In this study, we focused on 'Adaptive Reuse', which has the concepts of space reconstitution and social and cultural regeneration, so that previously constructed highrise buildings can accommodate various changes in modern society, and new spatial design was developed based on the characteristics of adaptive reuse. We sought to present the concept and direction, detailed strategies, and architectural response elements of space reconstitution for the purpose of realizing the typical spatial reorganization required by modern society and seeking directions and methods of space reconstitution that can be applied to future society. The purpose is to do so.
The stability of penicillin G potassium injection after reconstitution was evaluated in two different diluents of sodium chloride $0.9\%$ and dextrose $5\%$ in water stored at room temperature or refrigerated condition. The concentrations of penicillin G, stored for 24 hours at room temperature or for 10 days at refrigerated condition, were determined by HPLC. Also the pHs of the reconstituted solutions were monitored. The concentrations and pHs of penicillin G potassium 500,000 U/ml injection after reconstitution gradually decreased in all conditions. Stored at room temperature after reconstitution, a new peak which suspected as degradation products of penicillin G was detected in 5 hours in sodium chloride $0.9\%$, 4 hours in dextrose $5\%$ in water. At refrigerated condition, the new peak was detected in 4 days in both sodium chloride $0.9\%$ and dextrose $5\%$ in water. The degradation products of penicillin G allergy have been thought to be one of the substances responsible for evoking allergic reactions. In conclusion, the penicillin G potassium 500,000 U/ml injection after reconstitution was stable for 4 hours in sodium chloride $0.9\%$ 3 hours in dextrose $5\%$ in water solution at room temperature. At refrigerated condition, both solutions were stable for 3 days after reconstitution.

Recombinant human ferritin homopolymers (rHF and rLF) were successfully produced in the Saccharomyces cerevisiae Y2805, which was transformed with human ferritin H or L-chain genes, respectively. In order to characterize the molecular properties of the recombinant ferritins in relation to mineralization, the proteins were isolated and apoferritins were prepared. The apoferritins were reconstituted with 2000 Fe atoms per protein molecule under various experimental conditions (the concentration of the protein, the buffer concentration of the MOPS buffer, the total volume of the reaction and the reconstitution method). The structure and composition of the iron cores formed in the ferritins were examined using transmission electron microscopy. The recombinant ferritins behaved in a similar manner to other mammalian ferritins in accumulating iron in the core. Proteins of rHF and rLF showed varying reconstitution yields of 37-72% depending on the reaction conditions. In general, the rHF showed higher reconstitution yield than the rLF at the protein concentrations and the reaction volumes we examined. Iron cores with a similar mean particle size were obtained in the rHF, rLF and horse spleen ferritin reconstituted at a protein concentration of 1.0 mg/mL. Electron diffraction of all the three ferritins showed 2-3 diffuse lines, with d-spacings corresponding to those of the mineral ferrihydrite with a limited crystallinity.
Objective: To explore the immune reconstitution of $CD4^+T$ cells after allogeneic hematopoietic stem cell transplantation (Allo-HSCT) and its relationship with invasive fungal infection (IFI) in patients with hematological malignancies. Materials and Methods: Forty-seven patients with hematological malignancies undergoing Allo-HSCT in Binzhou Medical University Hospital from February, 2010 to October, 2014 were selected. At 1, 2 and 3 months after transplantation, the immune subpopulations and concentration of cytokines were assessed respectively using flow cytometry (FCM) and enzyme linked immunosorbent assay (ELISA). The incidence of IFI after transplantation and its correlation with immune reconstitution of $CD4^+T$ cells were investigated. Results: The number of $CD4^+T$ cells and immune subpopulations increased progressively after transplantation as time went on, but the subpopulation cell count 3 months after transplantation was still significantly lower than in the control group (p<0.01). In comparison to the control group, the levels of interleukin-6 (IL-6) and IL-10 after transplantation rose evidently (p<0.01), while that of transforming growth factor-${beta}$ (TGF-${beta}$) was decreased (p<0.01). There was no statistically significant difference level of interferon-${\gamma}$ (IFN-${\gamma}$) (p>0.05). The incidence of IFI was 19.2% (9/47), and multivariate logistic regression revealed that IFI might be related to Th17 cell count (p<0.05), instead of Th1, Th2 and Treg cell counts as well as IL-6, IL-10, TGF-${beta}$ and IFN-${\gamma}$ levels (p>0.05). Conclusions: After Allo-HSCT, the immune reconstitution of $CD4^+T$ cells is delayed and Th17 cell count decreases obviously, which may be related to occurrence of IFI.

