Study on the Resazurin test as a Rapid Method for the Estimation of Maturity of Kimchi (김치숙성도 판정기준을 위한 신속 검사법 Resazurin-test 에 관한 연구)

  • An attempt was made to establish a simple and rapid method for the evaluation of Kimchi quality. Four kinds of Kimchi included one home made traditional Kimchi were stored at $4^{\circ}C$ and$25^{\circ}C$ and biochemical changes, including pH, total acidity, volatile acidity, volatile basic nitrogen, vitamin C, NaCl, glucose, nitrite and resazurin value were determined and tested for the relationship with sensory evaluation. The change in pH, total acidity and resazurin value were highly correlated with the sensory maturity of Kimchi-Resazurin value was strongly influenced by the sample dilution and fermentation temperatures. It was concluded that resazurin test with sample solution of $2{\sim}4%$ could be used as a rapid method for the estimation of the maturity of Kimchi.

Resazurin Reduction Time Test to Determine Post-pasteurization Contamination and Shelf Life of Market Milk (시유의 2차오염과 저장가능기간을 결정하기 위한 Resazurin 환원시간검사)

  • The selective media including NPC agar, DHL agar, MacConkey agar, and Cetrimide desoxycholate agar were compared to determine selectivity for the growth of bacteria Cetrimide desoxycholate agar was better than NPC agar, DHL agar, and MacConkey agar for the growth of psychrotrophic grarn-negatve bacteria including Pseudomonas. and for the inhibition of gram positive bacteria The specificity of resazurin reduction time test was investigated to determine post-pasteurization contamination of market milk. Equal volume of Cetrimide desoxycholate broth was added to market milk, which was then incubated at $21^{\circ}C$ for 18 hours. The growth of bacteria in the incubated milk was detected in resazurin reduction time test. The results in resazurin reduction time test and total bacteria number count of market milk after storage at $7^{\circ}C$ were relatively correlated each other. Pseudomonas was isolated most frequently from the market milk stored at $7^{\circ}C$ for 10 days, and Acinetobacter and Aeromonas followed. Acinetobacter, Pseudomonas and Enterobacter were frequently isolated from the mixture of market milk and Cetrimide desoxycholate broth incubated at $21^{\circ}C$ for 18hours in resazurin reduction time test.

Development of Rapid Somatic Cell Counting Method by Using Dye Adding NIR Spectroscopy (색소첨가 NIR을 이용한 우유 체세포수 측정법 개발)

  • To develop the somatic cell counting NIR Spectrum method within a range of 400-2500 nm, eosin-Y, methyl red, methylene blue, resazurin and amido black 10B were tested at 0.01% in raw milk. The PLS (Partial Least Square) results are summarized as follows: Correlation coefficients of the calibration model measurements by NIR spectroscopy in raw milk for eosin-Y, methyl red, methylene blue, resazurin and amido black 10B were 0.78, 0.65, 0.63, 0.65, 0.98 and 0.99, respectively. The correlation coefficients of the prediction model measurements by NIR spectroscopy in raw milk for eosin-Y, methyl red, methylene blue, resazurin and amido black 10B were 0.49, 0.21, 0.36, 0.47, 0.95 and 0.98 respectively. Based on these results, amido black 10B was the best additive for the NIRS somatic cell count method.

Chemical changes in resazurin by probiotics and its application for evaluating living bacterial cell counts and their reduction potentials (프로바이오틱스에 의한 레자주린의 화학적 변화와 생균수 및 환원활성 측정에의 적용)

  • Resazurin, an oxidized blue dye, is reduced to resorufin, showing a peak absorbance change and emitting fluorescence due to the metabolic activity of living cells. In this study, the growth of Lactobacillus rhamnosus GG (LGG) and the redox potential of living probiotic bacteria were evaluated based on changes in the absorbance or fluorescence of resazurin. Fluorescence analysis is a more accurate and sensitive method for quantifying viable LGG than the colorimetric absorbance measurement of resazurin change. Fluorescence measurement could detect LGG of 6.5-9.5 log CFU/mL within 30 min with R2=0.99. No significant effect of further reduction of resorufin to dihydroresorufin by LGG was observed. Various probiotics showed different resazurin-reducing activities, and L. kimchicus had the highest reducing activity among the six probiotics tested. These findings suggests that fluorescence measurement in a resazurin-based assay is useful for analyzing bacterial growth and the redox potential of living probiotics.

  • We developed a method to measure the concentration of highly concentrated colored samples using color coordinates. We present a color analysis to estimate the concentration of highly concentrated binary solutions with an empirical function and apply it to estimate the kinetics of a catalytic irreversible reaction of resazurin to resorufin. The developed method enabled one to measure the concentration of solutions whose range of concentrations is nonlinear and beyond the limit of UV-vis spectroscopic measurement directly without dilution.

