Objectives : This study presents a high performance liquid chromatography - electrospray ionization-mass spectrometer (HPLC-MS/MS) methods for the quantitative and qualitative analysis of various active components in Samul-tang, which is composed of four crude herbs. Methods : HPLC-ESI-MS/MS for the determinations of paeoniflorin and 5-HMF (5-hydroxymethyl 2-furaldehyde) in the Samul-tang, the separation method was performed on an COSMOS1L 5C18-AR-Il (2.0 X 150 mm I.D.) column by gradient elution with 0.1% acetic acid and 5% CH3CN in water (A)-0.1% acetic acid and 5% H20 in CH3CN (B) as the mobile phase at a flow-rate of 300 ${\mu}L/min$ with detection at quadrupole mass spectrometer. The all marker substances were always detected as the base peaks in the positive ion mode. Results : The paeoniflorin of Paeoniae Radix in Samul-tang showed a strong base peak [M+H2O]+ in the positive detection mode to give the following as; paeoniflorin (498.109 [M+H2O]+, 479.8 [M]+, 301 [M-glucose]+, 179.3 [glucose]+). Based on the HPLC retnetion time and MS of standard compounds confirmed the identity of active compounds in Rehmanniae Radix Preparata as follows; 5-HMF (127.0[M+H]+, 109.3 [M-OH]+) in the positive ion mode. Conclusion : According to the above results, HPLC-ESI-MS method permits assignment of tentative structures such as paeoniflorin and 5-HMF in the Samul-tang.
Directional words like 'ascending(升, sheng)', 'descending(降, jiang)', 'floating(浮, fu)', 'sinking(沈, chen) are technical terminologies of Oriental Medicine to express the directions of material medica functioning-at the phase of Qi-superiorly, inferiorly, exteriorly, and interiorly in human body, and are constituting one of the major principles in prescribing medicines. Lacking the objective methodology to measure or, at least determine Qi, However, we can not expect to secure meaningful data to verify those directional terms. This problematic situation caused chaotic discrepancy between theory and practice of Oriental Medicine. Thus, adopting the method of dynamic meditation 'Hanalmomjit' of Lux Vitae meditation society, by which various state of Qi-sensational movements can be expressed, we compared its moves with the directional terms of traditional canons in Oriental Medicine. In this thesis, the experiment of expressing materia medica via 'Hanalmomjit' is resulted out and compared with the terms, confirmed to have a certain meaningfulness. The new method showed these results below: Herba Menthae, described to have the disposition of ascending and floating, showed 60% of ascending movements in 'Hanalmomjit' experiment. Radix Rehmanniae Praeparata, described to have the disposition of descending and sinking, showed 50% of descending movements. Radix Glycyrrhizae, described to have plain disposition, showed 60% of central movements. Rhizoma Atractylodis and Pericarpium Citri Reticulatae, described to have the disposition of ascending and floating, showed also ascending movements. Cortex Magnoliae Officinalis, described to have the disposition of descending and sinking, showed also descending movements. In conclusion, qi-sensational movement of 'Hanalmomjit' for materia medica showed sufficient relations described in classics, proving itself to be useful method for objective demonstration of material medica.
Objective : Recently, hair has become one of the important aspects of beauty. So, there are many studies about prevention and treatment of alopecia. Finasteride and minoxidil have been known to drug for alopecia treatment. However, these agents have side effects such as irritation, pruritus, and erythema when they were used for a long time. So, we assessed hair growth effect on herbal mixture (RAA) include in Rehmanniae Radix Preparata, Achyranthis Radix, and Acanthopanacis Cortex. Methods : 6 weeks aged C57BL/6 mice were divided three treatment groups. : CON group (vehicle solution), MNXD group (positive control, 3% minoxidil), RAA group (15% RAA solution). And we applied 200 ㎕ of three groups to shaved dorsal skin every day. Hair growth effects of treatment were determined through phototrichogram by folliscope and hair follicle morphometry by H&E staining. And we assessed hair growth-related gene (VEGF, IGF-1, TGF-β1) expressions by western blot and 5-α reductase II analysis of dermal (skin) or internal organ (prostate gland). Results : Hair density and hair follicle size in MNXD or RAA group was enhanced compared to those of CON group, respectively. Also, the protein expression levels in dermal of VEGF, IGF-1 increased but TGF-β1 decreased in RAA and MNXD group, compared to CON group, respectively. 5-α reductase II levels of tissues in MNXD or RAA group significantly decreased compared to those of CON group, respectively. Conclusion : These results suggest that RAA has the promoting effect on hair growth.
Objectives : Haedoksamul-tang (HSTE), a water extract from a mixture of Phellodendri Cortex, Coptidis, Scutellariae Radix, Gardeniae Fructus, Angelica acutiloba Radix, Cnidii Rhizoma, Paeoniae Radix, Rehmanniae Radix, has been traditionally used for allergic skin diseases such as atopic dermatitis and contact dermatitis in oriental countries. However, little is known about the effects of aqueous extract of HSTE on trimellitic anhydride (TMA)-induced contact hypersensitivity (CHS) in a mouse model. Methods : In this study, we investigate the pharmacological effects of HSTE on TMA-induced CHS in Balb/c mice. Contact hypersensitivity was induced in mice by topically sensitizing and challenging with TMA in flank skin and ears during oral administration (for 17 days) and topical treatment (30 min before challenge) with HSTE. We examined the effects of HSTE on IgE and IgG1 levels, inflammatory parameters in ear tissues, CD4+/CD8+ ratio, cytokine and chemokine production in sera, tissues, and immune cells from TMA-sensitized mice. Results : Oral and topical administration with HSTE reduced, in a dose dependent manner, thickness and leukocyte infiltration of ear tissues and IgE levels in serum from mice sensitized with TMA. In addition, auricula lymph node cells isolated from TMA-sensitized mice significantly elevated the expression ratio of CD4+/CD8+ as well as increased the production of IL-4, IL-5, IL-13 and IFN-γ by ex vivo stimulation with antibodies against CD3 and CD28, and these inflammatory indexes, except for IFN-γ, were significantly suppressed by orally and topically administration of HSTE. Furthermore, stimulation of auricula lymph node cells from TMA-sensitized mice with antibodies against CD3 and CD28 increased the production of MCP-1/CCL2 and MIP-1α/CCL3, and these effects were inhibited in a dose-dependent manner in cells from mice treated with HSTE. Conclusions : These results suggest that HSTE can be used for treating contact hypersensitivity by inhibiting leukocyte infiltration as well as production of serum IgE and chemokine/Th2 cytokine in an animal model.
