홍삼에 함유된 갈변물질 및 산성다당체에 대한 연구현황 (Current Studies on Browning Reaction Products and Acidic Polysaccharide in Korean Red Ginseng)

  • 우리나라의 건강기능식품법에서는 인삼 또는 홍삼의 기능성이 자양강장, 면역증진 및 원기회복으로 한정되어 있다. 지금까지 홍삼에 함유된 갈변물질과 산성다당체에 대해서 많은 연구가 수행되었는데, 갈변물질은 항산화 활성과 면역증진기능, 그리고 산성다당체는 소화기관암의 증상개선, 면역기능 증진, 혈압개선, 혈액응고방지, 조혈기능, 알콜해독(간기능 회복, 고지혈 개선 등) 등의 효능이 우수하다고 보고되었다. 이러한 관점에서 현재 인삼과 홍삼의 3가지 기능성외에 항산화활성이라든지 그 외 더 많은 기능성을 추가할 필요성이 있으며, 또 홍삼에 함유되어 있는 활성성분을 적극적으로 이용하여 기준규격형제품은 물론이고 질병위험감소에 대해서 기능성을 표시할 수 있는 개별인정형 제품이나 나아가서는 일반 또는 전문의약품으로 개발하여 홍삼의 이용 가능성을 극대화시키고 국제적으로 경쟁력이 있는 한국인삼으로 그 위상을 제고시켜야 한다고 생각한다.

STUDIES ON BIOLOGICALLY ACTIVE SUBSTANCES IN NON-SAPONIN FRACTION OF KOREAN RED GINSENG

  • Acidic polysaccharide from Korean red ginseng was found to inhibit pancreatic lipase activity and cause reduction of plasma triglyceride level after oral administration of corn oil emulsion to rats. Thus acidic polysaccharide may reduce plasma triglyceride through its inhibitory action on pancreatic lipase and successive inhibition of intestinal absorption of fat due to reduction of lipolysis. In the course of this experiment, we found an unknown ninhydrin positive substance in Korean red ginseng. The unknown substance was identified to be arginyl-fructosyl glucose(Arg - Fru - Glc). Coment of this new compound was $5.37\%$ in Korean red ginseng powder. Sucrase and maltase activities in mucous layer of rat jejunum were found to be inhibited by Arg-Fru-Glc. Physiological significance of the new compound was discussed based on these experimental results.

홍삼추출잔사로부터 항암면역조절 활성을 보여주는 홍삼산성다당체(RGAP)의 분리 및 이화학적 특성 (Isolation of Immunomodulatory Antitumor Active Polysaccharide (RGAP) from Red Ginseng By-Product and Its Physico-chemical Properties)

  • 홍삼으로부터 홍삼알콜 추출물을 제조하고 부산물로 생성된 홍삼추출잔사로부터 한외 여과를 이용하여 홍삼산성다당체(RGAP, red ginseng acidic polysaccharide)를 대량으로 분리할 수 있는 방법을 개발하였다. 본 방법으로 홍삼추출잔사로부터 분리된 RGAP의 수율은 20.9%이었고, RGAP로부터 지표성분인 활성성분(GFP)의 분리수율은 39.4%이었다. 분리된 RGAP는 macrophage에 의한 NO(nitric oxide)를 생성하여 암세포를 사멸시키는 것으로 나타났다. RGAP를 BALB/c 마우스에 복강투여한 후 분리된 복강 macrophage를 P815 암세포(mastocytoma)와 함께 배양하였을 때 배지내의 NO 농도는 대조군에서 4.3 $\mu\textrm{m}$이었으나, RGAP 100, 300 mg/kg 투여군에서는 각각 9.8 및 25.3$\mu\textrm{m}$로 증가하는 경향을 나타내었다. 아울러 암세포인 P8l5세포의 사멸율도 정상대조군일 경우 7.3%이었으나 RGAP 100, 300 mg/kg 투여군에서는 각각 22.7 및 31.8%로 증가하는 경향을 나타내었다. RGAP을 2M의 TFA로 10$0^{\circ}C$에서 1시간 가수분해한 후 당 표준품과 TLC로 비교한 결과 당 표준품과 Rf치 및 발색 색깔이 일치하는 4개의 spot, 즉 rhamnose(Rf 0.78, 노란색 ), glucose(Rf 0.61, 녹색), galactose(Rf 0.56, 녹색), glucuronic acid(Rf 0.36, 황갈색)를 관찰할 수 있었다. 가수분해된 RGAP 분해산물을 Gl(glucose)부터 G7(maltoheptaose)까지의 당류와 함께 TLC를 행한 결과 진한 색상을 나타내는 spot은 Rf 0.56~0.61로 G2(maltose, Rf 0.46) 이상의 단당류(monosaccharide)인 것으로 나타났다. RGAP의 일부 이화학적 특성을 조사한 결과 pH는 4.74, 건조감량은 4.72%, 조단백질 함량은 3.30%, 회분은 4.74%이었고 중금속인 Pb는 2.30 ppm이었다.

