Changes in Protein Contents and Activities of Proteolytic Enzymes in Medicago sativa During Regrowth

  • An expreiment with non-nodulating alfalfa (Medicago sativa L.) plants was designed to investigate the changes in protein contents and the activities of proteolytic enzymes during a regrowth period of 24 d. Shoot removal caused a depression of root growth and significantly reduced protein contents in roots. An initial decline of root proteins for the first 10 d was followed by a rapid recovery from d 11 to 24. The major increase of regrowing shoot weight occurred also from d 11. The activities of aminopeptidase and endoprotease slightly decreased in regrowing leaves, while protein contents remains stable after shoot removal. Roots exhibited source behaviour with a rapid increase of endoprotease activities for the first 10 d of regrowth; about a 370% increase over the initial level was observed. Increase in endoprotease activity in roots coincided with the time of protein remobilization after shoot removal, indicating the important role of endoproteases in protein degradation.

Dry Matter Production , Growth Characteristics and Nutritive Value of Wild Birdsfoot trefoil (야생 버어드풋 트레포일의 건물수량 , 생육특성 및 사료적인 가치)

  • Birdsfoot trefoil grows wild in some parts of Korea, but specific information is lacking as to its growth characteristics, nutritive value and dry matter production potential. The objective of this study was to evaluate the usefulness of wild birdsfoot trefoil(Lotus cornicubus L.) as a forage. Wild birdsfoot trefoil and Au Dewey were field sown at Keongsan, Keongbuk in the spring of 19%. Emergence for Au Dewey was better than for wild birdsfoot trefoil. It was observed that the flowering date of the wild birdsfoot trefoil was similar to Au Dewey. Regrowth of Au Dewey was better than that of the wild birdsfoot trefoil at each harvesting. AAer the last harvesting date, October 1, there was no regrowth of the wild birdsfoot trefoil, but regrowth of Au Dewey measured IOcm. Weed infestation in the wild birdsfoot trefoil plots was higher than in the Au Dewey plots. The dry matter yields were significantly(P< 0.05) higher for Au Dewey than for the wild birdsfoot trefoil.

Dry Matter, Nitrogen Distribution and Organic Reserves Accumulation as Affected by Nitrate Supply Level in Alfalfa (Medicago sativa L.) (질산태 질소의 공급수준에 따른 알팔파의 건물, 질소의 분포 및 저장영양소의 축적)

  • To investigate the effects of the exogenous N $O_3$$^{[-10]}$ supply level on the nitrate assimilation and growth during the vegetative growth stage, and on the accumulation of organic reserves during the successive regrowth period, dry matter (DM), the amount of nitrogenous compounds, total N and starch in alfalfa plants grown hydroponically with 0.2, 1.0 and 3.0 mM KN $O_3$ was estimated, respectively, during vegetative growth period and two cycle of regrowth. When compared with DMs and N contents in various N compounds in the organs grown with 1.0 mM N $O_3$$^{[-10]}$ , N starvation symptoms were found in 0.2 mM and a depressive effect was observed in 3.0 mM after 10 weeks of vegetative growth. Total starch content in root system gown with 0.2, 1.0 and 3.0 mM N $O_3$$^{[-10]}$ during the first regrowth was 50.96, 15.47 and 6.37 mg plant$^{-1}$, respectively. Starch was contained mainly in taproots. The starch content was not significantly changed by 24 days of the second regrowth with 1.0 mM N $H_4$N0$_3$. Total nitrogen content in root system grown with 0.2, 1.0 and 3.0 mM N $O_3$$^{[-10]}$ during the first regrowth was 6.66, 8.43 and 11.09 mg plant$^{-1}$ , respectively. Nitrogen was contained mainly in lateral roots; 80% (in 0.2 mM), 74% (1.0 mM) and 76% (3.0 mM) of total nitrogen in root system. Total N content in root system at the end of the second regrowth also closely affected by the N $O_3$$^{[-10]}$ supply level during the first regrowth. These results suggest that the level of N $O_3$$^{[-10]}$ may strongly influence the accumulation of organic reserves in root system, and that the initial level of organic reserves for the successive regrowth was one of the determinants for shoot regrowth.

