선행연구로서 꽃송이버섯 배지내 LED 조명색에 의한 푸른곰팡이 생육억제 효과는 모든 처리구에서 생육이 억제되었으나 blue 처리구에서 푸른곰팡이균의 포자발생량이 매우 높아 꽃송이버섯 배양에 부적합하다는 것으로 확인되었다. 배양 28일 후 LED 파장에 따른 꽃송이버섯의 균사생장 특성과 LED 파장 색상별 배양시간의 경과에 따른 균사생장 특성을 조사한 결과 모두 red 파장 처리구에서 균사생장이 높은 경향을 나타내었다. 이에 따라 red 파장 범위에서 광량을 달리하여 균사생장 조건을 확인한 결과, 광량 세기가 낮은 1.41 ${\mu}mol/m^2S$ 처리구에서 우수한 균사생장을 나타내었다. 계대배양 실시 여부에 따른 균사생장 특성에서는 실험 전 1회 계대배양을 실시한 처리구에서 유의적인 차이를 나타내었다. 배지조성을 달리한 톱밥배지 내 LED 광량별 균사생장은 미송 처리구에서 높은 경향을 나타내었다. 이를 미송 톱밥배지에서 LED red 2.11 ${\mu}mol/m^2S$의 광량을 처리하였을 경우 84일 배양한 처리구에서 유의적인 차이를 보였다. 상기의 결과를 종합하여 보았을 경우 red 파장 2.11 ${\mu}mol/m^2S$ 광량에서 84일 이상 미송 톱밥배지에서 배양하는 것이 꽃송이버섯 배양에 좋을 것으로 판단되었다.
The study was carried out to analyze wood quality of coffin board, the wood species of coffin excavated in Youngdong, Korea. All coffin woods were identified as hard pines, Korean red pine(Pinus densiflora S. & Z.). According to wood quality of coffins, inner coffins of Bae Cheon-Jo and his wife showed similarity and dissimilarity comparing with several literatures. The similarity was in the structure of panels, bottom lining board and charcoal layer, and the dissimilarity was in jointing. From the analysis of fibrous materials attached to coffin woods, the paper mulberry and cotton fibers were identified. In the inner coffin of Bae Cheon-Jo, tool traces by handle planer were observed.
Layered structures of fiber cell wall of Korean red pine (Pinus densiflora) were investigated by confocal reflection microscopy (CRM). CRM micrographs revealed detailed structures of the fiber cell wall such as S1, S2, and S3 layers as well as transition layers (S12 and S23 layers), which are present between the S1, S2, and S3 layers. Microfibril angle (MFA) measurement was possible for the S2 and S3 layer in the cell wall. The experimental results suggest that CRM is a versatile microscopic method for investigation of layered structures and MFA measurement in individual sub layer of the tracheid cell wall.
To evaluate the resistance of wood to decay caused by fungi, sapwood and heartwood of red pine (Pinus densiflora) and sapwood and heartwood of larch (Larix kaempferi) were conducted. Wood samples were immersed for 96 h in pyroligneous liquor. Then, the brown-rot fungus, Fomitopsis palustris, was used to examine the decay resistance of red pine and larch. Weight and density of wood from the all conditions increased after immersion treatment. Weight loss after decay resistance test was also dropped with a immersion treatment. The lowest weight loss indicated at immersion-treated heartwood of larch. Immersion treatment using pyroligneous liquor effectively increased the resistance of wood to decay caused by fungi.
Many kind tree species are growing in Korea. However most of them are inferior in wood qualities and production for commercial utilization except several species. Among the useful several species Red Pine (Pinus densiflora S. et. Z.) is growing in main tree species and most generally utilizing wood in korean forest. It is used widely as mine timbers, cross ties, boxes, floorings, pulpwoods, constructions and engineerings. Although this species is most useful and economical in this country, the basical properties on relation between wood and relative humidities were not studied certainly. This study was performed by need of inspection the hygroscopic and swelling properties to be apllied for physical manufacturing. According to the results the Equilibrium Moisture Contents of each relative humidity conditions are as table (3), and the swelling values of sap and heartwood are as table (4) and (5) in text. In this study sapwood moisture contents are higher than heartwood in each relative humidities conditions. Tangential and radial ratios of average swelling values are 1:1.971 in sapwood and 1:2.178 in heartwood.
To elucidate the structure of procyanidin in Korean red pine (Pinus densiflora S. et Z.), bark, the extractives were extracted with acetone-water mixture(7:3, v/v) from inner bark of Korean red pine. The extracts separated three fractions which were extracted by liquid-liquid extraction. The extracting solvents were chloroform and ethyl acetate and water. The part of ethylacetate soluble was chromatographed by liquid chromatography. The ethylacetate soluble portion yielded four natural procyanidin dimers, two known epicatechin-($4{\beta}{\rightarrow}6$)-catechin, catechin-($4{\alpha}{\rightarrow}8$)-catechin and two unknown catechin-($4{\beta}{\rightarrow}6$)-catechin and conformational isomer of epicatechin-($4{\alpha}{\rightarrow}6$)-catechin. The additional catechins was also isolated. The structures of these procyanidins were elucidated by their $^{13}C$-NMR spectra.