Antithrombin Active Polysaccharide Isolated from the Alkaline Extract of Red Ginseng

  • 홍삼으로부터 염기성 수웅액 추출, 음이온교환 크로마토그래피 및 겔여과 크로마토그래피를 순차적으로 수행하여 항트롬빈 활성을 보이는 다당체를 분리하였다. 이 다당체는 셀룰로스 아세테이트막을 사용한 전기영동에서 하나의 띠로 나타나 비교적 순수하게 정제되었음을 알 수 있었고, 겔여과 크로마토그래피에서 결정된 평균 분자량은 약 177kDa이었다. $40.2\%$의 유론산, $9.2\%$의 황산기 및 $1.5\%$의 단백질을 포함하는 산성다당체였고, 구성 중성당으로 rhamnose, mannose, galactose, arabinose, glucose, fucose, xylose를 1.00 : 0.88 :0.86 : 0.78 : 0.70 : 0.33 : 0.22의 비율로 포함하고 있었다. 이 다당체는 트롬빈에 의한 피브리노겐의 응고를 농도에 비례하여 저해하였고, 혈장을 사용한 실험에서 내인성 경로를 통해 혈액응고를 저해하는 것으로 나타났다.

홍삼 추출물의 임파구 증식 및 활성 촉진효과 (Stimulatory Effect of Korean Red-Ginseng Extract on the Proliferation and . Cellular Activity of Lymphocytes)

  • The effect of Korean red-ginseng extract on the proliferation and cellular activity of mouse B and T lymphocytes was examined in vitro. Both water and ethanol extract from red-ginseng increased the growth of normal B and T lymphocytes 1.5∼2.5-folds. Saponin and polysaccharide fractions from ginseng extract also stimulated the proliferation of normal lymphocytes much higher than several well-known immunostimulators. B and T lymphoma cell lines responded to the ginseng extract and fractions by growth, too, while non-lymphoid cell lines did not. Immunoglobulin production of unprimed B-lymphocytes was little affected by the ginseng extract and fractions, though the ethanol extract slightly enhanced Ini, production of B-lymphocytes. When cytolytic activity of T lymphocytes against tumor tells was induced in vitro, both of the saponin and polysaccharide fractions and the ginseng ethanol extract increased the cellular activity of cytotoxic T lymphocytes 4-5-folds, while the ginseng water extract did not. Especially, the saponin fraction exhibited 10-times higher stimulatory effect on the cytolytlc activity of cytotoxic T cells than the ethanol extract and the pclysaccharide fraction did. These results suggest that Korean red-ginseng contain potent immunomodulating components to stimulate the proliferation of B and T lymphocytes and the cellular activity of cytotoxic T lymphocytes.