Effect of Nitrogen Application Level and Regrowth Period on Composition of Fatty Acid in Tall fescue and Bermudagrass (질소시비 수준 및 재생기간이 톨페스큐와 버뮤다그라스의 지방산조성에 미치는 영향)

  • Two experiments were conducted to evaluate the effects of nitrogen (N) application level and regrowth period on the fatty acid (FA) concentration and forage production of tall fescue (Festuca arundinacea Schr.) and bermudagrass (Cynodon dactylon (L.) Pers.). N was applied at 0, 50 and 100 kg/ha, and swards were cut after regrowth periods (15d, 30d and 45d). Concentrations of Individual FA were determined by gas chromatography. FA composition of tall fescue contains a high proportion ($87{\sim}88%$) of total FA contents as palmitic (C16:0), linoleic (C18:2) and linolenic acid (C18:3). The FA composition was not affected by N application, but a longer regrowth period significantly (p<0.05) decreased the composition of linolenic acid (C18:3) and increased those of linoleic acid (C18:2). The FA composition of bermudagrass was not affected by N application, but a longer regrowth period significantly (p<0.001) decreased the composition of linoleic (C18:2) and linolenic acid (C18:3). Linoleic and linolenic acfd of tall fescue were higher than those of bermudagrass. Dry matter (DM) yield of bermudagrass was significantly (P<0.05) affected by N application level and regrowth period. DM yield was higher at high N application and was higher at longer regrowth period. The crude protein concentration was higher at high N application and was low at longer regrowth period. Application of nitrogen fertilizer Increases dry matter (DM) yield and crude protein (CP) content of bermudagrass. These studies demonstrate opportunities to affect the FA concentration and composition of FA in forage through management strategies, which could affect milk FA composition.

Differences in Regrowth and Terminal Flower Bud Formation of 'Fuji' and 'Jonagold' Apple Trees in Response to Summer Heading Back Pruning of Current Season's Shoots (신초의 하계 절단전정에 의한 사과품종 '후지' 및 '조나골드'의 2차생장과 정화아 형성의 차이)

  • Current season's shoots on 2-year-old branches of 'Fuji' and 'Jonagold' apple trees were heading back pruned to 5 leaves from early May to mid July at about 12-day intervals. The summer heading back pruning induced regrowth on the pruned shoots with different responses between the two cultivars. Generally, greater regrowth occurred on the pruned shoots of 'Fuji' trees than on those of 'Jonagold', irrespective of the time of the heading cut. The shoots of 'Fuji' trees pruned in late May or in June exhibited greater regrowth compared with those pruned in early May or in July, whereas the summer heading back pruning in June resulted in the greatest regrowth for 'Jonagold'. The heading cut induced terminal flower bud formation on the pruned shoots, the percentage of which was higher in 'Fuji' than in 'Jonagold'. The highest percentages of terminal flower bud formation for 'Fuji' and 'Jonagold' were obtained with the heading cut in late May and in mid June, respectively. Percent flowering of the buds was similar in both cultivars, but percent fruit set was slightly higher in 'Fuji' than in 'Jonagold'. The time of the heading cut did not affect percent fruit set in both cultivars. Our results demonstrate that summer heading back pruning of current season's shoots induces regrowth and terminal flower bud formation therefrom when done at appropriate time, but the specific responses to the heading cut are cultivardependent.

Hair Growth Promotion Effect of a Bio-Active Shampoo, Bonogen in C57BL/6 Mice (C57BL/6 마우스에서 기능성 샴푸 Bonogen의 양모 촉진 효과)

  • Bonogen shampoo is composed of several plant extracts which are known to be used in oriental medicine. This study was carried out to investigate the effects of Bonogen shampoo on hair growth in an alopecia model of C57BL/6 mice. There were eight male and female experimental groups including distilled water(DW: negative control), a commercial shampoo[M], 3% minoxidil (MXD) and Bonogen shampoo(BNG). Dorsal skin hair of six-week-old mice was trimmed with an electric clipper carefully not to damage the skin. The next day, mice without skin scratch were selected, randomized and separated in 10 mice per group. The test compounds were topically treated with 0.15 ml per mouse or dorsal skin for 21 days daily and then washed thoroughly with DW. The hair regrowth was determined photographically at 0, 4, 7, 10, 15, 18, and 21 days and histologically at day 21. No clinical signs were observed in all mice. Although body weight was slightly increased in 3% MXD group than other groups, it was not significant. Hair regrowth began to be promoted after 14 days and appeared a distinct regrowth pattern in all animals by topical treatment of test compounds at 18 days. In particular, the topical treatment of bonogen shampoo or 3% MXD for 21 days to dorsal skin accelerated hair regrowth faster than DW or M shampoo. At 21 days, the hair regrowth promotion speed was in order of 3% MXD>BNG>M>DW. The bonogen shampoo or 3% MXD also promoted hair follicle elongation compared to the negative control. These results suggest that bonogen shampoo has hair growth promoting activities and may be useful for treatment of bald or alopecia.

