Development of a Receptor Methodology for Quantitative Assessment of Ambient PM-10 Sources in Suwon Area (수원지역 대기 중 PM-10 오염원의 정량평가를 위한 수용방법론의 개발)

  • A total of 328 ambient PM-10 samples was collected by a PM-10 high volume air sampler during the periods of February 1997 to February 1999 from Kyung Hee University at Suwon Campus. The samples were analyzed for their bulk chemical compositions(Cu, Fe, Pb, Zn, Al, $Na^{+}$, $NH_{4}^{+}$, $K^{+}$, $Ca^{2+]$, $Mg^{2+}$, $Cl^{-}$, $NO_{3}^{-}$, and $SO_{4}^{2-}$ by both an atomic absorption spectrophotometer and an ion chromatograph. The purpose of this study was t develop a receptor methodology for quantitative assessment of PM-10 sources. The data obtained from this study were ex-tensively examined using the target transformation factor analysis(TTFA) and the chemical mass balance (CMB). When TTFA was initially applied seasonal basis. five sources(such as automobile-related, sulfate-related, incine-ration, soil and combustion-related) were identified both during winter and fall. Since the total number and the type of sources were resolved by TTFA for the four seasons, CMB was employed to cross-check the results of TTEA. The total of six source categories identified by TTEA was intensively investigated on the basis of source profiles acquired from various source libraries established both in Korea and abroad. The results of this study showed the applicability of two popular receptor models as a new methdology for quantitative assessment PM-10 sources in Korea. Seasonally segmented data sets with the combined application of TTFA and CMB yielded a physically reasonable source apportionment result and provided a mean to increase the number of potential sources. Furthermore, this study suggested the possibility of the CMB application to ambi-ent data from Korea after identifying potential sources through traditional factor analysis.

Determination of Source Contribution Based on Aerosol Number and Mass Concentration in the Seoul Subway Stations (분진의 개수농도 및 질량농도에 입각한 서울시 지하철 역사 내 오염원의 기여도 결정)

  • The subway play an important part in serious traffic problems. However, because subway system is a closed environment, many serious air pollution problems occurred in subway stations and injured passenger's health. Therefor, it is a necessary to identify sources and to estimate pollutant sources in order to protect passenger's health and to keep clean subway environment. The purpose of this study was to analyze a air quality in the subway stations and to apply a new receptor methodology for quantitatively estimate of PM10 sources. In this study, the size distributions of particulate matters has been measured by using Aerosizer LD (U.S.A., API, Inc.). It's real time measurement capability of time-of-flight technique offers a significant advantage of user convenience and air pollution management. Also, the mass concentrations of PM 10 has been measured by using mini-vol portable sampler (U.S.A., Airmetrics Co.). The sampling performed in Seoul subway stations during the period of February 2000 and April 2000. The number distribution data used in this study consisted of 26 raw data sets in the Jongno-sam-ga station. Correlation Analysis can be used in subway stations for source separation and identification. Then, number contribution from each source is determined by the particle number balance (PNB). The mass concentration data used in this study consisted of 31 raw data in the 8 different stations. The mass contributions of PM10 sources in the concourse by using PMF/CMB model.

Optimizing Oily Wastewater Treatment Via Wet Peroxide Oxidation Using Response Surface Methodology

  • The process of petroleum involves in a large amount of oily wastewater that contains high levels of chemical oxygen demand (COD) and toxic compounds. So they must be treated before their discharge into the receptor medium. In this paper, wet peroxide oxidation (WPO) was adopted to treat the oily wastewater. Central composite design, an experimental design for response surface methodology (RSM), was used to create a set of 31 experimental runs needed for optimizing of the operating conditions. Quadratic regression models with estimated coefficients were developed to describe the COD removals. The experimental results show that WPO could effectively reduce COD by 96.8% at the optimum conditions of temperature $290^{\circ}C$, $H_2O_2$ excess (HE) 0.8, the initial concentration of oily wastewater 3855 mg/L and reaction time 9 min. RSM could be effectively adopted to optimize the operating multifactors in complex WPO process.

