반응염료염액에서의 키틴의 염료흡착성능 (Dye Adsorption Ability of Chitin in Reactive Dyebath)

  • In order to decolorize the reactive dye wastewater, we investigated the dye-adsorption ability of chitin, which was natural polymer obtained from shrimp shell. Chitin particle(less than 250 ${\mu}{\textrm}{m}$n) was prepared from shrimp shells in the processes of decalcification in aqueous hydrochloric acid solution and deproteination in aqueous sodium hydroxide solution. The particle size of chitin was controlled to less than 250 ${\mu}{\textrm}{m}$. Three tripes of the reactive dyes-C.I. Reactive Red 120, C.I. Reactive red 241 and C.I. Reactive Black 5-were used. Dye adsorption ability of chitin was investigated by dipping the particle in the dyebaths of concentration of 0.0l%, 0.03% and 0.05% for various periods of time(1,3,5, 10,20,40,80,160minutes). The influence of addition of salt(Na$_2$SO$_4$) and alkali to the dyebaths on dye-absorption was also investigated. We obtained the following results fur the dye-absolution ability of chitin in the dyebaths of three types of reactive dyes. 1) The amount of dye uptake by chitin was increased by addition of salt to the dyebaths. 2) As the concentration of alkali became higher than 3g/I, the amount of dye uptake by chitin was increased. Chitin showed good dye-adsorption ability, when the alkali concentration was high. 3) Chitin showed equal dye uptake in the three types of dyebaths when the dye concentration was 0.0l%. Over 90% of dyestuffs was adsorbed from the dyebaths in ten minutes. When the dye concentration was higher, better adsorption ability was showed in a dye bath of Reactive black 5 than in the others. When the dye concentration was 0.03%, 90% of Reactive red 120 and Reactive red 241 was adsorbed in 40 minutes and the same of Reactive black 5 in 10 minutes. When the dye concentration was 0.05%, 9()% of Reactive red 120 was adsorbed in 80 minutes, and Reactive black 5 in to minutes.

트리페닐메탄계와 아조계 색소를 탈색할 수 있는 Klebsiella pneumoniae WL-5의 분리 및 특성 (Isolation and Characterization of Klebsiella pneumoniae WL-5 Capable of Decolorizing Triphenylmethane and Azo Dyes)

  • 여러 가지 난분해성 색소에 대하여 탈색능을 나타내는 Klebsiella pneumoniae WL-5이 염색폐수처리장의 활성슬러지로부터 분리되었다. 이 세균은 정치배양과 at pH 6-8 및 $30-35^{\circ}C$에서 높은 탈색능을 나타내었다. Congo Red색소에 대해서는 $200\;{\mu}M$ 농도에서 12시간 배양하였을 때 90% 이상이 탈색되었고, Malachite Green, Brilliant Green, Reactive Black-5에 대해서는 $10\;{\mu}M$ 농도에서 80% 이상이 탈색되었지만, Reactive Red-120, Reactive Orange-16, Crystal Violet에 대해서는 $10\;{\mu}M$ 농도에서 각각 46%, 25%, 13%의 비교적 낮은 탈색능을 나타내었다. 트리페닐메탄계 색소는 세포표면에의 흡착에 의한 탈색을 나타내었고, 아조계 색소는 지금까지 알려져 있지 않는 새로운 효소반응계에 의해서 탈색된다는 것을 제시하였다.

  • 석탄계 활성탄을 사용한 Reactive Red 120 (RR 120) 염료의 흡착특성을 활성탄의 양, pH, 초기농도, 접촉시간 및 온도를 흡착변수로 사용하여 조사하였다. 등온흡착평형관계는 Langmuir 식이 Freundlich 식보다 더 잘 맞았다. 흡착 메카니즘은 균일한 에너지 분포를 가진 단분자층 흡착이 우세하다고 판단되었다. 평가된 Langmuir 분리계수(RL = 0.181~0.644)로부터 이 흡착공정이 효과적인 처리영역(RL = 0~1)에 속하는 것을 알았다. Temkin 식과 Dubinin-Radushkevich 식에 의해 구한 흡착에너지는 각각 E = 15.31~7.12 J/mol과 B = 0.223~0.365 kJ/mol로 흡착공정은 모두 물리흡착(E < 20 J/mol, B < 8 kJ/mol)으로 나타났다. 흡착속도실험결과는 유사 1차 반응속도식에 잘 맞았다. CGAC에 대한 RR 120 염료의 흡착반응은 온도가 올라갈수록 자유에너지 변화값이 감소하였기 때문에 온도 증가와 함께 자발성이 높아지는 것으로 나타났다. 엔탈피 변화(12.747 kJ/mol)는 흡열반응임을 알려주었다. CGAC에 의한 RR 120의 흡착반응의 등량흡착열은 9.78~24.21 kJ/mol로 물리흡착(< 80 kJ/mol)임을 밝혔다.

