Optical Detection of Red Blood Cell Aggregation under vibration (진동장에서의 적혈구 응집성을 측정하는 광학적 방법)

  • Aggregability of red blood cells (RBCs) was determined by a laser backscattering light analysis in a microfluidic channel. Available techniques for RBC aggregation often adopt a rotational Couette-flow using bob-and-cup system for disaggregating RBCs, which causes the system to be complex and expensive. A disposable microfluidic channel and vibration generating mechanism were used in the proposed new detection system for RBC aggregation. Prior to measurement, RBC aggregates in a blood sample were completely disaggregated by applying vibration-induced shear. With the present apparatus, the aggregation indexes of RBCs can be easily measured with small quantities of blood sample. The measurements with the present aggregometer were compared with those of LORCA and showed a strong correlation between them. The aggregability of the defibrinogenated blood RBCs is markedly lower than that of the normal RBCs. The noble feature of this design is the vibration-induced disaggregation mechanism, which enables to incorporate disposable element that holds the blood sample.

Increased of the Red Blood Cell in Peripheral Plasma of Transgenic Pigs Harboring hEPO Gene

  • The present study were performed to analysis the hematocrit and the red blood cells content into the blood plasma of the transgenic pigs harboring recombinent human erythropoietin gene (rhEPO). Mouse whey acidic protein (mWAP) linked to rhEPO gene was microinjected into pronuclei of porcine one-cell zygotes. After delivered of offspring, PCR analyses identified one mWAP-rhEPO transgenic founder offspring(F$_{0}$). The first generation of transgenic pig (F$_{0}$) harboring mWAP-hEPO appeared to be a male, and the second generation (F$_1$) pigs were made by natural mating of F$_{0}$ with domestic swine, and male and female transgenic pigs (F$_1$) were identified by PCR. The blood samples from transgenic and normal pigs were collected for 50 days during lactation and were counted the red blood cell (RBC) numbers and Hematocrit (HCT) content into the blood. The transgenic pigs expressing rhEPO in their blood gave rise to higher RBC numbers and HCT contents than control animals. rhEPO was secreted both in the blood and milk of genetically engineered pigs harboring rhEPO gene. Therefore, this study provides a model regarding the production of transgenic pig carrying hEPO transgene for biomedical research.earch.

Live Blood Analysis on Interior Vascular Laser Irradiation Therapy and Exterior Vascular Laser Irradiation Therapy (혈관레이저 치료와 관련한 생혈액 검사의 진단 의미 고찰)

  • Objectives : The purpose of this study was to investigate the usability of live blood analysis on interior and exterior vascular laser irradiation therapy. Methods : We had analyzed the changing forms of the live blood sample with microscope before and after vascular laser irradiation therapy of blood. The live blood analysis was operated on Rouleau of red cell, erythrocyte aggregation, thrombocyte aggregation, uric acid crystals, red crystals, protoplasts. First, we analyzed all patients on each item, then did same thing classified two groups, Interior and exterior. Results : Rouleau of red cell, erythrocyte aggregation, thrombocyte aggregation, uric acid crystals, red crystals, protoplasts were decreased significantly, after interior and exterior aggregation, uric acid crystals. Interior vascular laser irradiation therapy was more effective than interior on Rouleau of red cell, erythrocyte aggregation, thrombocyte aggregation, uric acid crystals. Interior vascular laser irradiation therapy was more effective than exterior on red crystals, protoplasts. Conclusions : This study suggests that live blood analysis has the usability on vascular laser irradiation therapy. Then according to interior and exterior vascular laser irradiation therapy, the result has some different on each item. So it is better that choose the method, interior or exterior, for more effective therapy.

