Antioxidant Activity of Amino Acid-Xylose Browning Reaction Products 2. Isolation of Antioxigenic Substrates from Browning Reaction Products by TLC and Dialysis (Amino 산-Xylose 갈변반응 물질의 항산화성 2. TLC와 투석을 이용한 항산화성 갈변물질의 분리)

  • In order to isolate antioxigenic substrates, the browning reaction products of xylose and various amino acids were analysed by TLC and dialysis. Rf values of browning reaction products of xylose and hydrophobic amino acids separated on silica gel TLC plate were shown in the range of 0.38 to 0.56 and that of basic amino acids was around 0.2. Browning reaction products made from xylose and Trp were separated on TLC into four bands with Rf values of 0.25, 0.55, 0.81 and 0.91 respectively. Among these the bands with Rf values of 0.25 and 0.55 appeared having strong antioxidant activity. The band of Rf 0.55 which showed the highest activity was positive to Prochazka reagent and had an absorption maximum at 275 nm. In dialysis of the xylose-Trp browning reaction products, the undialysed fraction (inner solution) was repsponsible to the antioxidant activity, which was separated into two bands with Rf values of 0.25 and 0.55 on TLC. The inner fractions of the browning products of xylose and His or Arg were also apparent in antioxdant activity.

  • The HS (headspace)-SPME (Solid phase microextraction) as rapid and simple method was performed for the determination of volatile fatty acids (VFAs) from the aqueous samples. In-fiber derivatization of VFAs with 1-Pyrenyldiazomethane (PDAM) was applied to improve their sensitivity of detection. In SPME procedure, typical parameters such as effects of solution pH, and salting out reagent and ultrasonication were investigated to improve the extraction efficiency. Based on the developed method, VFAs in wastewater samples were determined by gas chromatography / mass spectrometry-selected ion monitoring (GC/MS-SIM) mode.

Development of a Passive Sampler using a Fluorescence Material for the Ambient Ozone (형광물질을 이용한 대기 중 오존 Passive Sampler의 개발)

  • The purpose of this study is to develop an ozone passive sampler and to evaluate its performance p-Acetamidophenol using as the reagent for ozone reacts specifically with the ambient ozone to produce a fluorescence material (p-acetamidopheonl dimer). The volume of absorbent solution and the extraction time determined at suitable conditions for measuring ozone were 100$\mu$L and 60 min, respectively. The changes of fluorescence were observed with incresing the storage period of passive samplers in ambient air. but the cool storage in a refrigerator did not remarkably influence the increase of fluorescence. The measurement for the precision oi the passive sampling was carried out with duplicate measurement of passive samplers. The intra-class correlation coefficients of passive samplers using dry and wet filters were 0.992 and 0.962, respectively The results from field validation tests indicated practical agreement (dry filter: r=0.963, wet filter: r=0.995) between the passive sampler and an UV photometric $O_3$ analyzer. The limit of quantification of ozone passive samplers with sampling time of 8 hr (wet filter) and 24 hr (dry filter) were 8.0 ppb and 2.7 ppb, respectively.

A New Chemiluminescence Method for Determination of Cytosine Arabinoside in Pharmaceutical Preparations

  • A novel chemiluminescence (CL) system was established for the determination of cytosine arabinoside (Ara-C) in pharmaceutical preparations. It was showed that a clear CL signal was observed when Eosin Y mixed with Fenton reagent. The CL intensity was decreased significantly when Ara-C was added to the reaction system and partially scavenged the hydroxyl radicals in the solution. The extent of decrease in the CL intensity had a good stoichiometrical relationship with the Ara-C concentration. Based on this, we developed a new method for the determination of Ara-C using a flow injection analysis (FIA) technique with CL detection. Under the optimal conditions, the linear range of Ara-C concentration was $6.0{\times}10^{-9}\sim1.0{\times}10^{-7}mol/L$ (R = 0.9982) with a detection limit of $7.6{\times}10^{-10}mol/L$ (S/N=3), the RSD was 5.6% for $6.0{\times}10^{-8}mol/L$ Ara-C (n = 11). The method was successfully applied to the determination of Ara-C in injection samples. The possible chemiluminescence reaction mechanism was discussed.

Fluorometric Quantitative Analysis of Al(III) Ion Using 5-Methoxy-2-phenyliminomethylphenol

  • A novel Schiff base ligand (N, O system) 5-methoxy-2-phenyliminomethylphenol ($5-CH_3O-PMP$) was synthesized. Using the synthesized ligand as a fluorescent reagent, a fluorometric method was developed for the quantitative analysis of Al(III) ion. The quantitative analysis of Al(III) ion was performed by making the complex compound between Al(III) ion and $5-CH_3O-PMP$ in ethanol-water solution (85/15, v/v, pH 6.2). The excitation wavelength (${\lambda}em$) of the complex compound was 397 nm while the emmision wavelength (${\lambda}em$) was 498 nm. The quantitative analysis of Al(III) ion was carried out by estimating the fluorescence intensity. The various calibration curves were used for the quantitative analysis in the range of 0.27$\sim$27 ng/mL Al(III) ion concentrations. The detection limit was 0.027 ng/mL. Using the fluorometric method developed in this study, satisfying results were obtained from various samples such as tap water, hot spring water, river water, sea water and waste water, which contained considerable amounts of interfering ions.

