Cryopreservation of Pine Wood Nematode, Bursaphelenchus xylophilus

  • Pine wood nematode isolates from Haman, Gyoungsangnam-do were cryopreserved with three different cryoprotectants (glycerol, ethylene glycol and dimethylsulfoxide), Nematode Growth medium (NGM) and M 9 buffer solution, respectively, and then survival rate, reproduction ability, and pathogenicity of stored nematodes were compared. Survival rates of juvenile was excellent in 15% glycerol solution as $72{\pm}5.3%$. The survival ability of nematodes at juvenile stage was more efficient than that of nematodes at adult stage. Pre-incubation treatment at $4^{\circ}C$ before storing in liquid nitrogen didn't affect the survival ability of pine wood nematode. When the reproduction ability of nematodes was confirmed on Botrytis cinerea agar plates at $28^{\circ}C$ incubator, there was no difference between cryopreserved and non-cryopreserved. Pathogenic ability of cryopreserved nematodes was also evaluated and confirmed by artificially inoculating them on 2-year old red pine seedlings.

Atmospheric Quality, Soil Acidification and Tree Decline in Three Korean Red Pine Forests

  • Although a forest damage of large area due to air pollution has not yet been found in Korea, declines of Korean red pine (Pinus densiflora Sieb. et Zucc.), the most common coniferous species, have been locally reported. To evaluate the effect of air pollution and acid deposition on the forests, SO$_2$ concentration, acid load, soil pH and tree decline were monitored for 13 years from 1988 to 2001 in Namsan, Doowang and Gyebangsan with the gradient of air pollution. During the study period, annual mean SO$_2$ concentration in Namsan, Doowang and Gyebangsan were 14 ppb, 13 ppb and 6 ppb, respectively. Annual mean acid loads in Namsan and Doowang were three to four times more than that in Gyebangsan. As respected, forest surface soils in Namsan and Doowang were acidified to pH 4.1 and 4.3, whereas that in Gyebangsan showed normal value as pH 5.4. On the other hand, decline degrees of Korean red pines in Namsan and Doowang in both 1996 and 2001 were higher than those in Gyebangsan. It is reasonable that the severer tree declines in Namsan and Doowang could be closely related with the higher air pollution, acid load, and the effects (possibly Ca deficit and Al toxicity) of soil acidification.

Biological Pretreatment of Softwood Pinus densiflora by Three White Rot Fungi

  • The effects of biological pretreatment on the Japanese red pine Pinus densiflora, was evaluated after exposure to three white rot fungi Ceriporia lacerata, Stereum hirsutum, and Polyporus brumalis. Change in chemical composition, structural modification, and their susceptibility to enzymatic saccharification in the degraded wood were analyzed. Of the three white rot fungi tested, S. hirsutum selectively degraded the lignin of this sortwood rather than the holocellulose component. After eight weeks of pretreatment with S. hirsutum, total weight loss was 10.7%, while lignin loss was the highest at 14.52% among the tested samples. However, holocellulose loss was lower at 7.81 % compared to those of C. lacerata and P. brumalis. Extracelluar enzymes from S. hirsutum showed higher activity of ligninase and lower activity of cellulase than those from other white rot fungi. Thus, total weight loss and changes in chemical composition of the Japanese red pine was well correlated with the enzyme activities related with lignin- and cellulose degradation in these fungi. Based on the data obtained from analysis of physical characterization of degraded wood by X-ray Diffractometry (XRD) and pore size distribution, S. hirsutum was considered as an effective potential fungus for biological pretreatment. In particular, the increase of available pore size of over 120 nm in pretreated wood powder with S. hirsutum made enzymes accessible for further enzymatic saccharification. When Japanese red pine chips treated with S. hirsutum were enzymatically saccharified using commercial enzymes (Cellulclast 1.5 L and Novozyme 188), sugar yield was greatly increased (21.01 %) compared to non-pre treated control samples, indicating that white rot fungus S. hirsutum provides an effective process in increasing sugar yield from woody biomass.