Modeling Methodology for Cold Tolerance Assessment of Pittosporum tobira (돈나무의 내한성 평가 모델링)

  • This study was carried out to develop a simple, rapid and reliable assessment model to predict cold tolerance in Pittosporum tobira, a broad-leaved evergreen commonly used in the southern region of South Korea, which can minimize the possible experimental errors appeared in a electrolyte leakage test for cold tolerance assessment. The modeling procedure comprised of regrowth test and a electrolyte leakage test on the plants exposed to low temperature treatments. The lethal temperatures estimated from the methodological combinations of a electrolyte leakage test including tissue sampling, temperature treatment for potential electrical conductivity, and statistical analysis were compared to the results of the regrowth test. The highest temperature showing the survival rate lower than 50% obtained from the regrowth test was $-10^{\circ}C$ and the lethal was $-10^{\circ}C{\sim}-5^{\circ}C$. Based on the results of the regrowth test, several methodological combinations of electrolyte leakage tests were evaluated and the electrolyte leakage lethal temperatures estimated using leaf sample tissue and freeze-killing method were closest to the regrowth lethal temperature. Evaluating statistical analysis models, linear interpolation had a higher tendency to overestimate the cold tolerance than non-linear regression. Consequently, the optimal model for cold tolerance assessment of P. tobira is composed of evaluating electrolyte leakage from leaf sample tissue applying freeze-killing method for potential electrical conductivity and predicting lethal temperature through non-linear regression analysis.

  • This experiment was canied out to determine the effects of 3cn1, 6cm, and lOcm of cutting height at harvest on the grass growth, dry matter(DM) yield, and weeds development in reed canarygrass(Phu1uris umndinacea L.) pasttire. The cultivars of reed canarygrass used in this study were Palaton. Veuture and Frontier(contro1). and the grass was harvested four times at soiling stage in 1992. The plant height at first harvest was 84, 96 and 94cm in Palaton, Venture and Frontier, respectively. The average regrowth height was same as 59cm in three cultivars. However, the regrowth height by cutting height was 61 -65cm in 6 and IOcm, and 51 -53cm in 3cm of low cutting height. Annual DM yield was not different in three cultivars of Palaton(l2.58lkg), Venture(l2,752kg), and Frontier (12,243kgIha). The yield at first harvmt wa5 significantly high in 3cm of height, however, the forage yields at second, third and fourth harvest were greatly high in 6cm of stubble height(P<0.05). Total yields by 3, 6 and lOcm of cutting height were 12.306. 14,094 and 11,342kg in Palaton, 12,794, 14,155 and 11,307kg in Venture, and 12,258, 12,940, and 11.535kglhg in Frontier, respectively. The highest yield was achieved in 6cm of stubble heigth(P<0.05). Daily DM production during grass regrowth was not affected by cultivars, and the best regrowth was observed by 6cm of cutting height. Development of weeds was high in 3cm of low stubble height, regradless of cultivars. The contents of crude protein, neutral detergent fiber, acid detergent fiber, and hemicellulose were not affected by cultivar and cutting height. In conclusion, it is suggested that the 6cm of cutting height is the most effective for grass regrowth, forage production, and weed control in reed canarygrass pasture. regradless of cultivm of Palaton, Venture and Frontier.

Urease and nitrification inhibitors with pig slurry effects on ammonia and nitrous oxide emissions, nitrate leaching, and nitrogen use efficiency in perennial ryegrass sward

  • Objective: The present study was conducted to assess the effect of urease inhibitor (hydroquinone [HQ]) and nitrification inhibitor (dicyandiamide [DCD]) on nitrogen (N) use efficiency of pig slurry for perennial ryegrass regrowth yield and its environmental impacts. Methods: A micro-plot experiment was conducted using pig slurry-urea 15N treated with HQ and/or DCD and applied at a rate of 200 kg N/ha. The flows of N derived from the pig slurry urea to herbage regrowth and soils as well as soil N mineralization were estimated by tracing pig slurry-urea 15N, and the N losses via ammonia (NH3), nitrous oxide (N2O) emission, and nitrate (NO3-) leaching were quantified for a 56 d regrowth of perennial ryegrass (Lolium perenne) sward. Results: Herbage dry matter at the final regrowth at 56 d was significantly higher in the HQ and/or DCD applied plots, with a 24.5% to 42.2% increase in 15N recovery by herbage compared with the control. Significant increases in soil 15N recovery were also observed in the plots applied with the inhibitors, accompanied by the increased N content converted to soil inorganic N (NH4++NO3-) (17.3% to 28.8% higher than that of the control). The estimated loss, which was not accounted for in the herbage-soil system, was lower in the plots applied with the inhibitors (25.6% on average) than that of control (38.0%). Positive effects of urease and/or nitrification inhibitors on reducing N losses to the environment were observed at the final regrowth (56 d), at which cumulative NH3 emission was reduced by 26.8% (on average 3 inhibitor treatments), N2O emission by 50.2% and NO3- leaching by 10.6% compared to those of the control. Conclusion: The proper application of urease and nitrification inhibitors would be an efficient strategy to improve the N use efficiency of pig slurry while mitigating hazardous environmental impacts.

  • The mechanisms of the meltback etching and regrowth processes are studied experimentally. The method to reduce the anisotropy and elliminate the gallium islands in the processes are developed. It is possible to fabricate the GaAs optical devices by the use of the process.