Evaluation of Acitivity of QoI Fungicide against Colletotrichum acutatum s. lat. Causing Pepper Anthracnose Using Resazurin-Based Respiration Assay (Resazurin 기반 호흡 측정법을 이용한 고추탄저병균에 대한 살균제의 효과 검정)

  • Resazurin-based microtiter assay was used to evaluate the inhibitory effect of fungicides on the respiration of Colletotrichum acutatum s. lat. 20JDS8 sensitive and 20CDJ6 resistant to strobilurin fungicides. The spores of C. acutatum s. lat. 20JDS8 were inoculated into potato dextrose broth (PDB) at densities of 1x104, 1x105 and 1x106 spores/ml, respectively. The relative fluorescence unit (RFU) of all treatments inoculated at each spore density started to rise after 12 hr of incubation, and were 1,965.5, 5,412.5, and 10,061.0, respectively, after 24 hr of incubation. To evaluate the inhibitory effect of fungicide on the respiration of the pathogen, the spores of the pathogen were inoculated into the PDB and treated with the fungicides 0, 6, 12, and 24 hr after incubation, respectively. After keeping the pathogen culturing for another 24 hr, PrestoBlue reagent was treated into the PDB culturing the pathogen. The RFU of each treatment was examined 1 hr after the reagent was treated. When dithianon, isopyrazam, pyraclostrobin, and fluazinam were treated at high concentrations in the stages of spores (immediately after inoculation [0 hr]), spore germination (after incubation for 6 hr), and hyphal growth (after incubation for 12 hr), the respiration of pathogens was inhibited by 90-100%. When the fungicides were treated after culturing the pathogen for 24 hr, the respiratory inhibitory effects were greatly reduced. With pyraclostrobin-resistant C. acutatum s. lat. 20CDJ6, azxoystrobin, trifloxystrobin and kresoxim-methyl, which have the same mode of action, had very little or no respiratory inhibitory effect in all growth stages of pathogens. Based on the above results, it was thought that the resazurin-based microtiter assay could quickly and accurately evaluate the inhibitory efficacy of the fungicides that inhibited respiration.

  • Nanoparticles play an important role as a catalyst in many chemical syntheses. Colloidal nanoparticles were usually synthesized with reducing, capping, and shape directing agents which induce surface poisoning of catalysts. A new green synthesis for silver nanoparticles was developed by utilizing less additives which could be a hazardous waste. A crystallization technique was employed to reduce the amount of reducing and capping agents during synthesis resulting in less surface poisoning of the nanoparticle. The synthesized Ag nanoparticles using monosaccharides and disaccharides as reducing agents could be used as a catalyst for the redox reaction of resazurin and the mechanism of the reaction using Ag nanoparticles was studied.

Studies on Rapid Microbiological Testing Method of Fresh Pork by Applied Resazurin Reduction Test(RRT) Method (Resazurin 환원법을 응용한 돈육의 신속 미생물 검사법에 관한 연구)

  • In order to search for reliable rapid methods of estimating bacterial counts in pork, this study was tried to measure resazurin reduction time which is simple in experimental method, low in analytical cost, able to estimate bacterial count within short time. The results were summarized as follows; Correlation coefficient between initial bacterial log count(25$^{\circ}C$/72hr, Y) and resazurin reduction time(X) from blue color to pink color during incubation at 25$^{\circ}C$ and 30$^{\circ}C$ was higher than other conditions as -0.95 and -0.94, respectively. Considering correlation coefficient and reduction time, incubation temperature was compatible at 30$^{\circ}C$, and regression equation(RE) was Y = -0.4386X + 7.7870. At a bacterial load of $10^2$cfu/$cm^2$, $10^3$cfu/$cm^2$ and $10^4$cfu/$cm^2$ in pork, reduction time was 13.2hr, 10.9hr and 8.6hr, respectively. Correlation coefficient between initial bacterial log count(30$^{\circ}C$/72hr, Y) and resazurin reduction time(X) from blue color to pink color during incubation at 30$^{\circ}C$ was highest among other conditions as -0.93, and RE was Y = -0.4171X + 7.5540. At a bacterial load of $10^2$cfu/$cm^2$, $10^3cfu/$cm^2$ and $10^4cfu/$cm^2$ in pork, reduction time was 13.3hr, 10.9hr and 8.5hr, respectively. Correlation coefficient between initial bacterial log count(35$^{\circ}C$/72hr, Y) and resazurin reduction time(X) from blue color to pink color during incubation at 30$^{\circ}C$ was highest among other conditions as -0.93, and RE was Y = -0.3514X + 6.7513. At a bacterial load of $10^2$cfu/$cm^2$, $10^3$cfu/$cm^2$ and $10^4$cfu/$cm^2$ in pork, reduction time was 13.5hr, 10.7hr and 7.8hr, respectively.

Development of Measuring Technique for Somatic Cell Count in Raw Milk by Spectroscopy (분광분석법을 이용한 우유의 체세포수 측정기술 개발)

  • The objective of this study was to develop models to predict SCC (somatic cell count) in unhomogenized milk by visible and near-infrared (NIR) spectroscopic technique. Total of 100 milk samples were collected from dairy farms and preserved to minimize propagation of bacteria cells during transportation. Reductive reagents such as methyl red, methylene blue, bromcresol purple, phenol red and resazurin were added to milk samples, and then colors of milk were changed based on SCC of milk. For optimal reductive reagents, reaction time was controlled at 3 level of reaction time. A spectrophotometer was used to measure reflectance spectra from milk samples. The partial least square (PLS) models were developed to predict SCC of unhomogenized milk. The PLS results showed that milk samples with reductive reagents had a good correlation between predicted and measured SCC at 5 minutes of reaction time in the visible range. The PLS models with resazurin reagent had the best performance in $400{\sim}600\;nm$. The prediction results of milk samples with resazurin had 0.86 of correlation coefficient and 14,184 cell/mL of SEP.
