The Therapeutic Effects of Optimal Dose of Mesenchymal Stem Cells in a Murine Model of an Elastase Induced-Emphysema

  • Background: Chronic obstructive pulmonary disease is characterized by emphysema, chronic bronchitis, and small airway remodeling. The alveolar destruction associated with emphysema cannot be repaired by current clinical practices. Stem cell therapy has been successfully used in animal models of cigarette smoke- and elastase-induced emphysema. However, the optimal dose of mesenchymal stem cells (MSCs) for the most effective therapy has not yet been determined. It is vital to determine the optimal dose of MSCs for clinical application in emphysema cases. Methods: In the present study, we evaluated the therapeutic effects of various doses of MSCs on elastase-induced emphysema in mice. When 3 different doses of MSCs were intravenously injected into mice treated with elastase, only $5{\times}10^4$ MSCs showed a significant effect on the emphysematous mouse lung. We also identified action mechanisms of MSCs based on apoptosis, lung regeneration, and protease/antiprotease imbalance. Results: The MSCs were not related with caspase-3/7 dependent apoptosis. But activity of matrix metalloproteinase 9 increased by emphysematous lung was decreased by intravenously injected MSCs. Vascular endothelial growth factor were also increased in lung from MSC injected mice, as compared to un-injected mice. Conclusion: This is the first study on the optimal dose of MSCs as a therapeutic candidate. This data may provide important basic data for determining dosage in clinical application of MSCs in emphysema patients.

The Effect of Silk Fibroin Particles Coated with Hydroxyapatites on Bone Regeneration in the Rat Calvarial Defect Model (백서 두개골 결손모델에서 하이드록시아파타이트 입자로 입혀진 실크단백이 골재생에 미치는 영향)

  • Purpose: This study evaluated the capability of bone formation of silk fibroin particles coated with hydroxyapatites (HA/SF), as bone graft material when put into the calvarial defect of rats. Methods: Twenty Sprague Dawley rats were used for this study and round shaped defects were formed in the center of parietal bones (diameter: 8.0 mm). The defect was filled with (1) HA/SF (experimental group), or (2) left as a vacant space (control group). The animals were sacrificed at 4 or 8 weeks, postoperatively. The specimens were decalcified and stained with Masson's trichrome for histomorphometric analysis. Results: The average of new bone formation was $33.18{\pm}3.10%$ in the experimental group and $20.49{\pm}5.79%$ in the control group at 4 weeks postoperatively. That was $42.52{\pm}7.74%$ in the experimental group and $25.50{\pm}7.31%$ in the control group at 8 weeks postoperatively. The difference between the groups was significantly higher at both 4 weeks and 8 weeks postoperatively (P<0.05). Conclusion: The rat calvarial defect was successfully repaired by HA/SF graft. The HA/SF graft showed more new bone formation compared with the unfilled control.

Analysis of Patched Cylindrical Shells with Circumferential Through-Wall Cracks (원주방향 관통균열을 갖는 원통형 쉘 구조의 패치보강 해석)

  • In this study, behavior of unpatched and patched cylindrical shells with through-wall cracks has been estimated using numerical experiments, and patching effect of them has been investigated according to various patching parameters. To show credibility of numerical models considered, two ways such as h- and p-methods have been adopted. Also, domain integral method and virtual crack extension method have been considered to calculate energy release rates based on linear elastic fracture mechanics. For examples, the unpatched cylindrical shells with circumferential cracks under remote tension have firstly been analyzed to show the validity of finite element modeling with h-method or p-method, and then the results have been compared with literature values published. Next, the sensitive analysis of patch repaired problems in terms of thickness of patch and adhesive, shear modulus of adhesive, composite material type of patch, crack length, etc. has been carried out.

Effects of the Methanol Extract of Houttuynia cordata $T_{HUNB}$ on the Accumulation of Cadmium in Rats (III) (어성초 메탄올 추출물이 흰쥐 장기내 카드뮴 축적에 미치는 영향 (III))

  • This study was conducted to investigate the antitoxic component in methanol extract of Houttuynia cordata $T_{HUNB}$. The results were as follows: Generally, detoxication effects by Houttuynia cordata $T_{HUNB}$ methanol extract increased in proportion to the methanol extract concentrations. When 40 mg/kg dosage of Houttuynia cordata $T_{HUNB}$ methanol extract was administrated, it showed the highest antitoxic effects in metallothionein induction. After the methanol extract treatment. Body weights increased in proportion to methanol extract concentrations. However, after 3 weeks, the body weight decreased insignificantly. From the above results, these results suggest that the methanol extract of Houttuynia cordata $T_{HUNB}$ increased metallothionein concentration and decreased the toxicity of cadmium in rats.

  • The studies were conducted to investigate the inflammation, gastric ulcer, analgesic and homothrmic of experimental animals by KYUNG OK-KO water extract and drink, which is a traditional preparation in Korea. 1. It was exhibited significantly anti-inflammatory effects to acute inflammation on carragennan edema, and preventive and therapeutic effects to chronic inflammation on Freund's complete adjuvant in rats(100, 200, 400 mg/kg). Especially, preventive effect of KYUNG OK-KO water extracts at doses of 100, 200, 400 mg/kg in rats were showed dose-dependantly. 2. Aspirin-induced gastric ulcer were remarkably repaired by all experimental groups of KYUNG OK-KO water extracts. 3. The number of writing syndromes by acetic acid induced also were decreased which MPI's test was increased by tail flick apparatus in mice remarkably, all doses of water extracts(100, 200, 400 mg/kg) and dose-dependantly. 4. Aminopyrine-induced homothermic effects of KYUNG OK-KO water extract(100, 200, 400mg/kg) and drink (0.7, 1.4, 2.1 ml/kg) were significantly evaluated dose-dependantly in 400 mg/kg, 2.1 ml/kg groups.

