Reduction of hexavalent chromium by pseudomonas aeruginosa HP014

  • Microbial reduction of hexavalent (VI) to trivalent (III) chromium decreases its toxicity by two orders of magnitude. In order to investigate the nature of Cr-reduction, Cr-resistant Pseudomonas aeruginosa HP014 was isolated and tested for its reduction capability. At the concentration of 0.5 mM Cr(VI), cell growth was not inhibited by the presence of Cr(VI) in a liquid medium, and Cr(VI) reduction was accompanied by ell growth. When cell-free extract was tested, the reduction of Cr(VI) showed a saturation kinetics with the maximum specific activity of 0.33 .mu.mol min$\^$-1/ mg$\^$-1/ cell protein, and an apparent K. of 1.73 mM Cr(VI). The activity required either NADH or NADPH as an electron donor. However, NADPH gave 50% as mush activity as sequently the supernatant and pelleted membrane fractions were tested for Cr(VI) reduction activity. The supernatant of the centrifugation showed almost the same Cr(VI) reduction activity as compared with that of the cell-free extract, indicating that the Cr(VI)-reducing activity of P. aeruginosa HP-14 is due to soluble enzyme. Moreover, the activity appeared to be the highest among the known activities, suggesting that the strain might be useful for remediation of Cr(VI)-contaminated sites.

Antimicrobial Activity of Dyed Fabrics with Gromwell Colorants (자초색소를 이용한 염색직물의 항균성)

  • Antimicrobial activity of silk and cotton fabrics dyed with gromwell colorants was examined by shake flask method. The effects of bacterial reduction rate on dye uptake and mordanting were explored. The silk(K/S value 8) and cotton fabric (K/S value 4) dyed with gromwell colorants showed high bacterial reduction rate of 99 % and 88 %, respectively. Post-mordanted silk fabric increased at K/S value, also showed higher bacterial reduction rate than unmordanted. But the effects of antimicrobial activity on mordants showed respectively different results. Pre-mordanted cotton fabric except Sn mordant increased at K/S value, but it was not significant. Pre-mordanted cotton fabric showed significantly higher bacterial reduction rate than unmordanted. Therefore, it was considered that dye uptake of cotton fabric dyed with gromwell colorants and mordants were involved in the bacterial reduction rate. It was concluded that gromwell colorants had antimicrobial activity.

  • The activity of taurine transporter is affected by various extracellular stimuli such as ion, hormone and stress. To assess effects of steroid hormones antral cyclosporin A (CsA) on the taurine transporter activity, murine monocytic RAW264.7 cell line was stimulated with dexamethasone (DM), triamcinolone (TA), cortisone (CS), hydrocortisone (HCS), prednisone (PSN), prednisolone (PSL) and methylprednisolone (MPSL) in the presence of 12-0-tetradecanoylphorbol-13-acetate(TPA). Treatment of TPA on the cell line led to significant reduction of taurine transporter activity. However, in case of stimulation of the cells with steroid hormones in the presence of TPA, all of them recovered TPA-induced reduction of the taurine transporter activity. Treatment of the cells with CsA led to significant reduction of the taurine transporter activity. Ionomycin (IM) recovered the reduced taurine transporter activity by CsA, but failed in the presence of EDTA, a calcium chelating agent. These results showed that glucocorticoid hormone recovered TPA-induced reduction of taurine transporter activity and that IM recovered CsA-induced reduction of the transporter activity by increasing intracellular free $Ca^{++}$ concentration.n.

A Continuous Spectrophotometric Assay for NADPH-cytochrome P450 Reductase Activity Using 1,1-Diphenyl-2-Picrylhydrazyl

  • NADPH-cytochrome P450 reductase (CPR) transfers electrons from NADPH to cytochrome P450, and catalyzes the one-electron reduction of many drugs and foreign compounds. Various forms of spectrophotometric titration have been performed to investigate the electron-accepting properties of CPR, particularly, to examine its ability to reduce cytochrome c and ferricyanide. In this study, the reduction of 1,1-diphenyl-2-picrylhydrazyl (DPPH) by CPR was assessed as a means of monitoring CPR activity. The principle advantage of DPPH is that its reduction can be assayed directly in the reaction medium by a continuous spectrophotometry. Thus, electrons released from NADPH by CPR were transferred to DPPH, and DPPH reduction was then followed spectrophotometrically by measuring $A_{520}$ reduction. Optimal assay concentrations of DPPH, CPR, potassium phosphate buffer, and NADPH were first established. DPPH reduction activity was found to depend upon the strength of the buffer used, which was optimal at 100 mM potassium phosphate and pH 7.6. The extinction coefficient of DPPH was $4.09\;mM^{-1}\;cm^{-1}$. DPPH reduction followed classical Michaelis-Menten kinetics ($K_m\;=\;28\;{\mu}M$, $K_{cat}\;=\;1690\;min^{-1}$). This method uses readily available materials, and has the additional advantages of being rapid and inexpensive.