산불 피해목의 재질변화에 관한 연구(II) - 산불 피해 소나무의 경시적 재질변화 - (Variation of Material Properties of Fire-killed Timber - Impact of Time on Degradation of Mechanical Properties -)

  • 동해안 지역에서 산불로 고사된 소나무의 5년에 걸친 경시적 재질 변화를 역학성능을 중심으로 조사하였다. 고사에 따른 재질열화로 가장 크게 영향 받는 역학성능은 충격 휨 흡수에너지였다. 따라서 고사 초기의 재질변화를 예측하는 지표로 활용이 가능할 것으로 밝혀졌다. 고사 경과에 따라 변재의 재질열화가 심재에 비해 두드러지게 진행되었다. 열화의 정도는 충격 휨 흡수에너지가 가장 컸으며 압축강도가 가장 작았다. 고사 이후의 외관 변화와 역학성능의 열화진행 정도로 볼 때 산불피해 소나무의 유효한 활용을 위한 적정 벌채 시기는 산불발생 1년 이내가 바람직한 것으로 밝혀졌다.

전라도지역 조선후기 목조불상의 수종 (Species of Wooden Buddhist Statues of the Late Joseon Dynasty in Jeollado, South Korea)

  • The objective of this study is to identify the species of 17 wooden Buddhist statues of the late Joseon Dynasty (the 17th and 18th century) in Jeollado, southwestern region of Korea. The bodies of statues were made of Ginkgo (Gingko biloba L.: 88%) and alder (Alnus spp.: 12%). The hands of statues were alder(64%), willow (Salix spp.: 27%) and Ginkgo(9%). The bottoms of hollow bodies were covered all with Japanese red-pine (Pinus densiflora S. et Z., 'sonamu') panels. The main species of statue body, Ginkgo tree was known to be introduced to Korea from China with Buddhism. The results indicated that Ginkgo trees of Korea in the 17th century had already become large and rich enough to be used for most of statues. Ginkgo wood has low shrinkage and even texture, which are crucial for carving sculptures. Alder and willow woods used for statue hands have fine and firm textures. The pedestals for these statues were also made of red pine wood. Red pine woods have rather high shrinkage and low hardness, but it is versatile woods strong enough to support heavy statues.

농촌 소나무림에서 산불에 의한 식생변화와 유용미생물의 적용 (Vegetation Change after A Forest Fire in a Rural Japanese Red Pine Forest and Applications of Effective Microorganism)

  • This study examined the natural restoration of vegetation through monitoring of the development of a vegetation community from 2006 through 2007 after a forest fire. Approximately 5,000 $m^2$ in a forest near Topyeon-ri, Kangnae-myeon, Chungcheongbuk-do with Japanese red pine (Pinus densiflora) forest and its floor vegetation had been completely burned by a fire in April 2005. This area and another nearby Japanese red pine forest were selected as the experiment site and the control site, respectively. Vegetation survey was conducted at the experiment site and the control site. A seed bank experiment was carried out in the greenhouse to examine underground vegetation. Effective microorganism(EM) was applied to the seed bank experiment to estimate its effects on the direction of ecological succession. According to the results, a total of 36 plant species including shrub and herbaceous species were discovered in the experiment site. Quercus serrata, Lespedeza cyrtobotrya, and Castanea crenata, Rubus crataegifolius, Oplismenus undulatifolius, and Carex lanceolata were among the most abundant species. Biomass in the experiment site reached 2.4 times biomass than those in the control site, indicating the productivities of shrub and herbaceous layers are better in the experiment site. According to the result of the soil seed bank experiment of the experiment site, a total of 182 plants of 14 species were recorded. In addition, a total of 13 plants of 2 species were found from soil seed bank of the experiment site applied by EM. If EM is applied to the burned site, it will control the budding of herbaceous plants, creating the gap between herbaceous plants. This loss of competition is expected to help the restoration of trees in the burned area.

산불로 인한 지표층 연소량 및 온실가스 배출량 추정 (Estimation of Biomass Loss and Greenhouse Gases Emissions from Surface Layer Burned by Forest Fire)

  • 산불은 전 세계적으로 다량의 온실가스를 배출하여 지구온난화에 기여한다. 본 연구에서는 산불로 인한 지표층 연료의 연소량과 이로 인한 온실가스 배출량을 분석하고자 하였다. 이를 위해 51개 산불을 대상으로 소나무림과 활엽수림으로 구분하고, 다시 지표화와 수관화 피해 지역으로 나누어 잔존 연료량을 채취하여, 미연소 지역의 연료량과 일원분산분석을 수행하였다. 그 결과 활엽수 지표화 지역은 8,361 kg/ha, 소나무 지표화 지역은 8,055 kg/ha, 소나무 수관화 지역은 12,333 kg/ha이 연소되어, 수종별 산불행태별로 차이가 있었다. 이때의 연소율은 각각 78, 59, 90%이었다. 이산화탄소, 메탄, 아산화질소 등의 온실가스 배출량은 활엽수 지표화 연소지역 15,856 kg/ha, 소나무 지표화 연소지역 14,834 kg/ha, 소나무 수관화 연소지역은 약 22,709 kg/ha이었다.

