The mainstay therapy against leishmaniasis is still pentavalent antimonial drugs; however, the rate of antimony resistance is increasing in endemic regions such as Iran. Understanding the molecular basis of resistance to antimonials could be helpful to improve treatment strategies. This study aimed to recognize genes involved in antimony resistance of Leishmania tropica field isolates. Sensitive and resistant L. tropica parasites were isolated from anthroponotic cutaneous leishmaniasis patients and drug susceptibility of parasites to meglumine antimoniate (Glucantime$^{(R)}$) was confirmed using in vitro assay. Then, complementary DNA-amplified fragment length polymorphism (cDNA-AFLP) and real-time reverse transcriptase-PCR (RT-PCR) approaches were utilized on mRNAs from resistant and sensitive L. tropica isolates. We identified 2 known genes, ubiquitin implicated in protein degradation and amino acid permease (AAP3) involved in arginine uptake. Also, we identified 1 gene encoding hypothetical protein. Real-time RT-PCR revealed a significant upregulation of ubiquitin (2.54-fold), and AAP3 (2.86-fold) (P<0.05) in resistant isolates compared to sensitive ones. Our results suggest that overexpression of ubiquitin and AAP3 could potentially implicated in natural antimony resistance.
The uplift capacity of a pile for improving the wind resistance of the 1-2 W type plastic film pipe on greenhouses was tested using the plane and corrugated piles with various shapes and diameters. First, the resistant uplift capacity was measured by using the uplift loading on plane piles. As the uplift loading on plane piles increased, the resistant uplift capacity also increased until the loading was reached to ultimate uplift capacity. After ultimate uplift capacity was appeared the uplift displacement, the uplift capacity was decreased gradually. Secondly, the resistant uplift capacity was measured by using the uplift loading on corrugated piles. After the uplift capacity was reached the uplift displacement, the uplift capacity was continually increased or decreased. In general, the ultimate uplift capacity was independent of pile shapes, pile diameter length, and embedded pipe depth. However, the ultimate uplift capacity of a corrugated pile was twice more than that of a plane pile without regard to its diameter and embedded depth. The ultimate uplift capacity per unit pile area was increasing in deeper embedded depth. However, the longer a pile diameter was, the less ultimate uplift capacity. The uplift capacity of a plane pile, used in conjunction with the design wind velocity (26.9m.s$^{-1}$ ) of the project area, was unsatisfiable without regard to diameters and embedded depths of piles, while most of corrugated piles were well appeared uplift capacity under various experimental conditions.

The outbreaks of blast, bacterial blight and viral diseases have been increasing in early maturing rice cultivating areas in the central northern regions, recently. As the occurrence of sudden insects pests and disasters increases due to global climate warming, it is urgent to develop a variety of disaster-tolerant, high-quality varieties in response. This study was carried out to elucidate the characteristics of early-maturing, high-quality and multiple disease resistant rice variety, Cheolweon109 that was adapted to cultivation in the mid-mountainous regions of the central northern regions. Cheolweon109 was derived from a cross between Suweon546, medium maturing variety, and Sangju44 which is early maturing and resistant to blast, bacterial blight and rice stripe virus. The heading date of Cheolweon109 was July 30, 3 days later than Odae. The culm length of Cheolweon109 was 79 cm, which was about 5 cm taller than Odae, and the ripening ratio was 85.1%, which was 10% higher than that of Odae. This variety had 5.54 MT/ha of milled rice productivity, which was 99% of the Odae. Although Cheolweon109 was tall, it was strong against lodging. It was strong against bacterial blight (K1, K2, K3 race), rice stripe virus, and the pre-harvest sprouting which rate was 2.4%. The appearance of the grains of rice was clean, the glossiness was 70.6, and the head rice ratio was 95.3% high. Because Cheolweon109 had superior disease resistance, disaster resistance, and high quality than Odae, it was expected that can be used to expand the diversity of early maturing and high-quality rice varieties in central northern regions.

