Solvent Extraction과 Gel Chromatography를 이용한 Phenoxyethanol Galactoside와 Chlorphenesin Galactoside의 정제 (Purifications of Phenoxyethanol Galactoside and Chlorphenesin Galactoside using Solvent Extraction followed by Gel Chromatography)

  • 재조합 ${\beta}$-galactosidase (${\beta}$-gal)을 이용하여 transgalactosylation 반응으로 합성된 2-phenoxyethanol galactoside (PE-gal)과 chlorphenesin galactoside (CPN-gal)의 정제를 실시하였다. 먼저 PE와 PE-gal이 포함된 반응물에 ethyl acetate (EA)를 넣고, EA/water 이상계 시스템에서 PE와 PE-gal을 분획하였다. 이 시스템에서 PE는 EA층으로 PE-gal은 water층으로 분획되었다. 그리고, 물층을 모아서 silica gel chromatography를 실시하였다. 최종적으로, silica gel chromatography만 실시하여 PE-gal을 정제한 경우와, EA 처리 후 silica gel chromatography를 실시하여 PE-gal을 정제한 경우의 정제 PE-gal에 대하여 HPLC (High performance liquid chromatography)와 TLC (thin-layer chromatography) 분석을 수행하였다. 그 결과 EA를 처리하여 분획한 후, silica gel chromatography를 수행한 시료에서 잔여 PE가 완전히 제거되는 것을 관찰하였고, silica gel chromatography만 실시하여 PE-gal을 정제한 경우에는 상당량의 잔여 PE가 관찰되었다. 이 때, mole 기준으로 약 21%의 정제 수율을 확인할 수 있었다. 마찬가지로 CPN-gal의 정제에서도 EA 분획 처리 후, silica gel chromatography를 수행하였더니, 잔여 CPN이 거의 없는 순수한 CPN-gal을 얻을 수 있었다.

수돗물속 생물막 형성의 초기 세균 (Initial Bacterial Groups in the Development of Biofilm in Drinking Water)

  • To clarify the pioneer group in the development of biofilms in high chlorine residual water, a semi-pilot model system was operated and 16S rDNA V3 targeted PCR-DGGE was submitted. Biofilm formation occurred rapidly in the model of a drinking water distribution system. It reached $10^3\;CFU/cm^2$ or more on the surface of stainless steel, PVC, and galvanized iron in chlorinated (1.0 mg/l) water within a week. Within a week, uncultured Proteobacteria- and Bacillales group-like sequences were detected and Sphingomonas-like sequences were identified from all season and all pipe materials tested. Hence Sphingomonas species were regarded as the potential pioneer group in the development of biofilm in drinking water and this results would be useful for the prevention of biofilm formation and safety of drinking tap water.

  • The residual treatment facilities in WTP(water treatment plant) play an important role in solid-liquid separation. At present, it is difficult to solve problems related with thickening and dewatering of WTP sludge, and discharging waste water to river. The quantity of residuals generated from water treatment plants depends upon the raw water quality, dosage of chemicals used, performance of the treatment process, method of sludge removal, efficiency of sedimentation, and backwashing frequency. Sludge production by the physical separation of SS(Suspended Solid) occurs under quiescent conditions in the primary clarifier, where SSs are allowed to settle and to consolidate on the clarifier bottom. Raw primary sludge results when the settled solids are hydraulically removed from the tank. In this study, Drawing characteristics of the sludge thickening in the thickener of Water Treatment Plants was simulated by Using CFD(Computational Fluid Dynamics.

광양만과 진주만 해역에서 담수 유입으로 인한 밀도성층 변화 (Variation of Density Stratification due to Fresh Water Discharge in the Kwangyang Bay and Jinju Bay)

  • 본 연구에서는 EFDC 모델을 이용하여 광양만과 진주만 해역에 대한 조석과 수온, 염분을 고려한 3차원 해수순환 양상을 재현하였다. 수치모델 결과는 관측 자료를 이용하여 검증하였고 그 결과 모델은 관측결과를 잘 재현하였다. 실험 결과, 연구해역의 해수순환은 노량수도와 대방수도 중간 해역에서 해수가 수렴 및 발산되는 양상을 보이고 잔차류는 전형적인 2층 순환구조를 보인다. 모델을 통해 광양만과 진주만 일대의 밀도성층 변화를 분석한 결과, 연구해역은 담수의 영향이 지배적인 해역으로 진주만의 경우 남강댐 방류의 영향을 지배적으로 받는 해역이지만 남강댐에서 방류를 하지 않는 경우에는 섬진강의 영향도 받는 것으로 나타났다. 한편 남강댐 방류시에는 광양만의 성층이 상대적으로 강화되는 결과를 볼 수 있었다.