The Issues and Problems of Housing Improvement Policies (주택개보수 정책의 현안과 과제)

  • The maintenance and improvement of housing become more important than quantitative supply. So this paper suggests the directions to solve the current issues of housing improvement policies. In 2015, the housing improvement for the low-income has been expanded with the revision of housing benefit policy. And each related government departments are steadily carrying out various housing improvement programs. Under the current improvement policies, individual houses are being repaired according to the criteria of the target selection. And the repair allowance of the housing benefit is limited to the ownership households except the rented. Hereafter, it has to considered the review of the detailed goal setting and application problems in housing improvement policy and to consider the extension to the rented households and the raise of the income criteria of targets. Also, it needs to carry out the improvement of several neighboring houses at the same time for synergy effect other than individual house, to change the selection way of repair contractors and to secure the quality and advancement of the improvement.

  • In recent years, carbon fiber-reinforced polymer (CFRP) has been widely used for repairing and/or strengthening structural elements in concrete. Not enough test data, however, are available to predict the long-term performance of the repaired and improved structures exposed to weathering. The objective of this research is to study the effect of freeze-thaw cycling on the behavior of reinforced concrete (RC) beams strengthened in shear with carbon fiber sheet. Six small-scale RC beams (100mm${\times]$100mm${\times]$400mm) were strengthened with CFRP in shear, subjected to up to 400 cycles freeze-thawing from -17${\sim}4^{\circ}C$, and tested to failure in four-point bending. Test result, there was no significant damage to carbon fiber sheet strengthened concrete beams had been suffered 30 cycles of freeze-thawing, and more over 60 cycles of freezing-thawing brought about a reduction in resistance of only 25% of the initial level.

Effects of Seungseup-tang on MIA-induced Osteoarthritis Rats (승습탕(勝濕湯)이 MIA로 유발된 골관절염 흰쥐에 미치는 영향)

  • Objectives This study was designed to evaluate the anti-inflammatory effects of Seungseup-tang (SST) on the monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods Osteoarthritis was induced by injection of MIA ($50{\mu}l$ with 80 mg/ml) into knee joint cavity of rats. Rats were divided into 4 groups (normal group, control group, indomethacin treated group, SST treated group, each n=6). Normal group was not injected with MIA and taken normal diet. Control group was injected with MIA and taken with distilled water. Indomethacin treated group was injected with MIA and taken indomethacin 5 mg/kg by oral administration. SST treated group was injected with MIA and taken SST 200 mg/kg by oral administration. We examined the weight-bearing ability of hind paw, biomarkers related to oxidative stress in serum, inflammatory proteins and inflammatory mediators and cytokines. Moreover, histopathological examination of knee joint structure was also performed by Hematoxylin & Eosin (H&E), Safranin-O staining method. Results In the present study, SST treated group showed a similar inhibitory effects alike indomethacin treated group, in most of the studied parameters of inflammation. The increased oxidative stress biomarker such as reactive oxygen species (ROS) and peroxy nitrite ($ONOO^-$) in the serum were reduced with SST. Especially, the level of $ONOO^-$ compared with control group significantly suppressed. Also, the expression of inflammatory mediators and cytokines induced by nuclear factor-kappa B (NF-${\kappa}B$) activation was modulated through inhibition of IkBa phosphorlation. In addition, histological analysis revealed that cartilage damage by MIA repaired markedly in SST treated group. Conclusions According to the results, Seungseup-tang may be effective for preventing the progression of osteoarthritis.

Reliability Analysis of Sloped-Coastal Structures with Sea-Level Rise (해수면 상승에 따른 경사식 해안 구조물의 신뢰성 해석)

  • A system of risk assessment is developed by using the reliability analysis which evaluate quantitatively both stability and performance of sloped-coastal structures according to several scenarios of sea-level rise. By using reliability functions on armor unit and run-up, the probabilities of failure can be straightforwardly calculated with respect to several design parameters such as nominal diameter of armor unit, slope of coastal structure, and freeboard height. By comparing the results before and after sea-level rise, it may be possible to exactly assess some ranges of decrease of stability and performance of sloped-coastal structure with respect to sea-level rise. Therefore, it can also be possible to make a decision which parameters should be repaired or strengthened in order to maintain the original stability and performance of sloped-coastal structures. Finally, The present results may be useful for designing some kinds of new sloped-coastal structures including the effect of sea-level rise.

Effect of tapered-end shape of FRP sheets on stress concentration in strengthened beams under thermal load

  • Repairing and strengthening structural members by bonding composite materials have received a considerable attention in recent years. The major problem when using bonded FRP or steel plates to strengthen existing structures is the high interfacial stresses that may be built up near the plate ends which lead to premature failure of the structure. As a result, many researchers have developed several analytical methods to predict the interface performance of bonded repairs under various types of loading. In this paper, a numerical solution using finite - difference method (FDM) is used to calculate the interfacial stress distribution in beams strengthened with FRP plate having a tapered ends under thermal loading. Different thinning profiles are investigated since the later can significantly reduce the stress concentration. In the present theoretical analysis, the adherend shear deformations are taken into account by assuming a parabolic shear stress through the thickness of both beam and bonded plate. The shear correction factor for I-section beams is also included in the solution. Numerical results from the present analysis are presented to demonstrate the advantages of use the tapers in design of strengthened beams.