Changes in the Red Cell Volume and the Plasma Chloride Level under the High $CO_2$ Concentration in vitro (고농도(高濃度)의 $CO_2$가 적혈구용적(赤血球容積) 및 혈장(血漿) Choloride 치(値)에 미치는 영향(影響))

  • The changes in the red cell volume and the plasma chloride level were measured when the blood $CO_2$ content was altered by equilibration with the atmospheric air or pure $CO_2$ for 20 minutes. The red cell volume was expressed in terms of hematocrit and mean corpuscular volume (M.C.V.). The results obtained were as follows. 1) On equilibration with the atmospheric air, the MCV and the plasma chloride level were $91.6{\pm}1.26\;c.{\mu}$ and $110.7{\pm}6.28mEq/L.$ respectively. 2) On equilibration with pure $CO_{2}$, the MCV and the plasma chloride level were $109.6{\pm}2.0\;c.{\mu}$ and $90.7{\pm}5.17\;mEq/L.$ respectively. 3) When the blood was subjected to equilibration with the atmospheric for air 20 minutes after equilibration with pure $CO_{2}$ for the same period of time the MCV and the plasma chloride level were $89.9{\pm}6.34\;c.{\mu}$ and $100.3{\pm}5.50\;mEq/L.$ respectively. From the above results it can be concluded that an increase of the blood $CO_2$ content in the experimental condition causes definitely a decrease of the plasma chloride level and a concomitant increase of the red cell volume, and that a decrease of the blood content $CO_2$ in the experimental condition causes definitely an increase of the plasma chloride level and a concomitant decrease of the red cell volme. Apparantly there exists a parallel relationship between the extent of the decrease of the plasma chloride level and that of the increase of the red cell volume when the blood $CO_2$ content increased in the experimental condition. When the blood $CO_2$ content decreased, the extent of the decrease of the red cell volume exceeds that of the increase of the plasma chloride level.

Explainable Machine Learning Based a Packed Red Blood Cell Transfusion Prediction and Evaluation for Major Internal Medical Condition

  • Efficient use of limited blood products is becoming very important in terms of socioeconomic status and patient recovery. To predict the appropriateness of patient-specific transfusions for the intensive care unit (ICU) patients who require real-time monitoring, we evaluated a model to predict the possibility of transfusion dynamically by using the Medical Information Mart for Intensive Care III (MIMIC-III), an ICU admission record at Harvard Medical School. In this study, we developed an explainable machine learning to predict the possibility of red blood cell transfusion for major medical diseases in the ICU. Target disease groups that received packed red blood cell transfusions at high frequency were selected and 16,222 patients were finally extracted. The prediction model achieved an area under the ROC curve of 0.9070 and an F1-score of 0.8166 (LightGBM). To explain the performance of the machine learning model, feature importance analysis and a partial dependence plot were used. The results of our study can be used as basic data for recommendations related to the adequacy of blood transfusions and are expected to ultimately contribute to the recovery of patients and prevention of excessive consumption of blood products.

  • This paper presents the development of a microsystem for whole blood purification and electrophysiological analysis of the purified cells. Magnetophoresis using continuous diamagnetic capture (DMC) was utilized for whole cell purification and electrical impedance spectroscopy (EIS) was utilized for electrophysiological analysis of the purified cells. The system was developed on silicon and plastic substrates utilizing conventional microfabrication technologies and plastic microfabrication technologies. Using the magnetophoretic microseparator, white blood cells were purified from a sample of whole blood. The experimental results of the DMC microseparator show that 89.7% of the red blood cells (RBCs) and 72.7% of the white blood cells (WBCs) could be continuously separated out from a whole blood using an external magnetic flux of 0.2 T. EIS was used as a downstream whole cell analysis tool to study the electrophysiological characteristics of purified cells. In this work, primary cultured bovine chromaffin cells and human red blood cells were characterized using EIS. Further analysis capabilities of the EIS were demonstrated by successfully obtaining unique impedance signatures for chromaffin cells based on the whole cell ion channel activity.