Comparisons of the physico-chemical properties of earthworm(Lumbricus terrestris) soil casts and top soil of red pine forests in a limestone area were carried out. The casts production durign August and September in 1990 amounted to 6∼7t/ha. The particle of top soil and casts ranged 40∼50% and 10∼20% for sand , 25∼30% and 30∼35% for silt, and 20∼25% and 55∼65% clay, respectively. Significant difference in pH value was not observed between casts and top soil. The casts had 1.4times of organic matter, 1.5times of N, 1.8times of available P, 2times of exchangeable K, 1.3times of exchangeable Ca, and 1.6times of exchangeable Mg than the top 10cm of soil did. Earthworms have altered the soil texture and increased nutrient availability through production of the soil casts in this limestone area.
This study demonstrates the utilization of light microscopy and Fast Fourier Transform-Peak Finding (FPF) method for microfibril angle (MFA) measurement from unstained sections of red pine (Pinus densiflora). To obtain an image with optimal contrast and resolution for MFA measurement, effects of numerical aperture (NA) of condenser lens and color filters were investigated. About 60% of NA of the maximum condenser NA produced an image with optimal contrast, but a color filter with short wavelength range (DAPI) created images with improved resolution. Manual angle measurement and the FPF method were applied to the image with optimal contrast for MFA measurement. The experimental results from the FPF method were considered to be more repeatable and less subjective than those from the manual angle measurement.
The effects of dried leaf powders and ethanol extract of persimmon, green tea and pine needle on lipid metabolism, lipid peroxidation and antioxidative activity were investigated in rats. Forty-nine male Spargue-Dawley rats weighing 107.8$\pm$1.8g were blocked into seven groups according to body weight. Rats were raised for four weeks with diets containing either 5%(w/w) dried leaf powders of one of three different Korea traditional teas, persimmon(Diospyros kaki Thunb), green tea(Camellia Sinensis O.Ktzc)or pine needle(pinus Koreansis Sieb. Et Zucc), or ethanol from equal amounts of each dried tea powder. Food intake, weight gain, food efficiency ratio, and weights of liver, kidney and epididymal fat were significantly higher in the green-tea-powder group, and significantly lower in the pine-needle-powder and pine-needle-extract groups. Persimmon-leaf powder was found to decrease plasma total lipid, triglyceride and cholesterol concentration by increasing fecal total lipid triglyceride and cholesterol excretions. Liver cholesterol concentration was significantly lower in the green-tea and pine-needle-extract groups. Red-blood-cell superoxide dismutase(SOD) and glutathione peroxidase(GSH-px) activities were significantly increased in rats fed green-tea extract. Liver SOD activity was increased in rats fed pine-needle powder or extract, and liver GSH-px activity was increased in rats fed greentea powder. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentration were both decreased in rats fed dried leaf posers or extracts of persimmon or green tea. It is believed that high vitamin E levels in persimmon leaf, and high flavonoid, beta-carotene and vitamin C levels in green tea effectively inhibited lipid peroxidation. In conclusion, persimmon and green tea leaves were effective in lowering lipid levels and inhibiting lipid peroxidation in animal tissue, while pine needles were effective and lowering body weight gain. From these results, persimmon and green tea leaves can be recommended in the treatment and prevention of chronic discorders such as cardiovascular disease, cancer and aging. As ethanol extracts from these teas were also effecitive in lowering tissue lipid levels and inhibiting lipid peroxidation, we recommend the use of discarded tea grounds for this.
사찰인근 소나무림은 문화경관림으로서 가치가 높으나 식생천이와 기후변화로 쇠퇴할 가능성이 높다. 본 연구에서는 사찰인근 소나무림을 대상으로 식생구조와 소나무 활력도, 입지환경 특성을 조사해 소나무 생육과 입지환경간의 상관관계를 밝히고자 했다. 소나무림이 양호하게 남아 있는 전남지역의 천은사, 원효사, 증심사, 태안사 4개소를 연구대상지로 선정했다. 이 대상지의 교목층에는 모두 소나무가 우점하고 하층에는 낙엽활엽수종이 주로 출현했다. 소나무의 수령이 많은 천은사 지역보다 증심사 원효사의 소나무 생육상태가 전반적으로 좋지 않았다. 토양의 총질소량은 증심사가 다른 지역보다 높은 편이었는데 하층식생 발달로 총질소량이 늘어난 것으로 보인다. 이로 인해 낙엽활엽수종과의 질소양분 경합에서 밀려 소나무의 생육이 악화된 것으로 추정된다. 해발고와 가지길이는 부(-)의 상관관계를, 경사도와 소나무 평균상대우점치도 부(-)의 상관관계를 보였다. 즉, 해발고 경사도가 상승할수록 토양환경을 포함한 생육환경이 열악해지면서 소나무의 생장량 우점도가 저조해지는 것으로 보인다. 경사도는 소나무 가지길이와 정의(+) 상관관계를 보였다. 소나무림이 주로 분포하는 입지환경 범위 내라면, 급경사지일수록 낙엽활엽수종과의 경쟁을 회피해 소나무 생육이 양호해지는 것으로 판단된다. 반면, 토양의 전기전도도가 증가할수록 소나무 생육이 더 악화되는 것으로 분석됐다. 토양양분 증가는 낙엽활엽수종의 식생 발달을 촉진해 소나무의 생육환경을 악화시켜 소나무림의 건전성 유지에 부정적 영향을 미친 것으로 보인다.
