Production and Consumption of Goat Milk Products in Korea (한국의 산양유제품 생산 및 소비 현황)

  • There has been a successful re-entry in the form of infant foods and as a product concept of "well-being" milk by feeding goat natural medicinal plants in high mountain lands and goats eating natural feeds. Typical composition of cow's milk and goat's milk are not significantly differ in major nutritional constituents. However, the noticeable differences between milks of the bovine and caprine species concern in the dimensions of the micelles, in casein composition, in size of the micelles and in the mineral charge of the micelle, but the ratio Ca/Pi in the micelle is very close for the two species The potential market in Korea could be expected to expand by keeping its freshness and nutritional benefits. The supply of goat milk products all year around is also an important to the consumers. In order to increase its market scale of goat milk, product manufacturers need extensive advertising promotion. Domestically, goat milk is currently manufactured at small scale dairy goat milk companies and consumed mainly in the form of fresh or fermented goat milk, while imported goat milk powder is used to produce infant goat milk formula by major dairy companies. Decreasing the unpleasant goaty flavour for the Korean consumers would be essential for the researchers who work for dairy science and technology.

  • Pichia pastoris and Hansenula polymorpha, the methylotrophic yeasts have been widely used as a host for the production of e eudaryotic proteins due to the advantages related to their inherited characters. This paper describes the method to enhance t the productivity of recombinant proteins by P. pastoris and H. po$\psi$morpha. In the production of recombinant proteins using a f fed batch fermentation system, the effects of specific growth rate on the specific expression rate of re$\infty$mbinant proteins w were studied. In both species, the expression system of recombinant proteins using the fed batch fermentation was optimezed.

Effects of Saengshik Supplementation on Health Improvement in Diet-Induced Hypercholesterolemic Rats (고지혈증 유발 흰쥐에 있어서 생식의 건강개선효과)

  • The study was conducted to investigate the effect of Saengshik supplementation on lipid metabolism in rats fed high cholesterol diets. Male Spraque-Dawley rats were administrated 1% cholesterol to induce hypercholesterolemia and were fed on diet containing Saengshik (30%, w/w). Serum and liver lipid profiles and fecal bile acids excretion were examined after 7 weeks of experimental diet. The feeding of diet containing 30% Saengshik significantly decreased total cholesterol (TC) contents in liver and plasma. Since cholesterol was balanced by entero-hepatic circulation through bile acid synthesis and reabsorption, inhibition of bile acid re-uptake in intestine decreases total cholesterol in liver and blood. In this point, significant increament of fecal bile acid excretion in Shaengshik group decreased TC and improved hypercholesterolemia. Also plasma high density lipoprotein-cholesterol (HDL-C) decreasing risk for coronary heart disease in Saengshik supplemented group was significantly higher than control group, whereas low-density lipoprotein-cholesterol (LDL-C) accumulation in arterial vessel wall was significantly lower than control group. In this result, we supposed that supplementation of Saengshik, uncooked natural food, may improve hypercholesterolemia through increment of cholesterol excretion.

Inhibitory effects on colonization of Streptococcus mutans by eighteen months xylitol consumption (18개월간 자일리톨 껌 저작에 따른 Streptococcus mutans의 집락 형성 억제효과)

  • The purpose of this study was to evaluate suppression of Streptococcus mutans(S. mutans) colonization by long-term xylitol consumption among pregnant women who can easily get bad oral hygiene status during pregnancy and breast feeding. Participants were voluntarily recruited for a pregnant women's oral health prevention program, classified into two groups(a control and a xylitol group), and then followed for 18 months. Stimulated whole saliva was collected in the afternoon(1:00-3:00 p.m.) at baseline, 6 months, 12months, and 18 months. Information regarding oral health behavior(tooth brushing frequency and regular dental visits) was obtained via a questionnaire. S. mutans counts of the xylitol group decreased considerably between baseline and the re-examinations at 6, 12, and 18 months. Although the S. mutans counts decreased for all time points compared to baseline in the control group, these differences did not reach significance except 18 months. Regular chewing of xylitol gum over a long period may lead to decrease the colony counts of S. mutans.

