국내분리 유아 로타바이러스의 혈청형과 염기서열 분석 (Serotype and Nucleotide Analysis of Human Rotavirus Isolates in Korea)

  • Rotaviruses belong to Reoviridae causes diarrhea in human beings as well as domestic animals. This study was conducted to see what type of human rotaviruses are distributed in Seoul and Kyung-gi province. Twenty two of 81 patients showed rotavirus positive with diagnostic kit and RNA electropherosis. We isolated all of rotaviruses from the patients. Electropherotypes of 22 isolates showed 4:2:3:2 pattern whereas those migration patterns were long type. All of those isolates belonged to group A. Twenty out of 22 isolates reacted with monoclonal antibodies specific to G1, P1A and subgroup II, whereas rest of them, A-29 and K-30 reacted with subgroup I specific monoclonal antibody. The nucleotide sequence of an isolate K-21 showed $98{\sim}100%$ and $90{\sim}96%$ homologies with those of Wa and KU strain, respectively.

Negative stain을 이용한 Rotavirus의 투과전자현미경적 구조 (An Electron Microscopic Structure of Rotavirus by Negative Stain)

  • The Rotaviruses are members of the family Reoviridae and are the major cause of severe childhood gastroenteritis worldwide. Recently, electron microscopy has been used to detect non-group A rotaviruses to determine a relatively high resolution structure of the rotavirion. Mature, infectious virions(double-shelled particles) have a diameter of approximately 70nm, and have a capsid structure composed of two concentric protein layers. We have studied patient's stool specimen by negative staining technique complete removal of sucrose suspension. This negative staining technique that could be carried out in about 30 minutes and that could be used with crude stool specimen was an advantage of major significance. Removal of sucrose in the sample by has been completed washing with distilled of sucrose and by washing with distilled water. Ultrastructurally, typical feature of rotavirus has a double capsid construction with an inner capsid of 55nm and on outer 65-70nm diameter can be clearly demonstrated.

동남아시아로부터 수입된 paradise fish Macropodus opercularis로부터 분리한 reovirus의 특성 (Characterization of an isolated reovirus from the paradise fish Macropodus opercularis imported from Southeast Asia)

  • 2008년 3월 인도네시아로부터 수입된 paradise fish에서 대량폐사가 발생하여 병원체 검사를 실시한 결과, megalocytivirus 및 Mycobacterium sp.가 검출되었으며, CHSE-214세포에서 미동정 바이러스가 분리되었다. 본 연구에서는 미동정 바이러스의 특성을 조사하였으며, 또한 병원성 실험을 통해 paradise fish의 대량 폐사와의 연관성을 조사하였다. 분리 바이러스는 CHSE-214, BF-2, GF, SSN-1, FSP 및 FFN 세포에서 합포체의 세포변성효과를 나타내었고 IUdR 및 chloroform 처리에 감염성이 상실되지 않으며, 산 (pH 3), 알카리 (pH 11) 및 열 ($56^{\circ}C$, 30분)에 안정하였다. 핵산 분석 결과에서는 적어도 10개의 segment (0.7-3.6 kb)를 지닌 RNA 바이러스로 확인되었으며, 전자현미경 관찰 결과, 약 65 nm 크기로 인벨롭이 없으며 2중의 capsid를 지닌 바이러스가 세포질에서 관찰되었다. 이상의 결과는 Reoviridae family에 속하는 바이러스의 전형적인 특성과 일치하므로 paradise fish에서 분리된 바이러스는 reovirus로 확인되었다. Reovirus를 사용한 감염실험에서는 병원성이 나타나지 않아, reovirus는 paradise fish의 대량 폐사의 직접적인 원인이 아닐 것으로 사료되었다.

A Cytoplasmic Polyhedrosis Virus Isolated from the Pine Processionary Caterpillar, Thaumetopoea pityocampa

  • A cytoplasmic polyhedrosis virus (CPV) was isolated from the larvae of Thaumetopoea pityocampa and shown to cause an infection of midgut cells. This viral infection revealed several important diagnostic symptoms, including discoloration of the posterior midgut, reduced feeding, and extended development time of the larvae. The virus infection is lethal to Thaumetopoea pityocampa, and with the increasing doses kills the larvae within 4-5 days post infection. Electron microscopy studies showed typical cytoplasmic polyhedral inclusion bodies that are icosahedral, and ranged from 2.4 to $5.3{\mu}m$ in diameter. Electrophoretic analysis of the RNA genome showed that the virus has a genome composed of 10 equimolar RNA segments with the sizes of 3,907, 3,716, 3,628, 3,249, 2,726, 1,914, 1,815, 1,256, 1,058, and 899 bp, respectively. Based on morphology and nucleic acid analysis, this virus was named Thaumetopoea pityocampa cytoplasmic polyhedrosis virus (TpCPV), and belongs to the genus Cypovirus, family Reoviridae.