대구 팔공산 지역의 소나무 수관층 연료 특성 (Crown Fuel Characteristics of Japanese Red Pine (Pinus densiflora) in Mt. Palgong, Daegu)

  • 본 연구에서는 우리나라에서 수관화에 취약하다고 밝혀진 소나무림을 대상으로 수관층의 연료특성을 잎과 가지 굵기 별로 구분하여 수분함량, 연료밀도로 제시하고자 하였다. 이를 위해 대구 팔공산에서 소나무 10그루를 벌채하여 각 부위별로 건중량을 측정하였다. 분석결과 수관층 살아 있는 부위의 평균 수분함량은 53%, 죽은 부위는 15.3%였으며, 생엽의 수분함수율은 평균 56%이었다. 수관화 확산시 실제적으로 연소되는 잎과 직경 1 cm 이하의 가지를 합한 연료량은 전체 바이오매스에서는 16.2%, 수관층 바이오매스에서는 55%를 차지하였다. 수관 연료밀도는 평균 0.24 kg/$m^3$이었으며, 잎과 1 cm 굵기 이하의 가지를 합한 유효 수관연료밀도는 0.1325 kg/$m^3$이었다.

Profiling Patterns of Volatile Organic Compounds in Intact, Senescent, and Litter Red Pine (Pinus densiflora Sieb. et Zucc.) Needles in Winter

  • This study was aimed to investigate the changes of chemical composition of the volatile organic compounds (VOCs) emitted from red pine needles in the process of needle abscission or senescence. The VOCs in intact, senescent, and litter red pine needle samples were analyzed by headspace-solid phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC/MS). And then, multivariate statistical interpretation of the processed data sets was conducted to investigate similarities and dissimilarities of the needle samples. Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were used to investigate the dataset structure and discrimination between samples, respectively. From the data preview, the levels of major components of VOCs from needles were not significantly different between needle samples. By PCA investigation, the data reduction according to classification based on the chlorophyll a / chlorophyll b (Ca/Cb) ratio were found to be ideal for differentiating intact, senescent, and litter needles. The following OPLS-DA taking Ca/Cb ratio as y-variables showed that needle samples were well grouped on score plot and had the significant discriminant compounds, respectively. Several compounds had significantly correlated with Ca/Cb ratio in a bivariate correlation analysis. Notably, the litter needles had a higher content of oxidized compounds than the intact needles. In summary, we found that chemical compositions of VOCs between intact, senescent, and litter needles are different each other and several compounds reflect characteristic of needle.

Effects of Pretreatment for Controlling Internal Water Transport Direction on Moisture Content Profile and Drying Defects in Large-Cross-Section Red Pine Round Timber during Kiln Drying

  • Round timber materials of 600 mm length, cut from large-cross-section round timber of red pine (Pinus densiflora S. et Z.) of 450 mm width and 4.2 m length, were prepared as the target of kiln drying in this study. After treating the target materials through end sealing (ES), end sealing - kerfing (ES-K), lateral sealing - end sealing - boring (LS-ES-B), or lateral sealing - partial end sealing (LS-PES), the effects of the treatment on the incidence of drying defects were determined. The target materials with exposed lateral surface and sealed cross surface were steamed at the initial temperature of 65℃ above the official pest control temperature of 56℃, followed by kiln drying toward the final temperature of 75℃. The target materials with sealed lateral surfaces, on the other hand, were dried at the initial temperature of 90℃ at almost the maximum temperature of conventional kiln drying, as there is no risk of early check formation caused by surface moisture evaporation. The final temperature was set at approximately 100℃. The drying time, taken for the target materials with initial moisture content of 70%-80% to reach the target moisture content of 19%, varied across treatment conditions. The measured drying time was 1,146 hours (approximately 48 days) for the timber with sealed cross surface and 745 hours (approximately 31 days) for the timber with sealed lateral surface, until the moisture content reached the target level. The formation of surface checks could not be prevented in the control and ES groups, but a definite preventive effect was obtained for the LS-ES-B and LS-PES groups.