Geotrichum candidum을 이용한 염색 염료의 색도제거 (Decolorization of Textile Dyes by Geotrichum candidum)

  • Geotrichum candidum (KCTC 6195)를 이용하여 색도제거를 위한 최적조건은 초기 pH 6, $30^{\circ}C$, glucose 농도 30 g/L이었으며 빛은 세포성장과 색도제거에 영향을 주지 않았다. 한편, 세포성장과 색도제거를 위해서는 세포의 성장원(glucose)이 필수적이었다. 염료의 종류에 따라 색도제거량과 속도는 차이가 있지만 분산염료, 산성염료, 반응염료에 대해 색도제거가 고체배치와 액체배치에서 가능했으며 Acid goange 10 염료의 경우 배양 후 120 시간 후에는 초기 100 ppm에서 91%로, 초기 500ppm에서 84%까지 색도제거되는 것을 알 수 있었다. 색도제거에서 Acid red 1: 19.8%, Acid red 88, 73%, Acid orange 10; 12.1%, Reactive blue 19; 14.6%가 흡착으로 제거되었다. 이로서 효소에 의한 색도제거뿐만 아니라 흡착에 의해 색도제거됨을 알 수 있었다. 2일간 배양하고 glucose를 첨가하여 1일간 추가 배양한 경우 97%까지 색도제거 되었다.

반응성 이색성 염료를 사용하여 제조한 폴리(비닐 알코올) 편광필름의 내구성 및 편광특성에 관한 연구 (Studies on Durability and Polarization Properties of Poly(vinyl alcohol) Polarizing Films Prepared with Reactive Dichroic Dyes)

  • 이색성 염료인 콩고레드, 다이렉트블랙 22 및 다이렉트블랙 4를 염화시아눌산과 반응시켜 3,5-이염화-2,4,6-트리아진 잔기를 갖는 반응성 이색성 염료를 합성하였다. 합성한 반응성 이색성 염료를 폴리(비닐 알코올)(PVA) 필름에 염착시키고 400% 일축 연신시켜 편광필름을 제조한 후 투과도 및 편광효율을 측정하였다. 비교를 위하여 요오드를 PVA 필름에 염착시키고 400% 연신시켜 편광필름을 제조하여 투과도 및 편광효율을 측정하였다. 내구성 실험을 $50^{\circ}C$의 온도에서 85% 상대 습도의 조건으로 120시간 동안 진행하였다. 결론적으로, 요오드 염착 PVA 편광필름과 비교하여 반응성 이색성 염료 염착 PVA 편광필름의 경우 내구성이 크게 증진되었음을 알 수 있었다.

  • 적색을 낼 수 있는 물질을 만들기 위해 비공액 주사슬로 되어 있는 고분자의 곁사슬에 스티렌 분자가 연결되어 있는 트리페닐아민, 반응성 있는 작용기를 가진 아미노벤즈알데히드 그룹, 및 PM(4-(dicyanomethylene)-2-(tertbutyl)4H-pyran) 그룹을 도입하였다. 이 고분자의 전자 흡수 스펙트럼은 용액과 필름 상태에서 비슷하였다. 모든 고분자는 전기화학적으로 활성을 보였으며, 전기 발광 소자를 작동하였을 때 700nm 근처에서 적색을 보였다. $ITO/PPV/P5-PM/BCP/Alq_3/Al$으로 구성된 소자는 $50mA/cm^2$의 낮은 전류 밀도에서 $120cd/m^2$의 밝기를 보였으며, 외부 양자 효율은 $0.67\%$를 나타내었다. 발광 고분자 층에서의 균형있는 전하의 재결합을 유도하여 소자의 발광 효율을 높일 수 있었다. 이중 기능성(bifunctionalized)을 도입함으로써 적색 발광을 내면서 효율이 높은 발광 고분자를 개발하였다.