Breed differences in adult animals are determined during fetal development. If interventions are to be developed that influence growth of muscle and fat, it is important to know at which time during gestation breed differences appear and are fixed. The objective of this study was to characterize fetal development in cattle of different breeds. Pregnant cows of 4 cattle breeds with different growth impetus and muscularity were slaughtered under normal processing conditions and the fetuses were removed. German Angus, a typical beef cattle; Galloway, a smaller, environmentally resistant beef type; Holstein Friesian, a dairy type; and Belgian Blue, an extreme type for muscle growth were used. Fetuses of each breed were investigated at 3, 6, and 9 mo of gestation. Fetuses were weighed and dissected into carcass, organs, and muscles. Body fat weight was obtained using the Soxhlet extraction method. Fetal weight increased most rapidly in the third trimester of gestation mainly due to the accelerated muscle and fat deposition. The organ weight to body weight (BW) ratios decreased and the muscle and fat weight to BW ratios increased. At 3 mo of gestation, Galloway fetuses had the significantly smallest BW, half-carcass weight, leg weight, organ weight, muscle weight and shortest leg length. In contrast, Holstein fetuses had the significantly greatest BW, liver, kidney, and lung weights and significantly longest leg length among the 4 breeds, but no differences between Holstein Friesian and Belgian Blue were detected in half-carcass and leg weight. Indeed, Belgian Blue fetuses had the significantly greatest half-carcass weight, leg weight, and muscle weight at 9 mo of gestation, and Galloway had a significantly greater body fat to BW ratio than Holstein Friesian and Belgian Blue. These differences were not evident at 3 and 6 mo of gestation. These data show that the profound increase of tissue and organ weights occurred in later gestation in cattle fetuses even though breed differences were evident as early as 3 mo of gestation. Depending on the tissue of interest, impacting fetal growth likely needs to occur early in gestation before the appearance of breed-specific differences.
The inheritance and varietal differences of resistance to bacterial leaf blight caused by Xanthomonas oryzae in rice were studied. Among eighteen cultivars used, 'IR2061-465-1-5-5', 'IR2061-552-6-9', 'IR 1561-228-9-3', ana 'Milyang42, were found to have a high level of resistance to the three isolates of X. oryzae at maximum tillering and flowering stage. Varietal differences in lesion development of the cultivars belonging to the same varietal group were clearly recognized. For the genetic study on bacterial leaf blight resistance, two rice breeding lines 'IR2061-465-1-5-5', 'IR1561-228-3-3' were crossed with susceptible cultivar 'Milyang 23'. The $F_1,F_2,\;and\;F_3$ progenies were evaluated at flowering stage using the bacterial isolate 'JN 7853'(II). Average lesion length of the $F_1$ plants of the crosses showed longer than that of resistant parents, and the frequency distribution of lesion length in the $F_2\;and\;F_3$ populations showed continuous variation.
Helices 4 and 5 of the Bacillus thuringiensis Cry4Ba $\delta$-endotoxin have been shown to be important determinants for mosquito-larvicidal activity, likely being involved in membrane-pore formation. In this study, the Cry4Ba mutant protein containing an additional engineered tryptic cleavage site was used to produce the $\alpha4$-$\alpha5$ hairpin peptide by an efficient alternative strategy. Upon solubilization of toxin inclusions expressed in Escherichia coli and subsequent digestion with trypsin, the 130-kDa mutant protoxin was processed to protease-resistant fragments of ca. 47, 10 and 7 kDa. The 7-kDa fragment was identified as the $\alpha4$-loop-$\alpha5$ hairpin via N-terminal sequencing and mass spectrometry, and was successfully purified by size-exclusion FPLC and reversed-phase HPLC. Using circular dichroism spectroscopy, the 7-kDa peptide was found to exist predominantly as an $\alpha$-helical structure. Membrane perturbation studies by using fluorimetric calcein-release assays revealed that the 7-kDa helical hairpin is highly active against unilamellar liposomes compared with the 65-kDa activated full-length toxin. These results directly support the role of the $\alpha4$-loop-$\alpha5$ hairpin in membrane perturbation and pore formation of the full-length Cry4Ba toxin.
Ganciclovir resistance of human cytomegalovirus is associated with mutations in the viral UL97 gene and poses severe problems for immunocompromised patients. In this study, PCR-based restriction fragment length polymorphism and sequencing analyses detected the UL97 D605E mutation in all five clinical isolates from patients with ganciclovir-resistant human cytomegalovirus infection during prolonged ganciclovir therapy, whereas the M460V mutation was only present in 1 of 5 isolates. On the other hand, the detection rates of the D605E mutation in the stored available DNA samples from the donor and allogeneic stem cell transplantation recipients were 66.7% and 93.7%, respectively, suggesting that the presence of D605E mutation was not associated with the ganciclovir exposure. Although the D605E mutation may not be related to ganciclovir resistance, we suggest that this mutation could be an important molecular marker of human cytomegalovirus evolution in East Asian countries. Moreover, the restriction fragment length polymorphism method using the restriction enzyme HaeIII, which is generally used to detect the UL97 A591V mutation, could also detect the D605E mutation and may therefore be a useful tool for future research on the investigation of UL97 gene mutations.