Expression and Characterization of a Single-Chain Variable Fragment against Human LOX-1 in Escherichia coli and Brevibacillus choshinensis

  • The single-chain variable fragment (scFv) against lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) is a promising molecule for its potential use in the diagnosis and immunotherapy of atherosclerosis. Producing this scFv in several milligram amounts could be the starting point for further engineering and application of the scFv. In this study, the abundant expression of the anti-LOX-1 scFv was attempted using Escherichia coli (E. coli) and Brevibacillus choshinensis (B. choshinensis). The scFv had limited soluble yield in E. coli, but it was efficiently secreted by B. choshinensis. The optimized fermentation was determined using the Plackett-Burman screening design and response surface methodology, under which the yield reached up to 1.5 g/l in a 5-L fermentor. Moreover, the properties of the scFvs obtained from the two expression systems were different. The antigen affinity, transition temperature, and particle diameter size were 1.01E-07 M, $55.2{\pm}0.3^{\circ}C$, and 9.388 nm for the scFv expressed by B. choshinensis, and 4.53E-07 M, $52.5{\pm}0.3^{\circ}C$, and 13.54 nm for the scFv expressed by E. coli. This study established an efficient scale-up production methodology for the anti-LOX-1 scFv, which will boost its use in LOX-1-based therapy.

Classification and Regression Tree Analysis for Molecular Descriptor Selection and Binding Affinities Prediction of Imidazobenzodiazepines in Quantitative Structure-Activity Relationship Studies

  • The use of the classification and regression tree (CART) methodology was studied in a quantitative structure-activity relationship (QSAR) context on a data set consisting of the binding affinities of 39 imidazobenzodiazepines for the α1 benzodiazepine receptor. The 3-D structures of these compounds were optimized using HyperChem software with semiempirical AM1 optimization method. After optimization a set of 1481 zero-to three-dimentional descriptors was calculated for each molecule in the data set. The response (dependent variable) in the tree model consisted of the binding affinities of drugs. Three descriptors (two topological and one 3D-Morse descriptors) were applied in the final tree structure to describe the binding affinities. The mean relative error percent for the data set is 3.20%, compared with a previous model with mean relative error percent of 6.63%. To evaluate the predictive power of CART cross validation method was also performed.

Comparative study of Ecological Risk Assessment : Deriving Soil Ecological Criteria (토양생태계 위해성평가기법 비교연구: 토양생태준거치 산정을 중심으로)

  • The purpose of ecological risk assessment in soil ecosystem is to protect ecological receptors and to provide a scheme of efficient management for soil contaminants. Developed countries have already prepared the methodologies of ecological risk assessment by considering their soil properties, land use, and ecological receptors. In this study, we compared the soil ecological risk assessment processes in the similarity and differences in methodology. Four countries, except for USA, adjusted the toxicological data for ecological risk assessment, based on their representative soil properties because the soil properties affect toxic effects to ecological receptors. The soil ecological risk assessment methodology of Netherlands and UK was based on 'Technical guidance document on risk assessment (TGD)' of European Chemical Bureau (ECB). Australia, USA, and Canada developed their autonomous methodology. In the Netherlands, UK, Australia, and Canada, they employed the species sensitivity distribution (SSD) approach if sufficient toxicity data are available. The USA determined the ecological soil screening level by obtaining the geometric mean of toxicological data for three species. Furthermore, all countries consider secondary poisoning in their soil ecological risk assessment. The latest risk assessment methodology of soil ecosystem that this study investigated can be used to explore what Korea needs to develop the Korean ecological risk assessment methodology of soil ecosystem in the future.

Estimation of Quantitative Source Contribution of Ambient PM-10 Using the PMF Model (PMF모델을 이용한 대기 중 PM-10 오염원의 정량적 기여도 추정)

  • In order to maintain and manage ambient air quality, it is necessary to identify sources and to apportion its sources for ambient particulate matters. The receptor methods were one of the statistical methods to achieve reasonable air pollution strategies. Also, receptor methods, a field of chemometrics, is based on manifold applied statistics and is a statistical methodology that analyzes the physicochemical properties of gaseous and particulate pollutant on various atmospheric receptors, identifies the sources of air pollutants, and quantifies the apportionment of the sources to the receptors. The objective of this study was 1) after obtaining results from the PMF modeling, the existing sources of air at the study area were qualitatively identified and the contributions of each source were quantitatively estimated as well. 2) finally efficient air pollution management and control strategies of each source were suggested. The PMF model was intensively applied to estimate the quantitative contribution of air pollution sources based on the chemical information (128 samples and 25 chemical species). Through a case study of the PMF modeling for the PM-10 aerosols, the total of 11 factors were determined. The multiple linear regression analysis between the observed PM-10 mass concentration and the estimated G matrix had been performed following the FPEAK test. Finally the regression analysis provided quantitative source contributions (scaled G matrix) and source profiles (scaled F matrix). The results of the PMF modeling showed that the sources were apportioned by secondary aerosol related source 28.8 %, soil related source 16.8%, waste incineration source 11.5%, field burning source 11.0%, fossil fuel combustion source 10%, industry related source 8.3%, motor vehicle source 7.9%, oil/coal combustion source 4.4%, non-ferrous metal source 0.3%. and aged sea- salt source 0.2%, respectively.