Toxicity Study of Red Ginseng Acidic Polysaccharide (RGAP) : Single and 2-week Repeated Oral Dose Toxicity Study in Rats

  • The present study was conducted to investigate the single and 2-week repeated dose toxicity of red ginseng acidic polysaccharide (RGAP) in Sprague-Dawley rats. The test article was administered orally to rats at dose levels of 0, and 2000 mg/kg/day for single dose toxicity study and at dose levels of 0, 250, 500, and 1000 mg/kg/day for repeated dose toxicity study. In both studies, there were no treatment-related effects on mortality, clinical signs, food and water consumption, ophthalmoscopy, urinalysis, hematology, serum biochemistry, necropsy findings and organ weights of all animals treated RGAP. Based on these results, it was concluded that the 2-week repeated oral dose of RGAP may have no toxic effect in rats at a dose level of 1000 mg/kg/day. In the condition of this study, the no-observed-adverse-effect level (NOAEL) was considered to be 1000 mg/kg/day for both sexes.

  • We have been isolating various physiologically active substances from non-saponin fraction of Korean Red Ginseng These are adenosine, gyro-glutamic acid, dencichine and acidic polysaccharide. Adenosine and gyro-glutamic acid are loom to inhibit epinephrine-induced lipolysis in fat cells and stimulate the insulin-mediated lipogenesis. In addition to these actions, adenosine was found to inhibit both norepinephrine- and histamine-induced aorta constriction, and pyre·glutamic acid inhibits angiotensin-converting enzyme. Dencichine stimulated histamine-induced aorta constriction. Finally, acidic polysaccharide was found to inhibit both lipolytic and anorexigenic actions of Toxohormone-L. Based on these experimental results, I presented a brief review on these compounds isolated from non- saponin fraction of Korea Red Ginseng. Effects of extrusion cooking on physicochemical properties of white and red ginseng (powder)

  • A systematic comparison of the physicochemical properties of white ginseng (WG), extruded white ginseng (EWG), red ginseng (RG), and extruded red ginseng (ERG) was performed. The aim of the present study was to identify the effects of the physicochemical properties of ginseng by extrusion cooking. The highest value of the water absorption index (WAI) was 3.64 g/g obtained from EWG, and the highest value of the water solubility index (WSI) was 45.27% obtained from ERG. The ERG had a better dispersibility compared with other samples. Extrusion cooking led to a significant increase in acidic polysaccharide and total sugar content but resulted in a decrease in crude fat and reducing sugar contents. Enzyme treatment led to a sharp increase in acidic polysaccharide content, especially the cellulose enzyme. Extrusion cooking led to a significant increase in 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and reducing power, and the increases in WG and RG were 13.56% (0.038) and 3.56% (0.026), respectively. The data of this study provide valuable information about the effects of extrusion on quality changes of EWG and ERG.

Inhibitory Activities of Red Ginseng Acidic Polysaccharide in Platelet Aggregation

  • Red ginseng acidic polysaccharide (RGAP), isolated from Korean red ginseng (Panax ginseng C.A. Meyer), has been shown to have a variety of biological functions such as immunostimulating and anti-tumor activities. In the present study, we investigated whether RGAP inhibited ligand-induced platelet aggregation. The washed platelet-rich plasma was prepared from male SD rats with successive centrifugation. The platelets $(10^8/ml)$ were preincubated with 1 mM of $CaCl_2$ for 2 min either in the presence or in the absence of RGAP $(10{\sim}50\;{\mu}g/ml)$ and were stimulated with collagen (2.5 ${\mu}g/ml$) and thrombin (0.1 U/ml). RGAP dose-dependently inhibited thrombin-induced platelet aggregation with $IC_{50}$ value of $26.2{\pm}2.0$ ${\mu}g/ml$. In collagen-induced platelet aggregation, RGAP inhibited the reaction with an $IC_{50}$ value of $31.5{\pm}3.0\;{\mu}g/ml$. RGAP potently suppressed the intracellular calcium ion, which was stimulated by thrombin (0.1 U/ ml). Among mitogen-activated protein kinase (MAPK) subtypes, the extracellular signal-regulated kinase (ERK) 1/2 and p38 MAPK were analyzed in the present study. RGAP inhibited the phosphorylation of ERK2 and p38 MAPK, which was activated by collagen (2.5 ${\mu}g/ml$). Finally, these results suggested that besides saponin fraction, RGAP take an important role in the preventive effect of Korean red ginseng against cardiovascular disease such as thrombosis and atherosclerosis.