We have conducted in vitro reconstitution study of ferritin from its subunits FerH and FerL. For the reconstitution, FerH was produced from an expression vector construct in Escherichia coli and was purified from a heat treated cell extract by using one-step column chromatography. FerL was expressed as inclusion bodies. The denatured form of FerL was obtained by a simple washing step of the inclusion bodies with 3 M urea. The reconstitution experiment was conducted with various molar ratios of urea-denatured FerH and FerL to make the ferritin nanoparticle with a controlled composition of FerH and FerL. SDS-PAGE analysis of the reconstituted ferritins revealed that the reconstitution required the presence of more than 40 molar$\%$ of FerH in the reconstitution mixture. The assembly of the subunits into the ferritin nanoparticle was confmned by the presence of spherical particles with diameter of 10 nm by the atomic force microscopic image. Further analysis of the particles by using a transmission electron microscope revealed that the reconstituted particles exhibited different percentages of population with dense iron core. The reconstituted ferritin nanoparticles made with molar ratios of [FerH]/[FerL]=l00/0 and 60/40 showed that 80 to $90\%$ of the particles were apoferritin, devoid of iron core. On the contrary, all the particles formed with [FerH]/[FerL]=85/ 15 were found to contain the iron core. This suggests that although FerH can uptake iron, a minor portion of FerL, not exceeding $40\%$ at most, is required to deposit iron inside the particle.
The purpose of this paper is to introduce a new and effective classroom speaking activity helping student's communication in real situation. It will be one of useful teaching techniques for teachers because it can be used with other various types of speaking activities together. The activity is designed by the researcher, and named as the Partner's Speech Reconstitution(PSR) in this paper. In chapter 2, Noticing and Output hypothesis which is the theoretic basis of the PSR will be described and the chapter 3 will explain activity methods and examples of the PSR, and then describe its merits and demerits. The researcher applied and practiced the PSR in the speaking class for international students in the K university for three semesters. This paper systematically introduces its organized activity. It helped learners elicit speaking performance of students who avoided talking in the speaking class, made the students concentrate in speaking activity, and helped the learners to talk sufficiently by inducing each student to reconstitute partner's speech production.
256 다중 검출기 전산화단층촬영 (multi detector computed tomography, MDCT)에서 두개부 전용 팬톰을 이용하여 분해능 파라메터와 재구성 필터의 영상 품질을 평가하고자 하였다. 사용한 장비는 256 MDCT 을 사용하였으며 philips system head phantom을 이용하여 Extended Brilliance Workspace에서 영상의 질 평가를 측정하였다. 장비에서 지원하고 있는 분해능 파라메터와 재구성 필터가 화질에 미치는 영향을 알아보기 위해서 검사조건을 $512{\times}512$ matrix, $128{\times}0.625$ mm beam collimation, 120 kVp, 250 mAs, 0.5 sec, 250 mm field of view (FOV), 절편 두께와 절편 간격은 5 mm, 1.0 pitch로 동일하게 적용하였다. 분해능 파라메터은 'Standard', 'High', 'Ultrahigh'로 구분하여 적용하였으며, 재구성 필터는 'A', 'B', 'C', 'D', 'UA', 'UB', 'UC' 등으로 바꿔가면서 영상을 재구성하여 노이즈, 균일도, 직선성, 변조전달함수 (modulation transfer function, MTF)의 50%와 10%를 측정하였다. 그 결과 'High' 분해능은 균일도, 직선성, MTF 50%와 10%에서 우수하였다. 'UA', 'UB' 재구성 필터는 균일도와 노이즈 평가에서 양호했으며 'C'재구성 필터는 직선성과 MTF 50%와 10%에서 양호하였다.
