Cloning and Functional Studies of Pro-Apoptotic MCL-1ES BH3M (세포사멸을 유도하는 새로운 단백질인 MCL-1ES BH3M의 클로닝 및 기능연구)

  • BCL-2 family members are essential protein for the regulation of cell death and survival consisting both antiapoptotic and pro-apoptotic proteins. In the present study, we designed and cloned a new apoptotic molecule MCL-1ES BH3M coding a modified protein of MCL-1L. Compared to MCL-1L protein, MCL-1ES BH3M lacks the PEST motifs known to be involved in MCL-1L protein degradation and has seven mutated residues in BH3 domain critical for dimerization with BCL-2 family members. Overexpression of MCL-1ES BH3M induced death of different cells, and its cell killing effect was not blocked by forced expression of the pro-survival protein MCL-1L. Expression of MCL-1ES BH3M protein led to the activation of caspase 9 and caspase 3, suggesting apoptotic cell death, and confocal fluorescent microscopic analyses showed that MCL-1ES BH3M was partially localized in mitochondria. In conclusion, we reported a new apoptotic molecule and determined its cell death activity in cells.

Cognitive-Motor Interaction-Based Instrument Playing for Improving Early Social Skills of Children With Autism Spectrum Disorder (ASD 아동의 초기 사회기술 향상을 위한 인지-운동 통합 기반 악기연주 중재)

  • This study aimed to investigate the effect of cognitive-motor interaction-based instrument playing on the early social skills of children with autism spectrum disorder (ASD). Nine children with ASD, averaging 5.6 years of age, participated in twelve individual sessions lasting 30 minutes each. The intervention comprised five stages: self-regulation, motivation to engage in social interaction, acceptance of a partner in co-playing activities, interpersonal coordination with a partner, and engagement in joint music playing. To evaluate changes in early social skills, joint attention and social interaction behaviors were observed, and the Social Responsiveness Scale (SRS-2) was administered pre- and post-intervention. Social synchronization was measured through a dyadic drum task, measuring synchronization accuracy, duration, and timing. Following the intervention, all nine children demonstrated increased early social behaviors, although there were no significant differences in SRS scores. Moreover, synchronized movement improved significantly in accuracy and maintained duration but not in reaction time. This study highlights the significance of recognizing the cognitive-motor interplay as crucial element in facilitating early social skills development in children with ASD.

A Study on Exploring Factors Influencing Military Security Level (Based on the Theory of Planned Behavior, Deterrence and Protection Motivation) (군(軍) 보안수준에 미치는 영향요인 탐색에 관한 연구 (계획 행동이론, 억제이론, 보호 동기 이론을 중심으로))

  • Recently, as the environment of the 4th industrial revolution has arrived, the opening, sharing and convergence of data are actively being achieved in any organization. However, the opening and sharing of data inevitably leads to security vulnerability and there is ambivalence that is a threat that can affect the existence of an organization operated in the 4th industrial revolution environment. Especially security issues in the organization of the military can be a threat to the state, not the military itself, so it is always necessary to maintain a high level of security discipline. In this paper, 14 variables were selected through structural equation model applying theory of planned behavior, deterrence and protection motivation to find out the security level development measures by extracting factors that can affect security level. As a result, the theory of planned behavior that the security knowledge embodied through the usual security regulation education and evaluation affects the behavior was adopted, and the theory of deterrence and protection motivation showed the significance of the rejection level. In addition, it was confirmed that the variables that have the greatest impact on the military security level through the measured values of the three-year security audit were commanders and mental security. In conclusion, in order to improve the security level, it is suggested that security education, definite reward and punishment, and security system upgrading should be firmly established and mental security posture should be secured.

Development of Promoters Inducing Gene Expression in Poultry Muscle Cells (가금 근육세포에서 유전자 발현을 유도하는 프로모터 개발)

