Production of Ginsenoside Rd from Ginsenoside Rc by ${\alpha}-{\small{L}}$-Arabinofuranosidase from Caldicellulosiruptor saccharolyticus

  • Ginsenoside Rd was produced from ginsenoside Rc using a thermostable recombinant ${\alpha}-{\small{L}}$-arabinofuranosidase from Caldicellulosiruptor saccharolyticus. The optimal reaction conditions for the production of ginsenoside Rd from Rc were pH 5.5, $80^{\circ}C$, 227 U enzyme/ml, and 8.0 g/l ginsenoside Rc in the presence of 30% (v/v) n-hexane. Under these conditions, the enzyme produced 7.0 g/l ginsenoside Rd after 30 min, with a molar yield of 100% and a productivity of 14 g $l^{-1}\;h^{-1}$. The conversion yield and productivity of ginsenoside Rd are the highest reported thus far among enzymatic transformations.

  • In order to realize the simple and economic 3rd order LPF using switched-capacitor(SC), we proposed the 3rd order RC LPF using a OP amplifier and realized the 3rd order SC LPF by replacing all the resistors in the RC LPF to SC. The frequency response of SC LPF was compared with the that of RC LPF. As a result, it was found that the response of realized SC LPF is almost similar to that of RC LPF.

Morphological Characteristics of the Rice (Oryza sativa L.) with Red Pigmentation (적색종피 돌연변이 벼의 형질특성조사)

  • Seven rice germplasms with red pigmentation within the pericarp were isolated from a large mutant collection. These red pericarp phenotypes resulted from the functions of the Rc, Rd and RdRc genes. Among them, two brown pericarp of the Rc type, four red pericarp of the RdRc type, and one white pericarp of the Rd type were identified. Morphological and agronomic characteristics of those rice germplasms were studied. The Rc type germplasms have the faint red or brown color pericarp and the Rd types produce the white pericarp, whereas the RdRc type germplasms have the dark red pericarp. Most of the important agronomic characteristics including plant stature, tillering ability, spikelet fertility, and total grain yield were lower in the colored rice than those of the wild-type control. All of the studied colored rice germplasms had a tendency of easy seed-shattering in comparison to the control. These characteristics of newly identified germplasms will be useful for identifying the genes responsible for pericarp color phenotype determination.

Ginsenosides Attenuate Formalin-Induced Pains Through Spinal and Supraspinal Sites

  • In previous studies we have demonstrated that several individual ginsenosides such as Rc, Rd, Re and Ri relieves formalin-induced pain following systemic treatment. But it is unknown where these single ginsenosides induce antinociception. We investigated the antinoiceptive effect of four individual ginsenosides on formalin-induced pain after intrathecal (i.t.), intracereventricular (i.c.v.), or subcutaneous (s.c.) administration using mice. We found that ginsenoside Rc, Rd, and Re except Rf attenuated both acute and tonic phase of pain. Ginsenoside Rf attenuated only tonic phase of pain after i.t. administration. The ED$\_$50/ was 1.0 (0.55∼l.75 mg/kg) for Rc, 1.15 (0.6∼2.25 mg/kg) for Rd, and 8.9 (3.9∼20.5 mg/kg) for Re in acute phase of pain. The ED$\_$50/ was 0.3 (0.1∼0.85 mg/kg) for Rc, 0.6 (0.35∼l.1 mg/kg) for Rd, 2.45 (1.25∼4.65 mg/kg) for Re, and 1.9 (1.5∼4.25 mg/kg) for Rf in tonic phase of pain. We also found that ginsenoside Rc, Rd, Re, and Rf after i.c.v. administration attenuated both acute and tonic phase of pain. The ED5o for acute phase of pain was 0.9 (0.55∼l.4mg/kg) for Rc, 0.9 (0.45∼1.7 mg/kg) for Rd, 0.93 (0.5∼l .75 mg/kg) for Re, and 1.85 (0.95∼3.5 mg/kg) for Rf. The ED$\_$50/ for tonic phase of pain was 0.7 (0.45∼1.05 mg/kg) for Rc,1.25 (0.7∼2.2 mg/kg) for Rd, 0.85 (0.45∼1.6 mg/kg) for Re, and 0.8 (0.4∼1.45 mg/kg) for Rf. Thus, the order of the analgesic potency was Rc$\geq$Rd>Re>Rf in both i.t. and i.c.v. administration routes. However, s.c. pretreatment of four ginsenosides did not reduce formalin-induced pain. These results suggest that analgesic effect of ginsenosides is achieved through spinal or supraspinal site(s) in formalin test.