Steel plate shear walls are recently used as efficient seismic lateral resisting systems. These lateral resistant structures are implemented to provide more strength, stiffness and ductility in limited space areas. In this study, the seismic behavior of the multi-story steel frames with steel plate shear walls are investigated for buildings with 4, 8, 12 and 16 stories using verified computational modeling platforms. Different number of steel moment bays with distinctive lengths are investigated to effectively determine the deflection amplification factor for low-rise and high-rise structures. Results showed that the dissipated energy in moment frames with steel plates are significantly related to the inside panel. It is shown that more than 50% of the dissipated energy under various ground motions is dissipated by the panel itself, and increasing the steel plate length leads to higher energy dissipation capability. The deflection amplification factor is studied in details for various verified parametric cases, and it is concluded that for a typical multi-story moment frame with steel plate shear walls, the amplification factor is 4.93 which is less than the recommended conservative values in the design codes. It is shown that the deflection amplification factor decreases if the height of the building increases, for which the frames with more than six stories would have less recommended deflection amplification factor. In addition, increasing the number of bays or decreasing the steel plate shear wall length leads to a reduction of the deflection amplification factor.
The selected five plant growth-promoting rhizobacteria (PGPR) strains, WR8-3 (Pseudomonas fluorescens), WR8-6 (P. putida), WR9-9 (P. fluorescens), WR9-11 (Pseudomonas sp.), and WR9-16 (P. putida) isolated in the rhizosphere of watermelon plants were tested on their growth promotion and control effect against gummy stem rot of watermelon. Strains, WR8-3 and WR9-16 significantly increased stem length of watermelon, and there was a little increase in leaf area, fresh weight and root length when strains, WR8-3, WR9-9 and WR9-16 were treated. Generally, seed treatment was better for plant growth promotion than the soil drench, but there was no significant difference. Seed treatment and soil drench of each bacterial strain also significantly reduced the mean lesion area (MLA) by gummy stem rot, but there was no significant difference between the two treatments. At initial inoculum densities of each strain ranging from 10$^6\;to\;10^{15}$ cfu/g seed, approximately the same level of disease resistance was induced. But resistance induction was not induced at the initial inoculum density of 10$^3$ cfu/g seed. Resistance was induced by treating the strains, WR9-9, WR9-11 and WR9-16, on all of four watermelon varieties tested, and there was no significant difference in the decrease of gummy stem rot among varieties. Populations of the strains treated initially at log 9-10 cfu/g seed, followed with a rapid decrease from planting day to 1 week after planting, but the population density was maintained above log 5.0 cfu/g soil until 4 weeks after planting. Generally no or very weak in vitro antagonism was observed at the strains treated excepting WR9-11. Rifampicin-resistant bacteria which had been inoculated were not detected in the stems or leaves, which suggesting that the bacterium and the pathogens remained spatially separated during the experiment. This is the first report of rsistance induction in watermelon to gummy stem rot by PGPR strains.
DNA marker is a powerful tool for plant genetics and breeding. In this study, 995 SSR markers were employed with chilling resistant cucumber, known as 'NC76', and chilling susceptible cucumber, known as 'GY14'. Using 2% agarose gel electrophoresis, 145 SSR markers were identified as length variation markers between 'NC76' and 'GY14'. The SSR markers that showed no length polymorphism were then screened using high resolution melting analysis technique and additional 30 polymorphic SSR markers were identified. As a preliminary evaluation for mapping, 20 markers among these 175 markers were employed to a $F_2$ population of 'NC76' x 'GY14' cross. Linkage analysis revealed 13 markers that joined into six linkage groups and seven markers that remained unlinked. This result indicates that these 175 markers could be used for construction of a genetic map using a cross between 'NC76' and 'GY14' for further investigation in developing markers related to resistance to chilling in cucumbers.