Re-evaluation of the optimum dietary protein level for maximum growth of juvenile barred knifejaw Oplegnathus fasciatus reared in cages

  • We determined the optimum dietary protein level in juvenile barred knifejaw Oplegnathus fasciatus in cages. Five semi-purified isocaloric diets were formulated with white fish meal and casein-based diets to contain 35, 40, 45, 50, and 60 % crude protein (CP). Fish with an initial body weight of $7.1{\pm}0.06g$ ($mean{\pm}SD$) were randomly distributed into 15 net cages (each size: $60cm{\times}40cm{\times}90cm$, $W{\times}L{\times}H$) as groups of 20 fish in triplicates. The fish were fed at apparent satiation level twice a day. After 8 weeks of feeding, the weight gain (WG) of fish fed 45, 50, and 60 % CP diets were significantly higher than those of fish fed 35 and 40 % CP diets. However, there were no significant differences in WG among fish fed 45, 50, and 60 % CP diets. Generally, feed efficiency (FE) and specific growth rate (SGR) showed a similar trend as WG. However, the protein efficiency ratio (PER) was inversely related to dietary protein levels. Energy retention efficiency increased with the increase of dietary protein levels by protein sparing from non-protein energy sources. Blood hematocrit content was not affected by dietary protein levels. However, a significantly lower amount of hemoglobin was found in fish fed 35 % CP than in fish fed 40, 45, 50, and 60 % CP diets. Fish fed 60 % CP showed the lowest survival rate than the fish fed 35, 40, 45, and 50 % CP diets. Broken-line analysis of WG showed the optimum dietary protein level was 45.2 % with 18.8 kJ/g diet for juvenile barred knifejaw. This study has potential implication for the successful cage culture of barred knifejaw.

Reliability of a Questionnaire for Women's Reproductive History (여성의 출산력과 초경 및 폐경 연령에 대한 설문지의 신뢰도)

  • Objectives : This study was performed to evaluate the reproducibility of a questionnaire concerned with reproductive history and to ascertain which characteristics of the subjects (age, the visit-revisit intervals, education and chronic disease) are associated with good reliability in the Korean Multi-Center Cancer Cohort (KMCC) study. Materials and Methods: A total of 19,688 participants were enrolled between 1993 and 2004. Among them, we selected 386 participants who were aged 40 or more and who re-visited within 8 years after the first visit. Reliability was measured by the percent agreement according to error range for the continuous variables and the percent agreement and kappa statistics for the categorical variables. Results : The pregnancy histories were reliable (kappa=0.67) and the reasons for being menopausal among the postmenopausal women were also reliable (kappa=0.92). The percent agreement of the breast-feeding history was high (96.1%), although the kappa statistic was low. For the continuous variables, when the error range of one variable was considered to be reliable, the percent agreement of the age at menarche and the age at the first full term pregnancy was good (69.4% and 83.6%), whereas that of the age at menopause was low (51.5%). The factors associated with high reliability were a younger age, the presence of chronic disease and a short visit-revisit time interval. Conclusions : The agreements for parity, the reasons for menopause, and the breastfeeding history in the reproductive history questionnaire used in the KMCC were relatively good. The questionnaire for the menarche age and the menopausal age might have lower reliability due to the difference between Korean age and American age. To obtain reliable information, more attention should be given to the items in questionnaire-based surveys, and especially for surveying old-aged women.

Protection of Infection and Eradication Activity of Culture Product by Pediococcus pentosaceus CBT SL4 Showing Antimicrobial Activity against Helicobacter pylori (Helicobacter pylori에 대한 항균활성을 나타내는 Pediococcus pentosaceus CBT SL4 배양물의 감염방어 및 제균활성)

  • New food ingredient was developed to eradicate and protect against re-infection of Helicobacter pylori in fermentation broth of lactic acid bacteria (LAB) showing antimicrobial activity against pathogenic microorganisms such as H. pylori and Listeria monocytogenes. LAB strain CBT SL4 was identified as Pediococcus pentosaceus by 16S rDNA sequencing and its culture broth showed antimicrobial activity of 800 AU/mL against H. pylori in optimized fermentation process. Using thin layer concentration system and spray-typed fluid bed drier system, concentrated powder product showing activity of 12,800 AU/g was harvested. Product showed eradication and protection activities against H. pylori infection on feeding test (50 AU/day) using Mongolian gerbil infection model. After 4 weeks therapy of 8,000 AU/day, ${\Delta}13CO_2$ level (DOB30) decreased about 40% in urea breath test on patient with H. pylori infection. Result show concentrated culture product of P. pentosaceus CBT SL4 has eradicating and protecting activities against H. pylori infection and can be used as food-active ingredient for prevention of gastric and duodenum ulcer caused by H. pylori.