Polyethersulfone (PES) ultrafiltration (UF) membranes loaded with silver nitrate for bacteria removal

  • PES UF membranes containing silver were prepared to impart antibacterial properties for waste water treatment. Asymmetric membranes for antibacterial application were prepared from polyethersulfone (PES) and silver nitrate ($AgNO_3$) (PES/$AgNO_3$=15/2 by weight) solution in N-Methyl-2-pyrrolidone (NMP) via simple wet phase inversion technique. These membranes were characterized by polyvinylpyrrolidone (PVP) and polyethylene glycol (PEG) of different molecular weights (1000 ppm in water) at room temperature and on operating pressure of 5 bars. It was observed that the water flux of PES-$AgNO_3$ membrane is slightly lower than virgin PES but still increased linearly with the increment of pressure applied. The morphology of the resulting membranes was examined using Field-Emission Scanning Electron Microscope (FESEM) coupled with Energy Dispersive Spectroscopy (EDS). Elemental analysis using EDS proved that silver is successfully loaded on the membrane surfaces. Due to the success of loading silver on membrane surfaces, antibacterial activities were evaluated via agar diffusion method against Escherichia coli (E.coli) and Staphylococcus aureus (S.aureus) culture. By incorporating 2 wt% of silver nitrate, PES-$AgNO_3$ showed significant inhibition ring on both E.coli and S.aureus. Filtration of E.coli solution (OD 0.31) showed satisfactory rejection data with ~100% inhibition growth after 24 hours incubation at $37^{\circ}C$. Resultant membranes also exhibit better tensile strength (compared to virgin PES) up to 71% may be due to the suggested interactions. The residual silver during fabrication was measured using ICP-MS and result showed that the residual silver content of PES-$AgNO_3$ membrane was only ~1% of the original silver added in the polymer solution. These studies have shown that PES-$AgNO_3$ UF membranes are potential in improving the filtration in water treatment.

하천수중 잔류 베타-락탐계 항생제의 동시분석법 (Simultaneous analysis of β-lactam antibiotics in surface water)

  • 환경 시료중에 잔류하는 베타-락탐계 항생제를 동시에 추출하는 방법을 확립하였다. pH 2 에서 HLB 고체상을 사용하여 추출 및 정제한 후, 7종의 베타-락탐계 항생제(amoxicillin, ampicillin, penicillin G, cefaclor, cefadroxil, cefatrizine 및 cephradine)를 LC/ESI-MS/MS를 사용하여 동시에 분석하였다. 새롭게 확립된 분석법은 하천수중에서 베타-락탐계 항생제의 농도가 0.01~1.0 ng/mL일 때 정량을 위한 검정곡선의 상관계수($r^2$)가 0.9911~0.9995 범위로써 우수한 직선성을 나타내었다. 바탕하천수로부터 구한 검출한계(LOD)와 정량한계(LOQ)는 각각 0.0003~0.0234 ng/mL 와 0.0046~0.0778 ng/mL이었다. 이 분석방법은 물 환경중에 잔류하는 베타-락탐계 항생제를 정성 정량하는데 빠르고 신뢰할 만한 방법이 될 것이다.

2-Methylisoborneol(2-MIB)제거를 위한 산화 및 흡착공정의 특성 (The Characteristics of Oxidation and Adsorption Processes for 2-Methylisoborneol(2-MIB) Removing)