  • The purpose of the study was to investigate the effect of ginseng saponins (panaxadiol, ginsenoside $Rb_1$, $Rb_2$, Rc, Rd) on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with gamma-ray. ICR mice were given each saponin (i.p. 50 mg/kg of body weight) at 24 hours before irradiation. The radioprotective effects of saponins were compared with the irradiation control respectively. The jejunal crypts were protected by pretreatment with ginsenoside Rc (p<0.05) and Rd (p<0.05). The spleen colony was increased by pretreatment with panaxadiol (p<0.05) and ginsenoside Rd (p<0.05). And the frequency of radiation induced apoptosis was significantly reduced by pretreatment with panaxadiol (p<0.05), ginsenoside Rb2 (p<0.05), Rc (p<0.05) and Rd (p<0.01). These results suggest that ginsenoside Rc, Rd might have a major radioprotective effect.

Tolerability and pharmacokinetics of ginsenosides Rb1, Rb2, Rc, Rd, and compound K after single or multiple administration of red ginseng extract in human beings

  • Background: We investigated the tolerability and pharmacokinetic properties of various ginsenosides, including Rb1, Rb2, Rc, Rd, and compound K, after single or multiple administrations of red ginseng extract in human beings. Methods: Red ginseng extract (dried ginseng > 60%) was administered once and repeatedly for 15 days to 15 healthy Korean people. After single and repeated administration of red ginsengextract, blood sample collection, measurement of blood pressure and body temperature, and routine laboratory test were conducted over 48-h test periods. Results: Repeated administration of high-dose red ginseng for 15 days was well tolerated and did not produce significant changes in body temperature or blood pressure. The plasma concentrations of Rb1, Rb2, and Rc were stable and showed similar area under the plasma concentration-time curve (AUC) values after 15 days of repeated administration. Their AUC values after repeated administration of red ginseng extract for 15 days accumulated 4.5- to 6.7-fold compared with single-dose AUC. However, the plasma concentrations of Rd and compound K showed large interindividual variations but correlated well between AUC of Rd and compound K. Compound K did not accumulate after 15 days of repeated administration of red ginseng extract. Conclusion: A good correlation between the AUC values of Rd and compound K might be the result of intestinal biotransformation of Rb1, Rb2, and Rc to Rd and subsequently to compound K, rather than the intestinal permeability of these ginsenosides. A strategy to increase biotransformation or reduce metabolic intersubject variability may increase the plasma concentrations of Rd and compound K.

  • This investigation was carried out to study the influence of pH and heat treatment on the ginsenosides in the white ginseng extract. Changes in ginsenosides (Rb1, Rb2, ,Rc, Rd) and free sugar were measured by the peak area variation of HPLC chromatogram during 25 hours heat treatment at the various level of pH. It was found that :(1) The peak areas of Rb1. Rb2, Rc and Rd on the HPLC chromatogram were decreased remarkably below pH 4.0 and more decrease was found as the temperature and heating time increased. (2) Those of glucose and arabinose were increased remarkably. It is considrered that the increase of glucose and the formation of arabinose result from the hydrolysis of ginsenoside( Rb1, Rb2, Rc, Rd) linked with sugars.