리오셀 직물 수지가공 효과와 염색성 (Dyeing Properties Resin Treatment Effects of the Lyocell Fabrics)

  • The effects of resin finish and dyeabilities of four kinds of Iyocell fabrics that were manufactured by four kinds of pulps were investigated. The dyeabilities of Iyocell fabrics were similar, but differed from cotton fabric. In early stage of 30 minutes, cotton fabric was shown higher dye exhaustion ratio than Iyocell fabrics, however after then cotton fabric did not increase dye exhaustion, Iyocell fabrics increased continuously. At last, the dye exhaustion ratio of Iyocell fabrics were about 75% and that of cotton fabric was 65%. Two kinds of experimental procedures were applied for Iyocell fabrics. One was what the fabrics were treated with resins and washed with cellulase, and then dyed with reactive dyes. The other procedure was the fabrics were dyed with reactive dyes, and then applied the resin treatments and cellulase washing. After fibrillation and washing the undyed Iyocell fabric and the Iyocell fabric that was dyed with C.I.Reactive Red 120, their weight loss ratios were 3.5% and 2.8%, respectively. Dyeing with reactive dyes caused the crosslinking between cellulose and dyes and the crosslinking decreased fibrillation. The weight loss by enzyme washing of Iyocell fabrics decreased by the glyoxal and melamine resin treatments. The reduction of weight loss can be caused by fibrillation decrease. Dyeing and resin treating can be showed the synergic effect on the reduction of fibrillation. The effect of glyoxal resin on the reduction of fibrillation was a little better than that of melamine resin.

The Purification and Characterization of a Bacillus stearothermophilus Methionine Aminopeptidase (MetAP)

  • Methionine aminopeptidase (MetAP) catalyzes the removal of an amino-terminal methionine from a newly synthesized polypeptide. The enzyme was purified to homogeneity from Bacillus stearothermophilus (KCTC 1752) by a procedure that involves heat precipitation and four sequential chromatographs (including DEAE-Sepharose ion exchange, hydroxylapatite, Ultrogel AcA 54 gel filtration, and Reactive red 120 dye affinity chromatography). The apparent molecular masses of the enzyme were 81,300 Da and 41,000 Da, as determined by gel filtration chromatography and sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE), respectively. This indicates that the enzyme is comprised of two identical subunits. The MetAP specifically hydrolyzed the N-terminal residue of Met-Ala-Ser that was used as a substrate, and exhibited a strong preference for Met-Ala-Ser over Leu-Gly-Gly, Leu-Ser-Phe, and Leu-Leu-Tyr. The enzyme has an optimal pH at 8.0, an optimal temperature at $80^{\circ}C$, and pI at 4.1. The enzyme was heat-stable, as its activity remained unaltered when incubated at $80^{\circ}C$ for 45 min. The Km and Vmax values of the enzyme were 3.0mM and 1.7 mmol/min/mg, respectively. The B. stearothernmophilus MetAP was completely inactivated by EDTA and required $Co^{2+}$ ion(s) for activation, suggesting the metal dependence of this enzyme.

Template-free preparation of TiO2 microspheres for the photocatalytic degradation of organic dyes

  • $TiO_2$ microspheres were successfully synthesised by simple solution phase method by using various amount of titanium butoxide as precursor. The prepared $TiO_2$ were characterized by X-ray diffraction (XRD), UV-vis diffuse reflectance absorption spectra (UV-DRS), X-ray photoelectron spectroscopy (XPS) and scanning electron microscopy (SEM). XRD analysis revealed that the as-synthesized $TiO_2$ microsphere poses an anatase phase. The photocatalytic degradation experiments were carried out with three different dyes, such as methylene blue, brilliant black, reactive red-120 for four hours under UV light irradiation. The results show that $TiO_2$ morphology had great influence on photocatalytic degradation of organic dyes. The experimental results of dye mineralization indicated the concentration was reduced by a high portion of up to 99% within 4 hours. On the basis of various characterization of the photocatalysts, the reactions involved to explain the photocatalytic activity enhancement due to the concentration of titanium butoxide and morphology include a better separation of photogenerated charge carriers and improved oxygen reduction inducing a higher extent of degradation of aromatics.