  • A simple, selective and sensitive fluorescence quenching method was developed to the determination Cr(VI). The method is based on the oxidation of $I^-\;to\;{{I_3}^-}$ by Cr(VI) in sulfuric acid solution followed by immediate formation of ion association compound between I3 − and rhodamine 6G in Tween-80 micellar media at room temperature. The influence of several surfactants on rhodamine 6G fluorescence signal was studied; particular attention was paid in the aggregation behavior of rhodamine 6G–Tween-80 system. The experimental parameters (e.g., type of surfactant, reagent concentration) were studied and the optimal conditions were established. The linear calibration graph was obtained in the range 2.0 - 100.0 ng m$L^{-1}$ Cr(VI). The detection limit of the method was 0.37 ng m$L^{-1}$. The relative standard deviation (R.S.D.) is less than 5% (n = 5). The efficiency of the method for the determination of Cr(VI) in the presence of Cr(III) in the sample was investigated. The method was applied successfully to the determination of Cr(VI) and total Cr in water, and liver tissue samples.

Anti-Oxidative and Anti-Inflammatory Effects of Cheongajihwang-Tang Extract on RAW264.7 Cells (청아지황탕(靑蛾地黃湯)의 RAW264.7 Cell에서 항산화 및 항염증 효능 연구)

  • Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects.

Effect of Cingulate Cortical Ablation upon Gastric Secretion in Rats (흰쥐에서 대상회전 제거가 위액분비에 미치는 영향)

  • This study was undertaken to investigate the effect of cingulate cortical ablation upon gastric secretion and its components in rats. 23 male rats were divided into the cingulate(N=9) and the operated control(N= 14) groups. Cingulate cortex was ablated through a slit-shaped opening(1 mm in width, 5 mm in length) which was made symmetrically on both sides of, and parallel to, the sagittal suture by removing a bone flap from frontal bone on each side. In the operated control group, the surgical procedure was ended by the removal of the bone flap. Under light ether anesthesia, experimental animals were placed in a restraining jacket of fine mesh wire and gastric juice was collected for 5 hours via a canula which had been inserted through the anterior abdominal wall into the antral portion of the lumen of the stomach. Volume of the gastric juice was measured, and total acid output and free acid output were titrated with 0.04 N NaOH solution by using phenolphthalein and Topfer's reagent at indicator, and chloride ion output was estimated by means of chloridometer. Results obtained were that volume, total acid output, free acid output and chloride ion output of the gastric juice were higher significantly in the cingulate group than in the operated control group. It is inferred from the above results that the cingulate cortex exerts a fascilitatory influence upon gastric secretion and acid output in rats.

Simple and Quantitative Analysis Method for Lactic Acid by TLC (젖산의 빠른 정량적 분석을 위한 TLC 최적 조건)

  • TLC condition was developed for its simple separation and quantitative analysis of lactic acid. Rapid and clear separation of lactic acid by silica gel TLC plate was obtained by using nitromethane : 1-propanol : $H_2O$ (2 : 5 : 1.5, v/v/v) and a suitable dipping solution of 40 mg bromocresol purple in 100 mL 5% ethanol (pH 10.0). The lactic acid was shown as a bright yellow spot on a light cinnabar background. The quantitatively detectable concentration range of lactic acid was between 0.5 and 4% with 99.4%, confidence. Quantitative TLC analysis result was confirmed with HPLC and with enzymatic Quantitative analysis methods (by using lactate dehydrogenase).

Anti-inflammatory Effects of Gagamtongsoon-San Extract on Lipopolysaccharide(LPS)-Induced Inflammation in RAW264.7 Cells (LPS로 유발한 RAW264.7 세포 염증반응에 대한 가감통순산(加減通順散)의 억제 효과)

  • Objectives This study was designed to investigate whether the Gagamtongsoon-San (GT) has an inhibitory effect and its mechanisms are associated with the iNOS and COX-2. Methods Cytotoxic activity of GT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Inflammatory condition was induced by LPS. NO production was measured using Griess reagent system. The expressions of iNOS and COX-2 mRNA and protein were determined by realtime PCR. The concentrations of PGE2 were measured by an enzyme immunoassay (EIA). Results The GT does not impair the cell viability in tested concentration $500{\mu}g/ml$ or below. GT significantly reduced the NO production in a dose-dependent manner. GT $500{\mu}g/ml$ also suppressed LPS-induced mRNA expressions of iNOS and COX-2. GT $500{\mu}g/ml$ reduced the PGE2 secretion in LPS induced RAW264.7 cells. Conclusions These outcomes show that GT extract has an anti-inflammatory activities. And also this conclusion can be the data that supports the GT's anti-inflammatory effect objectively.