벼오갈병 바이러스 P12 단백질의 벼 감염세포 내 소재양식 (In Situ Localization of Rice dwarf phytoreovirus P12 Protein in Infected Rice Plant)

  • 벼오갈병바이러스(Rice dwarf virus, RDV)는 Phytoreovius 속에 속하며 게놈은 double stranded RNA의 12분절로 이루어져 있는데 전기영동상에서 크기가 큰 것부터 S1에서 S12로 명명한다. 본 실험에서는 RDV 입자 및 P12 단백질의 세포질 내에서의 소재양식을 확인하였다. 바이러스 입자 및 P12 단백질 특이적 항혈청에 protein A-gold 입자를 immunolabelling하여 전자현미경에서 gold입자의 존재양상을 관찰하였다. 바이러스 입자 특이적 항혈청을 사용한 경우에는 gold 입자가 세포질내의 바이러스 입자에 특이적으로 반응한 것이 관찰되었다. P12단백질 특이적 항혈청의 경우 gold 입자가 세포질내의 세포소기관에 특이적으로 존재하지 않고 세포질전체에 산재하여 존재하였다. 이 결과로서 RDV S12가 코드하는 단백질 P12가 세포질내에 존재함을 확인하였다.

한국영아에서 분리된 로타바이러스의 VP7 유전자형 및 염기서열 분석 (Typing and Sequence Analysis of the VP7 Gene of Rotavirus Isolated from Infants in Korea)

  • Rotaviruses are the most common cause of severe vomiting and diarrhea in children worldwide and classified as a genus in the family Reoviridae. Rotavirus has eleven segmented dsRNAs and the virion consists of three shells. Outer capsid VP7 and VP4 induce neutralizing antibodies and are classified into G types (glycoprotein VP7) and P types (protease-sensitive VP4). Characterization of VP7 gene of Korean isolates of human rotavirus was performed using multiplex PCR and nucleotide sequence analysis. After RT-PCR amplification of full length (1,062 bp) of VP7 genes, the amplified PCR products were G typed by multiplex PCR and the nucleotide sequences were compared with those of reference rotavirus from GenBank. The G type analysis revealed that 25% (2/8) belong to G1, whereas 37.5% (3/8) benong to G2 and G4, respectively. The Korean isolates within the same serotypes showed high homology of nucleotide sequences and could be discriminated from foreign isolates exception with two strains (CAU009 and CAU022). But Korean isolates CAU009 and CAU022 were close related into japanease isolates 417 (99.2%) and indian isolates (97.6%) than Korean isolatese. Our results showed that these two strains were supposed to be originated from abroad. As a results, The G typing and nucleotide sequence analysis of VP7 gene of rotavirus isolated from infants in Korea could be used for identification, serotying and determination of novel or unusual strains of rotaviruses.

벼 흑조위축병 바이러스의 분자생물학적 연구 (A Molecular Study of Rice Black-Streaked Dwarf Virus)

  • 우리나라에서 발생하는 주요 벼 바이러스로써 저항성 유전자원이 없어 현재까지 저항성 품종이 육성되지 못하고 있는 흑조위축병(Rice Black-Streaked Dwarf Virus, RBSDV)에 대한 유전정보에 대하여 연구하였다. 매개충인 보독 애멸구를 이용하여 이병주를 생산한 후 바이러스 입자를 순수 분리하여 전기영동한 결과 10개의 band를 확인하였다. RBSDV RNA로부터 역전사 효소를 이용 cDNA를 합성한 후 ${\lambda}gt11$에 삽입하여 cDNA library를 만들었다. 이 library에서 6개의 단편을 선발하였으며 그중 한 개의 clone(pRV3)은 hybridization을 통해 RBSDV 게놈 조각 3번 유래인 것을 확인하였다. pRV3의 염기서열을 결정한 결과 2개의 ORF의 일부분들을 갖고 있었으며 이것은 바이러스 저항성 작물개발에 이용될 수 있을 것으로 생각된다.