Analgesic, anticonvulsive, sedative and antipyretic actions of Yangkyuksanwhatang which is composed of nine crude drugs including Rehmanniae Radix, and its effects on isolated ileum and blood vessel were investigated. The results of the studies were summerized as follows; 1. Analgesic and antipyretic effects were observed. 2. Suppressive actions were not shown on the convulsion induced by strychnine, but significant effects were noted on the convulsion induced by picrotoxin. 3. A prolongation of anesthetic time induced by pentobarbital sodium was significantly observed. 4. Relaxing actions were noted on the ileum of mice, and also, same effects were recognized on the contraction of the ileum due to acetylcholine chloride and barium chloride. 5. The expansion of blood vessels by relaxation of smooth muscle and hypotensive effects were noted.
Through analyzing the frequencies of medicinal herb combinations in the prescriptions of "Bangyakhappyeon(方藥合編)", we could understand the characteristics of prescription composition and author's medical thoughts. So we conducted this study that analyze combination of 324 medicinal herbs in 467 prescriptions, and found meaningful combinations of 5,045. Among these, the most meaningful combination is the one of 10 herbs, Glycyrrhiza uralensis Fischer, Angelica gigas Nakai, Ziziphus jujube Mille, Atractylodes japonica Koidzumi, Poria cocas Wolf, Zingiber officinale, Rehmanniae radix preparata, Cinnamomum loureirii, Panax ginseng C.A.Meyer, Astragalus membranaceus. This combination is Sibjeondaebo-tang(Shiquandabu-tang) without Cnidium officinale and Paeonia japonica which compose Samul-tang(Siwu-tang). It means this combination is kind of modified Sibjeondaebo-tang(Shiquandabu-tang), which is more effective in tonifying qi than tonifying blood. Prescriptions of "Bangyakhappyeon(方藥合編)" involve herbs mostly used to improve weak and frail health and the herbs which are more effective in tonifying qi than tonifying blood. Through this study, we know the author' s one of medical view points on tonifying qi and blood, attaching importance to tonifying qi relatively to tonifying blood.
Naetakcheonkeumsan(NCS) was a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effect of NCS on the anti-cancer and proliferation of lymphocytes in normal mouse group, L1210 cells-transplanted mouse group and anti-cancer drug (vincristine) 0.005mg/kg were injected mouse(Ll210 cells-transplanted) group. We used NCS extract with freeze-dried, 8wks-old male mice, and Ll210 cell lines for this Study, The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follow ; Group C(NCS plus Rehmanniae Radix Preparat administered group) inhibited proliferaion of lymphocytes in normal mouse group and Ll210 cells transplanted mouse group. Group A(NCS administered group) and Group B(NCS plus Cervi pantotrichum Cornu administered group) inhibited proliferation of Ll210 cells in Ll210 cells-transplanted mouse group and anti-cancer drug were injected mouse(Ll210 cells-transplanted) group. Group C incresed proliferation of L1210 cells in L1210 cells-transplanted mouse group, but inhibited in anti-cancer drug(vincristine) 0.005mg/kg were injected mouse(L1210 cells-transplanted) group.
Rehmannia Radix Preparata (RRP) used to nourish Eum and enrich blood for consumptive fever, aching, and limpness of the loins and knees, and to replenish essence for tinnitus, premature greying of beard and hair. In the present study, we studied about the protective effect of RRP on hydrogen peroxide-induced oxidative stress in human vascular endothelial cells. ECV304 cells were preincubated with RRP (100, 200, 300 and $400{\mu}g/m{\ell}$) for 12hr and then treated with $600{\mu}M$$H_2O_2$ for 12hr. The protective effects of RRP on $H_2O_2$-induced apoptosis in ECV304 cells was determined by using MTT assay, FDA-PI staining, flow cytometric analysis, caspase-3 activity assay, ROS assay and western blot. The results of this experiment showed that RRP inhibited $H_2O_2$-induced apoptosis and ROS production in ECV304 cells. Moreover, RRP increased ERK activation that decreased in $H_2O_2$-treated ECV304 cells, and inhibited p38 and JNK activation. Furthermore, RRP increased expression of heme oxygenase-1 (HO-1) in $H_2O_2$-treated ECV304 cells. Also, HO-1 protein expression induced by RRP was reduced by the addition of ERK inhibitor (PD98059) in $H_2O_2$-treated ECV304 cells. These results suggest that protective effect of RRP on $H_2O_2$-induced oxidative stress in ECV304 cells may be associated with increase of ERK activation and HO-1 protein, and reduction of p38 and JNK activation.
