Application of GC-SAW(Surface Acoustic Wave) Electronic Nose to Classification of Origins and Blended Commercial Brands in Roasted Ground Coffee Beans (GC-SAW(Surface Acoustic Wave) 전자코를 활용한 볶은 커피의 원산지 및 배합 커피의 상품별 분류)

  • The numerous varieties of coffee beans contain a wide range prices and qualities. While the varieties of green coffee beans can generally be distinguished by their appearance, this visual criterion is impossible after the roasting process. Therefore, we need to develop a classification method or device. In this study, the potential of a new type of electronic nose, fast gas chromatography based on a surface acoustic wave sensor(SAW), was evaluated for the classification of origins and blended commercial brands in roasted coffee beans. Eight blended commercial brands and the origins of four similarly roasted ground coffee beans(with no significant difference of color) were rapidly(90 sec/sample) classified. The reproductive results were easily understandable over the aroma image pattern by $VaporPrint^{TM}$. In conclusion, GC-SAW electronic nose can be applied to the classification of origins and commercial brands in roasted ground coffee beans and to e evaluation of the similarities and differences of volatile pattern between samples.

Development of the Artificial Insemination Instrument of Bumblebee Queens (뒤영벌 인공수정기 개발)

  • An artificial insemination instrument of bumblebee queens was firstly developed. This instrument consists of location tube, transfer tube, holding tube, head product, and probe apparatus for reproductive tract etc. This instrument was designed to minimize stress and damage of reproductive tract of bumblebee queens. The regulator handle apparatus in artificial insemination instrument was used the principle of lever, that manipulates easy, accurate and rapid insemination of bumblebee queens. By using this instrument, the insemination rate was over 90%. This instrument will be useful for the breeding and conservation of excellent character of bumblebees.

Comparison of Brood Productions in the Cold-Treated Pairing vs. Not Cold-Treated Pairing in a Stenochronous Spider

  • To understand whether experience of cold season in reproductive behaviors in the adults of Amaurobius ferox, the paired adults of a female and a male were exposed under 'cold-treated environment' and 'not cold-treated environment', respectively. I investigated effects of the cold treatment on the brood production of A. ferox. In not cold treatment in which male-female pairs were formed in October at a temperature of $20^{\circ}C$ (${\pm}2^{\circ}C$) and continuously kept under not cold-treated environment, only 3 of 50 pairs successfully reproduced (reproduction was defined as the emergence of spiderlings). In cold treatment where individuals were kept in cold conditions for 3 months prior to pair formation, 57 out of 60 couples succeeded in reproducing. Females which did not experience the low temperature displayed strong aggressiveness toward males. This behavioral inhibition might the primary barrier to copulation of A. ferox that decrease following a period of low temperature. The reproductive inhibition might help the females to allocate the maximum amount of energy in a given environment to reaching the adult stage and to delay reproduction in unfavorable wintering conditions.

First report on the reproduction of captive Chinese many-toothed snake (Sibynophis chinensis) in Jeju Island, South Korea

  • The Chinese many-toothed snake (Sibynophis chinensis) is an endangered species in South Korea. To determine its reproductive activity, here we captured a gravid S. chinensis from Mulyeongari Oreum Wetland Conservation Area, Jeju Island, on May 27, 2018, and reared it at Animal Taxonomy and Morphology Laboratory, Jeju National University, South Korea. The snake oviposited a clutch of six eggs between June 12 and 20, 2018, with an average size of 24.86 ± 1.36 mm in length and 9.86 ± 0.49 mm in width, which were artificially incubated at a constant temperature of 27 ℃. After incubating for 34-41 days, five of these six eggs successfully hatched. Average snoutvent length (SVL), tail length (TL), and body weight (BW) of hatchlings were 136.5 ± 5.17 mm, 47.76 ± 2.80 mm, and 1.66 ± 0.12 g, respectively. This study observed the reproductive activity of S. chinensis and characterized its eggs and hatchlings for the first time, providing valuable information for the long-term conservation plan of S. chinensis in South Korea.