Biotransformation of major ginsenosides in ginsenoside model culture by lactic acid bacteria

  • Background: Some differences have been reported in the biotransformation of ginsenosides, probably due to the types of materials used such as ginseng, enzymes, and microorganisms. Moreover, most microorganisms used for transforming ginsenosides do not meet food-grade standards. We investigated the statistical conversion rate of major ginsenosides in ginsenosides model culture during fermentation by lactic acid bacteria (LAB) to estimate possible pathways. Methods: Ginsenosides standard mix was used as a model culture to facilitate clear identification of the metabolic changes. Changes in eight ginsenosides (Rb1, Rb2, Rc, Rd, Re, Rf, Rg1, and Rg2) during fermentation with six strains of LAB were investigated. Results: In most cases, the residual ginsenoside level decreased by 5.9-36.8% compared with the initial ginsenoside level. Ginsenosides Rb1, Rb2, Rc, and Re continuously decreased during fermentation. By contrast, Rd was maintained or slightly increased after 1 d of fermentation. Rg1 and Rg2 reached their lowest values after 1-2 d of fermentation, and then began to increase gradually. The conversion of Rd, Rg1, and Rg2 into smaller deglycosylated forms was more rapid than that of Rd from Rb1, Rb2, and Rc, as well as that of Rg1 and Rg2 from Re during the first 2 d of fermentation with LAB. Conclusion: Ginsenosides Rb1, Rb2, Rc, and Re continuously decreased, whereas ginsenosides Rd, Rg1, and Rg2 increased after 1-2 d of fermentation. This study may provide new insights into the metabolism of ginsenosides and can clarify the metabolic changes in ginsenosides biotransformed by LAB.

Content Comparison of Proximate Compositions, Various Solvent Extracts and Saponins in Root, Leaf and Stem of Panax Ginseng (인삼의 근, 엽 및 경의 일반성분, 용매별 엑기스 및 사포닌 함량 비교)

  • This study was carried out to investigate the effective components, especially saponins, in aerial parts of Panax ginseng. The contents of methanol and ethanol extracts in ginseng leaf were 35.9% and 27.3%, much higher than 15.4% and 8.37% in ginseng root and 21.7% and 16.3% in ginseng stem. And ginseng stem showed as high content of crude fiber as 39.2% which is very high compared with other two parts of ginseng. The contents of total crude saponin were 4.78%, 2.38% and 19.58% in ginseng root, stem and leaf, respectively. In ginseng leaf seven root ginseno-sides-ginsenoside-Rgl(3.32%), -Re(3.24%), -Rd(2.32 %), -Rc(0.65%), -Rb2(0.92%), -Rbl(0.29%), and -Rf(0.11%)-were analyzed by HPLC, Seven gisneno- sides-ginsenoside-Rgl(0.28%), -Re(0.3%), -Rd(0.05%), -Rf(0.01%), -Rc(trace), -Rb2(trace) and -Rbl(trace)-were detected in ginseng stem. Ginseng leaf contained high percentage of saponin and especially of ginsenoside-Rgl, -Re and -Rd. Therefore, ginseng leaf was good resources for ginsenoside-Rgl, -Re and -Rd.

Amplitude dependent damping ratio of domestic tall building by RD method (국내 고층건물의 RD법에 의한 감쇠율의 진폭의존성)

  • The measured damping ratio was analysed to obtain amplitude dependence. Wind-induced vibration of 20 story steel-framed building was measured to investigate amplitude dependence by RD method. Micro-tremo vibrations of 20 RC bearing wall typed buildings were performed to analysis the amplitude dependence by formula proposed by Tamua and ESDU. Amplitude dependent damping in 17 story steel-framed building was showed clearly in the increasing rate of 9%. But Amplitude dependent damping of 17 RC bearing wall typed buildings was very low in the increasing rate of 2.5%. The tendency of dynamic properties of building obtained here are useful for the validation of dynamic properties of buildings in the evaluation of serviceability.