A study on the Structural Relationship among Innovation Activity, Innovation Performance, and Business Performance of Export Firms (수출기업의 혁신활동, 혁신성과, 경영성과 간의 구조적 관계에 관한 연구)

  • The purpose of this study is to examine the relationship among innovation activities, innovation performance, and management performance of export firms. I first undertake a review of previous studies related to innovation activity, innovation performance, and export firms' innovation. Based on this, I set up a research model and hypotheses. First, The innovation activity is classified into product innovation activity, process innovation activity, and administrative innovation activity. Innovation performance is divided into product quality improvement and cost reduction. And the business performance is regarded as financial performance of export's firms. This study examined whether innovation activity affect innovation performance such as product quality improvement and cost reduction. In addition, I examined whether these innovation performance affects business performance (financial performance). To achieve this, a total of 368 questionnaires are used in this study and PLS (Partial Least Square) was used to analyze structural equation. As a result, this study shows that product innovation activity have a positive effect on product quality improvement, and process innovation activity also have a positive effect on product quality improvement. However, it was found that neither product innovation activity nor process innovation activity had an effect on cost reduction. And it was found that administrative innovation did not affect product quality improvement, but had a positive effect on cost reduction. Also, it was found that quality improvement, which is an innovation performance, did not affect the financial performance of export's firms, and cost reduction, which is an innovation performance, had a positive effect on the financial performance of the export's firms.

A Continuous Spectrophotometric Assay for NADPH-cytochrome P450 Reductase Activity Using 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium Bromide

Various spectrophotometric assays have been performed to examine electron-accepting properties of CPR and its ability to reduce cytochrome $b_5$, cytochrome c, and ferricyanide. In this report, reduction of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) by CPR has been assessed as a method for monitoring CPR activity. The principle advantage of this substance is that the reduction of MTT can be assayed directly in the reaction medium by a continuous spectrophotometric method. The electrons released from NADPH by CPR were transferred to MTT. MTT reduction activity was then assessed spectrophotometrically by measuring the increase of $A_{610}$. MTT reduction followed classical Michaelis-Menten kinetics ($K_m\;=\;20\;{\mu}M$, $k_{cat}\;=\;1,910\;min^{-1}$). This method offers the advantages of a commercially available substrate and short analysis time by a simple measurement of enzymatic activity of CPR.

Effect of Electrochemical Oxidation-Reduction Cycles on Surface Structures and Electrocatalytic Oxygen Reduction Activity of Au Electrodes

  • Oxidation-reduction cycling (ORC) procedures are widely used for cleaning nanoparticle surfaces when investigating their electrocatalytic activities. In this work, the effect of ORC on the surface structures and electrocatalytic oxygen reduction activity of Au electrodes is analyzed. Different structural changes and variations in electrocatalysis are observed depending on the initial structure of the Au electrodes, such as flat bulk, nanoporous, nanoplate, or dendritic Au. In particular, dendritic Au structures lost their sharp-edge morphology during the ORC process, resulting in a significant decrease in its electrocatalytic oxygen reduction activity. The results shown in this paper provide an insight into the pretreatment of nanoparticle-based electrodes during investigation of their electrocatalytic activities.

  • Silver nano-particles have been synthesized by liquid phase reduction method and alcohol reduction process. Silver nano-particles of the size 30 ~ 40 nm were formed successfully by alcohol reduction process. The formation, structure, morphology and size of silver nano-particles have been studied using FE-SEM, TEM, XRD, UV-visible spectroscopy. In particular high dispersion stability of the synthesized silver nano-particles could be obtained by PVP binding. Antibacterial activity of Ag/PET master batch sample made from its nano-silver particles showed excellent antibacterial activity against S. aureus and E. coli.

  • The antibacterial activities of several types of chitosan were measured against Staphylococcus aureus and evaluated for their application to antibacterial textile finishing. The % reduction of bacteria of the chitosans prepared in our laboratory were between 72 and 87%. The two water-soluble chitosans with molecular weights 1,000 and 3,000 did not show antibacterial activities. The deacetylation of chitosan was appeared to increase antibacterial activity. The % reduction in bacterial density of the 86%-deacetylated chitosan solution was 56% where that of the 76%-deacetylated chitosan solution was only 17% at 0.1% chitosan concentration. Molecular weights of the chitosans seemed not to affect antibacterial activities of chitosans. The antibacterial activity of the acid-soluble, 86%-deacetylated chitosan with 4 cps showed 98% of the % reduction at the level of 0.2% chitosan. The % reduction of bacteria of this chitosan was higher at the higher concentration of acetic acid in the chitosan-bacterial mixture. The antibacterial activity was increased with the pH change over the range of 4.0 to 6.5. The 100% of the % reduction of bacteria was achieved within 4 hour incubation of the chitosan-bacterial mixture. According to the data obtained from the above experiments, the four chitosans among the six prepared in our laboratory were proved to be valuable for antibacterial textile finishing.

Low Temperature Selective Catalytic Reduction of NOx over V2O5/TiO2 Catalyst Doped with Mn (Mn이 첨가된 V2O5/TiO2 촉매상에서 질소산화물의 저온 SCR 특성)

  • $V_{2}O_{5}/TiO_{2}$ catalysts promoted with Mn were prepared and tested for selective catalytic reduction of NOx in $NH_3.$ The effects of promoter content, degree of catalyst loading were investigated for NOx activity while changing temperatures, mole ratio, space velocity and $O_2$ concentration. Among the various $V-{2}O_{5}$ catalysts having different metal loadings, $V-{2}O_{5}$(1 wt.%) catalyst showed the highest activity(98%) under wide temperature range of $200-250^{\circ}C.$ When the $V-{2}O_{5}$ catalyst was further modified with 5 wt.% Mn as a promoter, the highest activity(90-47%) was obtained over the low temperature windows of $100-200^{\circ}C.$ From Mn-$V_{2}O_{5}/TiO_{2}$, it was found that by addition of 5 wt.% Mn on $V_{2}O_{5}/TiO_{2}$ catalyst, reduction activity of catalyst was improved, which resulted in the increase of catalytic activity and NOx reduction. According to the results, NOx removal decreased for 10%, but the reaction temperature down to $100^{\circ}C.$