Analysis of Asiatic Black Bear's foods by using Scats in the Jirisan National Park (지리산국립공원 반달가슴곰의 배설물을 통한 먹이원 분석)

  • Since 2004, Asiatic Black Bear of the Jirisan National Park is subjected to the restoration project through the re-introduction by the Maritime Province of Siberia in Russia, North Korea, Northeastern part of China and Seoul Grand Park. The purpose of this study is to analyze diets with scat to identify Asiatic Black Bear's feeding habit and eating resources environment in the Jirisan National Park. 78 scats of Asiatic Black Bears were observed during the field studies of radio telemetry-marked bears and by chance in the Jrisan National Park, from 2005 to 2013. The relative frequency of occurrence demonstrated that the plants comprised the highest with 77%, followed by insects with 12.8%, mammals with 5.6%, birds with 0.5%, arthropoda with 0.5%, and others with 3.6%. Also, 52 scats among the collected scats that were measured for the ratio of food were subjected to dry weight, and plants were the highest with 91.2%, followed by mammals with 3.6%, insects with 1.0% and others with 4.2% in the order mentioned. Asiatic Black Bear's feed research demonstrated that the Asiatic Black Bear at Jirisan is omnivorous and that the vegetable food is the main feed instead of animal feed.

Study on Recovery of Au from Flotation Tailing of Gold (금(金) 浮選(부선) 광미(鑛尾)로부터 금(金)의 회수(回收)에 관한 연구(硏究))

  • S.M.C (DSME), only operating gold mine in Korea, is processing about 160 ton/day to recover gold and more than 150 ton/day of tailing is produced. Some portion of the tailings are used as a filler material after drying, but most of them are stored on the tailing dam. As a result of chemical analysis by a fire assay method, it contained Au 1.5~2.0 g/ton and 225~300 g per day of gold is getting discarded. It is urgent to develop a technology to recover and reutilize Au. In the present study, flotation tests were carried out to recovery gold for the tailings. Test results show that products with gold grade 21.31 g/ton(Au grade) and 62.73% (Au recovery) were obtained under the optimal conditions including KAX addition rate 97.2 g/ton, frother AF 65 (0.248 l/ton) and depressant sodium silicate (4 kg/ton), it's possible to recover one of the most valuable metal Au, by re-feeding to rougher flotation.

IRS-2 Partially Compensates for the Insulin Signal Defects in IRS-1-/- Mice Mediated by miR-33

  • Insulin signaling is coordinated by insulin receptor substrates (IRSs). Many insulin responses, especially for blood glucose metabolism, are mediated primarily through Irs-1 and Irs-2. Irs-1 knockout mice show growth retardation and insulin signaling defects, which can be compensated by other IRSs in vivo; however, the underlying mechanism is not clear. Here, we presented an Irs-1 truncated mutated mouse ($Irs-1^{-/-}$) with growth retardation and subcutaneous adipocyte atrophy. $Irs-1^{-/-}$ mice exhibited mild insulin resistance, as demonstrated by the insulin tolerance test. Phosphatidylinositol 3-kinase (PI3K) activity and phosphorylated Protein Kinase B (PKB/AKT) expression were elevated in liver, skeletal muscle, and subcutaneous adipocytes in Irs-1 deficiency. In addition, the expression of IRS-2 and its phosphorylated version were clearly elevated in liver and skeletal muscle. With miRNA microarray analysis, we found miR-33 was down-regulated in bone marrow stromal cells (BMSCs) of $Irs-1^{-/-}$ mice, while its target gene Irs-2 was up-regulated in vitro studies. In addition, miR-33 was down-regulated in the presence of Irs-1 and which was up-regulated in fasting status. What's more, miR-33 restored its expression in re-feeding status. Meanwhile, miR-33 levels decreased and Irs-2 levels increased in liver, skeletal muscle, and subcutaneous adipocytes of $Irs-1^{-/-}$ mice. In primary cultured liver cells transfected with an miR-33 inhibitor, the expression of IRS-2, PI3K, and phosphorylated-AKT (p-AKT) increased while the opposite results were observed in the presence of an miR-33 mimic. Therefore, decreased miR-33 levels can up-regulate IRS-2 expression, which appears to compensate for the defects of the insulin signaling pathway in Irs-1 deficient mice.
