An investigation of the effect of epigallocathechin-3-gallate on the renal dipeptidase release

  • The action of epigallocatechin-3-gi:lllale (EGCG). polyphenol compound from green lea, on the release pattern of glycosylphosphatidylinositol (GPI)-anchored renal dipeptidase (RDPase) from renal proximal tubules (PTs) was examined. EGCG had a stronger inhibitory effect on the release of RDPase than alkaline phosphatase (APase), another GPI-anchored ectoenzyme used as a reference protein. The effect of EGCG on cell viability as assessed by MTT test was found to be intact, and moreover, was indicative of potent cell activation or proliferation. (omitted)

  • Background: Human seminal plasma (HSP)-induced hypersensitivity is one of the serious complications with sexual intercourse. The clinical manifestations of HSP-induced hypersensitivity may be related to the release of vasoactive mediators from mast cell induced by HSP. It has recently been reported that HSP modulates immune systems and induces mast cell degranulation and histamine release from rat peritoneal mast cells (RPMC). Ketotifen and disodium cromoglycate (DSCG), anti-asthmatic and anti-allergic drugs, have a role of mast cell stabilization and inhibit mast cell-induced leukocyte rolling and adhesion. But the inhibitory agents of HSP-induced mast cell activation are unknown. This study was performed to investigate the effects of DSCG and ketotifen on the HSP-induced mast cell activation. Methods: For this, influences of DSCG and ketotifen on the human seminal plasma-induced degranulation, histamine release and morphological changes of RPMC were observed. Results: The mast cell degranulation and histamine release of RPMC by HSP were induced in a dose-dependent fashion. The HSP-induced cytomorphological changes such as swelling, intracellular vacoules, and interrupted cell boundary were significantly inhibited by pretreatment with DSCG or ketotifen. DSCG and Ketotifen inhibited the HSP-induced degranulation and histamine release from RPMC. Conclusion: From the above results, it is suggested that DSCG and ketotifen have a inhibitory effect of the HSP-induced mast cell activation. DSCG and ketotifen may be used for treatment of HSP-induced hypersensitivity.

Mucin 분비에 미치는 대청룡탕(大靑龍湯) 및 <석실비록(石室秘錄)> 급치법방(急治法方)에 대한 연구 (The Effect of Daecheongryong-tang and prescription A on airway mucin secretion)

  • Objectives : This study was done with intend to investigate whether two oriental medical prescriptions, daecheongryong-tang (DCRT) and prescription A (P-A) significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. Methods : Confluent HTSE cells were metabolically radiolabeled with $^3H$-glucosamine for 24 hrs and chased for 30 min in the presence of DCRT or P-A to assess the effect of each agent on $^3H$-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase (LDH) release. Also, the effects of DCRT and P-A on contractility of isolated tracheal smooth muscle were investigated. Results were as follows : 1. DCRT significantly inhibited mucin release from cultured HTSE cells, with significant cytotoxicity. 2. P-A significantly increased mucin release from cultured HTSE cells, with significant cytotoxicity. 3. DCRT inhibited Ach-induced contraction of isolated tracheal smooth muscle. 4. P-A also inhibited Ach-induced contraction of isolated tracheal smooth muscle. Conclusion: Results suggest that DCRT and P-A have regulating effects on mucin secretion from goblet cells. Further investigation is needed, because of the value in finding novel agents to this purpose, and these oriental medical prescriptions have potential for this role.

해양 퇴적물에서 인 용출 차단을 위한 반응성 피복 소재로서 제강슬래그의 적용성 검토 (Applicability Assessment of Steel Slag as Reactive Capping Material for Blocking Phosphorus Release from Marine Sediment)

  • We investigated the applicability of steel slag as a capping material in order to minimize phosphorus(P) release into seawater. Steel slag is a byproduct from the iron and steel industries and the use of steel slag has some advantages in respect of both cost and environmental concern. P removal by steel slag were studied in a batch system with respect to changes in contact time and initial concentration. Kinetic adsorption data were described well by pseudo 2nd order model, indicating rate limiting step for P adsorption to steel slag is chemical sorption. Equilibrium adsorption data fitted well to Langmuir isotherm model which describes for single layer adsorption. The maximum P adsorption capacity of steel slag was 7.134 mg-P/L. Increasing the depth of steel slag produced a positive effect on interruption of P release. More than 3 cm of steel slag was effective for blocking P release and 5 cm of steel slag was recommended as the depth for capping of P contaminated marine sediments. Increasing P concentration and flow rate had a negative effect on P removal ratio. It was concluded that the steel slag has a potential capping material for blocking P release from marine sediments.

폴리카프로락톤 매트릭스로부터 세파드록실의 방출에 미치는 BSA의 영향 (The Effect of BSA on the Release of Cefadroxil from a Polycaprolactone Matrix)

  • In order to investigate the effect of bovine serum albumin (BSA), as a pore former, on the controlled release of an antibiotic from a biodegradable polymeric device, polycaprolactone (PCL)-cefadroxil matrices were prepared by the solvent casting method. The amount of cefadroxil released from various formulations at $37^{\circ}C$ was measured by HPLC. The duration of antimicrobial activity of matrices against S. aureus was evaluated by measuring the diameters of the inhibition zone. The morphology of the matrices was investigated by scanning electron microscopy (SEM). The release rate and extent of cefadroxil from PCL matrix increased as the loading dose and particle size of BSA/cefadroxil mixture powder increased. Cefadroxil released from the matrix exhibited antibacterial activity for up to 4 days. SEM of the cross-section of matrix showed the typical channel formation after 3 days of release study. Thus, a biodegradable polymeric matrix loaded with antibiotic/BSA mixture can effectively prevent bacterial infection on its surface, thereby bringing about an enhancement of biocompatibility of biomaterials.

