Cell Recycled Culture of Succinic Acid-Producing Anaerobiospirillum succiniciproducens Using an Internal Membrane Filtration System

  • Cell recycled culture of succinic acid-producing Anaerobiospirillum succiniciproducens was anaerobically carried out using an internal membrane filter module in order to examine the physiological response of A. succiniciproducens to a high-cell-density environment. The optimal growth of A. succiniciproducens and its enhanced succinic acid productivity were observed under $CO_2$-rich conditions, established by adding $NaHCO_3$ and $Na_2CO_3$, in the cell recycled system. A. succiniciproducens grew up to 6.50 g-DCW/l, the highest cell concentration obtained so far, in cell recycled cultures. The cells did not change their morphology, which is known to be easily changed in unfavorable or stress environments. The maximum productivity of succinic acid was about 3.3 g/l/h, which is 3.3 times higher than those obtained in batch cultures. These results can serve as a guide for designing highly efficient cell recycled systems for succinic acid at a commercial level.

A Study on the Preparation of Oil Absorbent Using Waste Polyurethane (폐폴리우레탄을 이용한 오일 흡수제의 제조)

  • The preparation of oil absorbent using waste polyurethane was studied. And the effects of shape, size, and contents of waste polyurethane foam was investigated. The waste foam was treated in shape of powder, cube and bar type generated from rigid sandwich panel process. The tests for flexural strength, combustibility, and water absorptivity were carried out to investigate the mechanical and physical properties of the recycled foams. And the cell microstructure was evaluated through Scanning Electron Micrograph. The recycled foam containing powder-shaped underfilled and showed poor properties that was generated through reactivity of the resins and increasing of slurry viscosity. For the recycled foam with the cube and bar-shaped, the underfilling was caused by interference between the waste PUFs and increasing surface areas of PUF. Low cell density, non-uniformity of cell shape and size, and low adhesion of the boundary surface (new/recycled) was showed as a result of the poor properties. Considering underfilling and the properties of PUF (new/recycled), maximum recycle contents were less than 20 wt% for the powder and above 40 wt% for the cube and bar.

  • The optimal cell concentration and dilution rate for maximum ethanol productivity were obtained using dynamic simulation in cell recycled continuous bioreactor. The good control performance was observed using rule-based STR (self-tuning regulator) compared to conventional STR. Rule-base contained the scheme to implement the STR in an efficient on-off way and the scheme for the controlled variable to reach the optimal value in a short time. Since a mathematical model was used to analyze and estimate the changes of the state variables and the parameters, it was possible to understand the physical meaning of the system.

  • An internal membrane bioreactor system was employed for continuous succinic ac id production from glucose in order to prove its performance and practicality. Succinic acid-producing Anaerobiospirillum succiniciproducens required more $CO_2$ for the proper growth and succinic acid production in cell recycled continuous culture than in batch culture. The maximum productivity obtained in cell recycled continuous culture was about 3.3 g/L-h which was ca. 3.3 times higher than that obtained in batch culture.

  • The purpose of this stuy was to evaluated the cytotoxicity of brackets which were recycled thermally or chemically. New brackets and used brackets which had been in mouth for at least 2years were used as samples and human gingival cell culture and agar overlay technique was used to evaluate the cytotoxicity. From the experiment the following results were obtained : 1. New brackets in the as received state showed mild cytotoxicity. 2. Thermally recycled brackets except the used bracket not electropolished showed moderate cytotoxicity and among them new brackets showed greater cytotoxicity than used ones. 3. Used brackets which were thermally recycled without electropolishing showed mild cytotoxicity. 4. Among thermally recycled brackets, electropolished brackets showed greater cytotoxicity than not electro-polished ones. 5. Chemically recycled brackets showed moderate cytotoxicity, and among them, new brackets appeared to be more cytotoxic than used ones.