Effects of local conspecific abundance on seed set and seed predation, and control of Carpinus laxiflora (Betulaceae) population density

  • In this study, the dependence of local conspecific abundance on seed set and predation was examined, and determinates of population density of the temperate tree species Carpinus laxiflora (Betulaceae) in Inagi city, Tokyo, Japan were identified. During a good seed year (2013), seeds were sampled from 27 individuals and categorized as Sound, Empty, Predated, Immature, Decayed, or Broken. Empty seeds were identified as those that failed to fertilize and predation rates were defined as the proportion of seeds that were affected by predators. The proportion of the seed set that was fertilized was significantly positively correlated with local abundance of conspecific trees, while the proportion of seeds that escaped predation was significantly negatively correlated with local abundance of conspecific trees. Thereby, the production of Sound seeds was highest at sites with moderate local conspecific abundance. Although this phenomenon was only observed in a few cases, it clearly showed optimal reproductive success under conditions of moderate density of conspecific reproductive adults, suggesting a mechanism that controls the population density of this species.

Phosphorus and Base Cation Inputs through Litterfall Components in Pine Forests after Tree Removal Due to Pine Wilt Disease Disturbance

  • This study was conducted to measure litterfall and nutrient (P, K, Ca, Mg) inputs under varying degrees of disturbance by pine wilt disease in pine forests in southern Korea. Litterfall was collected to evaluate nutrient responses at different intensities of disturbance (various levels of basal area) by pine wilt disease across 2 years. Phosphorus, Ca, and Mg concentrations in needle litterfall were positively correlated (p < 0.05) with decreased disturbance intensities (increased basal area) depending on the time of sampling, whereas the nutrient concentrations in other litterfall components (branches, bark, reproductive organs, and miscellaneous litterfall) were not significantly correlated (p > 0.05) with the intensity of pine wilt disease disturbance. Dry weight and nutrient inputs through litterfall components decreased linearly with increasing intensity of disturbance by pine wilt disease (p < 0.05), except for the nutrient inputs of branch (K, Ca, Mg) and reproductive organ (K, Ca) litterfall. These results indicate that decreased litterfall across different levels of disturbance may be related to the reduced soil nutrients in pine wilt disease forests.

Mammalian Research Topics and Trends in Korea (국내 포유류 연구의 주제와 동향)

  • Mammals in Korea have been studied in various fields such as animal science, veterinary medicine, laboratory animal science, ecology, and genetics. As the importance of biodiversity has been emphasized recently, conservation and management of mammals have attracted much public attention. However, in spite of such an increase in scientific research and public interest, it is still difficult to find a report or summary to grasp the trend of mammalian research in Korea. The purpose of this study is to provide the basic data for future plans of the detailed research area and the related policies by grasping the research trends of mammals in Korea. Using text-ming and co-word analysis, we analyzed 392 mammalian research papers published in Korean national journals as of 2015. Our results showed that the number of mammalian research papers published in Korea has gradually increased and that the research target species have also become increasingly diverse. The major research areas identified through text-mining and co-word analysis are (1) evolution/phylogenetics/genetics, (2) environmental science/ecology, (3) embryology/reproductive biology/cell biology, (4) veterinary medicine related to parasites, (5) parasitology related to rodents, (6) bacteriology/virology, (7) anatomy/cell biology/laboratory animal science, (8) veterinary science related to morphology and anatomy, (9) animal science, (10) marine mammalogy, and (11) Chiroptera (bat) research. Environmental science/ecology has been the most active field among the 11 research areas in recent times, and the proportion of research has increased sharply compared to the past. Environmental science/ecology is the core of biodiversity conservation, and as the importance of biodiversity has been emphasized in recent years, researchers' interest in mammal ecology appears to have increased. We expect that the results of this study will be useful for future research plan and related policies on mammals in Korea.

Reproductive Ecology of the Freshwater Marsh Clam, Corbicula papyracea (Heude) in Bukhan River (북한강 엷은재첩, Corbicula papyracea (Heude) 의 번식생태)