Decolorization of Porcine Red Blood Cell Globin With Ion Exchanger Method and Modification of Its Protein Functionalities

  • Extended use of porcine blood in food ingredients depends on the decolorization of red blood cell concentrates and the modification of its functional properties. The purpose of this study is to compare the relative effect of cation ion exchanger for decolorization of porcine red blood globin. The globin extract is freeze-dried for determination of various functional properties, such as solubility, emulsion capability and foaming ability. Since the isoelectric point of blood globin is located at pH 6.8, which is the neutral pH ranges (6-8), so its functionalities are inferior around these pHs. This weakness has been the main reason, which limit the extended use of blood globin in food industry. Acetylation and succinylation of blood globin can be an alternative way to improve its functionalities. These results may provide new information to understand the decolorization mode by cation ion exchanger for the blood globin. With chemical, the functionalities of blood globin could be obviously improved. The above findings could enable food industry to extend the use of blood globin as a food ingredient.

Relation between Various Body Fluid Volumes and Body Weight or Lean Body Mass in the Rats (흰쥐의 체액량과 체중 및 무지방 체중 사이의 관계)

  • Relationships between red ceil volume $(^{51}Cr-cell)$, total blood volume (red cell volume divided by hematocrit ratio), and extracellular fluid volume (SCN distribution space) and body weight (ranging between 73 and 384 grams) or lean body mass were studied in 59 nembutalized rats. Lean body mass was determined by means of underwater weighing method on rats clipped and eviscerated. There were positive correlations between body weight or lean body mass and the absolute values (in milliliters) of body fluid volumes. Body fluid volumes expressed on the body weight or lean body mass basis, however, showed negative correlations between body weight (grams) or lean body weight (grams) with one exception. Red cell volume expressed as % lean body mass showed a positive correlation with lean body mass. The other results are summarized as follows: 1. Body density of rats was 1.0561 $(range:\;1.0123{\sim}1.0781)$ and 19.8% body weight of total body fat was obtained. The mean value of lean body mass was 80.2% body weight 2. The correlation between body weight and lean body mass was high, namely, coefficient of correlation was r=.99. 3. The correlation between the absolute value of red cell volume (ml) and body weight showed a high correlation, namely, r= 92 and between the lean body mass coefficient of correlation was r=.93. On a weight basis, red cell volume was 2.67 ml/100 gm body weight or 3.48 ml/100 gm lean body mass. The coefficient of correlation between body weight (grams) and red cell volume (% body weight) was r=-. 30. The coefficient of correlation between lean body mass (grams) and red cell volume (% lean body mass) was r=. 50. Thus, the following regression equation was obtained. Red cell volume (% lean body mass)=. 00243 Lean body mass (gm)+3. 12. 4. Total blood volume was 6.06% body weight or 7.83% lean body mass. The correlation between these blood volume values and body weight or lean body mass were negative, namely, r= -.43 and r=-.42 respectively. 5. Extracellular volume (SCN space) was 30.0% body weight or 37.2% lean body mass. These percentage values showed negative correlations between body weight or lean body mass and coefficients of correlation were r=-.40 and r=-.54 respectively. 6. The rate of increase in body weight or lean body mass is accompanied by a smaller rate of increase in blood volume and extracellular fluid volume. The rate of increase in red ceil volume paralled that of lean body mass.

Effect of Ginseng Alcohol Extract on Sodium Movements across the Red Cell Membrane (인삼이 적혈구막을 통한 $Na^+$ 이동에 미치는 영향)

  • In an attempt to explore the effect of Ginseng on the permeability of the biological membrane to cations we have investigated the effect of Ginseng-alcohol extract on the transport of $Na^+$ human red blood cell preprations. The $Na^+$ influx was measured in intact red cells using $^{22}Na$ as a tracer and the efflux in reseated red cells using $^{24}Na$ as a tracer. 1. The influx of $Na^+$ was not apparently changed by the Ginseng-alcohol extract of 20mg% in the incubation medium. 2. Similarly, 20mg% Ginseng-alcohol extract in the cellular space did not alter the efflux of $Na^+$ from the cell. However, 50mg% of Ginseng-alcohol extract in the cell resulted in a significant increase in the $Na^+$ efflux and this effect was magnified when the cell was suspended in the medium containing the Ginseng-alcohol extract in a concentration of 20mg %. The results suggest that Ginseng-alcohol extract over 50mg% increase permeability of red blood cell membrane to $Na^+$ by altering the membrane integrity.