Cell-Recycled Continuous Ethanol Fermentation of Molasses (당밀의 균체순환식 Ethanol 연속발효)

  • A cell-recycled continuous fermentation process was studied to produce ethanol from molasses using Sacchoromyces uvarum ATCC26602 at 35 $^{\circ}C$. The fermentation system was divided into two stages in order to reduce the inhibitions of ethanol and substrate for cell growth and fermentation rate. The first reactor was aerated at the rate of 0.12 vvm whilst the second was kept anaerobic. In medium composition studies, it was revealed that inorganic nutrient supplement to the diluted molasses with 14% fermentable sugar was not needed for the fermentation, however, phosphate limitation was observed when cell propagation was contemplated. By using the cell-recycled continuous fermentation system, 14.5 hour was required to produce 8.4-9.0% (v/v) of ethanol from diluted molasses containing 14% of fermentable sugar. The ethanol productivity was 6.2g/$\ell$hr with the yield of 88.1-94.4% to the theoretical value.

  • Used polyurethane was chemically degraded by treatments with flame retardants such as tris(3-chloropropyl) phosphate (TCPP), triethyl phosphate (TEP), and trimethyl phosphate (TMP). The structure of degraded products (DEP) was analyzed by FT-IR and P-NMR and it turned out to be phosphorus containing oligourethanes. Rigid polyurethane foam was produced by using the degraded products (DEP) as flame retardants. The flammability of recycled rigid polyurethane was investigated. The recycled polyurethane shows a reduced flammability over virgin polyurethane. In order to evaluate flame retardant properties of the recycled polyurethane foams with various amounts of DEP, the combustion parameters of the foam was measured by a cone calorimeter. Scanning electron micrograph of recycled PU shows the same uniform cell morphology as virgin PU.

A Study on the Performance of Recycled Cells for application to Residential BESS (주택용 BESS에 적용하기 위한 재활용 셀의 성능에 관한 연구)

  • To determine the performance of recycled cells for application to residential BESS, cells used over the past 5 years were selected. The basic specifications of the cell used in the test are nominal voltage of 3.7[V], nominal capacity of 2,200[mAh], charging voltage of 4.05[V], continuous discharge current of 1[C](2,200[mA]), continuous charging current of 0.5[C](1,100[mA]). For new cells, the internal resistance was 21.3±1[mΩ], but for recycled cells, the average internal resistance was 25.38[mΩ], an increase of about 19.1[%]. The charge·discharge capacity was approximately 18.9~19.3[%] lower than that of a new cell. Because internal resistance and charge·discharge capacity are closely related to cell aging, cells to be applied to BESS need to use products with an initial internal resistance of 1.5 times or less and a charge·discharge capacity performance of 70[%] or more.

Optimization of Cell Concentration and Dilution Rate in Cell Recycled Ethanol Fermentation (세포재순환 에탄올 발효에서 세포농도와 희석률의 최적화)

  • The rheological characteristics of the ethanol fermentation broth were pseudoplastic when the yeast concentration was above 150g/L. From the viewpoint of rheological properties, the cell concentration below 150g/L was recommended for ethanol fermentation. Since the cell floc was formed at the cell concentration of 100 g/L, yeast cells were not much plugged in the pores of the membrane. The cell concentration above 100g/L was desirable when considering the permeability of the membrane. Since ethanol productivity was the highest when the cell concentration was 130 g/L in cell recycled ethanol fermentation. The optimal dilution rate was determined at 1.3 h-1 at constant cell mass of 130g/L. At this dilution rate, the ethanol productivity and glucose conversion ratio ware 80 g/L$\cdot$h and 0.94, respectively.

  • To increase the volumetric productivity a contimuous cell-recycled system was implemented. Cell concentrations between 9.2 and 15.0 g/1 were obtatined in the continuous fermentor study. At a 4% xylose feed and a specific oxygen supply rate(SOSR) of 1.04 g O2.hr-g DCW the ethanol yield was 0.36% at dilution rate. This represented a 26-% increase over that of th batch fermentation.