  • The skeletal muscles of livestock play a crucial role as protein sources for humans, and the consumption of poultry meat is steadily increasing worldwide. Numerous genes, including myogenic regulatory factors, are involved in myogenesis, and precise regulation of them is essential. In this study, genes specifically expressed in muscles were selected, and their promoters were cloned and analyzed. The analysis of gene expression in various tissues of animals revealed that many genes exhibited specific expression patterns in skeletal muscles, with TNNT3, TNNC2, and MYF6 genes showing similar patterns in poultry. The promoter regions of three genes were amplified by polymerase chain reaction to sizes of 1.2 kb, 1.03 kb, and 1.43 kb, respectively. These fragments were then inserted at the front of the enhanced green fluorescent protein gene in vectors. It was confirmed that the sequences of three promoters closely matched the chicken genome sequences. Upon introducing vectors with each promoter into QM7 quail muscle cells, all three promoters successfully induced the expression of the green fluorescent protein. The brightness of the green fluorescence in each promoter was approximately seven times dimmer compared to the control, CMV-IE promoter. It is predicted that more than 230 transcription factors can bind to each promoter, especially various transcription factors expressed in muscles, including myogenic regulatory factors such as MYF5, MYOD, and MYOG. These promoters can be valuable for studying gene expression in poultry muscle cells, and further research is needed to precisely investigate the regulatory region of gene expression in promoters.

A Study on the Policy Direction of the Online Platform Industry: Focusing on PEST-SWOT-AHP Analysis for Scholars and Researchers (온라인 플랫폼 산업의 정책 방향성 연구: 학자 및 연구자 대상 PEST-SWOT-AHP 분석을 중심으로)

  • This study proposes a developmental policy direction for the online platform industry, moving away from the regulatory-centered discussions that have predominated thus far. To offer policy directions, the PEST-SWOT-AHP analysis model was employed. The study first categorizes the issues of the domestic online platform industry into political, economic, social, and technological aspects, which are then further categorized into 16 strengths, weaknesses, opportunities, and threats. The relative importance among these factors was measured, leading to the derivation of four final strategies. The analysis indicates that policy directions should prioritize addressing weaknesses, with 'improving regulations that hinder innovation' being the most important factor across all categories, while technological factors were consistently rated highly in importance apart from this. Accordingly, the policy direction for the domestic online platform industry suggests avoiding excessive regulation and instead emphasizing policy support centered around technological development. This study is significant in that it presents a macroscopic developmental direction for online platform policies that have not been discussed in existing academic research, and it provides professional and objective indicators through consensus among scholars and researchers. In the future, it is hoped that research will continue to propose detailed policy strategies and implementation systems based on a macroscopic perspective.

A Study on the Development of Training Model by Enforcement of the IP Code(SOLAS Chapter XV)

  • Through the 106th session of the International Maritime Organization(IMO)'s Maritime Safety Committee(MSC), a mandatory safety training requirement for all personnel transferred or accommodated for offshore industrial activities was established and adopted under the name of SOLAS Chapter XV, IP(Industrial Personnel) Code. This regulation mandates pre-boarding safety training to enable individuals to anticipate and mitigate hazardous risks in navigation and operational environments. Consequently, the IP Code includes provisions regarding the training content for industrial personnel and regulations for the refusal of master who has a full responsibility for individuals who have not completed the required training(non-qualified industrial personnel). Referred to as the IP Code, this agreement is set to enter into force in July 2024, necessitating the establishment and operation of safety education for industrial personnel boarding ships before that date. Accordingly, this paper reviews the legal requirements related to training within IP code and analyzes the details of models including training objectives, target audience, duration, and course structure of safety trainings such as STCW, OPITO, GWO training, and other delegated training related to current ships. Additionally, it aims to propose a curriculum model for IP training courses which consists of a total of 16 hours over 2 days, offered by the Korea Institute of Maritime and Fisheries Technology, including teaching objectives, duration, and course structure.

GRIM-19 Ameliorates Multiple Sclerosis in a Mouse Model of Experimental Autoimmune Encephalomyelitis with Reciprocal Regulation of IFNγ/Th1 and IL-17A/Th17 Cells

  • The protein encoded by the Gene Associated with Retinoid-Interferon-Induced Mortality-19 (GRIM-19) is located in the mitochondrial inner membrane and is homologous to the NADH dehydrogenase 1-alpha subcomplex subunit 13 of the electron transport chain. Multiple sclerosis (MS) is a demyelinating disease that damages the brain and spinal cord. Although both the cause and mechanism of MS progression remain unclear, it is accepted that an immune disorder is involved. We explored whether GRIM-19 ameliorated MS by increasing the levels of inflammatory cytokines and immune cells; we used a mouse model of experimental autoimmune encephalomyelitis (EAE) to this end. Six-to-eight-week-old male C57BL/6, IFNγ-knockout (KO), and GRIM-19 transgenic mice were used; EAE was induced in all strains. A GRIM-19 overexpression vector (GRIM19 OVN) was electrophoretically injected intravenously. The levels of Th1 and Th17 cells were measured via flow cytometry, immunofluorescence, and immunohistochemical analysis. IL-17A and IFNγ expression levels were assessed via ELISA and quantitative PCR. IL-17A expression decreased and IFNγ expression increased in EAE mice that received injections of the GRIM-19 OVN. GRIM19 transgenic mice expressed more IFNγ than did wild-type mice; this inhibited EAE development. However, the effect of GRIM-19 overexpression on the EAE of IFNγ-KO mice did not differ from that of the empty vector. GRIM-19 expression was therapeutic for EAE mice, elevating the IFNγ level. GRIM-19 regulated the Th17/Treg cell balance.

