the search for the development of new anti-cancer agents by employing specific inhibitors of histone deacetylase (HDAC)-6 to block signal transduction pathways in cancer cells. This study examined the effects of tubastatin A (TubA), an HDAC-6 inhibitor, on the growth and development of immature oocytes in murine ovaries using RNA sequencing analysis. The results from a gene set enrichment analysis (GSEA) indicated that the expression of most of the gene sets involved in the cell cycle and control and progression of meiosis decreased in the TubA-treated group as compared with that in germinal vesicle (GV) stage oocytes. In addition, an ingenuity pathway analysis (IPA) suggested that TubA not only caused increased expression of p53 and pRB and decreased expression of CDK4/6 and cyclin D but also caused elevated expression of genes involved in the control of the DNA check point in G2/M stage oocytes. These results suggest that TubA may induce cell cycle arrest and apoptosis through the induction of changes in the expression of genes involved in signal transduction pathways associated with DNA damage and the cell cycle of immature oocytes in the ovary.

Current Status and Prospect of Seaweed-based Biofuels as Renewable Energy Resource (재생가능 에너지원으로서의 해조류 유래 바이오 연료의 현황과 전망)

  • Research and development of biofuels as one of the means to mitigate global warming and to avoid fossil fuel depletion has occurred for more than 30 years. However, there has only been limited distribution of a few first- and second-generation biofuels, and widespread supply and consumption of biofuels is still far from a reality. Although a relatively recently studied third-generation biofuel derived from seaweed biomass has been shown to have many advantages, it is yet to be deployed in commercial-scale seaweed biorefineries. This review paper examines the advantages and disadvantages of seaweed biorefineries for the entire value chain covering from seaweed and its cultivation to biofuel production based on an extensive literature search and the author's experience of conducting feasibility studies pertaining to seaweed biorefineries for over 10 years. For this purpose, the literature survey will cover the current status of seaweed production and its research and development worldwide, conversion technologies for biofuel production from seaweed based on bench-scale experiments, and large-scale techno-economic feasibility studies for seaweed conversion to biofuels and bioenergy. In addition, the main problems expected with the commercialization of seaweed-based biofuels will be identified. Finally, the current status of seaweed biorefinery technology and the author's views on its promising future will be summarized.

Development and Utilization of Linked Data of Port Maintenance Information for Port Facilities Based on Port BIM Standards (항만 BIM 표준 기반 항만 유지관리 정보의 링크드데이터 구축 및 활용)

  • The importance of using construction data is increasing in accordance with the recent trend in the smart construction. However, construction project and maintenance information is distributed on the web, and the existing BIM(Building Information Modeling) information exchange and linking method using IFC(Industry Foundation Classes) cannot support connection with BIM data and web resources. This study aims to establish the BIM-based port facility data integration system using linked data(LD) technology in order to integrate BIM and heterogeneous data in the port maintenance domain. To this end, the port BIM-based ifcOWL and port facility maintenance ontology were designed, and LD was built for the BIM and maintenance information of Busan New Port 2-1 Pier3, a BIM pilot project. In addition, service prototypes such as search, statistics and SPARQL(SPARQL Protocol and RDF Query Language) endpoint functions were implemented using the issued LD. The LD-based information utilization system is expected to improve the reusability of information by converting the existing closed information system into an open system and BIM and maintenance data as a web resource in a standard format.

Development of Native Local Foods in Chungcheongnam-do by Storytelling (스토리텔링을 통한 충남향토음식 개발 연구 - '무령왕 수라상'과 '몽유도원 밥상'을 중심으로 -)

  • The objective of this study was to familiarize people with the native local food of Chungnam Province by developing its regional food as a culture resource. Native Local Food Search and luxury itemization based on regionally-characterized stories enhances self-perception of the national culture, promotes appropriate local images to the public, and contributes to the local economy by increasing regional tourism. Therefore, this study researched local stories of cultural significance, that is, those connected to the history and originality of Chungnam Province, and developed contents related to Chungnam native local food. Features of the native local food were introduced by a story telling method in order to appeal to the five senses. The story was composed for easy understanding of the value of food, and the brand image of Chungnam was developed based on representative historical stories of the region. In this study, the following were developed as representative images of Chungnam: 'Royal meal table of King Moo-ryung' in Kongju was presented by recomposing the story of King Moo-ryung, a famouns king of the Baekje era; 'Mong-yoo-do-won's rice meal table' was introduced via Mongyoo-do-won's painting by Kyeon An, a famous painter of the Chosun era who was born in Seosan. The 'Royal meal table of King Moo-ryung' was set with food made from local farm products, demonstrating the flavor and elegance of the Baekje era. 'Mong-yoo-do-won's rice meal table' resembled Kyeon An's Mong-yoo-do-won-do, which has a background featuring early spring. The rice table was designed to remind people of a flamboyant painting like a scattered peach blossom leaf in the early spring. To verify the health effectiveness of each rice table, the function of each ingredient was investigated through 'Sik-ryo-chan-yo:a dietary treatment', which was published by Soon-Ui Cheon during the Chosan era. According to the results, most of ingredients are certainly beneficial to health, as was recognized in the early Choson era.

  • This study is a study on the methodology of establishing an innovation strategy centering on the customer experience, which is essential in order to transform the existing collection and preservation-centered mutual aid company service into a visitor-centered service. To this end, we conducted literature research on environmental changes in the funeral industry from the perspective of service science and the significance and value of customer experiences within them, good customer experiences and bad customer experiences from the perspective of customer experience management. A study was conducted to present and prove a specific model. The customer experience-oriented innovation strategy of the funeral industry means to search for various alternatives that can reach the target state from the present state, focusing on the customer, and select the most appropriate transformation plan among them. As an effect of application, it was found that it is a source of differentiation by generating positive emotions to customers, and that customer experience data is highly helpful in making important decisions for the actual resource input of the parent company. This innovation model was presented, and its value was firstly proved by analyzing the difference from the existing evaluation method. Finally, as a result of analyzing the causal relationship through regression analysis using the customer experience measurement procedure, customer experience diagnosis/evaluation, customer experience innovation strategy, and cooperative company's performance as variables, the relationship proved to be significant.

Federated learning-based client training acceleration method for personalized digital twins (개인화 디지털 트윈을 위한 연합학습 기반 클라이언트 훈련 가속 방식)

  • Digital twin is an M&S (Modeling and Simulation) technology designed to solve or optimize problems in the real world by replicating physical objects in the real world as virtual objects in the digital world and predicting phenomena that may occur in the future through simulation. Digital twins have been elaborately designed and utilized based on data collected to achieve specific purposes in large-scale environments such as cities and industrial facilities. In order to apply this digital twin technology to real life and expand it into user-customized service technology, practical but sensitive issues such as personal information protection and personalization of simulations must be resolved. To solve this problem, this paper proposes a federated learning-based accelerated client training method (FACTS) for personalized digital twins. The basic approach is to use a cluster-driven federated learning training procedure to protect personal information while simultaneously selecting a training model similar to the user and training it adaptively. As a result of experiments under various statistically heterogeneous conditions, FACTS was found to be superior to the existing FL method in terms of training speed and resource efficiency.