취연탕(取淵湯) 및 치효산가미방(治哮散加味方)이 기도점액 분비 및 기관평활근 긴장도에 미치는 영향 (Effects of Chwiyeon-tang and Chihyosan-gainbang on airway mucus secretion and contractility oftracheal smooth muscle)

  • Objectives : In the present study, the authors intended to investigate whether two oriental medical prescriptions named chwiyeon-tang and chihyosan-gamibang significantly affect much release from cultured hamster tracheal surface epithelial HTSE cells. Methods : Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of chwiyeon-tang or chihyosan-gamibang to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase LDH release. Also, the effect of chwiyeon-tang and chihyosan-gamibang on contractility of isolated tracheal smooth muscle were investigated. Results : (1) Chwiyeon-tang significantly inhibited mucin release from cultured HTSE cells, with significant cytotoxicity ; (2) Chihyosan-gamibang significantly stimulated mucin release from cultured HTSE cells, with minute cytotoxicity ; (3) Chwiyeon-tang and Chihyosan-gamibang did not affect contractility of isolated tracheal smooth muscle. Conclusions : We suggest that the effects of Chwiyeon-tang and Chihyosan-gamibang with their components should be further investigated and it is of great value to find, from oriental medical prescriptions, noel agents which might regulate mucin secretion from airway goblet cells.

경항통 환자의 치료시 자세이완치료 효과에 대한 임상적 연구 (The Clinical Study on the Effect of Positional Release Therapy on Patients of Neck Pain)

  • Objectives: The purpose of this study is to evaluate the therapeutic effect of Positional Release Therapy on patients of neck pain, Methods: 12 patients was treated by Positional Release Therapy(PRT) combined with acupuncture, We measured the Visual Analog Scale(VAS) and Neck Disability Index(NDI) before and after treatment The statistical analysis was performed by using the Wilcoxon signed rank test. Results: 1. Patients showed significant improvement in VAS after treatrnent(p<0.05). 2. Patients showed significant improvement in NDI after treatment(p<0.05). Conclusion: These results imply that Positional Release Therapy combined with acupuncture would be effective on relieving pain and increasing functional ability in daily-life of patients of neck pain.

세파드록실의 방출에 미치는 폴리우레탄 매트릭스 제조방법의 영향 (The Effect of Fabrication Methods on the Release of Cefadroxil from a Polyurethane Matrix)

The morphology of the matrices was investigated by scanning electron microscopy (SEM). Changing the fabrication method could alter the release rate of cefadroxil from the matrix. The matrix fabricated by the freeze drying method had more porous inner structure and showed higher release rate than that prepared by the solvent casting method. However, the duration of antimicrobial activity was shorter when the matrix was fabricated by the freeze drying method.

참전복(Haliotis discus hannai) 방류효과 향상을 위한 연구 (Enhancement of settlement of artificially released abalone, Haliotis discus hannai)

  • Settlement of artificially released abalone, Haliotis discus hannai at the marine farming area in Jeollanam-do in 2007 was very efficient by showing the recapture of 2.0-5.2 ind./m2 and the survival rate of 60-67%. The growth of shell after 1 year was highest in Hwoenggando by 75.9 mm and the lowest in Hwasan by 64.6 mm. Our result especially showed that the effect of the stock recruitment was the most efficient when abalone was released. The shell growth of the released abalone was better when sea mustard (Undaria sp.) was provided as food (shell length was up to 23.9 mm) or predators were periodically removed (shell length was up to 23.8 mm) in order to enhance the effect of the artificial release than the control group. The recapture rate was also higher than the control. However due to the difficulty of managing the release areas, it will be more efficient to release the recruited stocks after considering various biological and ecological factors such as survey of suitable release areas, stock size, release density, and health of stocks.

  • Solvent fractions were prepared from traditional herbal drugs which of methanol extracts inhibited $CCl_4$-induced cytotoxicity in primary cultured rat hepatocytes and c ontinuously assayed their effects. Ethylacetate and n-buthanol fractions from Cibotii Rhizoma and chloroform fraction from Gelatina Nigra inhibited the release of LDH and GPT from $CCl_4$-treated hepatocytes, respectively. Water fraction (WAR) among solvent fractions from Astragali Radix showed the most potent inhibitory effect on the release of GOT or GPT by treatment with $CCl_4$. All of solvent fractions prepared from Eucommiae Cortex had no effect on $CCl_4$-induced cytotoxicity. Chloroform and ethylacetate fractions from Rehmanniae Radix Preparata increased the release of GPT from $CCl_4$-treated hepatocytes. n-Hexan, chloroform or ethylacetate fraction from 5 herbal drugs increased the release of LDH, GOT or GPT from normal hepatocytes at the dose of 1.Omg/ml. Administration of WAR suppressed the elevation of GOT, ALP activities and MDA contents in the serum as well as in the liver tissue of $CCl_4$-intoxicated rats. Based on these results, isolation of antihepatotoxic substances from WAR is under the process.