  • One of the Musty and earthy smell compounds in raw water is generally attributed to 2-methylisoborneol (2-MIB). It is well known that activated carbon and oxidants such as $O_3$, Cl $O_2$, are effective ways to control 2-MIB. In isotherm equilibrium experiments, 2-MIB in distilled water was much more adsorbed to the activated carbon(A/C) than raw water containing dissolved organic carbon (DOC). The Freundlich constants(k) of distilled water and raw water were 3.36 and 0.049, and 1/n values were 0.80 and 0.42, respectively. The 2-MIB residual rate were Y = $e^{-0}$.55x/~ $e^{-0}$.54x/ with Ozone( $O_3$) dose by 5 minutes contact time at the 241 and 353 ng/L initial concentrations. The 2-MIB residual rate were Y = $e^{-0}$.32x/~ $e^{-0}$.35x/ with Chlorine dioxide(Cl $O_2$) dose by 15 minutes contact time at the 89 and 249 ng/L initial concentrations. 2-MIB was decreased from 1911 ng/L to 569ng/L by post-ozonation(70%removal efficiency) and removal efficiencies of 2-MIB by the following 4 kinds Granular Activated Carbon(GAC) process such as coal base, coconut base, wood base and zeolite+carbon base were 95.8, 89.5, 88.4, and 93.7% respectively.ely.

  • 본 연구에서는 상수관의 개량사업을 보다 효율적으로 실시 할 수 있는 방법으로 국내 실정에 적합한 상수관로의 노후도 조사방법을 이용하여 매설된 관의 노후도를 예측 근거한 최적 개량 모형을 제시하였다. 노후도 예측 모형은 확률론적 신경망 이론을 바탕으로 관별 노후도 정도를 5개 등급으로 구분하며 산정된 관별 노후도 등급 및 관경을 바탕으로 최대 잔존수명을 산정하였다. 최적 개량 모형은 관의 유지보수, 갱생, 교체의 시기 및 비용을 산정하는 것으로 최단경로흐름을 응용한 정수계획법을 이용하여 제시하였다. 또한 개발된 최적 개량 모형은 보다 현실적인 모형을 구축하기 위해 예산제약 유무에 따라 2가지 구분하여 모형을 제시하였다. 따라서, 본 연구에서 제안된 모형은 상수관로의 관리를 위해 보다 정량적인 방법으로 활용할 수 있을 것이다.

경기남부권 농업용 저수지의 유해 남세균 발생 및 독소물질 잔류 특성 (Characteristics of Harmful Cyanobacteria Occurrence and Toxin Residual in Agricultural Reservoirs of Southern Gyeonggi)

  • Algal blooms occur seasonally in the eutrophicated rivers or reservoirs, and some harmful cyanobacteria species produce toxic substances, which are directly or indirectly harmful to the ecosystem and terrestrial animals. So, the monitoring and control of harmful cyanobacteria occurrence and toxins residual in the aquasystem are important to preserve the water environment and secure public health. In this study, the four harmful cyanobacteria occurrences and toxic substance concentrations of two agricultural reservoirs in the southern part of Gyeonggi Province were investigated from August to October 2022. Among four harmful cyanobacteria (Microcystis sp., Anabaena sp., Oscillatoria sp., Aphanizomenon sp.), three kinds of cyanobacteria except Oscillatoria sp. were observed, and Microcystis sp. was the dominant cyanobacteria except for Anabaena sp. dominant result of a sample collected on October at reservoir B. The cell density of cyanobacteria was influenced by season and weather due to the length of daytime and concentrations of organic carbon and nitrogen. Three kinds of microcystin and anatoxin-a were quantitatively analyzed as total (in the cell body and water) and extracellular (in water) concentrations. The maximum total concentrations of anatoxin-a, microcystin-LR, microcystin-RR, and microcystin-YR were 0.1291 ㎍/L, 0.2776 ㎍/L, 0.3721 ㎍/L, and 0.0306 ㎍/L, respectively, in reservoir A and 0.3274 ㎍/L, 0.1495 ㎍/L, 0.2037 ㎍/L, and 0.0153 ㎍/L, respectively, in reservoir B.

공기주입과 스와빙 피그를 이용한 상수관로 세척 효과 평가 (An assessment of the effect of air scouring and swabbing pig cleaning technique on water distribution pipes)

  • Deposits discharged through the cleaning mainly were cement mortar, bitumen paintings and rust pieces, and fragments of perforation, stones and gravels. Deposits were more removed through swabbing pig cleaning rather than air scouring cleaning on the whole. However, air scouring cleaning were not influenced by the constraint conditions such as a change in the diameter or the presence of the valve in water mains compare to swabbing pig cleaning. So, it was thought that air scouring cleaning might be more favorable to water distribution network cleaning in the future. After the cleaning, water quality including residual chlorine and turbidity also was improved because of the removal of a significant amount of the deposits. Therefore, if the cleaning is continuously and regularly implemented in water mains, it is expected that it will help to recover the reliability and to preserve the health of water quality.