Smart IoT Service Users' Compliance with Personal Information Protection Behavior: An Empirical Study on the Message Design Features to Induce Installation of Software Updates (스마트 IoT 서비스 사용자의 개인정보 보호 행동 준수: 소프트웨어 업데이트 유도를 위한 메세지 디자인 특성에 관한 실증 연구)

  • Smart home services are growing rapidly as the development of the Internet of Things (IoT) opens the era of the so-called "Connected Living." Although personal information leaks through smart home cameras are increasing, however, users-while concerned-tend to take passive measures to protect their personal information. This study theoretically explained and verified how to design effective software update notification messages for smart home cameras to ensure that users comply with the recommended security behavior (i.e., update installation). In a survey experiment participated in by 120 actual users, the effectiveness of both emotional appeals (i.e., security breach warning images for fear appeals) and rational appeals (i.e., loss-framed messages emphasizing the negative consequences of not installing the updates) were confirmed. The results of this study provide theoretical interpretations and practical guidelines on the message design features that are effective for threat appraisals (i.e., severity, vulnerability) of smart home camera users and their protection motivation.

Manual Development Research for the Diagnosis of the Introduction of Low-Floor Bus (저상버스 도입진단 매뉴얼 개발 연구)

  • With the recent revision of 「Act on Promotion of the Transportation Convenience of Mobility Disadvantaged Persons」 and the Enforcement Regulations of the Act, bus business operators must introduce low-floor buses when scrapping buses. On the other hand, in the case of routes where low-floor buses cannot be operated, bus business operators can be exempted from introducing low-floor buses with the approval of their transportation administrative agency according to Article 4-2 of 「Enforcement Regulation of the Act on Promotion of the Transportation Convenience of Mobility Disadvantaged Persons」. According to the data from the Korea Bus Transportation Associations Federation, approximately 5.9% of all city bus routes were surveyed as the exceptions to introducing low-floor buses. Nevertheless the proportion is expected to increase because some regions with difficulties introducing low-floor buses are not included when calculating the proportion. By confirming the process of approving exceptions for introducing low-floor buses through local governments, there was no specific examination method or standard for approval of exceptions. Hence, there is the problem that some routes are approved as exceptions to introducing low-floor buses, even though low-floor buses can be operated on those routes. Therefore, this study aims to develop a manual that can objectively diagnose the overall operation environment of low-floor buses, such as road geometry and road facilities. Future research plans to apply it to more cases and improve it for more precise application in various contexts.

Disease Resistance-Based Management of Alternaria Black Spot in Cruciferous Crops (병 저항성 기반 십자화과 작물의 검은무늬병 관리)

  • Alternaria black spots or blights in cruciferous crops have been devastating diseases worldwide and led to economic losses in broccoli, Chinese cabbage, kale, radish, rapeseed, etc. These diseases are caused by different Alternaria spp., including A. brassicae, A. brassicicola and A. raphani transmitted from infected seeds or insect vectors. Efforts to excavate disease resistance traits of cruciferous crops against Alternaria black spots or blights have been demonstrated. Genetic resource of disease resistance was investigated in the wild relatives of cruciferous crops, and different cultivars were screened under different inoculation conditions. Development of the disease-resistant lines against Alternaria black spots or blights was also tried via genetic transformation of the cruciferous crops using diverse plant defence-associated genes. Plant immunity activated by pre-treatment with chemicals, i. e. β-amino-n-butyric acid and melatonin, was suggested for reducing Alternaria black spots or blights in cruciferous crops. The disease resistance traits have also been evaluated in model plant Arabidopsis originating from different habitats. Various plant immunity-related mutants showing different disease responses from wild-type Arabidopsis provided valuable information for managing Alternaria black spots or blights in cruciferous crops. In particular, redox regulation and antioxidant responses altered in the Alternaria-infected mutants were discussed in this review.