Network Pharmacology: Prediction of Astragalus Membranaceus' and Cornus Officinalis' Active Ingredients and Potential Targets to Diabetic Nephropathy (네트워크 약리학을 통한 당뇨병성 신병증에서의 황기와 산수유의 활성 성분 및 잠재 타겟 예측)

  • The purpose of this study is to predict the effects of macroscopic and integrative therapies by finding active ingredients, potential targets of Astragalus membranaceus (Am) and Cornus officinalis (Co) for diabetic nephropathy. We have constructed network pharmacology-based systematic and network methodology by system biology, chemical structure, chemogenomics. We found several active ingredients of Astragalus membranaceus (Am) and Cornus officinalis (Co) that were speculated to bind to specific receptors which had been known to have a role in the progression of diabetic nephropathy. Four components of Am and eleven components of Co could bind to iNOS; two ingredients of Am and six ingredients of Co could docking to cGB-PDE; one component of Am and nine components of Co could bind to ACE; three ingredients of Co with neprilysin; three components of Co with ET-1 receptor; four ingredients of Am and fourteen ingredients of Co with mineralocorticoid receptor; one component of Am and seven components of Co with interstitial collagenase; one ingredient of Am and ten ingredients of Co with membrane primary amine oxidase; one component of Am and four components of Co with JAK2; two ingredients of Am and one ingredient of Co with MAPK 12; one component of Am and five components of Co could docking to TGF-beta receptor type-1. From this work we could speculate that the possible mechanisms of Am and Co for diabetic nephropathy are anti-inflammatory, antioxidant and antihypertensive effects.

Chronic and Low Dose Exposure to Nonlyphenol or Di(2-Ethylhexyl) Phthalate Alters Cell Proliferation and the Localization of Steroid Hormone Receptors in Uterine Endometria in Mice

  • Based on our preliminary results, we examined the possible role of low-dose and chronic-exposing of the chemicals those are known as endocrine disrupting chemical (EDC), on the proliferation of uterine endometrium and the localization of steroid receptors. Immunohistochemical or immunofluorochemical methodology were employed to evaluate the localization of antigen identified by monoclonal antibody Ki 67 protein (MKI67), estrogen receptor 1 (ESR1), estrogen receptor 2 (ESR2), and progesterone receptor (PGR). In $133{\mu}g/L$ and $1,330{\mu}g/L$ di(2-ethylhexyl) phthalate (DEHP) and $50{\mu}g/L$ nonylphenol (NP) groups, the ratio of MKI67 positive stromal cells was significantly increased but not in $500{\mu}g/L$ NP group. The ratios of MKI67 positive glandular and luminal epithelial cells were also changed by the chronic administration of NP and DEHP in tissue with dose specific manner. ESR1 signals were localized in nucleus in glandular and luminal epithelia of control group but its localization was mainly in cytoplasm in DEHP and NP administered groups. On the other hand, it was decreased at nucleus of stromal cells in $1,330{\mu}g/L$ DEHP group. The colocalization patterns of these nuclear receptors were also modified by the administration of these chemicals. Such a tissue specific and dose specific localization of ESR2 and PGR were detected as ESR1 in all the uterine endometrial tissues. These results show that the chronic lows-dose exposing of NP or DEHP modify the localization and colocalization of ESRs and PGR, and of the proliferation patterns of the endometrial tissues.

Study of the Emission Correcting Methods Using Source-receptor Relationship and Satellite Observing Data (배출원-수용지 관계와 위성 관측자료를 이용한 배출량 개선 방법에 대한 연구)

  • With the development of remote sensing technology, it becomes possible to measure the trace gas concentration by satellites, so the ministry of environment is executing 152.5 billion-won to develop the geostationary orbital environment monitoring satellite and ground segment. A variety of contents for the application to utilize environmental satellite has been sought to increase the benefits. This study is for the improvement the accuracy of emission inventories, to correct emission amount with top-down method using the satellite observing data instead of bottom-up method. Two methodologies to estimate $NO_2$ emission were analyzed, one is ignoring $NO_2$ transportation effect(method-1), and the other is considering transportation effect(method-2) with source-receptor relationship equation, and the results were compared. The methodology of this study can be applicable all components which are observed from satellites.