세포막을 구성하고 있는 지질의 물리학적 성상이 단백질의 세포막 속으로의 삽입과정 및 단백질의 기능에 미치는 영향을 관찰하기 위하여 골격근의 근세망(SR)으로부터 $Ca^{2+}-ATPase$ 단백질을 분리한 후 이를 세포막의 주 구성성분인 포스파티딜콜린(PC)과 포스파티딜에타노라민(PE)의 혼합지질과 재조합(reconstitution)시켰다. 이와같이 인공적으로 재조합된 구조물에서 $Ca^{2+}-ATPase$의 기능을 측정하기 위하여 칼슘지시색소인 아르세나죠III(AIII)를 이용한 분광방법과 방사선동위원소를 이용한 여과법으로 칼슘흡수율을 측정하였고 또한 ATP 가수분해 능력을 측정하였다. 실험결과 칼슘의 흡수율은 포스파티딜코린의 함량이 많은 혼합지질과 재조합시킬 때에 증가하였고, ATP 가수분해 능력은 포스파티딜함량이 25%까지는 포스타피딜코린의 양에 비례하여 증가하였으나 50%이상에서는 약간 감소하는 경향을 보였다. 한편 지질세포막속으로 단백질이 삽입되는 양은 포스파티딜 함량이 25%일 때 최고의 값을 보였으며 함량이 그 이하 또는 이상일 때는 감소하였다. 이상의 실험결과로보아 단백질의 기능은 세포막이 "bilayer" 구조를 갖출때에 증가하고 세포막속으로 단백질이 삽입되는 양은 세포막이 "non-bilayer" 구조를 형성할 때에 증가함을 알 수 있다.
The present experiment was conducted to assess the effect of reconstitution (R) on utilization of red sorghum (S) in diets of broiler chickens. Day-old broiler chicks (n=360) were randomly divided into 36 groups of 10 chicks each, and 9 dietary treatments were allotted to 4 groups (replicates) in a completely randomized design. Out of the 9 treatments, one was corn-soy based control (D1). The rest of the treatments were diets consisting of four levels (25, 50, 75 and 100% part of corn) of raw red sorghum (S25-S100) or four levels of reconstituted red sorghum (RS25-RS100). The tannin content reduced from 2.3% to 1.6% after reconstitution of red sorghum. Body weight gain reduced significantly (p<0.01) in diets containing unprocessed red sorghum beyond 33% in diet or reconstituted red sorghum at any level. However, during finishing growth phase the birds receiving either processed or unprocessed sorghum (barring S75) had statistically similar gains in body weight. During over all growth phase (0-6 wk), live weight gains in all the dietary treatments did not differ statistically. Feed intake during 0-6 wk was significantly higher (p<0.05) in diets containing sole red sorghum than corn-soy based control diets Feed conversion ratio during 0-3 wk period in control and unprocessed red-sorghum diets were similar but statistically poorer (p<0.01) FCR emanated from reconstituted groups, while during 3-6 wk of age FCR was poorer (p<0.05) in diets containing 75% red sorghum, either processed or unprocessed. FCR, in overall growth phases, in control diet was statistically similar to the groups fed diets containing up to 33% unprocessed or 16% reconstituted group. The carcass traits and yield of organs did not differ (p>0.05) due to the various levels of red-sorghum. It was concluded that though the tannin content was reduced by 30% by the reconstitution process, but this did not give any additional advantage in broiler performance. More over, red-sorghum can be used effectively up to 33% in diet replacing 50% of corn after proper adjustment of proteins, energy and amino acids.
The ryanodine-receptor $Ca^{2+}$ release channel protein in the sarcoplasmic reticulum membrane of rabbit skeletal muscle plays an important role in muscle exitation-contraction (E-C) coupling. Various types of detergents were tested, including Chaps, cholate, octylglucoside, Zwittergents, Mega-9, Lubrol PX, and Triton X-100 for solubilization of this protein. Among these, Chaps and Triton X-100 were found to optionally solubilize the channel complex. Optimum conditions for this solubilization were pH 7.4 with a salt concentration of 1 M. The addition of phospholipid in the solubilization step helped in stabilizing the protein. The purification of the receptor was performed using sucrose density gradient centrifugation. Various methods [dilution, freeze-thaw, adsorption (Biobeads), and dialysis] were investigated to incorporate the Chaps-solubilized and purified $Ca^{2+}$ release channel protein into liposomes made from different types of phospholipids. Of these, a combined method consisting of a dialysis, freeze-thaw and sonication steps yielded the best results. Reconstituted vesicles produced by this method with 95% phosphatidylcholine (from soybean extract) had good function.