  • Gonadal development and maturation mechanism were studied on the freshwater marsh clam Corbicula papyracea (Heude), which is the endangered species in Korea. The specimens were collected in the rearing ponds and waterway of NFRDI in Gapyeong-gun, Gyeonggi-do from January to December 2004, and then investigated by condition factor, relative growth, gonadal development phases and gonad histological characters based on 30-50 individuals every month. Comparing with the freshwater marsh clam, C. papyracea is small, light olive brown shell and violet interior. The hermaphrodite individuals of C. papyracea take an internal fertilization and fertilized eggs are stored in the foster-sack in the gills, then the hatched juveniles are released outside after an ovoviviparous process. The average water temperature of inhabit area was in range of $1.8-27.0^{\circ}C$ and usually took great effects on the gonad maturation of C. papyracea. The condition factor ranged from 0.14 to 0.21 throughout the year, which was the lowest during winter season (December-February), and gradually increased to the highest value of 0.21 in May. The ratio of meat weight to total weight was 25.9-38.7%, indicating the similar trend with condition factor. The highest values of condition factor and the ratio of meat weight appeared 1-2 months later than gonadosomatic index reached the peak value, it was probably because that the ovoviviparous eggs would spend a long period before hatching from the foster-sack in the gills. To synthesize the characters of meat weight, condition factor and gonad development by histological study, reproductive cycle of C. papyracea could be divided into five successive stages: multiplicative stage (December to February), growing stage (February to May), mature stage (June to August), spawning stage (August to November), recovery stage (November to December). The smallest shell length of matured C. papyracea was 12.6 mm, and individuals, larger than 16 mm, was formed the nursery in the gills.

  • The reproductive ecology of freshwater bivalve, Unio douglasiae in Lake Uiam were examined from March 2014 to February 2015. The gonads distributed in reticular connective tissue, the dioecious clams take an internal fertilization and fertilized eggs are stored in the foster-sack in the gills, then the hatched juveniles are released outside after an ovoviviparous process. The gonad index (GI) began to increase in April, reached the maximum value in July and to decrease in August. The reproductive cycle of U. douglasiae can be divided into five successive stages: multiplicative stage (November to January), growing stage (February to March), mature stage (February to May), spawning stage (April to September), recovery stage (August to December). The spawning periods were from April to September, and the main spawning occurred between April and July. The shell length at 50% group maturity was estimated to be 29.4 mm.

Adverse Reproductive Effects on Plasma Vitellogenin and Sex Steroid Levels, and Gonadosomatic Index in Juvenile Common Carps (Cyprinus carpio) Exposed to 17$\beta$-Estradiol and D-2-Ethylhexyl Phthalate

  • Environmental estrogens are natural or synthetic substances present in the aquatic environment, especially in effluent from sewage treatment. However, the adverse effects of these estrogenic substances on fish reproduction are unknown. Di-2-ethylhexyl phthalate (DEHP) is the most common phthalate, which Ps used as a plasticizer in polyvinylchloride (PVC), and it is widespread in the environment and has been found in aquatic organisms and sediments. Therefore, juvenile common carps (Cyprinus carpio) were exposed to nominal concentrations of 17$\beta$-estradiol (E2) (0.5, 5, 50 $\mu\textrm{g}$/L) and DEHP (10, 100, 500 $\mu\textrm{g}$/L) for 21 days, to determine the adverse reproductive effects of these compounds on plasma vitellogenin (VTG) induction, sex steroid level, and gonad weight. Electrophoresis (SDS-PAGE) revealed that much of VTG was induced in fish exposed to 5 and 50 E$_2$ $\mu\textrm{g}$/L, but none of DEHP exposure showed induction. Enzyme-linked immunosorbent assay (ELISA) revealed that VTG was significantly induced in fish exposed to 5 and 50 E$_2$ $\mu\textrm{g}$/L, and combination of 50 E$_2$ $\mu\textrm{g}$/L with 10 and 500 DEHP $\mu\textrm{g}$/L, but none of DEHP exposure showed induction. Analysis of sex steroid levels in some fish revealed that testosterone (T) was detected in both male and female fish of the control and DEHP exposures, but none of fish exposed to 22 concentrations had detectable testosterone level. On the other hand, E$_2$ exposure induced 17$\beta$-estradiol in plasma of male fish, but there was no induction of 17$\beta$-estradiol in plasma of male fish exposed to DEHP. Comparison of gonadosomatic index (GSI) revealed that maximal E$_2$ exposure inhibited ovarian growth, but maximal DEHP exposure stimulated testicular growth. The results indicated that those comparisons can be a useful bio-indicator for determining adverse reproductive effect of endocrine disrupting chemicals (EDCs).
